1.Correlation between matrix metalloproteinase-9 and Cystatin C at the early stage of diabetic nephropathy

Objective To observe the changes in the level of serum cystatin C and the expression of matrix metalloproteinase-9 (MMP-9) in the kidney after the development of diabetic nephropathy in streptozotocin (STZ) induced diabetic rats, and to study their correlation. Methods Sixteen female Wistar rats were used to reproduce diabetes by interperitoneal injection of STZ (60mg/kg). Immunohistochemical staining was employed to detect the expressions of MMP-9 in kidneys with image analysis system, and ELISA was used to determine serum cystatin C after 4 or 8 weeks. The correlation between MMP-9 and cystatin C was analyzed by SPSS 10.0. Results The expressions of MMP-9 in diabetic kidneys both at the 4th and the 8th week (8.180?0.905 and 3.948?0.623, respectively) were significantly lower than that of normal control rats (15.620?3.444 and 16.185?3.120, respectively. 2.Analysis of clinical curative effect of the therapy of goserelin combined with microwave on postoperative patients of endometriosis

Objective To observe the clinical efficacy of therapy of application of goserelin(Zoladex)com-bined with microwave on patients with endometriosis after laparoscopic surgery.Methods 188 patients with endome-triosis were randomly divided into two groups according to the randomized digital table method,with 94 cases in each group,as the observation group(Zoladex combined with microwave therapy group)and the control group(Zoladex treatment group).Among them there were 80 patients with infertility,45 cases in the observed group and 35 cases in the control group.After treatment,the clinical efficacy rate,recurrence rate and pregnancy rate of patients with infertil-ity were compared between the two groups.Results The efficiency of the observation group was significantly higher than that of the control group(80.12% vs.65.31%,χ2 =5.338,P <0.05),and the recurrence rate of the observa-tion group was lower than that of the control group(22.34% vs.38.56%,χ2 =8.325,P <0.05),the difference was statistical significance.The pregnancy rate of patients with infertility in the observation group were significantly higher than that of the control group,and the difference was statistically significant[64.38%(29 /45)vs.44.46%(16 /35)χ2 =4.531,P <0.05].Conclusion As for patients with endometriosis after laparoscopic surgery,the therapy of Zoladex drug treatment assisted with microwave treatment at the same time,could achieve good clinical curative effect and low recurrence rate,and could improve the pregnancy rate of patients with infertility.
3.Anatomical study of the proximal end of the radius in adults and its clinical application

Objective To provide the anatomical basis for treatment of comminuted fractures of the radial head with bending steel miniplates by observations of the proximal end of the radius (PEOTR) in adults and to investigate the effect of its application. Methods The PEOTR and the dynamic relationship between the articular circumference of the radial head and the ulnar radial notch were studied with 32 pairs of specimen of the dried radii and ulnae in adults. From Jan 1988 to Mar 1998, eleven cases of comminuted fractures of the radial head were treated with bending steel miniplates procedure; there were seven males and four females, with an average age of 35 years. According to Mason's grading, 9 cases were classified as Type Ⅲ radial head fractures, 2 cases as Type Ⅳ radial head fractures. Results The average length of the PEOTR was 41.6 mm. The radial neck deviated from the radial shaft at an angle of 164?. Then at the neutral position, the greatest range of pronation and supination of the forearm, the range of contact of radians and the range of non contact radian between the articular circumference of the radial head and the ulnar radial notch were 69?,108?,45? and 138?, respectively. The period of follow up in all cases was 20-38 months. All had bony union, with an average healing time of 8.5 months. Evaluation was done according to the criterion made by Broberg and Morrey. The result was excellent in 6 cases, good in 4 cases, and fair in 1 case. Conclusion The treatment of comminuted fractures of the radial head with bending steel miniplates is a safe and effective method.
4.Observation of effect of metformin and exercise therapy for IGT.

Objective To observe the effect of metformin and exercise therapy for IGT. Methods 86 patients with IGT were given metformin and exercise therapy,the range of glucose,BMI and symptoms improvement were observed before and after the therapy. Results After 6 months therapy,the fasting blood glucose and 2 h after oral administration of 75 g glucose levels were significantly reduced(P<0.01=,BMI reduced(P<0.05=.72 cases (83.7%)were back to normal glucose tolerance,14 cases maintained IGT(those unable to adhere to exercise and no diet controlled),0 case became DM. Conclusion Metformin and exercise therapy had good efficacy in curing IGT.
5.Establishment of hepatocellular carcinoma model in mouse by interventional ultrasound

