1.A study on the relationship between perioperative detection of Foxp 3 mRNA level and chronic allograft nephropathy

Objective To study the relationship between perioperative detection of forkhead box protein P 3(Foxp3) mRNA level in peripheral blood mononuclear cell (PBMC ) of patients with kidney transplantation and chronic allograft nephropathy (CAN ) . Methods 35 cases of renal allografts completed in this hospital defined clinically and confirmed histopathologically were observed as CAN group ,simultaneously a retrospective case-control study was performed on 35 renal transplant recipients with normal graft function observed(control group) .Foxp3 levels of PBMC in perioperative period were dynamically determined in two groups .Real time PCR was applied to detect the expression of Foxp3 mRNA .In order to find out other risk factors influencing chronic allograft nephropathy and eliminate disturbance .The related clinic factors were compared between the CAN group and the control group .Re-sults The Foxp3 mRNA level was lower in the CAN group than the control group [(24 .1 ± 16 .2) × 10-4 vs .(52 .3 ± 27 .4) × 10-4 ,P<0 .01] .Among risk factors influencing chronic allograft nephropathy ,only acute rejection(AR) was probably different in two groups .In order to avoid disturbance ,further classification was done according to AR .The statistical results show that the Foxp3 mRNA level in two groups was not be affected by AR .Conclusion The Foxp3 mRNA level in perioperative period is associ-ated with CAN which maybe a risk factor for CAN .Foxp3 may be an index to predict the occurrence risk of chronic allograft ne-phropathy after kidney transplantation and guide individual treatment .
2.Effects of endogenous testosterone on vascular endothelial cell function and structure in rabbits of balloon injury

Objective To study the influences of endogenous testosterone on vascular endothelial cell function and structure in rabbits of balloon injury. Methods Twenty male rabbits were randomly divided into two groups: castrated group and control group.The former group animals were castrated and the later group animals received sham operation. All animals were performed with deendothelializing balloon injury in the right iliac artery. Nitric oxide (NO) and endothelin-1 (ET-1) were measured at different intervals during the experiment. After 2 weaks of endothelial denudation all animals were killed, and endothelin structure and morphological changes were observed with electron microscope and HE staining. Results There weren′t siginificant differences of plasma NO and ET-1 levels between the two groups, but plasma NO and ET-1 levels of the castrated group 2 weaks after endothelial denudation recovered to the levels before castration, while they didn′t in the control group. The extent of reendothelialization and intimal area of the harvested segments were similar between the two groups. Conclusion The decrease of endogenous testosterone level did not affect endothelial cell structure, but tended to protect function of injured endothelium.
3.The chemical component dynamic changes in compatibility of Carthamus tinctorius L. with Glycyrrhiza uralensis Fisch

AIM: To study the dynamic change of chemical components in the compatibility of Carthamus tinctorius L.with Glycyrrhiza uralensis Fisch during decoction together. METHODS: By means of HPLC analysis and phytochemical isolation technique,focusing on the differences between chemical components in co-decoction and individual decoction.Two chemical substances which showed quantitative changes during co-decoction had been isolated and identified as Licochalcone A and Licochalcone B,respectively. CONCLUSION: From the method described in this article,it could be concluded that the chemical profiles of TCM drug-pairs prepared by co-decoction are different from that of extract mixture combined by the individual extract of Carthamus tinctorius L.and Clycyrrhiza uralensis Fisch,respectively.
4.Prevalence of hypertension among "three minority ethnic groups" residents in Inner Mongolia Autonomous Region.

