1.Effects of Survivin-T34A mutant on breast cancer cell in vitro and in vivo.

To study the effect of the proliferation and apoptosis of Survivin-T34A mutant on breast cancer MCF-7 cell, we adopted the method of cell culture in vitro to observe the proliferation and apoptosis of the cell. In the experiment, MCF-7 cells were randomly divided into three groups and transfected with normal saline, PORF-9-null and Survivin-T34A, respectively. Breast cancer nude mouse models were established to study anti-tumor effect of Survivin-T34A in vivo. The activity of the cells in the Survivin-T34A-transfected group was lower than that in PORF-9-null group. The increase of cell apoptosis was observed under electron microscopy, meanwhile the apoptotic rate was obviously higher than that in PORF-9-null control by flow cytometry. Tumor inhibition effects of the mouse which received the injection of Survivin-T34A intratumoral injection were apparent, and the inhibition ratio was as high as 47.1%. In conclusion, Survivin-T34A mutant has anti-tumor effect through efficiently inhibiting the growth of breast cancer MCF-7 cell and actively promoting apoptosis of cancer cells.

2.Early-stage interpersonal perception and selection in internet instant messaging

Objective: To discuss the university students early-stage interpersonal perception and selection in internet Method; Totally 362 university students who used internet instant chat system were surveyed with the self-designed questionnaires. They were required to assess the influence degree of internet information on their feeling and choosing in a five degree scale. The differences in gender, subject and lasting time were also compared. Re-sults: (1) Age (4. 39 ± 1.94), nick name (3.71 ± 1.92) and self-explained (3.67 ± 1.97) were the most important information for choosing a friend in internet, which were higher than three most unimportant information including blood type (1.23 ±0. 80), level (1.40 ±0.95), and operation (1.41 ±0.89) (P<0.01) . (2) All students trended to evaluate others or selves information veritable. In all items, the veracity rate of self evaluate was high than 78.2% , and the veracity degrees of evaluating others'information were between 2.05 ~ 2.91. (3) Bad manners (1. 33 ±0.72), rough speech (1.41 ±0.71), and bored dialogue (1.50 ±0. 80) were the most related factors to choose friend, which were higher than three most unimportant information including no meeting chance (4.47±0.94), slow typing (3.13 ±1.12), poor motivation (2.32 ± 1.05) (P<0.01) . Conclusion: This study suggests that peoples early-stage perception and selection in internet are based on the selection, confirmation and conjecture of users, which will have certain effects on the development of network interpersonal relationship.

Objective To explore the clinical significance of early biomarkers in neonatal asphyxia diagnosis with myocardial damage by detection of ischemia modified albumin in neonatal serum(IMA) and glycogen phosphorylase isoenzyme BB(GPBB) for screening sensitive markers with direct myocardial injury.Methods 166 neonates were selected in our hospital as the research object,in which 136 cases with myocardial injury dividend into the experimental group and 30 cases into the control group.The experimental group were divided into mild group and severe group according to the degree of asphyxia.All the children were tested for GPBB and IMA 1 h after admission.Results The levels of GPBB in neonatal asphyxia myocardial injury group and severe asphyxia group were significantly higher than those in the control group,the difference was statistically significant(P<0.05).The sensitivity of GPBB in diagnosis of asphyxia was higher than that of IMA,the difference was statistically significant(P<0.05).Conclusion The degree of asphyxia is closely related to serum IMA,GPBB level in neonatal asphyxia complicated with myocardial injury.The sensitivity and specificity of GPBB in diagnosis asphyxia is better than IMA in children complicated with myocardial damage.

