Objective To study the ER status in Adriamycin-sensitive and Adriamycin-resistant MCF-7 human breast cancer cell lines. MethodsThe status of ER of MCF-7/ADR and parental MCF-7 cells was detected by Western blot. The expression of ER mRNA was detected by RT-PCR .The growth and the sensitivity to Estrogen(E 2) and droloxifene(Dro) of cells were investigated by MTT assay, and the distribution of cell cycle was detected by flow cytometric assay. Results ER and ER mRNA were positive in MCF-7 cells, and negative in MCF-7/ADR cells. In comparison with MCF-7 cells, MCF-7/ADR cells showed lower growth rate, and the cell cycle was arrested at G 0/G 1 phase. E 2 at concentrations between 1?10 -12mol/L to 1?10 -7mol/L significantly stimulated the growth of MCF-7, but did not stimulate the growth of MCF-7/ADR.Dro at concentrations between 10*!?mol/L to 20*!?mol/L significantly inhabited the growth of MCF-7, and the inhibition was dose-dependent. Dro at concentrations below 20*!?mol/L did not inhibit the growth of MCF-7/ADR, dro inhabited the growth of MCF-7/ADR only at the concentration of 20*!?mol/L, and the inhibition was more effective than MCF-7. Conclusions ER was lost in MCF-7/ADR cells, probably at mRNA level. Compared with MCF-7, the growth rate of MCF-7/ADR decreased,MCF-7/ADR cells lost the dependence on estrogen and the sensitivity to endocrine therapy.

OBJECTIVE:To establish the method for the simultaneous determination of clopidogrel (CLO) and its active metabolites (CATM) and inactive metabolites (CCAM),and to conduct pharmacokinetic study. METHODS:The plasma sam-ple had been derivatized by 2-bromine-3′-methoxy acetophenone(MPB),and was precipitated by acetonitrile. Using carbam-azepine as internal standard,UPLC-MS/MS was adopted. The separation was performed on Waters ACQUITY UPLC HSS T3 column with mobile phase consisted of water(containing 0.1% formic acid)-acetonitrile(containing 0.1% formic acid)using a gradient elution program at the flow rate of 0.50 ml/min. The ESI was equipped and quantitative analysis was operated in posi-tive ion and MRM mode. The mass transition ion-pairs were followed as m/z 322.1→211.8(CLO),m/z 504.1→155.0(the alkyl-ation derivatives of CATM,CATMD),m/z 308.3→198.0(CCAM),m/z 273.2→194.3(internal standard). RESULTS:The lin-ear calibration curves for CLO,CATMD and CCAM were obtained in the concentration range of 0.03-20.00 ng/ml,0.30-200.00 ng/ml and 10.00-10 000.00 ng/ml in plasma,respectively;intra-day and inter-day RSD for them were all less than 15%,and relative error(RE)ranged from -3.5% to 5.7%. Main pharmacokinetic parameters of CLO,CATMD and CCAM in 5 healthy volunteers after oral administration of CLO 300 mg were as follows:cmax were(7.89±5.46),(15.58±8.08),(8 023.33± 1 047.39)ng/ml;tmax were(1.25 ± 0.43),(1.25 ± 0.43),(1.67 ± 0.29)h;t1/2 were (2.31 ± 0.61),(0.64 ± 0.08),(6.53 ± 2.55)h;AUC0-t were(17.19±14.59),(21.39±9.58),(30 648.85±8 026.63)ng·h/ml. CONCLUSIONS:The established method is sensi-tive,rapid and convenient, which is suitable for pharmacokinetic study and plasma concentration determination of CLO and its metabolites.

