On the basis of ARMT core processor, a biological signal acquisition system with USB2.0 interface is presented in this paper. It is self-developed and Lab VIEW7 is taken as a host communicating software platform for the entire data communication system. The study is mainly focused on the firmware programming for USB chip, the development of USB WDM Driver, and the method for the calling of DLL from Lab VIEW7 to communicate with self-made hardware device.
2.Curative effect of intravitreal injection of conbercept on macular edema in different types of OCT diabetes mellitus

AIM: To observe the efficacy of intravitreal injection of conbercept for diabetic macular edema(DME)with different patterns of optical coherence tomography(OCT).
METHODS: A total of 96 patients(96 eyes)with DME were classified as diffuse retinal thickening(DRT group, 35 eyes), cystoid macular edema(CME group, 33 eyes)and serous retinal detachment(SRD group, 28 eyes)according to the OCT. All patients were treated with intravitreal injection of 0.5mg(0.05mL)conbercept. The changes of best corrected visual acuity(BCVA), central foveal thickness(CFT), the injection times and vision improves eyes were compared between three groups after 1,3,6mo treatment.
RESULTS: After 6mo follow up, the BCVA of the three groups showed a significant downward trend(Ftime=205.880, Ptime<0.01), and there were significant differences among the three groups(Fgroup=3.472, Pgroup=0.042). The DRT group had the best BCVA after treatment. The CFT of the three groups showed a significant downward trend(Ftime=392.994, Ptime<0.01), and there were significant differences among the three groups(Fgroup=5.046, Pgroup=0.012). The reduction of CFT in the DRT group and CME group were better than that in the SRD group. The DRT group had the least injection times, and the highest proportion of vision improves eyes after 6mo follow up.
CONCLUSION: The intravitreal injection of conbercept could significantly improve the visual acuity and reduce the CFT of DME with different OCT patterns. And DRT is the most effective type with the least injection times.
3.Analysis on identification and traceability of one non-toxigenic Corynebacterium diphtheriae from a patient with diabetic foot in Hainan province.

There is a rare case of an elderly diabetic with diabetic foot infection at Hainan General Hospital in September 2021, which was diagnosed as Corynebacterium diphtheriae infection incidentally on routine culture with conventional methods and molecular biological approaches, to aid in diagnosis in clinical practice. Owing to smear staining, Albert staining and VITEK 2 system, automated identification systems viz matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) confirmed combing with 16S ribosomal RNA (16S rRNA) gene has been used for the taxonomic classification of bacteria. Otherwise, toxin gene tox was done for diphtheria toxin synthesis. The isolate was Gram-stain-positive, rod-like arrangement with irregular thickness, with characteristic metachromatic granules, ferment most sugars and homology of 16S rRNA analyses with C. diphtheriae NCTC11397T (MW682323.1) was greater than a 100% possibility, toxin gene tox was negative. The findings lay the foundation to clinical identify and trace of non-toxigenic C. diphtheriae. Moreover, this work provides insights into the non-toxigenic C.diphtheriae that contribute to recognized risk of non-toxigenic C.diphtheriae infections.

4.Normal Range of Head-to-body Delivery Interval by Two-step Delivery.

BACKGROUNDThe one-step method was routine practices in China, scientific evidence to support this intervention is scarce. The purpose of this study was to observe the natural process of head-to-body delivery interval by waiting for at least one contraction (two-step) after head delivered in normal birth.
METHODSFrom March 1 to March 30 in 2015 at Haikou Maternal and Child Hospital in China, normal vaginal birth with normal baby condition were recorded by video. Videotapes were transferred to computer then replayed and observed.
RESULTSNinety-two cases were enrolled in this study. The average head-to-body delivery interval by two-step delivery was 71.04 ± 61.02 s, (mean + 2 standard deviation = 193.07 s, 95% confidence interval [15.65-229.15] s). Fifty-one patients (51/92, 55.43%) were <60 s, 41 patients (41/92, 44.57%) were over 60 s. Shoulders delivered at the first contraction were 96.74% (89/92), 3.26% (3/92) had delivered by the second contraction. Shoulders emerged from perineum were 71.73% (66/92), 15.21% (14/92) transversely, and 13.04% (12/92) emerged from under pubic arch. Babies cried before the shoulder were 31.52% (29/92), cried after birth 52.17% (48/92), and 16.30% (15/92) did not cry after birth. Baby activities included as making faces, sucking, and bubbled from mouth and noses, and the lighter blue color of skin with good perfusion.
CONCLUSIONSThe average time of head-to-body delivery interval was longer than 60 s by two-step delivery. Majority shoulders were delivered at the first contraction. Majority shoulders emerged from perineum rather from under pubic arch. The routine one-step method of shoulder delivery where the downward force applied is not necessary and is not the right direction. Baby's breath, making faces, sucking, bubble from noses and mouth, and the light blue color of the faces, all those signs during shoulder delivery indicated a normal live birth.

