1.Prevention and treatment of nephrotic syndrome associated bone metabolic abnormality in children

Metabolic bone disease in nephrotic syndrome(NS) are increasingly being renal physician's attention.As calcium binding protein and VitD binding protein losing with a large number of protenuria, the bone metabolic biochemical abnormalities had happened at the beginning of the onset of the nephrotic syndrome, and is further exacerbated by therapeutic high-dose or long course of glucocorticoids (GC) application.The main mechanism of the glucocorticoid-induced osteoporosis (GIOP) is for GC to inhibit the activity of osteoblasts and promote apoptosis of osteoblasts and formation of osteoclasts, resulting in secondary hyperparathyroidism,leading to increasing the risk of osteoporosis,slow growth and fracture,seriously harm to children's physical and mental health.The biomarkers of bone transform can prompt the NS with bone metabolic abnormalities early;vertebral body bone dual-energy X-ray absorptiometry bone mineral density detection is the best method and position to determine GIOP.As the most commonly used and effective means to prevent and control metabolic bone disease, calcium supplements and VitD were always taken when GC was treated for NS, even the dosage of GC was very low.So far, it is still lack of guideline of prevention and treatment of bone metabolic abnormalities in NS in children.
2.Effects of EEF1 A2 on the growth and proliferation of pancreatic cancer cell

Objective To investigate the effects of EEF1 A2 on growth and proliferation of the human pancreatic cancer cell line SW1990. MethodsThe EEF1 A2 gene was introduced into pancreatic cancer cell line SW1990 by adenovirus vector. The effects of EEF1 A2 on the growth of human pancreatic cancer cell were measured by MTT. Cell cycle was detected by flow cytometry and cell growth rate was examined by soft agar cloning formation test. ResultsAfter EEF1A2 induction, the expression of EEFA1 A 2 mRNA in pancreatic cancer cell line SW1990 increased, value of A750 at 72 h was 1. 2996 ±0. 2091, number of cells was 81250, cloning efficiency at 14 d was 82%, all of these parameters were significantly higher than those in the groups of blank adenoviras vector and PBS groups (P <0.05 ) ; the percentage of G1 phase cell was 28.5%, which was significantly lower than those in the groups of blank adenovirus vector and PBS groups; the percentage of Sphase ceil was 60.9%, which was significantly higher than those in the groups of blank adenovirus vector and PBS groups (P < 0.05 ). ConclusionsEEF1 A2 gene significantly enhanced cell growth and proliferation in human pancreatic cancer in vitro.
3.Pharmacokinetics and Relative Bioavailability of Diclofenac Potassium Capsules

OBJECTIVE:To study the pharmacokinetics and relative bioavailability of diclofenac potassium capsules in normal volunteers.METHODS:A single oral 50mg dose of domestic diclofenac potassium capsules or imported diclofenac potassium tablets were given to 10 healthy male volunteers in an open randomized crossover study.Diclofenac potassium concentration in plasma were determined by HPLC.The pharmacokinetic parameters as well as relative bioavailability were measured.RESULTS:The concentration- time curves of domestic diclofenac potassium capsules and imported diclofenac potassium tablets were conformed to one compartment open model.The AUC(0~ 6)was (1 834.52± 711 06) μ g/( h· L) and (1 891.19± 859.08)μ g/( h· L) , Tmax(1.15± 0.77)h and (1.30± 0.97)h,Cmax(1 522.29± 1 063.87)ng/ml and (1 508.54± 892.44)ng/ml respectively.There were no significant differences between the two formulations in the AUC(0~ 6),Tmax,Cmax.CONCLUSION:The relative bioavailability of domestic diclofenac potassium capsule was(100.80± 16 59)% compared with the imported tablet.The result of the statistical analysis showed that the two formations were bioequivalent.
4.~(99)Tc~m-MIBI SPECT in prediction of response to chemotherapy in patients with small cell lung cancer

Objective To assess the prediction value of technetium-99m methoxyisobutylisonitrile(~(99)Tc~m-MIBI) for the effect of chemotherapy in small cell lung cancer (SCLC) patients. Methods Fifty-three patients with SCLC were divided into two groups according to the chest computed tomography (CT) examination, 39 patients in group A with complete or partial remission, and 14 patients in groups B with stable or progressive status. ~(99)Tc~m-MIBI was performed before chemotherapy. Following i. v. administration of 740 MBq ~(99)Tc~m-MIBI, SPECT imagings at 10 -30 minutes (early) and 2 -3 hours (delayed) were performed to obtain the uptake ratio of early phase tumor/normal lung tissue (ER) and the uptake ratio of delayed phase tumor/normal lung tissue (DR). The retention index (RI) was calculated as (DR-ER)/ ER × 100%. The differences of ER,DR and RI between the two groups were tested through t-test and rank sum test. Results ~(99)Tc~m-MIBI uptake was significantly higher in group A than group B: 2.33(SD:0.21) vs 2.02(SD:0.31) for the early ratio (ER) (t = -3.401, P<0.05) and 2.44(SD:0.19) vs 1.86 (SD :0.30) for the delayed ratio (DR) (t = - 6.724,P < 0.05). The median of RI in group A was signifi-cantly higher than that in group B (5.31% vs -9.26%,P <0.05). Conclusions ER,DR and RI of ~(99)Tc~m-MIBI SPECT may be helpful in predicting the response to chemotherapy in patients with SCLC.
5.XAF1 gene mediated by recombinant adenovirus induces apoptosis in pancreatic cell line BxPC-3

