1.Effect of IL-21 on Treg cells in peripheral blood mononuclear cells in pathogenesis of Graves disease

Objective:To explore the role of IL-21 in regulating the expression of peripheral blood Treg cells in the pathogenesis of Graves disease ( GD ) . Methods: Electrochemical luminescence detection was used to determine levels of thyroid function indexes and autoantibodies. Peripheral blood mononuclear cells (PBMCs) were extracted,then,cultured in the presence or absence of IL-21 in vitro. The level of IL-10 protein was measured by enzyme-linked immunosorbent assay ( ELISA) ,and expressions of Foxp3 and IL-10 mRNA were examined by Real-time PCR. Results: Compared to eGD and control group, there were significant differences in the levels of thyroid function indexes in GD group (P<0. 05),whereas there was no difference between control group and eGD group (P>0. 05). Before IL-21 stimulation,compared to eGD and control groups,expressions of Foxp3 and IL-10 mRNA and level of IL-10 protein were significantly higher in GD group ( P<0. 05 );but there were no significant differences between eGD group and control group (P>0. 05). After IL-21 stimulation,expressions of Foxp3,IL-10 mRNA and IL-10 protein levels were decreased markedly in all three groups,among which GD group showed the greatest change(P<0. 05). Conclusion:IL-21 may inhibit the differentiation of Treg cells and the production of IL-10,therefore decreasing the number of Treg cells and the ability of suppressing effector T cells ,thus should be involved in the pathogenesis of GD.
2.Inhibitory Effects of Astragalosides on Proliferation of Lung Adenocarcinoma SPCA-1 Cells

OBJECTIVE:To investigate the inhibitory effect of astragalosides on proliferation of lung adenocarcinoma SPCA-1 cells. METHODS:After the cells were cultured in 0 (blank control),7.8,15.6,31.2,62.5,125.0 and 250.0 μg/ml for 48 h, MTT method was used to determine cell viability and median inhibitory concentration(IC50)was calculated,and terminal deoxynu-cleotidyl transferase-mediated dUTP-biotin nick end labeling assay(TUNEL)and flow cytometry were employed to detect apopto-sis. RESULTS:Compared to the blank control,after the cells were cultured in 7.8,15.6,31.2,62.5,125.0 and 250.0 μg/ml as-tragalosides for 48 h,the cell viability was poorer(P<0.01),and IC50 was 61.75 μg/ml;after the cells were cultured in 15.6,31.2, 62.5,125.0 and 250.0 μg/ml astragalosides for 48 h,the apoptosis rate was higher. CONCLUSIONS:Astragalosides have an anti-tumor effect to some degree by a mechanism which may be related to inhibiting cancer cell proliferation and accelerating apoptosis.
3.THE ROLE OF TGF?2 IN MYOCARDIALIZATION OF CARDIAC PROXIMAL OUTFLOW TRACT SEPTUM IN MICE

Objective To investigate the expression of TGF?2 in proximal OFT septum in connexin(Cx) 43 knockout(KO) mice and illustrate its relationship with myocardialization and reveal whether exogenous TGF?2 could promote myocardialization in vitro.Methods Objects were C57/BL6 mice of E12.5 to E15.5 by the mating of 2 month old Cx43 heterozygous mice,which included Cx43 homozygotes(Cx43-/-),heterozygotes(Cx43+/-) and wild-types(Cx43+/+) genotyped by PCR method.?-sarcomeric actin(?-SCA) and TGF?2 were detected by immunohistochemistry.TGF?2 with 3 different dosages was used to be the intervent added to the heart culture system in which the wild-type hearts of E12.5 were cultured till E15.5 before detecting the expression of ?-SCA by immunohistochemistry.Results The expression of ?-SCA in the proximal OFT septum was delayed obviously in Cx43-/-predominantly at E13.5 and E14.5.As compared with the Cx43+/+,the expression of TGF?2 in proximal OFT septum decreased obviously in Cx43-/-mice predominantly at E14.5,and less obviously in(Cx43+/-) mice.None of the intervention groups showed the obvious promoted myocardialization.Conclusions Cx43 KO mice exhibited delayed myocardialization,and the decreased TGF?2 may involve in the abnormal myocardialization of Cx43 KO mice.Application of TGF?2 in vitro could not exactly mimic the in vivo expression pattern of TGF?2,or the process of myocardialization may require precise dosage of TGF?2 and stringency of cultured system.

