2.Assessment of alteration of ischemic myocardium′s motion during systole by Doppler tissue imaging

Objective To study the velocity pattern of ischemic myocardium change during systolic phase by Doppler tissue imaging(DTI). Methods Twenty-three dogs were investigated as two groups. In 13 open-chest dogs as group A, regional wall motion abnormalities were induced by occluding branches of left circumflex coronary arteries(LCX). Left ventricular(LV) wall systolic velocity patterns at the mid-wall portion of the middle of LV posterior walls in the short-axis direction were recorded with DTI before and after artery occlusion. Ten dogs as group B were operated to occlude branches of LCX. Before and one week after operation the same parameters were recorded by transthoracic method. Results There were positive velocity peak(IVC 1), negative velocity peak(IVC 2) during isovolumic contraction and positive velocity peak(S) during ejection in DTI spectra of all normal LV posterior walls. The peak velocities of IVC 1 and S decreased significantly, but the peak velocity of IVC 2 increased significantly after occluding branches of LCX in both groups (P
3.Supernatant of myocardiocyte induces differentiation of bone marrow-derived mesenchymal stem cells

BACKGROUND:Culture supernatant containing myocardiocyte has been demonstrated to induce differentiation of bone marrow-derived mesenchymal stem cel s into myocardiocyte-like cel s. This may associate with some or several cytokines in the culture supernatant.
OBJECTIVE:To explore if the supernatant of myocardiocyte induces bone marrow-derived mesenchymal stem cel s to differentiate into myocardiocyte-like cel s is associated with the different cytokine content in the supernatant of myocardiocyte.
METHODS:Bone marrow-derived mesenchymal stem cel s were isolated and cultured in vitro by the whole bone marrow adherent culture. Cardiocytes were isolated and cultured by enzyme digestion. Bone marrow-derived mesenchymal stem cel s (1×108/L) and cardiocytes (1×105/L) were cultured for 72 hours and the supernatant was col ected. Hepatocyte growth factor, insulin-like growth factor 1, platelet-derived growth factor, stem cel factor, fibroblast growth factor and vascular endothelial growth factor levels in culture supernatant of bone marrow-derived mesenchymal stem cel s and cardiocytes were detected by enzyme-linked immunosorbent assay.
RESULTS AND CONCLUSION:The content of insulin-like growth factor 1, platelet-derived growth factor and fibroblast growth factor in the supernatant of cardiocytes was significantly higher in cardiocytes group compared with bone marrow-derived mesenchymal stem cel s group (P<0.01). Results indicated that insulin-like growth factor 1, platelet-derived growth factor and fibroblast growth factor in the supernatant of cardiocytes may have capability to induce bone marrow-derived mesenchymal stem cel s to differentiate into myocardiocyte-like cel s, and insulin-like growth factor 1 may serve as the main cytokine.
4.Effect of immunotherapy of recombinant chimeric epitopes of major aller-gen group 1 from Dermatophagoides farina on asthma of mice

Objective To investigate the effect of immunotherapy of recombinant chimeric epitopes of major allergen group 1 from Dermatophagoides farina on asthma of mice. Methods Forty mice were randomly divided into 4 groups:a negative con?trol group,an asthma group,an immunotherapy group of Der f 1,and an immunotherapy group of Der f 1A. On the 1st,7th and 14th day,the mice in the asthma group,immunotherapy group of Der f 1,and immunotherapy group of Der f 1A were injected intraperitoneally with the extract of D. farina 3 times to sensitize;and on the 21st day,the atomized inhalation was carried out for 7 days. In the control group,phosphate buffer solution(PBS)was applied for sensitization and inhalation. In the immunother?apy groups,Der f 1 and Der f 1A were applied to carry out the specific immunotherapy respectively for 30 min before the inhala?tion. Then,the leukocytes in the bronchoalveolar lavage fluid(BALF)were numbered and the pathological sections of lung tis?sues were observed;IL?5 and IFN?γin BALF and spleen cell culture supernatants(SCCS)as well as the specific IgE,IgG2a in the sera were detected. Results Compared with the asthma group,the lung inflammation of mice in the immunotherapy groups was lightened,and the total numbers of leukocytes in BALF were significantly reduced;IL?5 was significantly reduced and IFN?γwas significantly increased in BALF and SCCS of mice in the immunotherapy groups;and the specific IgE was significantly re?duced and IgG2a was significantly increased in the sera of mice in the immunotherapy groups(all P<0.01). Conclusion The recombinant chimeric epitopes of major allergen group 1 from D. farina could effectively relieve the symptom of asthma in mice, so as to provide the evidence for specific immunotherapy.
5.Correlation Between Intravoxel Incoherent Motion and Prognostic Factors of Rectal Cancer

Purpose To explore the correlation between quantitative parameters from intravoxel incoherent motion (IVIM) with prognostic factors of rectal adenocarcinoma.Materials and Methods Eighty-six patients with rectal adenocarcinoma who were underwent surgery without neoadjuvant therapy in our hospital between September 2015 and July 2016 were selected in this retrospective study.The image data included multiple b-values diffusion weighted imaging (DWI) examination and the corresponding D,D* and f values of the lesions.Relationships between the quantitative parameters and tumor pathology indexes including histological differentiation grade,tumor T/N stage,lymphangiovascular invasion state,the expression level of epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor-2 (Her-2) were assessed.Results The average D values of different differentiation degree (high,middle and low) of rectal adenocarcinoma were (0.541±0.093)×10 3mm2/s,(0.490±0.156)×10-3mm2/s and (0.342 ± 0.147)× 10-3 mm2/s,and the difference was statistically significant (P<0.05).TheD values were significantly different between the lymphangiovascular invasion and non invasion state [(0.511 ±0.154)× 10-3 mm2/s vs (0.387±0.130)×10-3 mm2/s,P<0.05)].However,there were no significant differences in the mean D,D* and f values among different tumor T/N stage (P>0.05).The average f value of EGFR or Her-2 high expression group was higher than that of low expression group (0.379±0.076 vs 0.298±0.099,P<0.01;0.356±0.097 vs 0.298±0.098,P<0.05,respectively).Conclusion Quantitative parameters of IVIM in rectal adenocarci-noma can be used as noninvasive imaging biomarkers to predict the biologic behavior of tumor and the prognosis of the patients.
6.STUDY ON THE MECHANISMS OF GENISTEIN AFFECTING THE CHEMOSENSITIVITY OF HUMAN BREAST CANCER CELL LINE MDA-MB-453 TO PACLITAXEL IN VITRO

