Objective To observe the effect of foam dressing and polyurethane gel pad in prevention of pressure ulcer for surgical patients.Methods 140 patients in neurosurgery department were randomly divided into the control group and the experimental group with 70 patients in each group.The control group received foam-rubber cushion for conventional nursing,the experimental group used foam dressing and polyurethane gel pad to prevent pressure ulcer.Instantly after the operation and 30 min later,the situation of skin pressure ulcer was recorded respectively,the maximum diameter of erythema was measured and the process was put into phases.24 hours after the operation,patients of the above two groups were visited and the situation of skin pressure ulcer was recorded.The skin pressure ulcer and the maximum diameter of erythema were compared between the two groups.Results Cases with phase Ⅰ and phase Ⅱ pressure ulcer in the experimental group were less than those of the control group at three time points,which were instantly after the operation,30 min and 24 hours later.The diameter of erythema at three time points was also less than the control group.Conclusions Foam dressing and polyurethane gel pad could effectively prevent or alleviate pressure ulcer after operation and therefore is worthy of clinical application.

Objective To retrospectively analyze the effectiveness and safety of drug-eluting stents (DES) on prevention of restenosis in old female patients (≥65 years) with multi-vessel coronary artery disease. Methods 387 old female patients with multi-vessel coronary artery disease underwent percutaneous coronary intervention (PCI) were divided into the DES group (n=139) and the metallic stent (BMS) group (n=248). Acute and long-term outcomes were compared between the two groups. Results The ratio of patients with diabetic disease was higher in DES group than that in BMS group (P0.05). The rates of restenosis shown by angiography and major adverse cardiac events (MACE) were significantly lower in the DES group compared with BMS group (4.2% vs 15.8%, 7.5% vs 17.3%, P

Objective To evaluate long-term clinical effectiveness in old female patients with multi-vessel coronary artery disease (≥65 years) treated with percutaneous coronary intervention (PCI). Methods 601 female patients with multi-vessel coronary artery disease underwent PCI were divided into the elder group (≥65 years, n=354) and the younger group (0.05). Conclusion PCI can be performed in old female patients with multi-vessel coronary artery disease with high success rate and few complications. Long-term clinical outcomes of PCI in old females with multi-vessel disease are similar to younger females.

OBJECTIVE To study infection and drug resistance of mycoplasma from semen of infertility men. METHODS Mycoplasma from semen of infertility men was identified by cultivation,and the sensitivities to drugs were also performed. RESULTS In 267 cases the positive rate of mycoplasma was 46.07%.Simple infection of Ureaplasma urealyticum(Uu) accounted for 41.57%(11),and Mycoplasma hominis(Mh) 1.50%(4),and the mixed 3.00%(8).The result of drug sensitive test showed that sensitivities of mycoplasma to minocycline,doxycycline and josamycin were the highest,and then were roxithromycin and azithromycin.The drug resistance of mycoplasma to ofloxacin and clindamycin was the highest. CONCLUSIONS The infectious rate of mycoplasma from semen of infertility men is on big rise.It is important to culture and test the drug sensitivities of mycoplasma to use drugs rationally.

OBJECTIVE To study infection status and drug resistance of Ureaplasma urealyticum(Uu) and Mycoplasma hominis(Mh) in the female genital tract.METHODS The retrospective analysis of identification and the antimicrobial susceptibility testing of mycoplasma in 2263 female cervical secretions from Jan 2007 to Jun 2008 were conducted.RESULTS In 2263 cases,the positive rate of mycoplasma was 66.46%.The infection only by Uu accounted for 45.03%,by Mh for 1.46%,and the mixed infection for 19.97%.The results of drug sensibility test showed that drug resistances of mycoplasma were diffirent among three types of mycoplasma infections.CONCLUSIONS The infectious rate of mycoplasma from female cervical secretions is on big rise.It is important to culture and test the drug sensitivities of mycoplasma for choosing drugs rationally and control the resistant strains.

OBJECTIVE To investigate conditions and features of antimicrobial resistance in Streptococcus pneumoniae clincal isolates.METHODS Totally 203 strains of S.pneumoniae were isolated from Jan 2005 to Dec 2006 in Yuying Pediatric Hospital of Wenzhou Medical College.Then the antimicrobial susceptibility test to ten drugs and the MICs of penicillin were detected.RESULTS In 203 S.pneumoniae strains,all were sensitive to vancomycin,were sensitive to 98.0%,levofloxacin,the sensitivity of penicillin,erythromycin,tetracycline,clindamycin and SMZ was lower than 15.0%.The penicillin insensitivity of 203 strains reached 86.2%.There were 24 of the 203 strains for which penicillin MICs were ≥4 ?g/ml.CONCLUSIONS S.pneumoniae clinical strains are resistant to most of the commonly used antibiotics.The percentage of high level multidrug resistant strains is very high.Vancomycin and levofloxacin are the most sensitive drugs for S.pneumoniae clinical strains,the next are chlorophenicol.Erythromycin,SMZ,tetracycline and clindamycin aren′t useful in treating S.pneumoniae infection.