5.Improved B-Lynch suture in the treatment of cesarean section refractory hemorrhage

Objective To explore the application value of improved B-Lynch suture in the treatment of cesarean section refractory hemorrhage.Methods To analyze retrospectively the information of cesarean section refractory hemorrhage patients who have taken conventional treatment or improved B-Lynch suture treatment in our hospital,taken the conventional treatment.the uterus was remove when it's invalid.the improved B-Lynch suture treatment was taken.Results In the improved B-Lynch suture treatment group,the patients have apprently little blood loss,shorter time in the cesarean section operation(t =8.58,6.38,P < 0.05).And none of the cases took the hysterectomy or occured DIC.In the conventional treatment group,the blood loss was more,the operation time was longer,and 6 cases took the hysterectomy,5 cases occured DIC.The two groups were significantly different(x2 =6.98,4.59,all P <0.05).Conclusion In the cesarean section,if refractory hemorrhage happen and the conventional measures does not work,the improved B-Lynch suture should be taken into consideration.It has good hemostatic effect.
7.Determination of oxygen consumption in water by continuous flow injection analysis using potassium permanganate

Objective:
To establish a continuous flow injection analysis with potassium permanganate for determination of oxygen consumption in water.
Methods:
The water samples and acid potassium permanganate working solutions were mixed using the continuous flow bubble spacing, and subjected to online heating reaction at 97 ℃. The peak height of the electrical signal of potassium permanganate was measured at the maximum absorption wavelength of 520 nm, and the standard substance, sulfuric acid concentration and potassium permanganate concentration were optimized according to the peak height of the electrical signal. The standard curve was plotted to measure the limit of detection, the limit of quantification, and spiked recovery rate of the method. The CODMn concentration was determined in 40 drinking water samples using acid potassium permanganate titration and continuous flow injection analysis using potassium permanganate, and the determination results of the two methods were compared with paired t-test.
Results:
Glucose was selected as the standard substance, and the mixture of 17.5% sulfuric acid and 3.2 mmol/L potassium permanganate was selected as the working solution. CODMn had a good linear relationship at concentrations of 0 to 6.00 mg/L, with a correlation coefficient of 0.999 and higher, a detection limit of 0.013 mg/L and a quantitation limit of 0.043 mg/L, respectively. The spiked recovery rates were 90.00% to 105.00% in 40 drinking water samples, with relative standard deviations of 0.12% to 1.36%. The determination results of two permanganate index standard substances were all within the range of standard values. The relative errors of CODMn concentration were 1.55% to 9.26% between the continuous flow injection analysis using potassium permanganate and acid potassium permanganate titration, and there was no significant difference (t=2.023, P=0.185).
Conclusion
The established continuous flow injection analysis with potassium permanganate is feasible for batch determination of oxygen consumption in water.
8.Efficiency of temporary cardiac pacing in the acute poisoning patients with serious bradycardia

Objective To explore the efficiency of temporary cardiac pacing on the acute poisoning patients with serious bradycardia.Methods Thirty-eight cases of the acute poisoning patients with serious bradycardia were treated randomly with temporary cardiac pacing (pacing group, 18 cases) and drug therapy (control group,20 cases) and followed with treatment for poisoning. Results Right ventricular pacing were successfully performed in 18 cases and the temporary cardiac pacemaker were placed. With the further treatment of poisoning, in the pacing group, the efficiency of anit-bradycardia and the poisoning cure rate was 100% and 88. 9% , respectively, whereas being 60% and 65.0% in the control group, respectively. The efficiency of anit-bradycardia and the poisoning cure rate were significantly different between the two groups (P < 0.01 or P < 0.05 ). Conclusions The treatment of temporary cardiac pacing has effect on the acute poisoning patients with serious bradycardi. It can effectively improve the rate of patients' resuscitation.

10.Effects of radiation from ~(188) Re on smooth muscle cell proliferation

AIM: Although endovascular radiotherapy inhibits neointimal hyperplasia, the exact alterations induced by ?-particles irradiation remain to be elucidated. The objective of this study was to investigate the ability and the cellular mechanism of local ?-particles emission from 188 Re to inhibit vascular smooth muscle cells (SMCs).METHODS: The SMCs in vitro were irradiated by 188 Re with single doses of 2.6 Gy-25.8 Gy. The effects of ?-particles on SMCs, such as effective irradiate doses, the period of inhibition for SMCs proliferation, the changes of cell proliferation rate and DNA synthesis rate, cell cycle progression and related gene expression, were investigated by cell count,-TdR incorporation, cell cycle progression analysis, cell viability and immunocytochemistry, respectivecy.RESULTS: ?-particles irradiation with dose of 5.2 Gy could inhibit significantly SMCs proliferation. At dose of 20.6 Gy DNA synthesis inhibitory rate was 92%, SMCs proliferation rate was only 3%. Renoval of 188 Re did not abolish the inhibitory effects of ?-particles on SMCs proliferation. The expression of P53 was up regulation and PCNA was down regulation after irradiation. CONCLUSION: ?-particles from 188 Re was significantly effective and permanent in inhibiting SMCs proliferation, and inhibitory effect was in dose-dependet manner ED50 was 5 Gy, the best dose to inhibit SMCs proliferation was 20 Gy. ?-particles irradiation induced SMCs to occur G 0/G 1 arrest, damaged the ability of SMCs reproliferation and led to cell clonogenic death. P53 and PCNA had regulatiory effects on SMCs proliferation after ?-particles irradiation.
