[Objective] To explore the effects of the traditional Chinese drugs Chaiyuwendantang(CYWDT) and its component on the plasm levels of ACTH,COR and cAMP in hippocampus of depressed mode.[Methods] Eighty——four male rats were divided into seven groups: control group,depressed model group,group of CYWDT,group of HuaTan,group of TiaoQiXue,group of YangXinPi,group of maprotiline equally and made up models in chronic unpredictable mild stress(CUMS).The plasm levels of ACTH,COR and the level of cAMP in hippocampus were detected by radioimmunoassay.[Results] We found that the plasm levels of ACTH,COR were significantly increase and cAMP in hippocampus was significantly decreasd in depression model of rat.CYWDT and TiaoQiXue could significantly decreased the level of ACTH(P

Objective To explore effective ways for community health service institutions to change patients' health service seeking patterns. Methods Yuetan Community Health Service Center of Fuxing Hospital affiliated to Capital Medical University and 9 community health service stations under its subordination in the 1996～2002 period were selected as a representative of the community health service institutions to be studied and intervened while the headquarters of Fuxing Hospital in the same period was selected as a representative of the third-tier hospitals to be studied. A comparison was made between the two in outpatient volume change. Questionnaires were distributed randomly to residents of the Yuetan area where Fuxing Hospital is located in order to study their health service seeking behaviors each year from 1996 to 2002. Results As a result of hard work in the 6 years, Yuetan Community Health Service Center saw a steady increase in outpatient volume whereas the headquarters of Fuxing Hospital witnessed a steady decrease in outpatient volume. Survey via questionnaires sent randomly to permanent residents of the Yuetan area indicated a change in residents' health service seeking patterns. Conclusion It is effective and viable for community health service institutions to change patients' health service seeking patterns through providing social-psychological-biological community medical services, enhancing health education and setting up a two-way referral system with third-tier hospitals.

Objective To explore the pathological mechanism of brain function and structure in young patients with major depressive disorder by diffusion tensor imaging(DTI)and resting state functional magnetic resonance imaging(fMRI).Methods Sixteen participants diagnosed with major depression (MD) and 16 healthy age and gender-matched controls(HC) were recruited. Resting state fMRI and DTI brain scans were performed on all participants. A voxel-based method (VBM) was used to analyze the DTI datasets, and regional homogeneity (ReHo) approach was applied to preprocess the fMRI datasets. The value of fractional anisotropy (FA) and ReHo maps were obtained in the whole brain.Results FA values in the MD group were significantly lower than those of the healthy controls in the white matter of the left middle temporal gyrus, right middle frontal gyrus, left medial frontal gyrus, right precentral gyrus, left angular gyrus, left fusiform gyrus, left superior temporal gyrus, right middle temporal gyrus, right sub-gyral, left insula, and left pyramis (P＜0.01). ReHo in the MD group decreased in the left superior frontal gyrus, right superior frontal gyrus, left middle frontal gyrus, right middle frontal gyrus, left medial frontal gyrus, right medial frontal gyrus, left paracentral lobule, right paracentral lobule, right inferior parietal lobule, left postcentral gyrus, right postcentral gyrus, left middle occipital gyrus, left lingual gyrus, right putamen, right cingulate gyrus, right cuneus, left superior temporal gyrus, and right middle temporal gyrus (P＜0.01). Conclusion Abnormality of brain white microstructure and function exist widely in young patients with major depressive disorder. Abnormal connection of structures and function between the brain areas may be the key reason for the depression.

Ski, as an evolutionary conserved protein, is widely involoved in the proliferation and differentiation of many kinds of cells in different species. Ski also plays an irreplaceable role in many physiological and pathological processes of nervous system, including em-bryonic nervous system development, central and peripheral nervous system diseases, and so on, which may be assiciated with the signal pathways of transforming growth factor-beta and another family member bone morphogenetic protein.

Objective:To study the effects of effective herbs in Yinchenhao Decoction(YD)on fatty deposition and TNF-?secretion of hepatic lipo-toxicity model in vitro.Methods:TG and TNF-?as the inspection target,apply the steatosis with TNF-?secretion lipo-toxicity model in vitro of HepG2 induced by long-chain FFA and uniform design U7(76)form for YD,which had 3 herbs,to compound 6 kinds of different prescriptions,in order to look for the effective herbs or optimized group in YD.Results:The Chinese medicine Yinchen and its optimized group-Yinchen and Zhizi combination can remarkably reduce the TG and TNF-?content.Conclusion:The Chinese medicine Yinchen and its optimized group-Yinchen and Zhizi combination were effective herbs in YD for inhibiting fatty deposition and TNF-?secretion of hepatic lipo-toxicity model in vitro.

