1.Circulating RNA in thyroid cancer

Thyroid cancer is the most common endocrine gland malignanct tumor. Numerous studies have found that the content of peripheral blood circulating RNA in various cancer types is aberrantly expressed, which could be a potential biological diagnostic marker and therapeutic target. Tissue-speciifc messenger RNA has a dysregulated expression and may be used for the diagnosis and residual/metastatic detection of thyroid cancer. Recent studies have showed that non-coding RNA (ncRNA) could act as oncogene or tumor suppressor gene in bacteria, fungi and mam-mals. It plays a regulatory role in occurrence and development of tumors and stably exists in peripheral blood. It is hopeful that it will become a new marker for diagnosis of tumors. This review introduces some latest research progress on circulating RNA associated with thyroid cancer; and emphatically discuss the role of mRNA, microRNA (miRNA), long non-coding RNA (lncRNA) and circular RNA (circRNA) in thyroid tumorigenesis and metastasis.

Operation is one of the important ways in breast cancer treatment.With the deep basic medical research,new theory,new concept and new technology improve the operation therapy in breast cancer in more scientific and rational way Breast-conserving therapy and sentinel lymph node bipsy (SLNB) are applied,and the neoadjuvant chemotherapy make breast-conserving therapy more feasible.The progress of operation therapy in breast cancer make it critical to emphasize the individualized treatment in breast cancer.

4.The relationships of urinary albmnin excretion rate to ambulatory blood pressure and erythrocyte sodium-lithium countertransport in NIDDM

Objective To investigate the potential association among urinary albumin excretion rate(U_(AER)), 24h ambulatory blood pressure(ABP) and erythrocyte sodium-lithium countertransport(SLC) in non-insulin dependent diabetes mellitus (NIDDM). Methods U_(AER), SLC were determined with radioimmunoassay and modified method of Canessa respectively, and 24h ABP profiles were recorded in 69 NIDDM subjects. Results NIDDM patients with increased U_(AER) had higher prevalence of hypertension and higher prevalence of abnormal circadian rhythm of blood pressure. Blood pressure(BP), particularly nighttime systolic BP was also significantly increased. U_(AER) was positively correlated to mean nighttime BP, mean daytime and 24h systolic BP, while inversely related to day-night dip in systolic BP. However, there was no significant difference in SLC between NIDDM subjects with and without increased U_(AER). Futhermore, neither U_(AER) nor all ABP indices were correlated to SLC. Conclusion Close association exists among diabetic nephropathy and increased BP and abnormal diumal BP rhythm, while either SLC and U_(AER) or SLC and BP is significantly correleted. Increased rate of SLC is not likely a marker for diabetic nephropathy in NIDDM, and it seems not to be involved in the pathogenesis of diabetic hypertension.
5.Effects of glucosidorum tripterygii tororum on cytokine productions in graft-versus-host disease mice

AIM: To observe the effect of glucosidorum tr ipterygii tororum (GTT) on cytokine productions in acute graft-versus-host disea se (aGVHD) mice. METHODS : C 57 BL/6 mice were exposed to radiation delivered by a linear accelerator . To es tablish a aGVHD model, the cell suspensions, which were obtained from bone marro w and spleen of the BALB/C mice, were transplanted to the radiated C 57 BL/6 mice. The recipients were treated with GTT, GTT+CsA and CsA+MTX. The serum conc entrations of IL-2, TNF-?, IL-4 and IL-10 were determined by ELISA. RES ULTS: The survival rate on day 11 in GTT group (9/10) was higher than in allogeneic bone marrow transplatation (allo-BMT) group (8/19). The concentratio ns of IL-2 and TNF-? in GTT group were significantly lower, but the concentrati on of IL-10 was remarkably higher than that in allo-BMT group ( P 0 0 5). CONCLUSION: GTT inhibited aGVHD development by regulating th e production of cytokines in the host.
6.Expression and significance of secretory phospholipase A2 in gastric adenocarcinoma

Objective To investigate the expression and signficance of secretory phospholipase A2(sPLA2>) in gastric adenocareinoma. Methods Sixty-five samples of gastric adenocarcinoma (11 with high differentiation, 10 with median differentiation and 44 with low differentiation) and 11 samples of normal gastric mucosa had been obtained from the Second Affiliated Hospital of Jilin University from January 2006 to April 2007. Fifty samples of gastric adenocarcinoma were found with lymph node metastasis. The expression of sPLA2> in gastric adenocarcinoma and normal gastric mucosa was detected. The relationship between sPLA2> and the differentiation of gastric adenoearcinoma, lymph node metastasis and helicobacter pylori infection was detected. All the data were processed with chi-square test or Spearman rank correlation. Results The positive rates of the sPLA2> expression in normal gastric mucosa and gastric adenocarcinoma were 36% (4/11) and 78% (51/65). The positive rates of sPLA2> expression in low, median and high differentiated gastric adenocarcinoma were 84% (37/44), 70% (7/10) and 64% (7/11), respectively. The expression of sPLA2> was positively correlated with the malignancy of gastric adenocarcinoma (r =0.272, P <0.05). The positive rates of sPLA2> expression in gastric adenocarcinoma with and without lymph node metastasis were 88% (44/50) and 47% (7/15), respectively. The expression of sPLA2> was correlated with lymph node metastasis (X2 = 9. 347, P < 0.05). The positive rates of sPLA2> expression in gastric adenocarcinoma with and without helicobacter pylori infection were 79% (38/48) and 76% (13/17), respectively. The expression of sPLA2> was not correlated with helicobacter pylori infection (X2 = 0. 000, P > 0.05). Conclusions The activation of sPLA2> gene may be correlated with the genesis of gastric adenocarcinoma. sPLA2> may influence the invasion of gastric adenocarcinoma, and can be used as an indicator in predicting poor prognosis.
7.Application of EWGSOP Consensus in Evaluating Elderly Female Patients with Sarcopenia Ranging in age from 70 to 79 from Communities of Shanghai

