1.Reduced expression of ADAM8 and its role in breast cancer

ObjectiveTo investigate the expression of a disintegrin-like and metalloproteinase 8 ( ADAM8 ) in breast cancer and in normal breast tissues and its negative regulation role in tumorigenesis and progress of breast cancer.MethodsThe expression of ADAM8 in breast cancer and normal breast tissues was detected by immunohistochemistry (IHC),qRT-PCR,and Western blot.The relation between ADAM8 expression and the clinicopathological parameters of breast cancer patients was analyzed.ResultsADAM8 was expressed in breast cancer and normal breast tissues.The expression of ADAM8 mRNA and protein was significantly lower in breast cancer than in normal breast tissues (qRT-PCR:P =0.015,IHC:P =0.044,Western blot:P =0.000).The expression rate of ADAM8 was related to lymph node metastasis,tumor stage and tumor size,although the difference had no statistical significance.IHC results showed that ADAM8 expression level was lower in stage Ⅲ + Ⅳthan in stage Ⅰ + Ⅱ ( P =0.574 ).qRT-PCR showed ADAM8 mRNA expression was lower in stage Ⅱb + Ⅲ than in stage Ⅰ + Ⅱ a ( P =0.247).ADAM8 expression was lower in the breast cancer tissues with lymph node metastasis than in those without lymph node metastasis (P =0.560 by IHC and P =0.592 by qRT-PCR).ADAM8 expression was lower in tumors whose size > 2 cm than in tumors whose size ≤ 2 cm,however,the difference had no statistical significance ( P > 0.05 ).ConclusionsADAM8 is significantly lower-expressed in breast cancer than in normal breast tissues,which is associated with clinical stages and lymph node metastasis.The reduced expression of ADAM8 may play a role in the pathogenesis and progress of breast cancer.
2.The value of carbon nanoparticles in total thyroidectomy combined with central lymph node dissection

Objective To explore the value of carbon nanoparticles in patients with papillary thyroid carcinoma (PTC) undergoing total thyroidectomy combined with ipsilateral central lymph node dissection.Methods 43 patients with unilateral PTC were retrospectively analyzed.All patients underwent total thyroidectomy combined with central lymph node dissection.Patients were divided into carbon nanoparticles group and the control group according to whether carbon nanoparticles were used in the operation.The operation time,postoperative hospitalization time,the serum calcium level and its rate of change,the parathyroid hormone and its rate of change on the 1 st day after surgery,the number of central lymph node and the transfer rate,and the postoperative complications were compared between the 2 groups.Results There was no statistical difference between the 2 groups in operation time(P > 0.05),while the postoperative hospitalization time of the carbon nanoparticles group was shorter than that of the control group(P < 0.05).The serum calcium and the parathyroid hormone on the 1 st day after surgery in the carbon nanoparticles group was (2.31 ± 0.13) mmol/L and (33.45 ± 14.37) pg/ml respectively,higher than those of the control group (P < 0.05).The low blood calcium rate (3/20 (15%)),the temporary hypoparathyroidism rate (2/20 (10%)) and the decline degree of parathyroid hormone (47.3 ± 14.31)% in the carbon nanoparticles group were lower than those of the control group(P <0.05).The number of central lymph node dissected (9.45 ± 2.33) pieces/case in the carbon nanoparticles group was more than that of the control group (P < 0.05).The number of lymph node in the right recurrent laryngeal nerve (3.12 ± 0.65) pieces/case was more than that of the control group(P < 0.05).The lymph node metastasis rate had no significant difference between the 2 groups(P > 0.05).The incidence of postoperative complications in the carbon nanoparticles group was lower than that of the control group (P < 0.05).Conclusion The application of carbon nanoparticles in total thyroidectomy combined with central lymph node dissection can contribute to the recognition and protection of parathyroid glands and its blood supply,improve the rate of central lymph node dissection,and reduce the incidence of postoperative complications.
3.Expression of Survivin and Bcl-2 in sinonasal inverted papilloma