AIM: To investigate the effect of purifried L-amino acid oxidase (LAO) from bungarus fasciatus snake venom on apoptosis and growth of HUVCE cell line. METHODS: The L-amino acid oxidase was purified by SP-sepharose HP column followed by Heperin-Sepharose (FF) column. The homogeneity of the preparation was examined by SDS-PAGE and the molecular weight of LAO was determined by SDS-PAGE and high performance liquid chromatography (HPLC) gel-filtration. The MTT assay was used to detect the viability of cells. Flow cytometry and laser confocal microscopy were used to identiyfy the cell cycle and apoptotic morphology after cells treated with LAO. RESULTS: An L-amino acid oxidase (BF-LAO) was successfully purified from the venom of bungarus fasciatus. It showed a single band in SDS-PAGE under both reduced and non-reduced conditions. The apparent molecular weight was determined to be 60 kD by SDS-PAGE and 70 kD by HPLC gel filtration. LAO inhibited growth and induced apoptosis of HUVCE cell line in a dose-dependent manner after 12 h incubation, with the 50% inhibitory concentration (IC_ 50 ) being of 2.8 mg/L. Flow cytometry and laser confocal microscope showed a typical apoptotic peak and morphological changes of these cells. CONCLUSION: The L-amino acid oxidase from bungarus fasciatus snake venom could inhibit the HUEVC cell growth and induce the cell apoptosis.

Objective To investigate the human papillomavirus 16/18 (HPV16/18) infective status and its relationship with the expression of p16,CK17,Ki-67 in immature squamous metaplasia(IM),cervical intraepithelial neoplasia (CIN) and cervical carcinomas (CC).Methods In situ hybridization (ISH) and immunohistochemistry were applied to detect the expression of HPV16/18,p16,CK17,Ki-67 on tissues from 30 IM,60 CIN and 30 CC.Results The expression rates of HPV16/18 in LSIL,HSIL and CC were 30 ％ (9/30),60 ％ (18/30) and 77 ％ (23/30),higher than that in IM [6.7 ％ (2/30)].HPV16/18 was positively expressed with the progression of cervical squamous epithelial lesions.The expression rates of CK17 in IM was 90 ％ (27/30),higher than that in LSIL,HSIL and CC [17 ％ (5/30),10 ％ (3/30),6.7 ％ (2/30)].The expression rates of p16,Ki-67 were 83 ％ (25/30) and 87 ％ (26/30) in LSIL,90 ％ (27/30) and 93 ％ (29/30) in HSIL,97 ％ (29/30) and 97 ％ (29/30) in CC,higher than that in IM [13 ％ (4/30) and 10 ％ (3/30)].The expression of p16,Ki-67 were particularly seen in HPV16/18-positive HSIL and CC,and the correlation were observed.Conclusion HPV16/18 infection is highly associated with the degree of squamous intraepithelial neoplasia and carcinomas of cervix and upregulated p16 and Ki-67,which suggested that HPV16/18 maybe cause mutation of p16 gene.With the progression of cervical squamous epithelial lesions,positive expression rates of CK17 were decreased compared with increased of HPV16/18,p16,Ki-67.Combined detection of HPV16/18,p16,Ki-67 is helpful for diagnosis and classification of cervical squamous epithelial lesions.

Objective To establish differential protein expressing profiles of human gastric adenocarcinoma cell in primary or metastatic lymph node tissues by two-dimensional differential gel electrophorosis (2-D DIGE) so as to investigate the metastatic molecular mechanism of the gastric cancer.Methods After obtaining 6 samples of human primary gastric cancer and metastatic lymph node tissues,with manual no-staining frozen sections microdissection,human gastric adenocarcinoma cells from primary or metastatic lymph node tissues were isolated,and then the total proteins were extracted and purified.Highly sensitive 2-D DIGE was used to separate the total protein differentially expressed in the cells.The proteins were visualized by using a fluorescence scanner at appropriate wavelengths for Cy2,Cy3 and Cy5 dyes (Typhoon 9400).Image analysis was carried out with the DeCyder-Differential analysis software (Biological Variation Analysis version 5.0).Results Not only can the study procure defined adenocarcinoma cell populations from gastric primary or metastatic lymph node tissues,but also can resolve the problem of the change in 2-D DIGE patterns because of the varying in protein changes owing to dyeing.All these showed that the technique was simple,easy to perform,versatile and of particular usefulness when laser capture microdissection (LCM) was practically unavailable.The 2-D DIGE patterns with high resolution and reproducibility from adenocarcinoma cells in gastric primary or metastatic lymph node tissues were obtained.The number of spots in Gel1,Gel2 were 1 416 (similar 1 062,decrease 277,increase 77),1 299 (similar 1 050,decrease 157,increase 92),respectively.A total of 11 differential proteins were acquired by image analysis with DeCyder-Differential analysis software (Biological Variation Analysis version 5.0).Conclusions In this report,a simple,easy to proform method of protein epuration,manual no-staining frozen sections microdissection is described,and have used the highly sensitive 2-D DIGE for the identification of proteins differentially expressing in human gastric adenocarcinoma cells from primary or metastatic lymph node tissues.These results provide a fundamental basis for further study of metastatic mechanism of gastric cancer and screen its specific markers.