ObjectiveTo study the feasibility and stability of hepatocellular carcinoma model established in mouse with the guidance of ultrasound, and to explore the value of ultrasonography in hepatocellular carcinoma model.MethodsThe type of H22 cells, whose concentration was 2×107/ml, were injected into the left liver of 30 mice with the guidance of ultrasound. After injection, the mice were examined by ultrasound on the tenth and twentieth day, and implemented laparotomy on the twentieth day. The tumors were examined pathologically. The formation rate and growth of tumors were evaluated, and the ultrasonic features of tumors were observed.ResultsThe formation rate of implanted tumors was 96.7% (29/30), and the peritumoral blood flow could be detected. The diameters measured by ultrasound and explorative laparotomy were (8.4±0.3) mm and (8.6±0.2) mm, respectively (P>0.05). The pathological examination showed that the tumors were hepatocellular carcinoma.ConclusionEstablishing mouse model of hepatocellular carcinoma by ultrasound-guided is simple and minimally invasive, and it provides an ideal model of high formation rate for the study of hepatocellular carcinoma.
6.Treatment of infantile hemangioma in parotid gland region with pingyangmycin injection

Objective To evaluate the clinical efficacy of pingyangmycin (PYM) injection on infantile hemangioma located in the parotid gland region. Methods Twelve patients were treated by intralesional injection of PYM. When necessary, the injections were repeated at an interval of one week, but not more than 3-4 sessions within a therapeutic period. Normally, the secondary therapeutic period was performed 1 month later. The general and local adverse responses were recorded and the clinical outcomes were assessed with a follow-up of 1 to 3 years. Results Complete clinical resolutions were achieved in 10 patients. 2 patients received one injection, 3 patients received 2 injections, 3 patients received 3 injections, and 2 patients received 4 injections. The remaining 2 patients with partial resolution received 6 and 7 injections respectively. No clinical recurrence was observed during the follow-up of 1 to 3 years. No ulcerations or postoperative sears in injection regions were presented. The function of facial nerve was remained normality in all patients. The systematic side effects included transient pyrexia and poor appetite appeared in partial patients. No allergy cases were found. Conclusion Treatment of infantile hemangioma located in parotid gland region with PYM injection reveals a high rate of complete clinical resolution, with fair cosmetic results and short treatment time, and it does not damage the facial nerve or form local scar.The treatment time of PYM injection seems to be positively related to size of the lesions.
7.Neuroprotective effects of blonanserin on H2O2-induced injury in PC12 cells

Objective To explore neuroprotective effects of blonanserin on H2O2-induced injury in PC12 cells. Meth?ods PC12 cells were divided into four groups:control group (C group), H2O2-treated group (H group), blonanserin pretreat?ed group (B group) and positive control group (vitamin E- pretreated, E group). The effects of different concentrations of blonanserin (0, 5, 10, 20, 40, 80 and 160 μmol·L-1) on cell proliferation in PC 12 cells were observed. MTT assay was used to detect the cell activity of different groups. The apoptotic rates of different groups were measured by TUNEL assay. The mor?phological changes were observed using inverted microscope and Hoechst 33258 staining. The superoxide dismutase (SOD) vi?ability and malondialdehyde (MDA) levels were detecded by biochemical methods in four groups. Results The appropriate concentration of blonanserin (0-20 μmol·L-1) can promote the growth of PC12 cells. Comparing with the C group, the apoptot?ic rate and MDA level were increased in group H, while the cell viability and the SOD viability were decreased obviously ( P<0.05). Compared with H group, the cell viability, SOD viability were significantly increased, while the MDA level and apoptotic rate were decreased (P<0.05). Conclusion Blonanserin shows neuroprotective effect on H2O2-induced injury in PC12 cells.
8.Effect of recombinant human growth hormone on height and serum IGF-1 in children with idiopathic short stature

Objective:To study the effect of recombinant human growth hormone on height and serum insulin-like growth factor 1(IGF-1) in children with idiopathic short stature.Methods:Fifty children with idiopathic short stature admitted to Weihai Central Hospital from September 2015 to February 2017 were randomly divided into treatment group(25 cases) and control group(25 cases) according to the random digital table method.The control group received nutritional support treatment.The treatment group was treated with subcutaneous injection of recombinant human growth hormone on the basis of the control group.All the children were treated and observed for one year.The height, weight, growth rate, bone age, total thyroid hormone level, fasting blood glucose level, serum IGF-1 level and adverse reactions were compared before and after treatment.Results:After 1 year of treatment, the height, growth rate, and IGF-1 of the two groups were significantly higher than those before treatment ( t=4.427, 2.987, 7.459, 5.963, 31.389, 21.790, P<0.001, 0.004, <0.001, <0.001, <0.001, <0.001). which in the treatment group were significantly higher than those in the control group ( t=2.914, 2.480, 12.090, P=0.006, 0.017, <0.001). The body weight, bone age, total thyroid hormone levels, and fasting blood glucose levels in the two groups were not significantly different from those before treatment ( t=0.548, 1.931, 0.300, 1.367, 0.735, 0.759, 0.693, 0.920, P=0.586, 0.060, 0.765, 0.179, 0.466, 0.452, 0.492, 0.362), and there were no statistically significant differences between the two groups ( t=0.371, 0.141, 0.059, 0.318, P=0.713, 0.889, 0.953, 0.752). There was no statistically significant difference in adverse reactions between the two groups during treatment (χ 2=2.083, P=0.149). Conclusion:Recombinant human growth factor can effectively accelerate the growth rate of children with idiopathic short stature, increase the height of children, increase the level of serum IGF-1, and has no obvious side effects.
9.In vitro isolation and cultivation of human scalp dermal papilla cells by two-step enzyme digestion of small specimens

