1.Study on the Preparation Technique and Quality of Xuelian Freeze-dry Sterile Injection powder

OBJECTIVE:To prepare xuelian freeze-dry sterile injection powder and to establish a quality control for this drug.METHODS:The best formulation and preparation technique of the powder for injection was optimized by screening the variety and quantity of vehicle;the contents,which were determined by UV,together with the finger print were taken as the main index to study the quality.The stability was observed through long test.RESULTS:Take the40%mannitol as excipient;The mean recovery results was99.4%(RSD=0.90%);The freeze-dry sterile injection powder could be reflection the finger print character of extract fluid better than injection.The preparation was stable after12months storage at room tempera-ture.CONCLUSION:The formulation is reasonable,the preparation technique is feasible and the quality is controllable.
2.Evaluation of left ventricular global and regional systolic function in patients with systemic lupus erythematosus ;by three-dimensional speckle tracking imaging

Objective To evaluate left ventricular(LV)global and regional systolic function by three-dimensional speckle tracking imaging (3D-STI)in patients with systemic lupus erythematosus (SLE). Methods Twenty-two patients with SLE were selected randomly as SLE group and twenty-two healthy people were included as control group.3D-STI was performed in all participants to get global and regional strain parameters,including global longitudinal peak systolic strain(GLS),global circular peak systolic strain (GCS ),global radial peak systolic strain (GRS ),global area peak systolic strain (GAS ),segmental longitudinal peak systolic strain (SLS),segmental circumferential peak systolic strain (SCS),segmental radial peak systolic strain (SRS),and segmental area peak systolic strain (SAS).The sensitivity and specificity of GCS and GAS reflecting LV systolic function in patients with SLE were analyzed under the receiver operating characteristic (ROC)curve.Results ①The global strain:compared with the control group,GCS and GAS decreased significantly in SLE group (P <0.01),there was no significant change in GLS and GRS(P >0.05).②The regional strain:compared with the control group,the SCS in segments of basal anteroseptal,basal inferoseptal,mid anterior,mid inferosptal,mid anterolateral,apical inferior,apical lateral decreased significantly in SLE group.The SAS of basal anterior,basal inferoseptal,mid inferosptal, mid anterolateral,apical anterioe,apical lateral decreased significantly in SLE group (P <0.01).The SCS in segments of apex and the SAS of mid anterior,apical septal,apex decreased in SLE group (P <0.05).③The ROC curve:the sensitivity and specificity of GCS on detecting the LV systolic dysfunction were 59.1%and 87.5%,and those of GAS were 77.3% and 83.3%,respectively.Conclusions 3D-STI is a convenient and accurate way to early detect LV global and regional systolic dysfunction in SLE patients.
3.The efficacy and safety study of JCS-01 non-invasive focused ultrasound fat reduction machine.

OBJECTIVETo evaluate the safety and efficacy of using the JCS-01 focused ultrasound fat reduction machine for noninvasively breaking fat cells and abdomen sculpting treatment.
METHOD40 qualified subjects were enrolled in the study per the IRB-approved protocol and randomly assigned into trial and control groups. The fat reduction treatments were performed using the JCS-01 focused ultrasound fat reduction machine made by Beijing 3H Medical Technology Co., Ltd. The trial group received ultrasound energy treatment in their abdominal subcutaneous adipose tissue on the 1st and 7th days of the trial. The control group followed the similar procedure except that the ultrasound energy was turned off during treatments. Both the trial and control groups were followed-up on 1,7,14, 35 days post treatment.
RESULTSThe waist circumference measured on 14, 35 days were reduced 2.52 cm +/- 1.04 cm and 3.31 cm +/- 0.67cm in comparison with the baseline values in the trial group, reaching the protocol hypothesis of at least 1.5 cm abdominal circumference reduction, which demonstrated the treatment efficacy. The adverse events included minor and transient thermal sensation and mild pain in the treated abdomen area in the trial group. No other adverse events were observed. Clinical biochemistry assay testing showed that the subjects liver and kidney function indicators had no significant change.
CONCLUSIONIt is safe and effective to use the JCS-01 focused ultrasound fat reduction machine for abdomen fat reduction treatment.

4.Effects of ingredients from Chinese herbs with nature of cold or hot on expression of TRPV1 and TRPM8.

OBJECTIVETo study the effects of the ingredients from Chinese herbs with the nature of cold or hot on the expression of TRPV1 and TRPM8.
METHODThe effects of ingredients from herbs on primary culture DRG neurons are observed in vitro. The expression quantity of gene is detected by the method of real time PCR. the 2 (-deltadeltaCT) method is applied to analyze the data.
RESULTIngredients from herbs with the nature of cold up-regulate the expression level of TRPV1 and down-regulate that of TRPM8, especially under the temperature condition of 39 degrees C; while ingredients from herbs with the nature of hot up-regulate the expression level of TRPM8 and down-regulated that of TRPV1, which is more significant under the temperature condition of 19 degrees C.
CONCLUSIONThe regulatory changes of TRPV1 and TRPM8 mRNA expression induced by the chemical ingredients might be related to the cold and hot natures of the herbs from which the ingredients are extracted. And this could be one of the therapeutic mechanisms for the treatment of Chinese herbal medicines to cold- and heat-related diseases.

5.The application value of urine γ-synuclein in the diagnosis of bladder cancer

Objective:To explore the application value of urine γ-synuclein (SNCG) in the diagnosis of bladder cancer.Methods:A total of urine samples from 129 patients with bladder cancer (malignant lesion group), 157 patients with urinary system benign lesions (benign lesion group), and 177 healthy people (the healthy control group) from January 2017 to April 2020 in the Fifth Clinical Medical College of Shanxi Medical University and Shanxi Provincial Cancer Hospital were collected. The concentration of SNCG in the collected urine was detected by using enzyme-linked immunosorbent assay. The receiver operating characteristic (ROC) curve was drawn to determine its sensitivity, specificity and accuracy for the diagnosis of bladder cancer.Results:The urine SNCG concentration in malignant lesion group [4.28 ng/ml (0.53-8.79 ng/ml)] was higher than that in healthy controls [1.44 ng/ml (0.56-3.51) ng/ml, H = 122.9, P < 0.01] and benign lesion group [1.97 ng/ml (0.51-5.87) ng/ml, H = 88.2, P < 0.01], and the concentration of urine SNCG in benign lesion group was higher than that in healthy controls ( H = 17.1, P < 0.01). ROC area under the curve (AUC) of urine SNCG in differentiating benign lesion group from healthy controls was 0.871(95% CI 0.819-0.923, P < 0.01), the best cut-off value was 2.79 ng/ml, the diagnostic sensitivity and specificity was 0.798 and 0. 977, respectively. AUC of urine SNCG in differentiating malignant lesion group from benign lesion group was 0.823(95% CI 0.769-0.877, P < 0.01), the best cut-off value was 3.54 ng/ml, the diagnostic sensitivity and specificity was 0.713 and 0.917, respectively. AUC of urine SNCG in differentiating malignant lesion group from healthy controls plus benign lesion group was 0.848 (95% CI 0.797-0.899, P < 0.01), the best cut-off value was 2.87 ng/ml, the diagnostic sensitivity and specificity was 0.791 and 0.901, respectively. Conclusions:The concentration of SNCG in urine of patients with bladder cancer is higher than that of patients with benign urinary lesions and healthy people. Urine SNCG has a good application value in the diagnosis of bladder cancer.