Objective: To investigate the role of Smads signaling pathway in the transdifferentiation of airway epithelial cells into myofibroblasts induced by rhTGF-?1 in vitro.Methods: We exposed cultured 16HBE cells to rhTGF-?1(10?g/L),detected the levels of Phosphorylated Smad2 and Smad3 in nucleus proteins by Western blot at different time points,treated with rhTGF-?1 the 16HBE cells pre-transfected with either the expression plasmid of Smad7(pCDNA3.0/Smad7) or the blank vector(pCDNA3.0),and determined the expression of ?-SMA by immunofluorescent staining and RT-PCR and that of E-cadherin by RT-PCR.Results: The expression of Phosphorylated Smad2 was observed and that of Smad3 increased in the TGF-?1-treated group,with the levels of Phosphorylated Smad2 and Smad3 elevated in a time-dependent manner.The level of ?-SMA was significantly decreased while that of E-cadherin markedly increased in the Smad7 transfection group as compared with the blank vector transfection group 24 h after TGF-?1 stimulation,and no differences were found between the two groups at 36 h.Correspondingly,the number of ?-SMA-positive cells was reduced in the Smad7 group compared with the blank vector group(P
5.Effects of Tiaoxin Formula on Spatial Learning and Memory Ability and LTP in Hippocampus of APP/PS1 Double Transgenic Mice with Alzheimer Disease

Objective To investigate the effects of Tiaoxin Formula on spatial learning and memory ability and long-term potentation (LTP) in the hippocampus of APP/PS1 transgenic mice with Alzheimer disease; To discuss its mechanism of action. Methods Totally 54 three-month-old APP/PS1 transgenic mice were randomly divided into model control group, Tiaoxin Formula group and positive control group, with 18 mice in each group. Another 18 three-month-old C57BL/6J wild mice were chosen as normal control group. All administration groups receive relevant medicine. 12 weeks later, Morris water maze test was used to test behavior and in vitro electrophysiology record. Results The Morris water maze test showed that in place navigation test, compared with the model control group, escape latency time in Tiaoxin Formula group was significantly reduced (P<0.01). In the space experiments, compared with the model control group, the number of crossing the platform quadrant and the time staying at the platform quadrant in Tiaoxin Formula group were significantly increased (P<0.05, P<0.01). In vitro hippocampal electrophysiological induction, compared with the model control group, fEPSP slope in the hippocampus in Tiaoxin Formula group were significantly increased (P<0.01), the PPF had no significant difference (P>0.05). Conclusion Tiaoxin Formula can improve the spatial learning and memory ability and LTP of APP/PS1 transgenic mice with Alzheimer disease, thus can realize cognitive protection effects.
6.Adverse reactions from stereotactic body radiotherapy for advanced hepatocellular carcinoma

Objective To investigate the adverse reactions by stereotactic body radiation therapy(SBRT)for advanced hepatocellular carcinoma and summarize nursing experience.Methods Forty-one patients with advanced hepatocellular carcinoma from the radiation department of Sun Yat-sen University Cancer Center from July 2010 to May 2012 were enrolled in this retrospective study.The adverse reactions were closely observed and the patients were given pertinent nursing.Results The effectiveness rate was 56.2%.During the therapy,36 patients developed nausea/vomiting of grade 1-2,taking up 87.8%,18 had grade 1-3 elevation of liver enzymes,taking up 43.9%,16 had grade 1-2 decrease of white blood cells,taking up 39.0%,8 had grade 1 anemia,taking up 19.5%and 21 had grade 1-2 decrease of blood platelet,taking up 51.2%.The adverse reactions were contained satisfactorily through careful observations and pertinent nursing.Conclusions The toxic reactions by SBRT are nausea,vomiting,enzymes elevation and decrease of whole blood cell.Therefore,nurses need to observe these toxic reactions carefully and give pertinent care to the patients so as to prevent the complications,especially radiation-induced liver injury.
7.The effect of 3'-meisoindigo on the function of splenocytes and thymocytes of C57BL/6 mouse