Objective:To develop an efficient and rapid method for the isolation and cultivation of human scalp dermal papilla cells from small specimens.Methods:Hair-bearing skin specimens measuring 0.5 cm × 0.5 cm -0.5 cm × 1 cm in size were obtained from the scalp of 3 patients with pigmented nevus and 6 with sebaceous nevus during surgery in Department of Dermatology, the First Hospital Affiliated to Army Medical University from September 2018 to January 2019. The subcutaneous fat layer containing hair follicles was cut out of the specimens, and hair follicles were sorted with ophthalmic forceps, which were subsequently digested with 0.6% dispase Ⅱ for 30 minutes, then with 0.2% collagenase Ⅳ at 37 ℃ for 30 - 60 minutes, and were centrifuged to obtain hair papillae. Morphological observation was performed on the isolated hair papillae, and dermal papilla cells were cultured, passaged and identified.Results:Under the microscope, the hair papillae isolated by two-step enzyme digestion of small scalp specimens were intact, and showed an inverted pear-like shape, and residual dermal sheaths could be observed around some hair papillae. However, no hair papilla was isolated by one-step enzyme digestion. With the two-step enzyme digestion method, the hair papilla separation rate was 60.8% ± 2.1%, the adherence rate of the dermal papilla cells at 72 hours was 86.6% ± 3.9%, the time for cells to emigrate out of hair papillae was 0.5 - 3.0 days, the total operation duration was 2.0 - 3.0 hours, and the actual operation duration after subtraction of digestion duration was 1.0 - 1.5 hours. The dermal papilla cells isolated by the two-step enzyme digestion method could grow in an aggregative pattern in early stage, but grew in a non-aggregative pattern after 8 passages.Conclusion:The two-step enzyme digestion of small specimens is a simple and efficient method for isolating human scalp dermal papilla cells.
10.Evaluation of symptomatic middle cerebral arterial atherosclerotic plaque by three dimensional-sampling perfection with application optimized contrasts using different flip angle evolutions of high-resolution magnetic resonance imaging

Objective To analyze characteristics and clinical significance of affected and unaffected side plaques in patients with symptomatic middle cerebral artery (MCA) atherosclerosis by three dimensional-sampling perfection with application optimized contrasts using different flip angle evolutions (3D-SPACE) of 3.0 T high-resolution magnetic resonance imaging (MRI).Methods During the period from September 2016 to June 2017,31 patients admitted to the Peking University Aerospace School of Clinical Medicine were enrolled in this study.Routine MRI and high-resolution MRI were performed respectively.Patient's affected and unaffected sides were distinguished based on the conventional MRI on the presence or absence of signs of ischemia.The characteristics of bilateral MCA plaques were studied by high-resolution MRI 3D-SPACE sequences.The stenosis rate,plaque load,eccentricity index were quantitatively analyzed,and the degree of plaque enhancement and remodeling were qualitatively analyzed.Results Thirty-one MCAs in affected and unaffected sides were respectively collected,and 124 MCA plaques and reference plane images were obtained.Among all the patients,tihe MCA in unaffected side was larger in lumen area and had atherosclerotic plaques,and some MCAs had varying degrees of thickening of the wall.The stenosis rate,plaque burden and significantly enhanced plaque proportion of MCA in affected side were significantly higher than those in unaffected side (stenosis rate:76.2% ± 11.5% vs 51.7% ± 13.1%,t =7.801,P<0.01;plaque burden:81.6% ±8.0% vs 56.7% ±13.9%,t =8.614,P<0.01;significantly enhanced plaque proportion:50.0% (13/26) vs 0 (0/26),x2 =17.333,P< 0.01).There was no statistically significant difference in plaque distribution between the two sides.Positive remodeling blood vessels had more vessel area than negative remodeling blood vessels ((13.8 ± 2.9) mm2 vs (11.3 ± 2.8)mm2,t =2.389,P =0.024).Conclusion The high resolution MRI 3D-SPACE sequence clearly shows the MCA wall and plague,and can analyze the enhancement degree and morphological features of affected and unaffected side atherosclerotic plaques simultaneously,thus it is beneficial for risk stratification assessment and individualized treatment for stroke Patients in clinical practice.