Objective:To investigate the mechanisms of genistein (GEN) affecting the chemosen- sitivity of human breast cancer cell line MDA-MB-453 to paclitaxel (PTX) in vitro. Method:HER2/neu- overexpressing breast cancer cells MDA-MB-453 were treated by GEN, PTX alone or combined in vitro. Cell cycle was measured by flow cytometry. The expression of HER2/neu protein was observed by immunocytochemistry and. Akt, p-Akt, cyclin B1 and CDK1 protein by Western blot. Results:Cell cycle of MDA-MB-453 cells was blocked at G1/S after treatment of GEN, while at G2/M after treatment with PTX alone. Both GEN and PTX did not change the expression of HER2/neu, total Akt and CDK1 in MDA-MB-453 cells, but GEN significantly decreased p-Akt and cyclin B1 level, and PTX obviously increased cyclinB1 level. GEN antagonized the effects of PTX on level of cyclin B1 protein and blockage of G2/M in MDA-MB-453 cells after treatment with GEN and PTX in combination. Conclusion:The antagonism effects of GEN on the increase of cyclin B1 and blockage of G2/M induced by PTX may be one of the mechanisms of GEN affecting the chemosensitivity of MDA-MB-453 cells to PYX.
7.Effects of cholesterol-inhibitor lovastatin on the activity of MAPK in MCF-7 breast cancer cells

Objective To investigate the effects of lovastatin on MAPK activity in MCF 7 breast cancer cells. Methods After MCF 7 cells were treated with 4, 8 and 16 ?mol/L lovastatin for 48~72 h, the expressions of ERK1 and p38 MAPK proteins were detected by Western blotting. Their phosphorylation levels were observed by kinase activity assay. Results MCF 7 cells treated with different doses of lovastatin for 48~72 h had no significant effect on the protein level of ERK1, but the activities of ERK1 and the P38 MAPK proteins were down regulated at 72 h after treatment. Conclusion Lovastatin may down regulate the activity of MAPK in MCF 7 cells by means of inhibiting the mevalonate metabolism pathway and/or affecting phosphorylation levels.
8.Application of medical image processing, transmission and on-line learning system in medical imaging teaching

Using the Medical Imaging Experimental Teaching Demonstration Center of Nanjing Medical University and related web site as a platform, we have established a case library and online learning system which fused medical imaging, pathology and anatomy images according to the human organ system. The system has achieved a“medical imaging”teaching real-time, diversified, systematic and clinical simulation technology. Medical students can perform image processing, simulate writing reports and discuss online using this system. Teachers can use the system to carry out teaching reform-problem-based learning (PBL), and the integration of teaching with horgan systems and case based curriculum.
9.Diazepam and sodium valproate infusion with continuous renal replacement therapy (CRRT) in treatment of patients with severe tetramethylene disulfotetramine poisoning

Objective To study the effects of diazepam and sodium valproate infusion with CRRT in treatment of patients with tetramethylene disulfotetramine poisoning. Methods Diazepam and sodium valproate were continuously infused to patients by infusion pump. Meanwhile, CRRT was used to eliminate tetramethylene disulfotetramine. Results Convulsion was effectively controlled within one hour in 7 patients. After treatment with CRRT for 1 to 4 times, all patients recovered. Conclusion CRRT with diazepam and sodium valproate infusion were effective in patients with tetramethylene disulfotetramine poisoning.
10.Relationship between myocardial perfusion impairment and dysfunction in diabetic rats using myocardial contrast echocardiography and velocity vector imaging

Objective To investigate whether myocardial dysfunction and perfusion impairment had happened in diabetes mellitus(DM)rats,and to assess the relationship between them by using myocardial contrast echocardiography(MCE)and velocity vector imaging(VVI).Methods MCE and VVI were performed from the short-axis views of the mid-left ventricular level both at rest and after dipyridamole stress in control rats and DM rafs(12 weeks after induction with streptozotocin).MCE-derived myocardial blood flow(MBF)and myocardial flow reserve(MFR)and VVI-derived circumferential strain rate(SRc)and SRc reserve were obtained.Results SRc(absolute value)and MBF in the DM group were significantly lower than those in the control group at rest(P =0.03 for SRc and P =0.005 for MBF).SRc reserve and MFR in the DM group were significantly lower than those in the control group after dipyridamole stress (P =0.000 for SRc reserve and P =0.014 for MFR).There was no significant correlation between SRc and MBF at rest in the DM group(r =0.189,P =0.453).However,significant negative correlation existed between SRc reserve and MFR(r =-0.658,P =0.003).Conclusions Both myocardial systolic function and perfusion are impaired in DM rats.The decrease of MBF might not be the predominant determinant of myocardial systolic dysfunction at rest.However,the impairment of MFR could be an important contributor to the decrease of myocardial contractile reserve.