OBJECTIVETo survey the current situation of hypertension among "three minority ethnic groups" in Inner Mongolia Autonomous Region.
METHODSHypertension epidemiological survey among Three Minority Ethnic Groups and Han nationality aged ≥ 18 years was performed from June to December 2010.
RESULTSThe prevalence rate of hypertension among the surveyed population was 29. 4% (630/2 146) ( standardized prevalence was 34.6%). The prevalence rate of hypertension in male was 33. 8% (359/1 062) ( standardized prevalence was 39.9%), and the prevalence rate in female was 25.0% (271/1 084) (standardized prevalence was 26.6%) and the prevalence rate in male was higher than in female (P < 0.01). The prevalence rate of hypertension for the Oroqen nationality, Ewenki nationality and Daur nationality was 33.8% (92/272), 32.4% (170/524), and 30.2% (174/576) respectively (standardized prevalence was 33.7%, 33.1%, and 31.3%), and which was significantly higher than in the Han nationality (25.1% (194/774), P < 0.01) (standardized prevalence was 25.8%). The awareness, treatment and control rate of hypertension in the Oroqen nationality residents was 64.1% (59/92), 56.5% (52/92), 27.2% (25/92) (standardized rate was 63.5%, 56.8% and 27.4%), and 60.0% (102/170), 53.5% (91/170), 24.1% (41/170) ( standardized rate was 62.9%, 56.7%, 26.6%) in the Ewenki nationality residents, and 59.2% (103/174), 54.0% (94/174), 20.7% (36/174) (standardized rate was 50.3%, 54.7%, 21.4%) in the Daur nationality residents, and 65.0% (126/194), 57.7% (112/194), 27.3% (53/194) (standardized rate was 63.3%, 56.5%, 27.1%) in the Han nationality residents. Awareness rate, treatment rate and control rate were similar among different nationalities (all P > 0.05).
CONCLUSIONThe prevalence rate of hypertension among " three minority ethnic groups" residents in Inner Mongolia Autonomous Region is high and comprehensive prevention and therapy strategies are warranted to reduce the hypertension burden in these residents.

5.Simultaneous Determination of 6 Residual Organic Solvents in Omeprazole Magnesium by Headspace GC

OBJECTIVE:To establish a method for the simultaneous determination of 6 kinds of residual organic solvents in Omeprazole magnesium,such as methanol,isopropanol,acetonitrile,dichloromethane,ethyl acetate and toluenein. METHODS:Headspace GC was adopted. The determination was performed on the column of DB-624 column by temperature programming. The inlet temperature was 200 ℃,and hydrogen flame ionization detector was adopted with the temperature of 250 ℃;nitrogen gas was used as carrier gas with flow rate of 2.0 mL/min;the splitting-radio was 5 : 1,and sample size was 1 mL;the headspace tem-perature was 80 ℃ and the equilibrium time was 20 min. RESULTS:The linear range were 12.56-628.00 μg/mL for methanol(r=0.9997),20.22-1011.20 μg/mL for isopropanol(r=0.9999),1.96-97.76 μg/mL for acetonitrile(r=0.9997),3.10-154.88 μg/mL for dichloromethane(r=0.9998),20.69-1034.56 μg/mL for ethyl acetate(r=0.9998),and 3.53-176.72 μg/mL for toluene(r=0.9998);the limits of quantitation were 1.00,0.91,0.47,0.93,0.41 and 0.35 μg/mL respectively;the limits of detection were 0.31,0.30,0.14,0.31,0.12 and 0.11 μg/mL respectively;RSDs of precision,stability and reproducibility tests were lower than 3%;the recoveries were 94.53%-101.29%(RSD=2.15%,n=9),97.78%-103.42%(RSD=1.77%,n=9),96.99%-105.76%(RSD=2.59%,n=9),96.83%-102.05%(RSD=1.86%,n=9),97.98%-101.13%(RSD=0.88%,n=9)and 97.80%-102.40%(RSD=1.41%,n=9). CONCLUSIONS:The method is sensitive and accurate,can be used for the simultaneous determination of 6 kinds of residual organic solvents in Omeprazole magnesium,such as methanol,isopropanol,acetonitrile,dichloromethane,eth-yl acetate and toluenein.
6.Content Determination of Magnesium in Esomeprazole Magnesium by Inductive Coupled Plasma Atomic E-mission Spectrometry