BACKGROUND: The effect of Chinese traditional medicine 3'-meisoindigo as well as indimbin derivatives on normal immunocytes is less reported while it is used for antitumor.OBJECTIVE: To investigate the effect of 3'-meisoindigo on the proliferation of the splenocytes and thymocytes of C56BL/6 mouse.DESIGN, TIME AND SETTING: The randomized cytopathology observation was performed between August 2007 and January 2008 at Zhujiang Hospital of Southern Medical University, Guangzhou, Guangdong Province, China.MATERIALS: C57BL/6 mice, clean, male, 6-8 weeks old, weighing (20±2) g.METHODS: The thymus gland and the spleen of C57BL/6 mice were ground to get the single-cell suspension and cells were treated by 5, 10, 15, 20 and 25 μ mol/L 3'-meisoindigo,respectively. Cells without any drug treatment were used as blank control and cells treated by concanavalin A were used as positive control.MAIN OUTCOME MEASURES: Proliferation of splenocytes and thymocytes detected using MTT method; IL-12 activity in culture supematant detected using ELISA method; the cell cycle, apoptosis rate, cell death rate and intracellular reactive oxygen species level detected using flow cytometry; the mRNA level of bcl-2 and cdk2 detected using reverse transcription-polymerase chain reaction; the expression rate of Bcl-2, CDK2 and Bax detected using fluorescence microscope.RESULTS: After treating for 24 hours, 15, 20 and 25 μ mol/L 3'-meisoindigo can significantly inhibit the proliferation of thymocytes and splenocytes (P < 0.05) and the inhibition was dose-dependent and time-dependent. Cells resumed proliferation after removing the 3'-meisoindigo, although they had been treated by 25 μ mol/L 3'-meisoindigo for 72 hours. The secretion of IL-12 was markedly reduced in all 3'-meisoindigo groups versus control groups at each time point (P < 0.05). 15 μ mol/L 3'-meisoindigo could decrease the mRNA expression level of apoptosis-related protein bcl-2 and cyclin cdk2 gene, decrease the expression level of BCL-2 protein and CDK protein, increase Bax expression level, decrease Bcl-2/Bax ratio markedly and started apoptosis.15 μ mol/L 3'-meisoindigo arrested cells in the G2/M stage of the thymocytes and splenocytes, and intracellular reactive oxygen species level elevated dose-dependently and time-dependently.CONCLUSION: In certain concentration range, 3'-meisoindigo can reversibly inhibit the proliferation of thymocytes and splenocytes of C57BL/6 mouse and can inhibit IL-12 secretion in parallel, and start apoptosis.
8.Application of detection of tumor markers CEA ,AFP ,CA19-9 and CA72-4 in digestive malignant tumors

Objective To explore the significance of combined detection of tumor markers cancer embryo antigen (CEA) ,alpha fetoprotein (AFP ) ,carbohydrate antigen 19-9 (CA19-9 ) and carbohydrate antigen 72-4 (CA72-4 ) in digestive tract tumors . Methods The electrochemical luminescence method was adopted to detected the CEA ,AFP ,CA19-9 and CA72-4 levels in 106 cases of digestive tract malignant tumor confirmed by pathological examination (malignant tumor group) ,110 cases of digestive tract be-nign diseases (benign disease group) and 60 persons undergoing the healthy physical examination (control group) in our hospital from January to December 2015 .The differences were compared among various groups and the positive detection rates of 4 markers in the patients with digestive tract tumors were compared .Results The serum levels of CEA ,AFP ,CA19-9 and CA72-4 levels in the malignant tumor group were significantly higher than those in the benign disease group and control group(P<0 .05);the benign disease group and normal control group had no statistically significant difference (P>0 .05) .In the single indicator detection of ser-um CEA ,AFP ,CA19-9 and CA72-4 ,AFP had the highest detection rate in hepatocellular carcinoma (HCC) ,which was 74 .19% ;the positive detection rate of CA 72-4 in gastric cancer was highest ,which was 60 .71% ;the positive detection rate of CA 19-9 in pan-creatic cancer was highest ,which was 75 .00% ;the positive detection rate of CEA was not high without specificity in various tumor diseases .The positive detection rate of 4-item combined detection was significantly higher than that of single item detection (P<0 .05) .Conclusion The detection of tumor markers CEA ,AFP ,CA19-9 and CA72-4 is conducive to the identification of tumor types ,meanwhile they can improve the detection rate of gastrointestinal cancer and conduces to early diagnosis and early treatment for the patients .
9.Characteristics of glucose metabolism in non-obese and obese women with polycystic ovarian syndrome

