1.Sodium valproate inhibit the expression of SCN3A gene

Objective To determine the role of valproate in the expression of SCN3A and provide new evidence for the clinical treatment of epilepsy. Methods We treated the N1E-115 cells with sodium valproate and examine the expression of SCN3A mRNA. We also constructed the expression vector of the SCN3A gene promoter then transfected the vector into N1E-115 cells ,respectively detected the level of SCN3A mRNA and the expression level of firefly luciferase of N1E-115 cells with valproate treated and untreated. Results Valproate inhibited the expression of SCN3A significantly including the level of SCN3A mRNA and luciferase protein. Conclusion Valproate inhibits the seizures by reducing the expression of the SCN3A, which provides a new understanding of the treatment of epilepsy.
2.Effects of Kangshuling Gel on Wound Treatment and Infection Prevention inⅡDegree Burns

OBJECTIVE:To discuss the effects of Kangshuling gel on wound treatment and infection prevention inⅡdegree burns. METHODS:60 patients with Ⅱ degree burns were randomized intro observation group and control group,with 30 cases in each group. Based on routine treatment (cleaning,disinfection,necrotic tissue removal,antibiotics use),observation group was given Kangshuling gel,and control group was treated with sulfadiazine. The average healing rate,healing time,surgical dressing exudate,inflammatory reaction around wound,the times of dressing change and inner layer gauze change,and wound infection rate were observed in 2 groups,and satisfaction survey was conducted in 2 groups. RESULTS:On 14th and 18th day after treat-ment,average rate of wound healing in observation group was significantly higher than in control group;the time of wound heal-ing was significantly shorter than control group,with statistical significance(P<0.05). Within 2 weeks after treatment,the number of gauze layer wetted by surgical dressing in observation group was significantly less than in control group,and case number of in-flammatory reaction around wound was significantly less than control,with statistical significance(P<0.05). The times of dressing change and inner layer gauze change in observation group was significantly less than in control group,and the rate of wound infec-tion was significantly less than control group,with statistical significance(P<0.05);the satisfaction of observation group was bet-ter than that of control group,with statistical significance(P<0.05). CONCLUSIONS:Kangshuling gel in the treatment of Ⅱ de-gree burns can effectively prevent and treat wound infection,reduce the rate of wound infection and the times of dressing change, shorten the time of wound healing and improve the rate of wound healing so,high satisfaction degree is obtained.

Objective:To investigate the specific immune act ivated effects in the mouse immunized by MHC I-restriced tumor antigen peptide pulsed dendritic cells(DC) with IL-2 gene modified.Methods:DC were transfected with IL-2 gene via adenovirus vector(DC-IL-2) the mRNA of mIL-2 was examined by RT-PCR.Syngeneic mice were immunized by DC-IL-2 pul sed with MHC-I-restricted Mutl tumor antigen peptiede of 3LL Lewis lung carcin oma (DC-IL-2-Mutl),the effects of lymphocyte subsets in the draining lymphoid node from the tumor antigen pulsed DCs with gene modified immunized mice by flo w-cytometric analysis(FACS) analysis.Results:The mRNA of mIL-2 was expressed in the DCs.The ratio of CD8+T cells was hosted in the dra ining lymphoid node from Mutl pulsed DCs(DC-Mutl) immunized mice.The ratio of C D8+T cells and NK cells were all hosted obviously in the draining lymphoid nod e from DC-IL-2-Mutl immunized mice .Conclusion:Immunized mice with DC-IL-2-Mutl can induce the mouse specific antitumor immunized affect in vivo and can activate a variety of immunized effect in the mouse.
4.Protective effect of isoflurane preconditioning on the brain against ischemia-reperftision injury in gerbils

