1.The analysis for allelic polymorphism of HLA-A,B and DRB1 in Han race population in Xining region of China

Objective:To investigate the HLA-A,B,DRB1 allele polymorphism of the Han race population in Xining region of China.Methods:Polymerase chain reaction and sequence-specific olignucleotide probe hybridizations were used to detect HLA-A,B and DRB1 alleles in 73 unrelated heath Han individuals from Xining region,and the results were compared with those of Han populations in China.Results:Eleven of alleles were detected and identified for HLA-A,Twenty for HLA-B,and seventeen for HLA-DRB.HLA-A02,A11,A30,A33;HLA-B13,B15,B37,B40,B46,B58;HLA-DRB103,DRB104,DRB109,DRB112,DRB113,DRB3,DRB4 were the most common alleles.Conclusion:The allelic polymorphism of HLA loci of Han race population in Xining area has owns characteristics.
2.The effect of tropisetron combined with sulpiride on chemotherapy-induced cisplatin program nausea and vomiting

Objective To investigate the efficacy and safety of tropisetron combined with sulpiride in treatment of chemotherapy-induced cisplatin program nausea and vomiting,so as to explore the effect to motilin and gastrin.Methods A total of 84 patients with chemotherapy-induced cisplatin program nausea and vomiting were divided into study group (44 cases) and control group (40 cases) by random digits table method,the patients in two groups were given tropisetron hydrochloride intravenous injection,and the study group was added sulpiride.The efficacy and side effects and effect to motilin and gastrin were observed.Results The fully control rate and efficient rate in acute nausea was 59.09% (26/44),37.50% (15/40),and 90.91% (30/44),72.50% (29/40) in study group and control group,and there was significant difference (P < 0.05).The fully control rate and efficient rate in acute vomiting was 61.36 % (27/44),37.50% (15/40),and 88.64% (39/44),67.50% (27/40) in study group and control group,and there was significant difference (P <0.05).The fully control rate and efficient rate in delayed nausea was 54.54% (24/44),32.50% (13/40),and 79.55% (35/44),57.50% (23/40) in study group and control group,and there was significant difference (P < 0.05).The fully control rate and efficient rate in delayed vomiting was 45.45 % (20/44),22.50% (9/40),and 75.00% (33/44),52.50% (21/40) in study group and control group,and there was significant difference (P< 0.05).The plasma motilin after treatment was lower than that before treatment in study group and control group[(308.35 ± 14.59) ng/L vs.(370.59 ± 15.72) ng/L and(341.87 ± 18.38) ng/L vs.(365.36 ± 23.72) ng/L],gastrin was higher than that before treatment in study group and control group [(206.97 ± 12.29) ng/L vs.(164.56 ± 14.17) ng/L and (171.58 ± 13.53) ng/L vs.(158.42 ± 17.29) ng/L],and there was significant difference (P < 0.05).There was significant difference in the plasma motilin and gastrin after treatment between two groups (P < 0.05).There was no significant difference in the occurrence of adverse drug reactions between two groups (P > 0.05).Conclusion Tropisetron combined with sulpiride than the routine use of tropisetron can obtain the better the antiemetic effect.

4.Advances of long non-coding RNA UCA1 in cancers

Long non-coding RNA urothelial carcinoma associated 1 (UCA1) is initially discovered and named in bladder cancer tissue,which is highly expressed in multi types of tumor tissues,such as bladder cancer,ovarian cancer,lung cancer,suggesting that UCA1 acts as oncogene.UCA1 is confirmed to regulate tumor cell proliferation,apoptosis,invasion and migration,which plays an important role in the occurrence and development of cancers.UCA1 is expected to become a new biomarker for diagnosis,prognosis and drug susceptibility,which may be a promising therapeutic target of cancer.
5.HLA class-Ⅰ antigens in Gansu Chinese Han nationality with leukemia

Objective:Host genetic factors are known to contribute to disease susceptibility.They may also be important in defining the pattern of disease presentation and progression,as well as its overall prognosis.However,no consistent HLA class-Ⅰ associations have been established in leukemia by PCR/SSOP in Gansu Chinese Han.Such studies have been reported in other counties,with conflicting results.This is the first PCR-based HLA class-Ⅰ association study in northwestern Chinese Han nationality leukemia.Methods:Compared HLA class-Ⅰ in 43 Chinese leukemia patients and 66 healthy Chinese controls as determined by polymerase chain reaction and sequence-specific olignucletide probe hybridization(PCR/SSO) DNA analysis.The present findings imply that HLA-associated genetic factors influence the risk for the development of leukemia.
7.Effect of RASSF1A gene promoter methylation on its expression level in cervical cancer tissue and its clinical significance

