1.Bone marrow mesenchymal stem cell transplantation combined with edaravone inhibits neuronal apoptosis after cerebral infarction
Chinese Journal of Tissue Engineering Research 2014;(10):1615-1620
BACKGROUND:Several studies have confirmed that bone marrow mesenchymal stem celltransplantation exerts a certain neuroprotective role in cerebral infarction. Edaravone is a novel potent smal-molecule hydroxyl radical scavenger, which play a protective role in the brain by eliminating free radicals and inhibiting nerve celldamage after cerebral infarction. OBJECTIVE:To explore the influence of bone marrow mesenchymal stem cells combined with edaravone on expressions of aquaporin-4, Bcl-2, and brain-derived neurotrophic factor after cerebral infarction in rats. METHODS:Eighty Wistar rats were selected to establish models of cerebral infarction by right middle cerebral artery occlusion, and then randomly divided into control group, bone marrow mesenchymal stem cells group, edaravone group and edaravone+bone marrow mesenchymal stem cells group (combination group). After 6 hours of modeling, rats from these four groups were respectively injected via the tail vein with PBS, bone marrow mesenchymal stem cells, edaravone and edaravone+bone marrow mesenchymal stem cells. After 72 hours, the rats were decapitated to take brain tissues. Consequently, expressions of aquaporin-4, Bcl-2, and brain-derived neurotrophic factor mRNA and protein were determined by RT-PCR and western blot methods. After 12, 24 and 36 hours, TUNEL method was used for the determination of cellapoptosis. RESULTS AND CONCLUSION:The expressions of Bcl-2 and brain-derived neurotrophic factor were higher in the combination group than the other three groups (P<0.05), but the expression of aquaporin-4 was lowest in the combination group (P<0.05). The number of apoptotic cells in the combination group was significantly lower than that in the other three groups. These findings suggest that the combination of bone marrow mesenchymal stem cells and edaravone can improve expressions of Bcl-2 and brain-derived neurotrophic factor in the injured site, and significantly inhibit neuronal apoptosis. Meanwhile, the combination therapy can decrease expression of aquaporin-4 and mitigate cerebral edema, which is superior to bone marrow mesenchymal stem celltransplantation and edaravone alone.

4.Effects of peroxisome proliferator-activated receptor gamma agonists on fibronectin expression after SiO2 stimulation
Chinese Journal of Tissue Engineering Research 2011;15(21):3972-3974
BACKGROUND: The effect and mechanism of peroxisome proliferator-activated receptor gamma (PPARγ) agonists on transforming growth factor (TGF)-β1-induced fibrotic responses in lung fibroblasts remain unclear, and seldom reported.OBJECTIVE: To study the effects of PPARγ agonists on TGF-β1-induced fibrotic responses in lung fibroblasts, so as to investigate its effects in preventing pulmonary interstitial fibrosis of silicosis. METHODS: The pulmonary alveolar macrophage was prepared with 200 mg/L SiO2 and the supernatants were got. Chinese hamster lung fibroblasts were incubated with the prepared supernatants. Then a cellular model in vitro for studying pulmonary fibrosis induced by SiO2 was developed. Chinese hamster lung fibroblasts were cultured to observe the effects of the PPARγ ligand (15 d-PGJ2) and its agonists (troglitazone and ciglitazone) on the expression of TGF-β1-induced fibronectin. RT-PCR was used to detect the mRNA expression of fibronectin and Western blotting was used to detect the protein expression of fibronectin. RESULTS AND CONCLUSION: (1) The expression of TGF-β1 mRNA showed an effect in a dose- (0-5 μg/L) and time- (0-24 hours) dependent manner. (2) PPARγ ligands and agonists reduced the mRNA and protein expressions of fibronectin. Results suggested that PPARγ agonists can inhibit TGF-β1-induced fibronectin synthesis after stimulation by SiO2 and may play a potential role in preventing pulmonary interstitial fibrosis.

The occurrence rate of adverse reactions in two groups were compared, the difference was not statistically significant [57.6%(19/33) vs.50.0% (15/30)] (P>0.05). Conclusion Three dimensional-conformal radiotherapy combined with oral capecitabine in the treatment of stage Ⅱ,Ⅲrectal cancer after operation has satisfactory effect and acceptable adverse reaction, it is worth generalizing and applying.
6.The value of ultrasonography in diagnosing of benign and malignant ovarian teratomas
Chinese Journal of Ultrasonography 2001;10(5):265-266
Objective To evaluate the value of ultrasound in the diagnosis of benign and malignant ovarian teratomas. Methods The ultrasonographic imaging features of eighty-four cases of benign and malignant ovarian teratomas were viewed by two dimensional and color Doppler ultrasonography. Results Eighty-four cases were confirmed by pathology report that seventy-eight cases were ovarian teratomas. The accurate rate of ultrasound diagnosis was 92.9%(78/84).Among these cases, tumors which on the single side existed in sixty-nine cases (two tumors existed on the same side in two cases),and which on double sides existed in nine cases.Eighty-nine tumors were checked out all together. The ultrasonographic images of the ovarian teratomas displayed three types: mix(65 masses) , false solid (14 masses) and cyst (10 masses) type. Mix type was the major (occupied 65/89,73.0%). Multiplicate signs are shown in all of them. Malignant teratomas (3 masses,4 masses) not only appeared similar, but also more complex than the benign. Conclusions Ultrasonography is the first examination to be selected to diagnose teratoma of the ovary. Their ultrasonographic images are charaeteristic. If the images appears disorderly inside, the malignancy should be predicted with other clinical materials.
