AIM:Toinvestigatetheassociationbetweenmethylationstatusofapoptosis-relatedgenesandche-mosensitivity in the lung adenocarcinoma cell line P 15.METHODS: Methylation-specific PCR was applied to detect the methylation status of p73, p14ARF, p16INK4a and bax genes of P15 cells in untreated control group and decitabine (DAC) treatment group.RT-PCR was used to detect the expression of p 73, bcl-xL, bad, bax, p14ARF and p16INK4a at mRNA level. Colony formation assay and cell growth inhibition assay were used to detect the sensitivity of P 15 cells to cis-diaminedichlo-roplatinum ( C-DDP) before and after DAC treatment .DAPI staining was used to determine the apoptosis of P 15 cells ex-posed to C-DDP before and after DAC treatment .RESULTS:p73, p16INK4a and bax were expressed in the methylation sta-tus.After DAC treatment, p16INK4a expression was decreased , and the expression of p73 and bax disappeared .The expres-sion of p73, p16INK4a and bax in the unmethylated status was weak , but the enhanced expression was observed following DAC treatment. After P15 cells were treated with DAC and C-DDP, the colony formation rate of the P15 cells was signifi-cantly decreased as compared with untreated control group .The apoptotic P15 cells in DAC+C-DDP treatment group were significantly higher than those in untreated control group (P<0.05).CONCLUSION:After treated with DAC, the sensi-tivity of P15 cells to C-DDP is increased due to the activation of silenced pro-apoptotic genes .DAC and C-DDP synergisti-cally promote tumor cell apoptosis .They have significant anti-tumor effect .

Journal of Integrative Medicine 2004;2(1):75-7
Parkinson's disease is a commonly encountered central nervous retrograde affection in elder persons. According to the theories of traditional Chinese medicine, Parkinson's disease is characterized by deficiency in the Ben (root) and excess in the Biao (branch). The Ben (root) is insufficiency of liver and kidney and deficiency of qi and blood; and the Biao (branch) is wind, fire, phlegm and stasis. Good therapeutic effects have been obtained by treatment based on syndrome differentiation, treatment with specific prescriptions and acupuncture. The further study on literatures, standardized criterion of symptoms differentiation and therapeutic effect determination as well as the screening and further research on the effective prescriptions and herbs should be paid more attention, meanwhile, the integration of herbs and acupuncture will be conducive to raise the therapeutic effect.

Objective To study the effect of rhu-IFN-? on immune state and vertical transmission in pregnant mice infected with Toxoplasma gondii. Methods Sixty pregnant BALB/c mice were randomly divided into three groups: a control group, infected group and treatment group. In the infected group and treatment group, each mouse was injected with T.gondii tachyzoites peritoneally on the day 8 of gestation. In the treatment group, each mouse was treated with 1 000 U rhu-IFN-? on the day 7,8,9 of gestation. Blood was collected from the tail veins of all the mice on the day 10,12 of gestation. The levels of blood CD4+ and CD8+ T cells were detected by flow cytometry. Meanwhile,on the day 12 of gestation, all mice were anatomized to observe live embryo rate and the infection status in fetal brain tissue. Results Compared with the control group, the levels of CD4+ T cells in the infected group and treatment group were low,and the levels of CD8+ T cells high on the day 10,12 of gestation,so the ratio of CD4+/CD8+ was inversed. However, compared with the infected group, the levels of CD4+ T cells in the treatment group were high and the levels of CD8+ T cells low on the day 10,12 of gestation,so the ratios of CD4+/CD8+ were high on the day 10,12 of gestation. Meanwhile, the live embryo rate was high and the infection rate of intrauterine embryonic low. Conclusion A proper dose of rhu-IFN-? could improve the function of immunity system and reduce the vertical transmission probability in pregnant mice infected with T.gondii.