Objective:To establish a method for the content determination of magnesium in esomeprazole magnesium. Methods:The content of magnesium in esomeprazole magnesium was determined by inductive coupled plasma atomic emission spectrometry. Re-sults:The linear range was 10.0-80.0 μg·ml-1(r=0.999 8). The average recovery was 99.55% with RSD of 1.35% (n=9). The detection limit was 0. 000 75 μg·ml-1 and the quantitative limit was 0. 002 5 μg·ml-1 . Conclusion:The method is accurate, sensitive and reproducible, which can be used for the content determination of magnesium in esomeprazole magnesium.
7.The correlation and significance of gene polymorphisms of C1236T, G2677T/A and C3435T with molecular subtypes of breast cancer

Objective To investigate the distribution of the MDR1 exon12 (C1236T), exon21 (G2677T/A) and exon 26 (C3435T) gene polymorphisms in breast cancer patients, and to analyse their relationship with molecular subtypes of breast cancer. Methods The genotyping of C1236T, G2677T/A and C3435T were detected by polymerase chain reaction (PCR)-high resolution melting (HRM) method in 400 cases of breast cancer. The Hardy-Weinberg equilibrium test was used for ge?netic equilibrium distribution of genotype. The molecular subtypes of breast cancer were classified based on St.Gallen Con?sensus 2013. The genotype distributions of C1236T, G2677T/A and C3435T in breast cancer were analyzed. Their relation?ship with molecular subtypes in breast cancer was analyzed as well. Results ①In 400 cases of breast cancer, there were 2, 3 and 2 specimens did not get genotyping results in C1236T, G2677T/A and C3435T genotype detection. The CC, CT and TT genotypes of C1236T accounted for 16.08% (64/398), 44.22% (176/398) and 39.70% (176/398). GG, GT, GA, TT and AT genotypes of G2677T/A accounted for 16.62%(66/397), 44.33%(176/397), 7.05%(28/397), 27.46%(109/397) and 4.54%(18/397). CC, CT and TT genotypes of C3435T accounted for 21.11%(84/398), 56.03%(223/398) and 22.86%(91/398) re?spectively. Hardy-Weinberg genetic equilibrium testing showed that polymorphisms of C1236T, G2677T/A and C3435T had group representation (P<0.05).②Eleven cases of HER-2 (2+) were excluded because they were not verified by FISH de?tection when performed molecular subtype of breast cancer. Luminal A subtype accounted for 41.90%(163/389), Luminal B subtype accounted for 32.65%(127/389), HER-2 over-expression subtype accounted for 13.62%(53/389) and triple nega?tive subtype accounted for 11.83%(46/389).③CT/TT genotype frequency of C3435T was significantly higher in breast can?cer patients with Luminal A subtype than that in breast cancer patients with HER-2 over-expression subtype and triple neg?ative subtype (χ2=12.011, P=0.001;χ2=13.976, P<0.001), while there was no statistical difference in C1236T and G2677T/A gene polymorphism between different molecular subtypes of breast cancer (P>0.05). Conclusion C3435T gene polymor?phism can explain more accurately heterogeneity of breast cancer. CT/TT genotype in different molecular subtypes of breast cancer may be more sensitive to drug treatment.
8.Influence of ABCB1 gene polymorphisms on breast cancer with paclitaxel-based chemotherapy