Objective To research the promoter methylation level of RAS association domain family 1A (RASSF1A) and RASSF1A gene mRNA expression level in cervical cancer tissue, and to analyze their relationships with clinicopathological parameters of cervical cancer and the clinical significance.Methods The RASSF1A gene promoter methylation and RASSF1A gene mRNA were detected respectively by methylation specific PCR and quantitative real-time PCR method in 40 cases of cervical cancer tissues and corresponding adjacent tissues.Results RASSF1A mRNA expression level in cervical cancer (0.26±0.05) was significantly lower than that in adjacent tissues (0.28±0.03), and the difference was statistically significant (t=2.27, P=0.026).The methylation rate of RASSF1A gene promoter region (0.71%±0.04%) was significantly higher than that in adjacent tissues (0.66%± 0.03%), and the difference was statistically significant (t=6.78, P=0.000).The expression of RASSF1A mRNA was significantly correlated with pathological differentiation (t=3.31, P=0.002), International Federation of Gynecology and Obstetrics (FIGO) stage (t=2.13, P=0.040), lymphatic metastasis (t=2.56, P=0.015).The promoter methylation level of RASSF1A gene was significantly correlated with pathological differentiation (t=2.08, P=0.045), FIGO stage (t=2.66, P=0.011), lymphatic metastasis (t=2.22, P=0.033), depth of invasion (t=2.12, P=0.041).Conclusion The RASSF1A gene promoter region methylation level and the RASSF1A gene mRNA expression level are associated with the malignant degree of cervical carcinoma.The RASSF1A gene promoter region methylation level may be used as a reference indicator for predicting the risk of metastasis of cervical cancer.
8.Effect of Rebamipide on the quality of gastric ulcer healing in rats

Objective To evaluate the effects of Rebamipide,Omeprazol and their combination in treatment of gastric ulcer.Methods Forty gastric ulcer models were induced by acetic acid and randomly divided into 4 groups with 10 in each.The rats in control,Rebamipide,Omeprazol and combination groups were received saline (3 ml/d),Rebamipide (60 mg/kg),Omeprazol (10 mg/kg) as well as Rebamipide combined with Omeprazol for 7 days,respectively.After sacrifice,size of the ulcer and focal layer structures were measured in vitro by using miniature probe ultrasonography(MPS),mucosal sections were stained with H-E for histological examination,levels of interleukin-8/ prostaglandin E2 (PGE2) and methylenedioxyamphet amine (MDA) were evaluated by ELISA and TBA,respectively. Results ① Miniprobe ultrasonography revealed that comparing to control group[(22.3 ± 1.8)mm2], the maximal length of ulcer in Rebamipide [(9.2 ± 1.0) mm2 ],Omeprazol [(9.8 ± 1.3) mm2] and combination [(4.8 ± 1.2 )mm2 ] groups were significantly reduced ( P<0.05 ).② In vitro,MPS examination revealed that layer echo structure of gastric wall was partially rebuilt in three medication groups,③ Histopathologically,comparing to control group,ulcer range and inflammatory infiltration in all medication groups were less important.④ The mucosal level of IL-8 in control group [(1387.8 ± 132.6)pg/ml] was higher than that in Rebamipide group[(970.0 ± 91.6) pg/ml],Omeprazol group [(1102.2±76.9)pg/ml] and combination group[(934.4 ± 110.2) pg/ml] (P<0.05).⑤ The mucosal level of MDA was (9.4±1.4)nmol/ml and (8.5 ± 4.8)nmol/ml in Omeprazol and Rebamipide groups, respectively,which was higher than that in combination group[(4.6±1.4)nmol/ml-] and lower than that in control group[(13.0±2.6)nmol/ml].⑥ The PGE2 level was significantly increased in Omeprazol, Rebamipide and combination groups [(96.9 ± 7.0) pg/ml,( 103.5 ± 12.5 ) pg/ml,and(235.5 ± 26.0) pg/ m,respeetively] compared to control group[(55.0±22.5)pg/ml,P<0.05)].Conclusions Rebamipide, Omeprazol and combination regimen may improve quality of ulcerative healing by increasing level of PGE2 and decreasing levels of IL-8 and MDA in gastric mucosa,and this will result in potentially reducing the rate of ulcer recurrence.Combination regimen shows more anti-ulcer effect comparing to monotherapy.
9.Histopathologic study of lens epithelial cells in different types of cataracts