BACKGROUND: The study about the multiple differentiation potentials of the mesenchymal stem cells is still on the stage of the animal experimentation. Can mesenchymal stem cells have the ability to differentiate into certain tissues and develop the corresponding functions after they are transplanted into certain tissues?OBJECTIVE: To observe the differentiation of the bone marrow mesenchymal stem cells into the nerve and its effect on the nerve functional recovery after they are transplanted into the peripheral zone of the ischemic infarction focus of the cerebral cortex.DESIGN: A randomized controlled study.SETTING: The Department of Anatomy of the School of Basic Medical College of Sun Yat-sen University; the Department of Neurology of the First Hospital of Sun Yat-sen University; Department of Pathology of the Medical College of Jinan University.MATERIALS: The experiment was conducted at the Medical College of Sun Yat-sen University from November 2002 to November 2003. Forty-eight male SD rats were chosen and randomly divided into two groups, with 32 rats in cerebral infarction model group and 16 in the non-model control group. In the cerebral infarction group, the rats were randomly divided again into two groups: 16 rats in the transplantation group and 16 in the phosphate buffered fluid group. The anterior fontanel taken as the reference point, 5 μL(5 × 104 L-1) of the bone marrow mesenchymal stem cells or the phosphate buffered fluid was respectively transplanted at the site 3 mm away on the caudal side and 1.5 mm aside at the depth of 2. 0 - 3. 0 mm.METHODS: The bone marrow mesenchymal stem cells were obtained through the separation and purification of the bone marrow of the ribs taken away in the thoracic surgery from the patients without the hematological diseases, and then the cells underwent in vitro culture, the amplification and the identification. At the 2nd and 6t1 weekend after the transplantation,the rats of every group were anesthetized, and the samples were taken from the transplantation site and made into the 25 μm of continuous frozen section. Then, the immunohistochemical method was used for the detection of the expressions of neuron specific enolase, neurofibril protein, glial fibrillary acidic protein, and nidogen to evaluate the ability of the bone marrow mesenchymal stem cells to differentiate into neurons and glial cells. Eight rats of the transplantation group and 8 rats of the phosphate buffered fluid group were randomly taken out, and 2 and 6 weeks before and after the transplantation the bar walking test evaluation method was used to identify the general status and reaction ability of the rats. Sixteen rats of the control group were assessed at the same time.enolase, neurofibril protein, glial fibrillary acidic protein and nidogen in the bar walking test.2nd weekend after the transplantation, there were positive expressions of glial fibrillary acidic protein and nidogen at the transplantation site of the bone marrow mesenchymal stem cells. At the 6th weekend there were positive expressions of neuron specific enolase and neurofibril protein at the transplantation site of the bone marrow mesenchymal stem cells. While in the phosphate buffered fluid group, there were negative expressions of neuron specific enolase, neurofibril protein, glial fibrillary acidic protein and nidosymptoms in the control group and the evaluation scores were all 9. 2 weeks after the transplantation, and the evaluation scores of the motor function in the transplantation group were higher than the ones in the phosphate buffered fluid group, [(6.7±0.9), (5.3-±1.0), (P ＜0.05)]. Six weeks after the transplantation, the evaluation scores of the motor function in the transplantation group were also higher than those in the phosphate buffered fluid group[(8.9±1. 1),(7.2±0.8),(P ＜0.05)].CONCLUSION: After their transplantation into the central nervous system,the bone marrow mesenchymal stem cells showed the ability to differentiate into neurons and glial cells, in which the characteristics of some neurons and glial cells were found. Bar walking test found that the evaluation scores of the motor function in the transplantation group were higher than those in the phosphate buffered fluid group, which suggests that the bone marrow mesenchymal stem cells have a significant effect on restoration of the functions of the nerves.
