1.An experimental study of the influence of impact to lamina on spinal cord blood flow

Objective To study the influences of impact of different loads to lamina on spinal cord with a closed spinal cord injury model and to explore the mechanisms of blood vessels in spinal cord injury. Methods 120 normal male SD rats were randomized into 4 groups, with 30 in each. Group A served as the control group which received no impact. In Groups B, C and D,impacts of 10, 15 and 20 N were given to lamina respectively. The blood flow in the spinal cord central gray matter and the dorsal white matter, the blood pressure and the heart rate were observed at different experimental time points. Results No obvious change was observed in the blood pressure and heart rate in every group at different time points. The spinal cord blood flow (SCBF) in Groups C and D varied in a regular pattern. Conclusions The impact load to lamina can affect the spinal cord mainly by altering the spinal cord microcirculation. The influence is the most obvious on the blood flow of microcirculation of the gray matter.

[Objective]To research the influence of spinal cord blood flow(SCBF) and the evoked potential(EP) by the extramarrow impacting and to explore the correlation and regularity between the spinal cord blood flow and evoked potential after spinal cord injury.[Method]Normal 120 SD rats were randomized into 4 groups,with 30 in each.Group A was the control group.In groups B,C,D,impacts of 10,15,20N were given to lamina respectively.The blood flow of impacting position and evoked potential were observed at different time points including 0 min,1 h,6 h,1 d,6 d,12 d,24 d.[Result]The blood flow of spinal cord represents a dimning trend with the impacting load raising.The blood flow of group A,had no marked change,the SEP and MEP also.The blood flow of group B represented a temporaly lowing,then recovered normal,the SEP and MEP also.The blood flow of group C and D represented a continuous lowing,then slowly raising.The group C recovered normal at 24d,the group D still lower than normal.[Conclusion]The impact load to lamina can influence the spinal cord function through altering the spinal cord blood flow.There is evident correlation between the SCBF and EP.

[Objective]To investigate the changes and correlation of bone collagen and biomechanical property in osteoporosis rats.[Method]Normal 30 nullimating female 7 month SD rats were divided randomly to two groups,each included 15.The sham group was unovariectomized,and the OVX group was ovariectomized.To execute the rats after 12 weeks,the author get the 5th lumbar vertebra,and then detected the biomechanical property and HYP.The author observed the collagen morphology of the 4th lumbar vertebra using the microscope after the masson triad color dyeing.[Result]Twelve weeks later,HYP in the sham group was(0.197?0.035)mg/g.The bone collagen was regular,compact,and closed succession.HYP in the OVX group was(0.182?0.022)mg/g.The bone collagen was incompact,thin,collapse and poor succession.There was significant change of the HYP in two groups(P
5.Expressions of p75NTR, Bax and Bcl-2 and cell apoptosis in rat cortical neurons following mechanical injury

ObjectiveTo study the expressions of p75NTR,Bax and Bcl-2 and cell apoptosis in rat cortical neurons following mechanical injury and discuss the mechanism and mutual action way during the apoptosis of rat neurons after mechanical injury.Methods Cortical neuron cultures were prepared from the brain tissues of day 17 rat embryos and were exposed to mechanical injury seven days after seeding.After the traumatic neuron injury models were created,the apoptosis ratio of neurons was tested at several time points.The models were divided into the minor,moderate,severe injury groups according to the injury severity and the control group.The expressions of Bax and Bcl-2 in each group were detected by immunohistochemistry method and that of p75NTR by Western-blot.Combining with cellular apoptosis ratio in each group shown after FCM analysis,the correlation between the expressions of p75NTR,Bax and Bcl-2 and the apoptosis in rat neurons after mechanical injury could be analyzed.ResultsThe apoptosis ratio of the neurons in all the injury groups was obviously higher than that in the control group,with significantly higher apoptosis ratio of the neurons in the severe injury group than the minor and moderate injury groups (P <0.05).P75NTR,Bax and Bcl-2 were all expressed in all the injury groups,with statistical differences between groups.The expression of Bax in the severe injury group was significantly higher than that in the minor and moderate injury groups(P < 0.05 ). Conclusions p75NTR expression and Bax/Bcl-2 ratio are closely correlated with neuron apoptosis.The early expression of p75NTR may be one of mechnisms for neuronal apoptosis after neuron injury,when Bax and Bcl-2 may be involved.
6.Application of lumbo-iliac fixation in pelvic ring reconstruction following communited Denis Ⅲ sacral fractures