Objective To investigate the protective effect of isoflurane preconditioning on the brain against ischemia-reperfusion (I/R) injury and the underlying mechanism.Methods Seventy-five male gerbils weighing 60-80 g were randomly divided into 5 groups ( n = 15 each): group A sham operation (sham) ; group B I/R; group C isoflurane + I/R (ISO); group D 5-HD (mito-KATP channel blocker) + I/R (5-HD) and group E 5-HD + isoflurane + I/R (5-HD+ISO). The animals were anesthetized with intraperitoneal (i.p.) 10% chloral hydrate 0.35 g?kg-1 . Global cerebral I/R was produced by occlusion of bilateral common carotid arteries for 5 min and confirmed by isoelectric potential on EEG. In ISO group the animals inhaled 1.2%-1.5% isoflurane for 30 min followed by 30 min wash-out period before I/R. In 5-HD group 5-HD 10 mg?kg-1 was injected i.p. 30 min before I/R. In 5-HD+ ISO group 30 min after i.p. 5-HD 10 mg?kg-1 , isoflurane was inhaled as in ISO group. The animals were killed 24 h after global cerebral ischemia and their forebrains were excised for (1) electron microscopic examination of mitochondria and (2) measurement of mitochondrial permeability transition pore (MPTP) (ultraviolet spectrophotometer) and (3) calcium content in mitochondria (spectrophoto-fluorometer) . Results Marked swelling of mitochondria with disrupted cristae and damaged matrix were observed in group I/R (B) , 5-HD (D) and 5-HD + ISO (E) while in sham (A) and ISO (C) groups relative intact mitochondria were seen. 5.Simultaneous Determination of Minoxidil and Tretinoin in Compound Minoxidil Gel by HPLC

Objective:To establish an HPLC method for the simultaneous determination of minoxidil and tretinoin in compound minoxidil gel. Methods:A Shim-Pack VP-ODS C18(250 mm × 4. 6 mm,5 μm)column was used with the mobile phase of methanol and 2% acetic acid solution by gradient eleution . The detection wavelength was respectively set at 280 nm and 350 nm,and the flow rate was 1. 0 ml·min -1 . The column temperature was 35℃ and the injection volume was 10 μl. Results:Minoxidil and tretinoin had a good linear relationship within the range of 4. 0- 240. 0 μg·ml-1 and 0. 05- 3. 00 μg·ml-1(r =0. 999 6 and 0. 999 3),respectively. The average recovery was 99. 56%(RSD = 0. 42% ,n = 9)and 99. 36%(RSD = 0. 50% , n = 9),respectively. Conclusion:The established method is simple,and suitable for the simultaneous determination of the two constituents in compound minoxidil gel.
6.The evaluation of taking the opportunity of family planning B ultrasound check of married women in childbearing age to carry out anti AIDS education

Objective To explore the married women of childbearing age education on HIV/AIDS intervention method, curb the spread of HIV.Methods Use family planning check B to exceed three op-portunity, for married women of childbearing age to carry out the publicity and consultation, and design a u-nified questionnaire, respectively in the survey of 500 were statistically analyzed before and after the inter-vention, effect evaluation in description epidemiological methods.Results Before and after the interven-tion, the AIDS knowledge awareness rate of married women of childbearing age was 62.75%(2183/3479) , 97.36%(3360/3451),attitudes to AIDS positive response rate was 65.19%(324/497), 95.94%(473/493), middle school students knowledge of AIDS awareness rate was 90.48% (5130/5670), 96.99%(5110/4956 ) , the villagers'awareness rate of AIDS related knowledge was 69.61% ( 2183/3136 ) , 97.36% (3360/3451) .After the intervention, the AIDS knowledge awareness and attitude to HIV positive answer rate of married women of childbearing age were significantly improved,middle school students, vil-lagers AIDS awareness rate were significantly higher than those before intervention.Conclusions The use of B-three examinations opportunities for married women in AIDS prevention knowledge of educational inter-vention improves the life skills of women of childbearing age to prevent AIDS, and to take advantage of the core and the influence of women of childbearing age in the family will spread AIDS prevention knowledge, radiation to family members and the community to improve AIDS prevention capacity of the whole society, to curb the AIDS epidemic.
7.Evaluation of intravenous and intraperitoneal chemotherapy in advanced gastric cancer