5. Rapamycin-induced β-Globin Family Locus Remodeling Regulates the Gene Switching Expression

β-Thalassemia caused by abnormal coding of the β-globin gene is the most common hemoglobinopathy in many Asian countries. The in-depth study of the molecular basis and epigenetic mechanism of globin gene expression is the key to explore a new treatment for thalassemia. In this study, FAIRE (formaldehyde-assisted isolation of regulatory elements), 3C (chromosome conformation capture) and ChIP (Chromatin Immunoprecipitation) were used to investigate the three-dimensional interaction network of β-globin family gene loci and the molecular mechanism of functional regulation of gene expression during rapamycin-induced chromatin remodeling in CD4+ T cells. The results showed that the opening degree of globin gene chromatin, the interaction frequency between the gene promoter region and the regulatory element LCR (Locus control regions), and the enrichment efficiency of CTCF (CCCTC-binding factor) in the gene promoter region changed differently during the change of rapamycin treatment concentration from low to high, which led to the same change trend of the gene expression pattern. At the 10 nmol/ L concentration, chromatin accessibility and gene expression decreased (P < 0. 05). At 20 nmol/ L and 50 nmol/ L concentrations, chromatin accessibility increased and gene expression was up-regulated (P < 0. 05). In this study, the molecular mechanism of gene expression regulation of the β-globin family was expounded through this dynamic change process. Our work provides a theoretical and clinical practice basis for clinical precision treatment.
6.Effect of Jiajian Fuyuan Huoxuetang Combined with Electroacupuncture on Neurological Rehabilitation After Spinal Cord Injury

Objective:To observe the effect of Jiajian Fuyuan Huoxuetang combined with electroacupuncture on neurological rehabilitation of patients with spinal cord injury, and to investigate its effect on neurotrophic and inflammatory factors. Method:One hundred and twenty patients were randomly divided into control group (59 cases) and observation group (61 cases) by random number table. Both groups’ patients got monosialotetrahexosylganglioside by intravenous drip for 6 weeks, 200-400 mg/time, 1 time/day.Rat nerve growth factor for injection by intramuscular injection for 4 weeks, 20 μg/time, 1 time/day. Patients in control group additionally got electroacupuncture treatment,and tongluo huatan capsule,3 caps/time,3 time/days. The patients in observation group additionally got Jiajian Fuyuan Huoxuetang combined with electroacupuncture, 1 dose/day. The courses of treatment were 12 weeks in two groups. Scores of degree of spinal cord neurological impairment were graded by using impairment score of American Spinal Injury Association (ASIA). Lower extremity motor ability was discussed by using the walking index of spinal cord injuryⅡ (WISCⅠ Ⅱ) and 10 minutes’ walking time (10 MWT). Before and after treatment, functions of daily life were evaluated by using Barthel index (MBI). To review the background and advance of a strategy of bioethnopharmaceutical analytical pharmacology(BAP).BAP is an idea for elucidating absorbed bioactive compound(ABC)in traditional Chinese medicine(TCM)formula or herbal remedy.It is also a mode of active comparison between absorbed compounds and the parent formula including determination of ABC in blood,the ABC dosage identical to its content in formula,and the active comparison of ABC and the parent formula.The ABC of Guan-Xin-Er-Hao formula elucidated by BAP strategy has been reviewed and the significance of BAP strategy will be further discussed.

9.ODNSIRUCTION AND ANALYSIS OF cDNA LIBRARY OF NECATOR AMERICANUS THIRD STAGE LARVAE

[Objective] To obtain the genetic information on Necator americanus and to search for the purpose genes.[Methods] Mrna was isolated from the third stage larvae of Necator americanus maintained in hamsters.Double strand Cdna was synthesized and ligated to ΛzapⅡ vector to construct the Cdna library.Expresed se-quence tages (ESTs) were obtained by single pass sequencing of randomly isolated Cdna clones from the es-tablished library.[Results] A Cdna librazy of N.americanus was successfully constructed with high recombi-nant efficiency.The titer of unamplified library was 1×107.The insert size was about 750~3000bp.Of 11 ESTs obtained from the library,7 have a significant homology with certain functional genes.[Conclunsion]A high quality and high representative Cdna library of N.americanus was constructed at the first time and ome functional genes were identified from the library by ESTs.
10.Efficacy and Safety of Fluorouracil Combined with Paracetamol in the Trea tment of Knee Osteoarthritis and Its Effects on Related Indexes

OBJECTIVE: To observe the efficacy and safety of fluorouracil combined with paracetamol in the treatment of knee osteoarthritis and its effects on related indexes. METHODS: Totally 115 patients with knee osteoarthritis admitted to our hospital from March 2015 to March 2018 were divided into control group (57 cases) and observation group (58 cases) according to medication plan. Control group was given Paracetamol tablets 0.3 g orally, 3 times a day, for consecutive 8 weeks. Observation group was additionally given intra-articular injection of Fluorouracil injection 0.075 g on the basis of control group, once a week, 4 times as a treatment course, for 2 courses in total. Clinical efficacies, VAS scores, Lysholm knee scores, Fugl-Meyer Assessment (FMA) scores, Barthel indexes, WHO QOL-BREF scores before and after treatment and the occurrence of ADR of 2 groups were observed. RESULTS: The total response rate of observation group was 94.83%, which was significantly higher than 78.95% of control group (P<0.05). Before treatment, there was no statistical significance in VAS score, Lysholm knee scores, FMA scores, Barthel index or QOL-BREF scores between 2 groups (P>0.05). After treatment, VAS scores of 2 groups were significantly lower than before treatment, and the observation group was lower than the control group; Lysholm knee scores, FMA scores, Barthel indexes and QOL-BREF scores were significantly higher than the same group before treatment, and observation group was significantly higher than control group (P<0.05). There was no statistical significance in the incidence of ADR between 2 groups (P>0.05). No severe ADR was found in 2 groups during treatment. CONCLUSIONS: Fluorouracil combined with paracetamol can significantly improve therapeutic efficacy of patients with knee osteoarthritis, and can significantly improve knee function, motor function, self-care ability and quality of life without increasing the occurrence of ADR.