The concept of epigenetics was first proposed by Waddington in 1942,referring to phenotypic changes that are not related to the underlying DNA sequence, yet alterations in the level and function of gene expression can be heritable, such as DNA methylation, histone modifications, microRNA interference, etc.Phenotype changes while genotype remains unchanged, this controlled mechanism can be kept stable throughout cell division, proliferation, and subsequent process of development. This process thus may cause corresponding changes in the pathophysiology, which are correlated with the occurrence of cancer, immune disorder, cardiovascular disease, metabolic syndrome and so on.Diabetes mellitus is the third heaviest chronic diseases in the world, and does serious harm to human health, especially the diabetic vascular diseases.Even if the blood glucose level is controlled in an ideal state, vascular inflammation still persists, indicating the existence of metabolic memory.Studies have suggested that the pathogenesis can be elucidated at the level of epigenetics. This paper mainly focuses on DNA methylation, histone modifications, and is an overview of epigenetics research progress in diabetic vascular diseases.

Chinese Journal of Tissue Engineering Research 2014;(4):493-498
BACKGROUND:There is a high risk for the elderly cervical spine surgery combined with cerebral infarction, whereas conservative treatment is often unable to resolve serious cervical lesions.
OBJECTIVE:To discuss the surgical effects of anchor fixation via posterior approach on cervical spinal canal stenosis combined with cerebral infarction.
METHODS:A total of 21 patients with cervical spinal canal stenosis combined with cerebral infarction who were admitted over the past 5 years accepted cervical posterior expensive open-door laminoplasty and fixation with wire anchors. Therapeutic effects were evaluated according to the Japanese Orthopaedic Association (JOA) scores.
RESULTS AND CONCLUSION:Al the patients were fol owed up 6 to 24 months, averagely 15 months. Al patients were smoothly through the perioperative period. There were no acute severe cerebral infarction cases. Preoperative JOA score was (7.6±2.0) points averagely, and postoperative JOA score was (13.3±1.8) points averagely, showing a significant difference (P<0.01). Of the 21 patients, excellent effects were in 11 cases and good in 7 cases. The effective rate was 95%(20/21), and the excellent-good rate was 86%(18/21). The risk of surgical treatment of patients with cerebral infarction is higher, but it is not the absolute contraindication. Choosing the proper operation way is highly important. Cervical posterior expensive open-door laminoplasty and fixation with wire anchors can be used to reduce the risk of cerebrovascular accidents to some extent, and obtain an excellent clinical effect.

Objective:To study the effects of ~(125)I-(α_v)ASODN on the in vitro invasive ability of heptocellular carcino-ma cell line(HepG2) through PEI-RGD-mediated receptor process. Methods: Intergrin α_v-specific antisense oligonucle-otide was labeled with ~(125)I, and PEI-RGD/~(125)I-(α_v)ASODN complex was prepared by combining ~(125)I-(α_v)ASODN with polyethyleneimine derivative PEI-RGD. PEI-RGD/~(125)I-(α_v)ASODN complex was transferred into HepG2 cells through the receptor-mediated process. The effect of PEI-RGD/~(125)I-(α_v)ASODN complex on the invasive ability of HepG2 cells was examined by Boyden chamber invasive assay. Results: (1) The labeling yield and radiochemical purity of ~(125)I-(α_v) ASODN were(73.78±4.09)% and(96.68±1.38)%, respectively, and the labeled compound had a good stability in vitro after 48 h at 37℃; (2) The ability of HepG2 cells to uptake PEI-RGD/~(125)I-(α_v)ASODN reached its peek ([12.77±0.85] % ) when PEI-RGD/~(125)I-(α_v)ASODN was at 4 μl/2 μg ([12.77±0.85] %), and then gredually decreased thereafter. So the dosage of PEI-RGD/~(125)I-(α_v)ASODN for the following experiment was chosen as 2 μl/1 μg; (3) The invasive capacity of HepG2 cells was significantly reduced in PEI-RGD/~(125)I-(α_v)ASODN group compared with those in other experiment and control groups (P <0.01 ). Conclusion: ~(125)I-(α_v)ASODN mediated by PEI-RGD can effectively inhibit the invasive capacity of HepG2 cells.
