Objective To study the apoptosis-inducing effect of recombinant adeno-associated virus serotype 8 with soluble TRAIL gene on human colon cancer cell line HCT116. Method Recombinant adeno-associated virus serotype 8 with TRAIL gene was generated by triple plasmid transfection of HEK293 cells with phosphate calcium precipitation. After infected by recombinant virus, cell growth assay was carried out by counting alive cells after trypan blue exclusion. Alternations of cell morphology was examined by microscopy. Cell apoptosis was detected by DNA laddering. The protein expressions of apoptosis-3 and apoptosis-8 were detected by Western blotting. Result The growth rate of the AAV8-sTRAIL infected HCT116 cells was significantly inhibited. Recombinant virus induced apoptosis in colon tumor cells by triggering caspase cascade, but not in normal human hepatocytes. Conclusion AAV8-sTRAIL efficiently suppresses the growth of human colon cancer cell line HCT116 by inducing apoptosis, with no evident toxicity to normal cell.

Objective To investigate sensitivity of colon cancer cells to 5-fluorouracil after downregulation of XIAP gene expression. Method Colon cancer cells HCT-8 and HCT116 were transfected with a short hairpin RNA targeted to XIAP by liposome, cells viability were examined.5-fluorouracil was applied into two kinds of colon cancer cells. Tumor cells sensitiviy to chemotherapeutic drug was evaluated. Caspase-3 activity in tumor cells was examined by Western blot. Result After downregulation of XIAP expression, cell growing viability of these two kinds of colon cancer cells was restricted, HCT-8 resistance to 5-fluorouracil was reversed ( P < 0. 01 ), HCT116 sensitivity to 5-fluorouracil was enhanced (P < 0.05), caspase-3 expression in colon cancer cells was highly activated, apoptosis inducing activity of 5-fluorouracil was increased significantly. Conclusions XIAP expression was a important mechanism in colon cancer cells HCT-8 and HCT 116 resistant to 5-fluorouracil, sensitivity to 5-fluorouracil of HCT-8 and HCT-116 was increased by downregulation of XIAP expression.

Objective To investigate the diagnosis and treatment of acute rejection of orthotopic liver transplantation (OLT). Methods From July 1999 to April 2000, two piggyback liver transplantations were performed on two patients with Wilson's disease. Results Cyclosporine A, azathioprine and methylprednisolone were the baseline immunosuppression management. Acute rejection occured 5 times in the 2 patients. The correct diagnosis was obtained through clinical inspection, liver function test and biopsy. The rejections were controlled by intensive steriod therapy plus OKT3 or FK506. Conclusions Clinical inspection and liver function test can suggest the episode of acute rejection in time. Hepatic biopsy is the key point for diagnosis of acute rejecton, Reasonable use of immunosppression is critical for the treatment.

Objective To detect the synergetic effect and mechanism of arsenic trioxide(As2O3)and Trichostatin A(TSA)during inducing apoptosis of HL-60 cells. Methods MTT method was used to test the proliferation of HL-60 cells.Cell cycle and apoptosis were detected by FCM.Semi-quantitative RT-PCR was used to detect the mRNA expression of Bax and Bcl-2 in the cells treated by As2O3 and(or)TSA.Results As2O3 combined with TSA could inhibit proliferation and induce cell cycle arrest at G0 and G1.The percent of apoptosis induced by combination of As2O3 and TSA was obviously higher than that of either As2O3 or TSA.Bax gene expression was increased, while Bcl-2 gene expression was decreased,Bax/Bel-2 ratio was up-regulated.Conclusion Synergetic effect by As2O3 and TSA is remarkable in inducing apoptosis of HL-60 cells.Cell cycte arrest and Bax/Bcl-2 ratio play an important role in apoptosis of HL-60 cells induced by As2O3,TSA or their combination.

Objective. To investigate the effects of cold preservation on rat fatty liver.Methods.We observed the changes of portal perfusion pressure,endothelin-1,enzymes relesase in the effluent and mortality of sinusoid lining cell after 0h,6h,or 12h preservation respectively and a subsequent 30 min reperfusion in rat fatty liver groups and control groups by using isolated perfused rat liver model. And we compared fatty liver groups with control groups by these indices.Results.There was no obvious difference between mildly fatty liver group and control group after long time(12h)preservation,between moderately fatty liver group and control group after short time(6h)preservation,between severely fatty liver group and control group without preservation(0h),while preservation reperfusion injury was more severe in moderately fatty liver group than in control group after long time(12h)preservation and in severely fatty liver group than in control group after short time(6h)preservation.Conclusions.The authors suggested that a mildly fatty liver donor could be used in the same way as nonfatty liver and a moderately fatty liver donor could be used depending on the time of preservation and the balance of the emergent needs of recipient and donor organ supply, while severely fatty liver donor should be discarded without hesitation.