Objective To detect gene polymorphisms of ABCB1/MDR1 in breast cancer and to analyze the association between ABCB1 gene polymorphisms and curative effect of paclitaxel-based chemotherapy in breast cancer.Methods Genotyping of ABCB1exon12 (1236),exon21 (2677)and exon26 (3435)was determined by polymerase chain reaction (PCR)-high resolution melting (HRM)method in 146 cases of female stage IV breast cancer to explore the relationship between the efficacy of chemotherapy in breast cancer patients with paclitaxel chemotherapy.Results In 146 patients with stage IV breast cancer,C1236T CC genotype accounted for 15.86%, CT genotype 44.83%,TT genotype 44.83%,G2677T/A GG genotype 16.67%,GT genotype 45.83%,GA genotype 6.25%,TT genotype 28.47%,AT genotype 2.78%,C3435 CC genotype 22.22%,CT genotype 56.25%,and TT genotype 21.53%.ABCB1 gene polymorphisms did not significantly differ between patients of Hui and Han Nationalities (P>0 .0 5 ).Hardy-Weinberg genetic equilibrium testing showed that polymorphisms of C1236T,G2677T/A and C3435T had group representation (P>0.05).In 146 stage IV breast cancer patients who had received paclitaxel-based chemotherapy,CT/TT genotype of C3435T site had a better response rate (74.11%) (χ2 =12.556,P<0.05),better curative effect than CC genotype (OR=4.183,95% CI 1.838 -9.521,P<0.05),T allele carriers more efficient than C allele carriers with paclitaxel-based chemotherapy (χ2 =8 .5 0 7 ,P<0 .0 5 ). Conclusion Detection of ABCB1 C3435T gene polymorphisms has a significantly clinical value in predicting the efficacy of taxanes chemotherapy in breast cancer patients.
9.The effects of WeChat platform nursing intervention on complications of ear expanded skinflap among children

Objective Comparative effect of WeChat platform nursing intervention and routine nursing intervention on reducing the incidence of complications of children skin expansion in the process of external ear reconstruction. Methods 132 cases of congenital microtia received skin expander implantation at our institution from Jan 2014 to Aug 20 14 were included as control group. 122 cases of congenital microtia received skin expander implantation at our institution from Sep 2014 to Apr 2015 were included as the intervention group. Children and their families in the control group were treated with routine propaganda and education, and the intervention group were added with internet nursing intervention. Collection and analysis the family health knowledge degree and the occurrence of complications in children of the two groups. Results The score of health knowledge about skin expansion in the intervention group was (33.67 ± 2.44),which was significant higher than that of in the control group (25.07 ± 2.02), t=9.71, P < 0.01. The incidence rate of complication in the intervention group was 2.46%(3/122), which was significant lower than that of in the control group, 8.33%(11/132),χ2=4.20, P<0.05. The satisfaction rate of patients′family member in the intervention group was 95.08%(116/122), which was significant higher than that of in the control group, 84.09%(111/132 ),χ2=15.65, P<0.05. Conclusions WeChat platform nursing intervention can reduce expansion flap complications occurrence rate in the periods of expander. It is worthy of clinical application.
10.Determination of Residual Organic Solvents in Erlotinib Hydrochloride by Headspace Gas Chromatography

Objective:To establish a headspace capillary gas chromatography method for the determination of residual solvents in erlotinib hydrochloride .Methods:A DB-624 capillary column (30 m ×0.53 mm, 3.0 μm) was used and the carrier gas was nitro-gen.The flow rate was 2.0 ml· min-1 and the inlet temperature was 190℃.The FID detector temperature was 230 ℃.The column temperature program was with the initial temperature of 35℃( maitaining 8 min) , risen to 170℃with the rate of 28℃· min-1 ( main-taining 8 min) , and then risen to 200℃with the rate of 32℃· min-1 ( maintaining 7 min) .The headspace vial temperature was 100℃and the time was 30 min.Results:Ethanol, isopropanol, methylene chloride and n-butanol had a good linear relationship within the range of 0.68-409.14 μg· ml-1 (r=0.999 8),0.67-404.88 μg· ml-1 (r=0.999 8),1.71-51.31μg· ml-1 (r=0.999 7) and 0.72-431.12 μg· ml-1(r=0.999 8), respectively.The average recovery was 99.0% (RSD=0.41%, n=9), 100.2%(RSD=0.52%, n=9),97.1%(RSD=1.75%, n =9) and 102.5% (RSD=1.08%, n=9), respectively.Conclusion: The method is simple and accurate , which can be used for the determination of four residual organic solvents in erlotinib hydrochloride .
