2.Preliminary study on lumbar facet joint derived inflammatory factors in degenerative lumbar spinal canal stenosis

[Objective]To initially approach the role of lumbar facet joint derived inflammatory factors in degenerative lumbar spinal canal stenosis. [Methods]Totally 75 cases of degenerative lumbar spinal canal stenosis(LSCS)(n=41)and lumbar intervertebral disc herniation(LDH)(n=34) undergoing posterior lumbar spinal surgery in our department were evaluated in terms of the extent of degenerative arthrosis according to the Weishaup grading criteria.The grading of backleg pain,melosalgia and functional impairment were recorded.The excised lumbar facet joints were collected as species.The content of interleukin-1? and tumor necrosis factor-? in the species were determined by ELISA.[Results]There was no TNF-? detected in both of the two groups.More IL-1? was detected in degenerative lumbar spinal canal stenosis group than that in lumbar intervertebral disc herniation group.It was demonstrated that the content of IL-1? in the species increased as the degeneration of lumbar facet joint sharpened.IL-1?-positive cases in degenerative lumbar spinal canal stenosis group showed higher grading of backleg pain,melosalgia and functional impairment.[Conclusion]The cartilage of degenerative lumbar spinal canal produced more IL-1?.Lumbar facet joint derived inflammatory factors might be one of the reasons that cause backleg pain and melosalgia and functional impairment in degenerative lumbar spinal canal stenosis patients.
3.Effect of Yangfei Huoxue Prescription on ERK1/2 and NF ?-B of Bleomycin-induced Pulmonary Fibrosis in Rats

Objective To study the effect of Yangfei Huoxue Prescription on ERK1/2 and NFK-B of bleomycin-induced pulmonary fibrosis in rats. Methods All of rats were randomly divided into seven groups and were intratracheally infused with bleomycin to build the model. Sham operation group and model group were given 5‰ CMC, dexamethasone group was given dexarnethasone solution, and each treatment group was given corresponding Chinese medicine mixture. After histological process, the expression of ERK1/2 and NFK-B were examined by SABC immunohistochemical method. 4.The influence of statins on the blood inflammatory factors and fatty acid of erythrocyte membrane inhypercholesterolemic patients.

3.64 mmol/L)received 8-week double blind comparisons treatment.After treatment we randomly selected 15 patients for each group.The fasting serum lipid was measured at the end of 4-week and 8-week,and the serum hs-CRP and IL-6 were measured too.Meanwhile seven hypercholesterolemic patients from rosuvastatin group and seven ortholiposis out-patients were included.These patients were treated with statins for 4 weeks.The erythrocytes before and after treatment were kept to be measured the FA.Results (1)Compared with the pretreatment,the level of LDL-C,TC,hs-CRP remarkably reduced in two treatment groups at the end of 4-week therapy.These inflammatory parameters did not further reduced,but hs-CRP reduced further at the end of 8-week therapy.(2)The percent age of SFA was obviously higher in hypercholesterolemic group than ortholiposis group.On the other hand,PUFA and ?-3 FA were obviously lower.After 4-week treatment the percent age of SFA reduced,while PUFA and ?-3 FA increased significantly in hypercholesterolemic group.Conclusion (1)Statins reducing inflammatory factors is independent of lipid-modifying ability.(2)Rosuvastatin treatment can effectively reduce the level of LDL-C and TC,and no significant difference in the effect is observed between two treatment groups.(3)The erythrocyte membrane FA proportion was obviously different in hypercholesterolemic group from ortholiposis group.Statins treatment could make the proportion of FA in erythrocyte membrane restore normal.
5. Quantitative evaluation of baicalin Radix Scutellariae granules by near-infrared spectroscopy

Objective To determine the content of baicalin in Radix Scutellariae granule from different manufacturers using near infrared spectroscopy(NIRS)technology. Methods Utilizing NIRS combined with partial least squares, and simultaneously optimizing the pretreatment methods and the range of spectrum, the quantitative model of baicalin in Radix Scutellarie granules was established. Results In the model correlation coefficient(R2)was 0.9702, root-mean-square error of calibration(RMSEC)was 0.555, and root-mean-square error of predict(RMSEP)was 1.05. Conclusion In this paper, rapid quality analysis method of Radix Scutellariae granules from different manufacturers is studied using NIRS technology. The model can predict the content of baicalin nondestructively and rapidly. It can give some reference for researching the quality control of Radix Scutellariae granules.
7.Application of free-form deformation algorithm in fast three-dimensional bone reconstruction.

