Journal of Central South University(Medical Sciences) 2010;35(6):566-571
Objective To explore the effect of E-Flow imaging on the radiotherapy of cervical lymph nodes metastasized from nasopharyngeal carcinoma. Methods A total of 52 nasopharyngeal carcinoma patients with 94 invasion neck lymph nodes were treated with conventional radiotherapy. Lymph node size, hemodynamic parameters, and blood supply were detected by E-Flow imaging and two-dimensional ultrasound before and after the radiotherapy. The relationship between those examinations and radiotherapy effect was analyzed. Results Of the 94 metastatic cervical lymph nodes from nasopharyngeal carcinoma before the radiotherapy, 40 disappeared, 52 reduced, and 2 maintained the original size after the radiotherapy. Blood flow in all residual lymph nodes decreased or disappearaed. Artery flow spectrum in the lymph node was changed from the high-speed high impedance to low-speed low resistance. The size of cervical lymph node and blood flow before the radiotherapy were associated with clinical efficacy (P<0.05). Conclusion E-Flow imaging plays an important role in assessing the efficacy of radiation therapy by changing the blood supply of cervical lymph node and its size before and after the radiotherapy.

Objective To study the effect of Schwann cell enclosed by antibody of neural cell adhesion molecule L1 on injured spinal cord. Methods Over 98% of the purity of Schwann cells obtained from bilateral sciatic nerves of 2 days newborn SD rats, the concentration of Schwann cells was about 2.5?104 /?l. The Schwann cells were enclosed by the antibody of neural cell adhesion molecule L1 using co-culture. The adult SD rats (weight 200-250 g) were used to establish the model of spinal cord injury by hemi-transection at the left side of T10 level. The animals were divided into three groups; the SC group was transplanted with 20 ?l suspension Schwann cells; the anti-L1 group with 20 ?l Schwann cells enclosed by antibody of neural cell adhesion molecule L1; and the control group was injected solely with normal saline to the injured cord. Eight weeks later regenerated neural axons were investigated through horseradish peroxiase HRP retrograde trace immunohistochemistry of neurofilament and Western blot. Results Few regenerated neural axons appeared in the control group; some of regenerated neural axons could be observed in anti-L1 group; plentiful and bulky regenerated neural axons were found in SC group. The group with antibody had significant less HRP positive neurons and neural axons than the group without antibody. Western blot showed that the quantity of neurofilament in the anti-L1 group was only two thirds of the SC group. Conclusion Schwann cell-derived neural cell adhesion molecule L1 is able to enhance the neural axon regeneration of injured spinal cord.

Objective To study the clinical features, treatment strategy and prognosis of children and adolescent with spinal bone tumors. Methods Forty-three patients were retrospectively investigated from January 1987 to June 2002, thirty-one of which were male, and the others were female. The age of patients ranged from 3 to 16 years with an average of 11.8 years. The pathological examination suggested that tumor-like conditions were 11 cases (25.6%), benign tumors 13 cases (30.2%), and malignant tumors 19 cases (44.2%). The cervical vertebrae tumors were 15 cases, thoracic vertebrae 16 cases, lumbar vertebrae 6 cases, and sacral vertebrae 6 cases. Treatment included: 1 case of malignant lymphoma was only subjected to chemotherapy; and the others were undertaken tumor resection and bone grafting to fusion with or without internal fixation. All patients of malignant tumors were performed adjuvant radiotherapy or chemotherapy after operation. Results The follow-up period was from 1 to 10 years with an average of 4.3 years. All patients of benign tumors obtained satisfied outcomes with local symptom and neural deficits improved, and no tumor recurrence happened. 6 cases (2 cases of primitive neural ectodermal tumor, 1 case of Ewing's sarcoma, 1 case of neuroblastoma, 1 case of plasma cell sarcoma and 1 case of chondrosarcoma) of malignant patients were died from metastasis and systemic failure, and the ratio was 14% and 31.6% of total and malignant tumor respectively. One case of bone giant cell tumor and 1 case of Schwannoma got recurrence at 2.5 years and 10 months after operation respectively. Conclusion For patients of children and adolescent with spinal bone tumors, the prognosis of the benign is better if tumors were removed radically. However, the patients with malignant tumors had worse prognosis with higher recurrent or death rate, and its prognosis is closely correlated to tumor's characters, operation strategy, and extending postoperative therapy.