Objective:To investigate the expression of Survivin and it's relationship with the expression of Bcl-2 in sinonasal inverted papilloma (SNIP).Method:Immunohistochemical method was used to determine the expression of Survivin and Bcl-2 in 30 cases of SNIP,10 cases of sinonasal squamous cell carcinoma(SCC) and 10 cases of normal inferior concha tissues.Result:Survivin was expressed in 22 of 30(73.3%)cases of SNIP,8 of 10(80.0%)cases of SCC and not expressed in 10(0%)cases of normal inferior concha tissues.Expression of Survivin was significantly higher in SNIP and SCC than in normal tissues.Bcl-2 was expressed in 9 of 10(90.0%)cases of SCC and 2 of 10(20.0%)cases of normal inferior concha tissues.Expression of Bcl-2 was significantly higher in SCC than in normal tissues.Bcl-2 was expressed in 14 of 30(46.7%)cases of SNIP,higher than normal tissues.Expression of Bcl-2 was positively related to expression of Survivin.Conclusion:Survivin may play an important role in the pathway of progression of SNIP and SCC.It may be identified as a new therapeutic target.Bcl-2 may play a synergic role with Survivin in progression of SNIP.
4.The clinical analysis of thyriod carcinoma showing thymus-like differentiation.

OBJECTIVE:
To investigate the clinical diagnosis, treatment and prognosis of carcinoma showing thymus-like differentiation (CASTLE) of thyroid, and to improve the understanding of CASTLE.
METHOD:
In 6 patients with CASTLE, we performed region VI lymph node dissection, the lesion resection of thyroid lobe and isthmus. After the operation, except one case of 68 years old patient, the rest patients were treated with radiotherapy 60 Gy in anterior cervical region after third week of operation and 5 courses of chemotherapy with cisplatin, each interval of 2 weeks. Insist on the oral Euthyrox.
RESULT:
All patients were alive without recurrence and metastasis.
CONCLUSION
The diagnosis of CASTLE depends on the histology and immunohistochemistry. Operation was the main treatment. CASTLE is a rare type of thyroid carcinoma with slow progress and good prognosis.

CONCLUSION
Survivin may play an important role in the pathway of progression of SNIP and SCC. It may be identified as a new therapeutic target. Bcl-2 may play a synergic role with Survivin in progression of SNIP.

6.Emotional experience and processing characteristics of long-term plateau immigrant undergraduates

Objective:To investigate the subjective experience and processing characteristics of positive and negative emotions in undergraduates who have immigrated to the plateau for a long time.Methods:SPSS 22.0 was used for data analysis.Using independent sample t-test to compare various data from the plateau group(58 undergraduates who had moved to an altitude of 3 658 m for over 2 years after reaching adulthood) with the plain group (58 undergraduates who had never been to the plateau), including the state anxiety inventory(SAI), self-rating depression scale(SDS), positive and negative affect schedule (PANAS), and the affective valency and arousal on the positive, neutral, and negative emotional pictures.And χ 2 test was conducted for the positive detection rates of state anxiety and depression of the two groups.Then, Pearson correlation analysis was performed on SAI, SDS and PANAS scores. Results:The positive detection rates(state-anxiety: 67.24%, depression: 56.90%) and scores of state anxiety(44.45±8.57) and depression(50.12±10.11) in the plateau group were all higher than those in the plain group(46.55%, 32.76%, (40.19±9.58), (44.64±9.17) respectively, χ 2=5.06, 8.12, t=2.52, 3.06, all P<0.05). In addition, positive affect scores of the plateau group (28.16±5.39) tended to be lower than those of the plain group (30.28±6.51), but the difference was not statistically significant( t=-1.91, P=0.059). SAI, SDS and PANAS scores were moderately correlated each other( r= 0.69, -0.52, 0.67, -0.62, 0.53, all P<0.01). Positive affect was mildly negatively correlated with negative affect( r=-0.27, P<0.01). The correlation between positive emotion and depression( r=-0.62) was greater than that between positive emotion and state anxiety( r=-0.52, t=2.57, P<0.05). The correlation between negative emotion and state anxiety( r=0.67) was greater than that between negative emotion and depression( r=0.53, t=1.71, P<0.05). The valency score of neutral images in the plateau group (3.06±0.20) was higher than that in the plain group ((2.99±0.20), t=2.01, P=0.047). Conclusion:The long-term plateau immigrant undergraduates are prone to state anxiety and depression, with depression being the primary and anxiety secondary, and exhibit a positivity bias to neutral stimulus processing.