Background Cataract was directly associated with the damage to the structure and function of lens epithelial cells (LECs).In those patients who suffer from cataracts,morphologic changes of LECs is the most compelling evidence confirming loss of cellular structure and function of LECs.So,learning about the morphological changes of LECs of the different types of cataracts is very important for study on biological behaviors of LECs in different environments or diseases.Objective This study was to evaluate the morphological and ultrastructural changes of the LECs in different types of cataracts.Methods Anterior capsular member from age-related cataracts,diabetic cataracts and high myopia complicated cataract were obtained during the cataract surgery and 15 pieces for each.Trypan blue and alizarin red (TB-AR) stain,haematoxylin and eosin stain were performed in the samples to assess the viability and morphology of LECs.The ultrastructural change of LECs was observed under the transmission electron microscope.The features of the LECs were compared among the different types of cataract.Results TB-AR stain showed that LECs were polygon in shape with the mosaic arrangement and round cell nucleus,and a few dead cells were seen in the samples age-related cataract.In the diabetic samples,LECs largened from swelling with different sizes.More dead cells were found in the high myopia complicated cataract.Haematoxylin and eosin stain exhibited that the anterior capsular membrane presented a homogeneuous membrane,and monolayer LECs attached firmly anterior capsular membrane in the samples of related cataract.Majority of the cells had the intact structure.However,the interspaces between cells and capsular membrane were found in diabetic cataract.Also,smaller LECs were seen in high myopia complicated cataract with the irregular cell morphology.Under the transmission electron microscope,LECs presented the normal shape,intact intercellular tight junction and well attachment between cells and capsular membrane in the samples of the age-related cataract.In the samples of the diabetic cataract,edema of LECs and large intercellular spaces were seen.In addition,the jogged pump and vacuolar degeneration of cytoplasm were revealed in the high myopia complicated cataract.Conclusions The degeneration,necrosis and apoptosis was a common pathological basis of age-related cataract,diabetic cataract and high myopia complicated cataract.However,the damage of LECs was more serious in diabetic cataract and high myopia complicated cataract than that of agerelated cataract.
10.Inhibition of homo sapiens eukaryotic translation elongation factor 1 alpha 2 expression induces apoptosis in pancreatic cell line and its possible mechanisms

Objective To elucidate whether down-regulation of homo sapiens eukaryotic translation elongation factor 1 alpha 2 (EEF1A2) expression induces apoptosis in pancreatic cancer cells and its possible mechanisms. Methods Two siRNAs targeting human EEF1A2 were synthesized and the siRNA/liposome complexes were transfected into the pancreatic cancer cell line BxPC-3. RTPCR and Western blot were used to analyze the change of EEF1A2 expression and the apoptosis rate of BxPC-3 cells was studied using Annexin-V/PI assay. To identify the mechanisms involved, the apoptosis associated proteins such as caspase-3, caspase-8, caspase-9, PARP, cytochrome C and Bid were detected by Western blotting. Results Both EEF1A2-targeting siRNAs reduced the EEF1A2expression, and the No. 2 siRNA inhibited EEF1A2 expression to less than 25 % in mRNA and protein levels. Down-regulation of EEF1A2 expression in BxPC-3 cells enhanced cell apoptosis (15.28% ±3.65%) at a greater level than negative siRNA-expressing cells (10. 11% ± 3. 05%) or mock cells (9.41 % ±4.14 %). Furthermore, reduction of EEF1A2 activated the pro-caspase-8, pro-caspase-3,pro-caspase-9,PARP and Bid to their active forms, and increased the expression of cytochrome C.Conclusions These data suggest that EEF1A2 down-regulation could significantly induce apoptosis of pancreatic cancer cell line BxPC-3, which is likely mediated by the death receptor and mitochondrial apoptotic pathways.