Objective To investigate the role of ski in proliferation of astrocytes and the molecular mechanisms in rats. Methods Astro-cytes were obtained from cerebral cortex of a three-day old rat and cultured in vitro. siRNA targeted to ski and negative control sequences were prepared. The astrocytes were divided into ski-siRNA group, siRNA negative control group and untreated control group, while the spe-cific siRNA targeting ski negative control sequences were transfected into astrocytes with Lipofectamine? RNAiMAX Reagent. The protein levels of ski, glial fibrillary acidic protein (GFAP) and Cyclin D1 were determined with Western blotting. The proliferation of astrocytes were measured with CCK8 assay. The cell-cycle of astrocytes were analyzed with flow cytometer. Results The protein level of ski (F=38.611, P<0.01), GFAP (F=7.547, P<0.05) and Cyclin D1 (F=3.901, P<0.05) reduced in ski-siRNA group, the proliferation of astrocyte was significantly inhibited since twelve hours after culture (F>30.507, P<0.01), and less cells were in S phase and more in G1/G0 phase (F>48.425, P<0.01), compared with the control groups. Conclusion ski knocking down by siRNA significantly inhibits the proliferation of astro-cytes, which may associate with the down-regulation of Cyclin D1 expression.

Objective To investigate the transcriptional autoregulation of PhoP/PhoQ under different growth conditions in Yersinia pestis.Methods The entire promoter region of YPO1635 was amplified and cloned into the pRW50 vector containing a promoterless lacZ reporter gene.The recombinant LacZ reporter plasmid was transformed into the wild-type strain (WT) and the phoP mutant strain (ΔphoP),respectively,to measure the promoter activity (the β-galactosidase activity) of the target gene in WT and ΔphoP by using the β-galactosidase enzyme assay system.Total RNAs were extracted from WT and ΔphoP strains,and primer extension assay was employed to detect the promoter activity by examining the amount of primer extension products of YPO1635 in WT and ΔphoP.Results The LacZ fusion results showed that the transcription of YPO1635 was positively regulated by PhoP under L-TMH and brain-heart infusion(BHI) conditions,but it was not regulated in H-TMH medium.The primer extension assay detected two transcriptional start sites located at 90 and 118 bp upstream of the translation initiation site of phoP,named P1 and P2,respectively.Under low Mg2+ TMH conditions,the promoter activity of P1 rather than P2 was positively regulated by PhoP.Under high Mg2+ TMH conditions,the promoter activities of both P1 and P2 showed no obvious difference in the WT and ΔphoP strains.Under rich BHI conditions,both promoters were under negative control of PhoP.Conclusion Different autoregualtion patterns of PhoP/PhoQ under different growth conditions would help Y.pestis to quickly adapt to the changing living environment.

Objective To investigate the correlation of trace elements in maternal blood,breast milk and infant blood.Methods Atomic absorption spectrographic method was used to measure blood and milk trace elements [calcium (Ca),ferrum (Fe),zinc (Zn),magnesium (Mg),copper(Cu)] of maternal of natural delivery and infant in 250 cases,then to analyze the correlation of trace elements in maternal blood,breast milk and infant blood in postpartum 42 days.Results There were statistically significant differences in trace elements between the maternal blood,breast milk and infant blood (P ＜ 0.05).There was a positive correlation of Ca,and Fe between maternal blood and infant blood [Ca (r =0.221,P =0.047),Fe (r =0.107,P =0.043)];Fe had a positive correlation between breast milk and infant blood (r =0.139 P =0.035);There was a positive correlation of the trace elements between maternal blood and milk,but no correlation in Mg.Conclusions There was the gradient of trace elements in maternal blood and breast milk in postpartum 42 days.It can keep breast milk in a certain percentage of trace element and contribute to absorption of trace elements and growth and development in infant.The regular determination of trace elements during pregnancy and postpartum in women and their infant can direct a balanced diet and advocate breastfeeding.It can also prevent the lack of calcium,iron,zinc and other trace elements in infant.

Objective To investigate the induction and function of circulating CD4+ CD25+ regulatory T cells(CD4+CD25+ Treg cells)in patients with chronic hepatitis B,and their role in the progress of chronic HBV infection.Methods Twenty two patients with chronic hepatitis B(CHB),11 patients with acute hepatitis B(AHB)and 20 healthy blood donators were enrolled in the study.After stimulating with recombinant HBV antigen or anti-CD3 mAb,the fraction of CD4+ CD25+Treg cells in peripheral blood mononuclear cells(PBMC)from different subjects was analyzed using flow cytometry.CD4+CD25+ Treg cells and effector cells(PBMC-Treg)were sorted out and cultured alone or together in vitro,and the cell proliferation and cytokines production were measured. Serum HBV titers,HBV markers and liver function were also evaluated.Results The fraction of circulating CD4+CD25+ Treg cells induced in PBMC from CHB patients under the stimulation of recombinant HBsAg was much higher than that in AHB patients(F=3.47,P＜0.05)and normal controls(F=4.98,P＜0.05).With HBsAg stimulation,cell proliferation and cytokines production (IFN-γand IL-10)of CD4+ CD25+ Treg cells either from patients or from normal controls was very low when cultured alone,and there was no significant difference between two groups(P＞0.05).Cell proliferation and IFN-γproduction of effector cells from AHB patients were higher than those from CHB patients,however,when effector cells were cultured with CD4+Cd25+Treg cells at a ratio of 5:1,the cell proliferation and IFN-γ production were significantly inhibited (F=4.66,5.24,P＜0.05).Conclusion Circulating nificantly suppress the production and IFN-γ production of effector T Cells,which may be associated with the progress of chronic HBV infection.