Objective: To observe the synergetic effect of folic acid and genistein on apoptosis in the early stage of neuron and its possible mechanism. Methods: Cyclophosphamide was added to primary cultured neurons to induce apoptosis. Three different dosages of folic acid and genistein were used to inhibit apoptosis. The results were observed by flow cytometry and scanning electron microscopy (SEM). Results: 1.Folic acid and genistein might play a role in early stages of neuron apoptosis, and the synergetic effect was demonstrated when folic acid and genistein were administered together. 2. The membrane changes in early stages of neuron apoptosis observed by scanning electron microscope were also in agreement with the flow cytometry data. Conclusion: Genistein might reinforce the protective effect of folic acid on neural tube defects by restraining the apoptosis of the early stage of neuron.

Objective To investigate the changing of endothelial progenitor cells and nitric oxide in patients with pulmonary arterial hypertension secondary to chronic obstructive pulmonary disease.Method Mononuclear cells from 20 healthy persons (the control group, n = 20) , COPD patients without PAH (COPD non-PAH group, n =30) and patients with PAH secondary to COPD (COPD and PAH group, n=30) were investigated. Total mononuclear cells were isolated from peripheral blood by Ficoll density gradient centrifugation. EPC number and migration were assayed by colony forming unit-EPCs (CFU-EPCs) assay and modified Boyden chamber assay , respectively. EPC adhesion assay was performed by replanting those on fibronectin-coated dishes , then adherent cells were counted. The concentration of NO was measured with method of nitrate reductase.Result The numbers of CFU and migration, adhesion activity of circulating EPCs in COPD[ (21.9±3. 9)CFU-EPC] and PAH group[ (14. 2 ±3. 5)CFU-EPC] were significantly lower than that in non-COPD group and COPD non-PAH group [ ( 24.9 ±4.1) CFU-EPC ]. It was also observed that a strong negative correlation between the levels of PAH and the numbers of CFU and adhesion, migration activity of circulating EPCs. The NO level in the PAH group [ (43. 6 ±4. 8)ng/ml] was significantly lower than that in control group [ ( 67. 17±4.9 ) ng/ml ] ( P < 0.01). The NO level was positively correlated with number and migration ability of EPCs( r =0. 77,0.71, P <0.01) , but not correlated with adhesion ability. Conclusion The number of CFU and migratory, adhesive activity of EPC in patients withPAH secondary to COPD was significantly decreased. These changes may be associated with low level of plasma NO.

Objective To evaluate biodistribution and pharmacokinetics pattern of ~(131)I-labeled rch24which is the region-grafted (humanized) anti-carcinoembryonic antigen (CEA) monoclonal antibody in nude mice. Methods Nude mice bearing cancer xenografts received intravenous injections of ~(131)I- rch24, then blood, plasma, heart, liver, spleen, lung, kidney, tumor and other tissues were taken at different time point for determination the concentration of radioactivity and calculate the T/NT value. Nude mice were packeted randomly to four group of high, medium, low dose and continuous administration, blood drug concentration was detected by ELISA method at the different intervals. Then, draw the concentration-time curve and calculate the pharmacokinetics paramete. Results After administration, radioactivity of the tumour was significantly enhanced whereas radioactivity of normal tissues decreased gradually. For single administration, at the dose of low to medium, pharmacokinetics pattern was linearity -kinetics whereas for high dose group, pharmacokinetics paramete shown some behavior of non-linearity-kinetics. Conclusion Our results suggest that the ~(131)I-labeled region-grafted (humanized) anti-CEA monoclonal antibody rch24 exhibit a considerable targeting activity so as to ~(131)I radioisotopes can be concentrated specifically in tumor. The pharmacokinetics pattern of this medicine was different at different dose.