Objective To investigate characteristics of glucose metabolism of non-obese and obese women with polycystie ovary syndrome(PCOS).Methods From March 2008 to July 2011,118 PCOS patients were enrolled in this study and were divided into 53 cases (body mass index(BMI) ≥25 kg/m2) in obese group and 65 cases (BMI < 25 kg/m2) in non-obese group.Participants' clinical data,glucose tolerance and insulin release test results were retrospectively reviewed.The prevalence of type 2 diabetes mellitus (T2DM),results of oral glucose tolerance test,impaired fasting glucose (IFG),impaired glucose tolerance (IGT) were compared between the two groups.Results (1) Blood glucose levels:at the time points of 30,60,120 and 180 minutes,the levels of fasting glucose were (5.2 ± 1.1),(8.5 ± 2.8),(8.1 ± 2.4),(6.3 ± 2.0) and (4.8 ± 1.5)mmol/L in non-obese group and(5.4 ±0.9),(9.1 ± 1.8),(9.3 ±0.6),(7.6 ± 1.0) and (5.4 ±0.8) mmol/L in obese group.Statistical difference was observed between obese and non-obese groups at each time point (t =-6.125,-4.005,-6.074,-6.751 and-4.512 respectively,P <0.01).(2) The level of insulin:at the time points of 30,60 and 120 min,the level of fasting insulin were (8 ± 4),(55 ± 21),(65 ± 14) and (45 ±18)mU/L in non-obese group and (13 ± 8),(85± 30),(105± 54) and (76 ± 46)mU/L in obese group.There were significant statistical difference between the two groups at each time point (t =-17.024,-12.540,-15.791 and-16.149 respectively,P < 0.01).However,at the time point of 180 minutes,the level of insulin did not exhibit significant difference between obese and non-obese groups (P > 0.05).(3) The prevalence of abnormal glucose metabolism:The rates of IGT were 13.85 % (9/65) in non-obese group and 24.53 % (13/53)in obese group,which also showed remarkable difference (x2 =18.446,P < 0.01).The rates of T2DM were 1.54% (1/65) in non-obese group and 7.55% (4/53) in obese group,which reached significant difference (x2=16.005,P < 0.01).Conclusion Abnormal glucose metabolism was observed more frequently in overweight or obese PCOS women.
10.Clinical analysis of malignant hematological disease combined with septicemia

Objective To study the clinical characteristics,pathogens,drug sensitivity and prevention measures in malignant hematological disease combined with septicemia.Methods The clinical data of 95 malignant hematological disease combined with septicemia patients from January 2009 to June 2013 were retrospectively analyzed.Results One hundred and ten episodes of septicemia occurred in all the 95 patients,among which acute leukemia patients accounted for 69.1% (76/110),patients with neutrophil counts <0.5 × 109/L accounted for 82.7% (91/110),and patients with agranulocytosis time >7 days accounted for 66.4%(73/110).Major pathogenic bacteria was gram negative bacteria,including Escherichia coli 30.9%(34/110),Klebsiella pneumoniae (14.5%,16/110),Pseudomonas aeruginosa (12.7%,14/110).Carbapenems and amikacin was the most sensitive in susceptibility testing.Ninety-eight episodes of septicemia cured or improved,and 12 patients died,with a mortality of 12.6% (12/95).Fifteen patients had septic shock,of whom 7 patients were dead.Conclusions Septicemia is very serious for malignant hematological patients,which may lead to rapidly progress,high incidence to septic shock and high mortality.Early prevention and treatment is important for patients,and strong anti-infection and supportive treatment is needed.