This paper presents a unit free-form deformation (FFD) method applied to rapid three-dimensioanl (3D) bone reconstruction, which was based on traditional FFD. With the femur as an example, we reconstructed a 3D model of femur from two X-ray images and a standardized model by taking advantage of unit FFD algorithm. The X-ray images and its parameters were taken by C-arm device. Those parameters and X-ray contour are contributed to 3D reconstruction. The out contours of X-ray image and standard model were connected by point matching algorithm. The unit-FFD lattice was built to reconstruct standard model and finally made the contour of X-ray image and standard model exactly the same. Experiments on shape accuracy, robustness and time consuming, carried out by 35 specimen from cadaver, showed that mean error of shape (0. 52 mm) and mean construction time (112 s) were lower than those using traditional method (0.7-2.6 mm, 8-20 min). The method proposed in this paper shows a good prospect in clinical application and related research.

9.The value of gemstone spectral imaging in reducing metallic implants artifacts

Objective To evaluate the value of gemstone spectral imaging (GSI)and multi artifact reduction system(MARs)in re-ducing metallic implants artifacts,and find the best monochromatic images.Methods Twenty-one patients with metal implants in our hospital receievd GSI examination ,MARS reconstruction and polychromatic images(140 kVp).Eleven groups of monochromatic and MARS images were obtained from 40 keV to 140 keV every interval of 10 keV by the special GSI viewer ,respetively.Subjective image quality of two methods were assessed by 3 radiologists in consensus.For every patient one group monochromatic images and blend enery images were selected two heaviest artifacts level and no artifacts level,the CT value and the standard deviations(SD)value of the region of interest(ROI)were meatured of artifacts.Contrast-to-noise ratio(CNR)and artifacts index(AI)were also caculated and were analyzed with paired-t test.Results There was significant statistical differences of CNR between energy spectral images and blend energy images in energy level of 40 to 120 keV(P <0.05),while there was no difference in energy level of 130 to 140 keV (P >0.05),and the optimal CNR was acquired at the energy level of 70 keV(CNR=8.75±3.28).The value of AI for two mehtods has no difference in energy level scope 40 to 60 keV(P >0.05),there was significant statistical differences of AI between energy spectral images and blend energy images in energy level of 70 to 140 keV(P <0.05),and the lowest of AI(AI=26.95±9.64)was ac-quired at 1 10 keV images.Conclusion The energy spectral CT possess superiority in decreasing the artifacts and will provide high image quality in the metal implants.Gemstone spectral CT has great significance for postoperative patients with metal implants in imaging diagnosis.
10.Effect of α-GalCer-activated natural killer T cell on survival of allograft with high-risk rejection after retrobubar injection

BackgroundCorneal graft reject is a major cause of corneal transplantation failure.Although many immune-suppressing drugs have been utilized to reduce the reject response,their adverse effects on organ and tissue are still insoluble.The tolerance induction of natural killer T (NKT) cells is currently under investigation.However,the study on the application of NKT cells in high risk corneal transplantation is seldom.ObjectiveThe present study was to explore the effects of α-GalCer-activated NKT cella on allografts survival after high-risk corneal transplantation surgery via retro-bubar injection.Methods The lymphocytes were picked up from the spleen of SPF Lewis rats and cultured in RPMI 1640 medium with 100 mg/L α-GalCer.After one week,NKT cells were sorted by the FACSVantage system as CD161+ TCR-α+ cell from the lymphocytes with the cell densities 5×106/ml.Ten SPF Fisher344 rats were used to prepare the donor corneas,and 20 Lewis rats served as recipients.The high risk corneal transplantation models were created by corneal suturing in 20 recipient rats.Penetrating keratoplasty (PKP) was performed in the model rats.0.1 ml NKT cells or the same volume of normal saline solution were retro-bubarly injected at the end of surgery respectively.The corneal allografts were observed and scored based on Holland criteria at the three-day interval under the slit lamp for 30 days.Two weeks after surgery,three rats from each group were sacrificed by excessive anesthesia method and the eyeballs were obtained for histopathological examination.The inflammatory cell infiltration ( CD4+ and CD8+ ) in grafts was evaluated by immunochemistry and flow cytometry.The use of the animals complied with the Statement of ARVO.ResultsThe mean survival time of the allografts was (7.90± 1.37) days in normal saline solution group and (14.70± 1.49) days in NKT cell group,showing a statistically significant difference between the two groups ( t =10.61,P =0.00 ).Two weeks after surgery,all the allografts showed the severe opacity with lots of new blood vessels and edema in normal saline solution group.However,the corneal grafts were clear in NKT cell group.Abundant CD4+ and CD8+T lymphocytes were seen in the allografts in normal saline solution group,but the inflammatory cells were obviously less in NKT cell group.The percentage of NKT cells in the spleen was (5.67±0.25)% in NKT cell group and ( 1.21±0.19)% in normal saline solution group ( t =8.43,P =0.00 ).Conclusionsα-GalCer-activated NKT cells can prolong the survival time of allografts in high-risk corneal transplantation.Retro-bubar injection of α-GalCer-activated NKT cells probably is a new approach to the prevention of the rejection of corneal transplantation.