Purpose:To evaluate the immediate therapeutic effects and toxicities of intraperitoneal versus intravenous chemotherapy in advanced gastric cancer.Methods:Sixty one patients were treated with intravenous chemotherapy of leucovorin (CF) , fluorouracil ( 5 Fu) etoposide (Vp16) and intraperitoneal chemotherapy of cis platinum (DDP) in advanced gastric cancer. The short term effects and toxicities were observed.Results:The overall response rate was 57.4% and the complete response rate was 9.8% of the 61 patients .Leukopenia was observed in 95.1%, thrombocy topentia were observed in 27.8% of patients . Other side effects were uncommon.Conclusions:Intravenous chemotherapy of ELF and intraperitoneal chemotherapy of DDP were effective for advanced gastric cancer and are worthy to be further studied.

Editor's Notes: The basic theory of traditional Chinese medicine is the theoretical foundation of Chinese medicine , modernization of Chinese medicine needs of the modern theory of Chinese medicine urgently . 9.Preventive and therapeutic effects of valsartan on hepatic fibrosis in rats

Objectives The purpose of this study is to observe the effects of valsartan on hepapetic fibrosis. Methods Thirty male Sprague-Dawley rats were randomly divided into three groups: valsartan -prevetive group (A), modle group of hepatic fibrosis (B)and valsar-tan-treating group (C). The model of hepatic fibrosis in rats was induced by intraperitoneai injection of dimethylnitrosamine (DMN) for 4 weeks(2ml/kg everyday, three times a week). Valsartan (10mg/kg everyday) was given together with injection of DMN per intrngastric (Ig) in group A for 8 weeks. After stop injection of DMN, the S valsartan(10mg/kg, everyday)was given per Ig in group C for 4 weeks. After modeling, normal saline were given per Ig everyday in group B. At the end of eighth week, the histomorphylogic structure of the liver was ob-served with light microscope. Immunohistoebemical staining was used to evaluate the expression of a-SMA. Results In group B, there was a large necrotic area and a number of pesudolobes appeared in the liver tissue. In group A, there were normal hepatic cords. In the group C, there was fibrosis interval formation and portal area expansion and fibrotie intervals extending to the lobule. The quantitative analysis of Mas-son staining showed that the collagen quantities in group B was higher than that of other group(P<0.01). The collagen quantities in group A was lower than that of group C(P<0.05). The results of immanohistochemical staining showed that the expression of a-SMA in group B was strong positive, middle positive in group C, and weak positive in group A (P<0.05). Conclusion The valsartan has preventive and treatment effects on hepatic fibrosis in rats of hepatic fibrosis model induced by DMN, and the preventive effect of valsartan is better than its treatment effect. The valsartan can ameliorate the hver cirrhosis by partly suppressing the activation of HSC.

OBJECTIVE:To evaluate the human body pharmacokinetics and bioequiavailability of two kinds of oral single dose of hydrochloric itraconazole capsules.METHODS:A randomized,crossover study of20healthy volunteers receiving sin-gle oral dose of200mg itraconazole was conducted with in vivo blood concentrations determined by HPLC-fluorescence de-tection.RESULTS:The pharmacokinetic parameters for the testing itraconazole and the reference itraconazole were as fol-lows,t 1/2 were(29.3?5.62)h and(29.3?5.81)h,respectively;C max were(81.4?60.0)?g/L and(77.8?45.2)?g/L,respec-tively;t max were(3.9?0.70)h and(4.2?0.70)h,respectively;AUC 0~72 were(1199.4?649.6)(?g?h)/L and(1174.3?701.9)(?g?h)/L,respectively;AUC 0~∞ were(1414.0?815.2)(?g?h)/L and(1386.1?735.8)(?g?h)/L,respectively.there were no significant differences in main pharmacokinetics parameters between2preparations,except in t max from analysis of variance and one-side&two-sides t-tests.The relative bioavailability of trial itraconazole capsule was(105.3?23.4)%.CONCLUSION:These two kinds of itraconazole capsules are bioequivalent.