Objective To explore the feasibility of using the consensus of European Working Group on Sarcopenia in Older People(EWGSOP) to diagnose sarcopenia among community-dwelling women aged from seventy to seventy-nine in Shanghai.Methods Twenty-two healthy community-dwelling women were recruited as the subject group(group S),and another ten young girls were randomly selected as the control group(group C).The anthropometry,bioelectrical impedance analysis(BIA)and dual energy X-ray absorptionmetry(DEXA)were used to evaluate the circumferences of upper and lower limbs,and the dimension of muscle mass,according to relative appendicular skeletal mass(RASM)and skeletal muscle index(SMI).The grip dynamometer and isokinetic dynamometer were used to measure the muscle strength of upper and lower limbs.The ten meters gait speed,functional reach test,timed up and go test,Berg balance scale and short physical performance battery were all applied to assess the physical performance.The criteria of EWGSOP consensus were applied to diagnose the situation of sarcopenia.Results Firstly,the average biceps circumference and fat mass of upper limbs in group S were significantly higher than group C (P<0.01),while no significant differences were observed in the muscle mass(P>0.05).Secondly,RASM values gained from BIA and DEXA in S group were significantly higher than C group(P<0.05),while the SMI values of the former were significantly lower than the latter(P< 0.01).Thirdly,absolute grip strength,relative grip strength and other bio-mechanical indexes of lower limbs in the s group were significantly lower than the C group(P<0.01);the relative grip strength showed significant correlation with most bio-mechanical indexes of lower limbs.Fourthly,the physical performance in group S declined significantly compared with that of the c group(P<O.05),and no significant correlation was found between muscle strength and physical performance of upper limbs except the absolute grip strength and gait speed.The prevalence was 0% according to the RASM index,while the prevalence was 63.6% and 45.4% according to the BIA and DEXA respectively.Conclusion The assessing pattern from EWGSOP consensus could not be simply copied to applying on Chinese old people due to serious difference between RASM and SMI.The combination of SMI+relative grip strength+ TUG/SPPB/Berg may be better than the classical mode of RASM+absolute grip strength+gait speed in diagnosing sarcopenia.
8.RNA interference inhibits the secretion of IL-1? in mice spleen lymphocytes

AIM: To decide the effect that selected siRNA degrades mRNA of IL-1? specifically and suppression of its expression after connected with target site with homology complementary sequence. METHODS: Synthesized DNA expression box aimed directly at target site through PCR reaction in vivo was purified, and transfected into lymphocytes stimulated by LPS. siRNA was transcribed by cellular endogenous RNA polymerase Ⅲ and then evoke the degradation of target mRNA. After 48 hours of transfection, the cell culture supernatant was collected and the concentration of IL-1? was assayed using ELISA. RESULTS: Compared with blank-control and negative-control, selected sequence decreased the expression of IL-1?. Rate of the suppression was about 15%. CONCLUSION: RNAi technology produces specific interference effect in mouse spleen lymphocytes in original culture and inhibits the excretion of IL-1?.
9.Effects of hyperbaric oxygen and cyclosporin A on the levels of active oxygens and nitric oxide in spleens of skin transplanted mice

AIM: To study effects of hyperbaric oxygen (HBO) and cyclosporin A (CsA) on the contents of active oxygens and nitric oxide (NO) in spleens of skin transplanted mice.METHODS: The donor mice BALB/C and receptor mice C 57BL/6 were tested for skin transplantation. The HBO group mice were treated with 99.2% oxygen under 0.25 MPa for 1.5 hours, while CsA group mice were treated with CsA 0.5 mg?kg -1?d by abdomen injection. After 14 days, the spleen were extracted the contents of malondialdehyde (MDA) and NO and the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), catalase (CAT) and NO synthases (NOS) were determined.RESULTS: (1) Compared with the control group, the transplantation group, HBO group and CsA group have markedly increased the content of MDA and the activities of GSH-PX and CAT; Compared with the transplantation group, the CsA group have markedly increased activity of SOD and reduced activities of GSH-PX and CAT; the HBO group have markedly reduced the activity of GSH-PX and increased the activities of CAT and SOD (P
10.Effects of rehabilitation exercises on quality of life of patients undergoing revascularization for coronary heart disease

ObjectiveTo investigate the effects of treadmill testing guided-rehabilitation exercises upon quality of life (QOL) of patients who underwent revascularization for coronary heart disease (CHD).Methods All the participants received routine therapy and rehabilitation for 6 weeks.A 36-item short-form (SF-36) was used to assess QOL of the patients.Results Injured functioning of patients who received revascularization for CHD were physiological function ( 37.86 ),social function ( 43.67 ),vitality (45.58),physical performance ( 65.45 ),mental health ( 68.73 ),role emotion ( 71.30 ),physical pain ( 79.52 ),and general health ( 86.75 ).After intervention,physiology function ( 37.86 ± 8.49 vs 62.13 ± 10.53 ),social function (43.67 ±26.23 vs 56.98 ± 18.89),vitality (45.58 ±27.01 vs 77.42 ±23.48),and physical pain (79.52 ±9.05 vs 87.32 ± 13.14) were significantly improved.Conclusion Rehabilitation exercises and behavioral psychological treatment may be beneficial to improve QOL of patients undergoing revascularization for CHD.