Objective To investigate whether expression of XAF1 mediated by edenovirus vector AdS/F35 could induce apoptosis in pancreatic cancer cell line BxPC-3 and its possible mechanisms.Methods Preconstructed recombinant Ad5/F35-XAF1 virus and negative control Ad5/F35-Null was tranfected into BxPC3:the expression of XAF1 mRNA and protein before and after tranfection was,analyzed by semi-quantitative RT-PCR and Western blot.The expressions of proteins including Caspase-3,PARP,Caspase-8 and bcl-2 were detected by Western blots.Cell apoptosis was assessed by Annexin-v/PI and TUNEL staining.Results After Ad5/F35-XAF1 tranfection,XAF1 mRNA and protein expression significantly increased,and the difference was statistically significant when compared with control group and Ad5/F35-Null group(P<0.05).The apoptosis rate was(19.90±3.09)%and(9.29±2.13)%,which was significantly different(P<0.01)from those in the Ad5/F35-Null group[(6.72±0.7)6%,(2.73±0.51)%]or in the control group[(7.22±1.53)%,(1.56±0.47)%].The expression of Caspase-3,PARP and Caspase-8 significantly increased,but the expression of bcl-2 decreased.Conclusions XAF1 plays a major role in the apoptosis of pancreatic cancer that acts thriugh the activation of death receptor pathway and mitochondrial pathway of apoptosis.
6.Effects of EEFIA2 gene on the invasion and migration of pancreatic neoplasms

Objective To observe the change of invasion and migration of the pancreatic carcinoma cell line SW1990 transfeeted with EEF1A2 gene.Methods Pancreatic carcinoma cell line SW1990 was transfected with EEF1A2 by recombinant adenovirus vector.The alteration of motility、invasion and adhesion property of SW1990 was evaluated by wound healing assay,transwell With or without Matrigel basement membrane and adhesion assay.Results Wound healing assay revealed that EEF1A2 enhanced cell motility and transwell assay with Matrigel indicated that the average numbers of transwell cells with EEFlA2 was increased from 23.25±5.23 to 65.42±8.24(P<0.05).The adhesive rate was substantially increased in EEF1A2 transfected SW1990 cells compared with control cells.Conclusions EEF1A2 gene can promote the migration.invasion and adhesion ability of pancreatic cancer cell in vitro.It is indicated that EEF1A2 may involve in the development of human pancreatic cancer by influencing cell biological characteristics.
7.Survey of Family Members of Stroke Patients on Negative Emotions and Psychological Treatment

Objective To investigate the awareness of the negative emotions on the physical rehabilitation and psychological treatment of family members of stroke patients in Gansu. Methods 1100 stroke patients from 10 hospitals of different levels in Gansu were surveyed. Results Family members who thought negative emotions can affect the recovery of limb function accounted for 77.17%, and who thought psychological treatment was needed accounted for 83.46%. Conclusion Family members of stroke patients think that negative emotions affect the recovery of limb function and psychological treatment is needed.
8.Effect of Integrated Psychotherapy on Activities of Daily Living in Post-stroke Depression

ObjectiveTo investigate the effect of integrated psychotherapy on activities of daily living (ADL) in patients with post-stroke depression. Methods60 stroke patients were divided into 2 groups: the treatment group and the control group, each including 30 cases. Both groups were treated with routine medicine and rehabilitation, while the treatment group was also treated with psychological approaches in addition. Before and 4 and 12 weeks after treatment, all the patients were assessed with the Zung Self-rating Depression Scale (SDS) and the modified Barthel Index (MBI). ResultsThere was no significant difference between these 2 groups in all the assessment parameters before treatment. The scores of SDS and MBI improved more in treatment group than in control group (P<0.05) 4 and 12 weeks after treatment. ConclusionThe integrate psychotherapy is effective on ADL in patients with post-stroke depression.
9.Awareness of Risk Factors for Stroke in Stroke Patients: 1043 Cases Survey

Objective To investigate the knowledge about risk factors for stroke in stroke patients. Methods 1043 stroke patients in 10 hospitals were investigated with self-designed questionnaire. Results The rate of awareness of risk factors for stroke was: hypertension 61.55%, hyperlipidemia 40.27%, drinking 32.21%, smoking 30.11%, diabetes mellitus 28.67%, heart disease 23.11%. 6.42% knew all the 6 risk factors, and 20.13% did not know any risk factor. The rate of awareness of diabetes mellitus was different among various aged groups (P<0.01), and the rate of awareness of hypertension, hyperlipidemia, and diabetes mellitus increased with the years of education. Conclusion The stroke patients know less about the common risk factors for stroke, who need more health education.