Objective To study the anatomic morphology of posterior column of ilium(PCI)and valne of lumbo-iliac fixation(LIF)in pelvic ring reconstruction following communited Denis Ⅲ sacral fractures.Methods A total of 17 hemi-sectional pelvis and dry ilia were selected to observe the length,width and height(safety and standard)of PCI,when a detailed description was done on the methods determining entry point and operation techniqus for iliac screws anchoring.Nine patients with eommunited sacral fractures involving Denis Ⅲ areas combined with neurologic deficits were decompressed for recovery of neurological function and reconstruction of pelvic ring by using LIF.Postoperative follow-up was done on role of LIF and result of neural rehabilitation.Results PCI was mean 110.57 mm in length,24.24 mm in width,14.54 mm in standard height and 31.46 mm in safe height.Nine patients could turn over in bed immediately following operation and walk one month later.A follow-up for 13-37 months in eight patients showed stable reconstructed pelvic ring,sound function of movement and bone graft fusion,which met the target of keeping stability of fractured extremity and pelvis.Conclusions pCI can provide anatomic support for LIF.When decompression and fixation is needed for sacral fractures combined with anterior pelvic ring disruptions,LIP can provide stable fixation and play an important role in reconstruction,nursing and rehabilitation of pelvic ring.
7.The curative effect and prognostic analysis of transcatheter arterial chemoembolization combined with radiofrequency ablation in treatment of liver cancer

Objective To investigate the clinical efficacy and prognosis of transcatheter arterial chemoembo-lization(TACE)combined with radiofrequency ablation(RFA)in treatment of liver cancer.Methods 64 patients of liver cancer were selected as the research subjects,and they were divided into two groups by randomized single blind method.32 cases in the control group adopted the TACE treatment,while 32 cases in the observation group were given TACE combined with RFA.The tumor shrinkage,complete necrosis rate,local recurrence rate,AFP levels and adverse reactions of the two groups were compared.The patients were followed up for 3 years to record survival.Results After treatment,the tumor shrinkage rate and complete necrosis rate of the observation group were 90.63% and 75.00%, which were significantly higher than 68.75% and 50.00% of the control group,the differences were statistically significant (χ2 =8.453,8.203,all P <0.05).The local recurrence rate of the observation group was 21.88%,which was significantly lower than 43.75% of the control group,the difference was statistically significant(χ2 =7.887,P <0.05).After treatment,the AFP level of the observation group was (70.11 ±6.45 )ng/L,which was significantly lower than (157.76 ±10.42)ng/L in the control group,the difference was statistically significant (t =7.433,P <0.05).The survival rates of 1 year,2 years and 3 years in the observation group were 84.38%,71.88%,56.25%, which were significantly higher than 65.63%,53.13%,31.25% in the control group (χ2 =8.677,8.203,8.985,all P <0.05).The incidence rate of postoperative adverse reaction had no significant difference between the two groups (χ2 =1.337,P >0.05 ).Conclusion TACE combined with RFA in treatment of liver cancer can significantly improve the effect of tumor necrosis rate,reduce the local recurrence rate and prolong the survival time of the patients, and it had less adverse reaction,higher safety,and which should be applied in clinical.

ObjectⅣe To investigate the nephroprotectⅣe effect of mycophenolate mofetil (MMF) on 5/6 nephrectomized rats and explore its mechanism. Methods The 5/6 nephrectomized SD rats were used. After the operation, 15 mg/kg MMF, 25 mg/kg fosinopril (F) and 15 mg/kg MMF+25 mg/kg fosinopril were respectⅣely delⅣered daily by gavage for 8 weeks. Urinary protein, BUN, serum creatinine were measured at the end of the study. Renal pathological changes were evaluated and immunohistochemistry was used to examine the expression of fibronectin (FN), collagen Ⅳ, macrophage chemoattractant protein 1 (MCP-1) and proliferating cell nuclear antigen(PCNA) . RT-PCR was used to examine the TGF-?1 mRNA and TIMP-1 mRNA in the cortex of the kidney. Objective To prove the prophylactic function of absorbable gelatin sponge and bioprotein glue for prevention of epidural and nerve root scar adhesions after laminectomy. Methods We excised the L6 vertebral plates of 36 adult New Zealand male rabbits,to make vertebral plate excision model and to find nerve root. Then divided them into 3 groups based on different materials:gelatin sponge and bioproten glue (group A),gelatin sponge (group B) and saline solution (group C). Polo-box domain 1 (PBD1) is a characteristic domain of polo-like kinase 1 (PLK1), which locates in C-terminal and can influence the catalytic activity and specific subcellular locations of PLK1. At present, most PLK1 inhibitors are developed to occupy the ATP pocket or its close sites. However, this kind of PLK1 inhibitors is difficult to pursue target selectivity and may encounter cross drug resistance with other kinase inhibitors due to the conserved sequence of ATP pocket. Recently, PBD1, with aberrant specificity in sequence and structure, has attracted enormous interests as the alternative target to the discovery of corresponding inhibitors for anti-tumor drugs. The structure and function of PBD1 as well as the advances of its inhibitors are reviewed in this paper.
