2.In vitro activity of three kinds of azole antifungal agents against Malassezia species

4.Clinical research of sentinel lymph node imaging for contrast-enhanced ultrasonography in colorectal cancer during operation

Objective To study the feasibility of contrast-enhanced ultrasonography (CEUS) in detecting sentinel lymph node(SLN) in patients with colorectal cancer during operation and their clinical significance.Methods Twenty-one patients who underwent radical resection of a primary colorectal cancer were enrolled.During operation,1 % methylene blue was injected around the tumor,SLN mapping was performed.Then the SonoVue was injected at the same sites,CEUS was performed to observe changes of SLN in real time,and marked them.The SLN detection rate and accuracy of CEUS were compared to those of conventional dye guided method.Results The rectal cancer of below peritoneal reflex,the detection rate of CEUS for SLN was 92.31 %,and the accuracy was 88.89%.The detection rate of dye guided method for SLN was 84.62%,and the accuracy was 93.10%,there was no statistically significant difference between CEUS and dye guided method (P >0.05).Conclusions When injected SonoVue in submucosal layer,the good mapping of SLN of patients with rectal cancer of below peritoneal reflex can be obtained,it will play an important role in clinical application to map SLN of patients with rectal cancer of below peritoneal reflex.
6.Efficacy of itraconazole pulse therapy in treatment of onychomycosis:a multicentre open study

Objective:To observe the efficacy and safety of itraconazole in treatment of onychomycosis and its influence on nail growth speed. Methods: In a multicentre open study involving 15 medical units nationwide, patients with onychomycosis were treated with orally itraconazole pulse therapy. At each stage after treatment, the clinical efficacy,mycological efficacy, and side-effects of the therapy and the net growth length of normal nail deck were observed.Results: The therapy had a satisfactory efficacy and prolonged effect in treating onychomycosis for both affected fingers and toe nails. The therapy was effective for onychomycosis infected with dermatophyton, yeast or non-dermatophyton fungus and had a fungus-eliminating-rate of 97.86%. The average net growth length of normal nail deck in affected toe nails was slightly longer than that in finger nails on 6 months and 9 months after treatment.No severe side effects were found. Conclusion: Itraconazole pulse therapy is effective and safe for onychomychosis and can increase nail growth length,especially for toe nails.
7.Prospect of development of military dermatology

Dermatology is closely related to the health of officers and soldiers both in peace time and war time. Under special circumstances, skin diseases have a direct impact on efficiency and combat effectiveness of the military. Research work on fungal infections and psoriasis in the PLA has been playing a leading role domestically, as well as reaching international advanced standards. In the past 20 years, PLA dermatologists had been granted 32 second awards of Army Science and Technology Advancement Prize. Among them, the results of a series of research on fungal infections accomplished by the Second Military University was granted a second award of the State Science and Technology Advancement Prize. In the past 5 years, the team of dermatology profession has been growing rapidly in the PLA. Up to now, there are 6 departments with 11 tutors who are entitled to train postgraduate students seeking doctorate degree, and about 100 of them have succeeded in thesis defense and conferred the doctoral degree. However, at present, there are still many problems unsolved and further research is necessary. The most important task is, under the guide of bio-physic-social medical modality, various sophisticated technologies should be applied in basic research, prevention, diagnosis, treatment etc. in the field of dermatology, in order to assure the physical and psychological health of the servicemen.
8.Research on multiple-drug resistance expression of CNE1 and CNE2 cell lines induced by radiation

Objective To study multiple-drug resistance expression of CNE1 and CNE2 cell lines induced by radiation and its reversal by cyclosporine A and interferon. Methods Drug-resistant CNE1 and CNE2 cell lines were induced by radiation, and expression of P-gp, MRP, TOPⅡ and GST-? on protein and mRNA level were analyzed by flow cytometry and reverse transcription-polymerase chain reaction methods before and after radiation. At the same time, effect of cyclosporine A and interferon on multiple-drug resistance expression induced by radiation was studied. Results The results showed that expression of P-gp, MRP and GST-? proteins on CNE1 and CNE2 cell lines were up-regulated by radiation, and cyclosporine A and interferon could reverse the up-regulation of P-gp and MRP proteins induced by radiation. Conclusion Radiation can induce multiple-drug resistance in CNE1 and CNE2 cell lines, and it can be reversed by cyclosporine A and interferon.
9.Trichophyton rubrum: relationship between susceptibilities to antifungal agents and species specificities

Objective:To study the relationship between susceptibilities of Trichophyton rubrum strains to antifungal agents and their species specificities.Methods: The susceptibilities of Trichophyton rubrum strains to itraconazole,ketoconazole,fluconazole,terbinafine,naftifine,5-flucytosine and amphotericin B were evaluated using a modified microdilution method.The relationship between susceptibilities and genotypes and phenotypes of Trichophyton rubrum strains with different origins was analyzed by random amplified polymorphic DNA(RAPD).Results: The Trichophyton rubrum strains showed narrow minimum inhibitory concentration(MIC) ranges to terbinafine(0.016-0.032 ?g/ml),naftifine(0.032-0.063 ?g/ml) and itraconazole(0.25-1 ?g/ml),whereas they showed broader MIC ranges to ketoconzole(0.25-2 ?g/ml)and fluconazole(1-32 ?g/ml).MICs of Trichophyton rubrum strains to terbinafine(M_0=0.032 ?g/ml) and naftifine(M_0=0.032 ?g/ml) were the lowest among 7 antifungal agents.Wilcoxon test(Kruskal-Wallis test) suggested that there was no significant relationship of MICs to terbinafine,naftifine,itraconazole and amphotericin B with the genotypes,phenotypes and origins of the Trichophyton rubrum strains.Conclusion: The antifungal susceptibility of Trichophyton rubrum strains may not be related to their genotypes,phenotypes or from which part of the body they are isolated.
10.Effects of 3 carbohydrates on CAP10 promoter activity of Cryptococcus neoformans capsule-associated gene

Objective:To study the effects of glucose,mannose and galactose on CAP10 promoter activity of Cryptococcus neoformans capsule-associated gene.Methods: Yeast cells were transfected with plasmid containing a 951 bp length of 5′ upstream flanking sequence of CAP10 coding region and a reporter gene,chloramphenicol acetyl transferase(CAT);the transfectants were treated with different concentrations(10,20,40,60,80 mg/ml) of glucose,mannose and galactose.CAT activity was assessed by ELISA method and CAT activities of different groups were compared.Results: Different concentrations of glucose and mannose had no obvious influence on CAT activity;different concentrations of galactose had obvious influence on CAT activity and the influence was positively dependent with its dose within the experimental concentration range.Conclusion: Glucose and mannose have no obvious effect on the activity of CAP10 promoter;galactose has obvious inductive effect on activity of CAP10 promoter,suggesting that CAP10 gene might be related with galactose metabolism.
