Objective:To investigate the mechanism of immunoglobulin Y antibodies(IgY) against tumour necrosis factor alpha(TNF-α) and Interleukin-1beta(IL-1β) in treating allergic bronchial asthma through nebulization inhalation. Methods:The allergic bronchial asthma model was established with Hartley guinea pig by ovalbumin nebulizating inhalation. The animals were randomly divided into 4 groups: normal control group(group A),allergic bronchial asthma group(group B),0.1% anti-TNF-α and IL-1β IgY treating group(group C),1.0% anti-TNF-α and IL-1β IgY treating group (group D).The animals were killed after treatment being accomplished for 2 h,4 h,8 h,24 h and the lungs were made pathological, which were then stained by hematoxylin-eosin(H.E.).The bronchoalveolar lavage fluid (BALF) was collected and the deposited cells were stained by Wright's.Results:①The histological appearance of lung: In group B the histological structure of alveolar ducts and alveolar walls was damaged, the alveolar space was full of transudate and lots of alveolar epithelial cells and leucocytes. The pulmonary interstitial edema, inflammatory cells infiltration, distorted or dilated capillaries and reducing capillary numbers of effective blood stream were observed in alveolar walls. In group C and group D the damage degree of alveolar ducts and alveolar wall was slighter than that in group B and there were few inflammatory cells in alveolar space. In bronchial lumen and pulmonary alveoli the sticky mucus plug was obviously less in group C and group D than in group B.Moreover, inflammatory cell infiltration was seldom observed aroud bronchia, and restoration of bronchial tunica mucosa epithelium was obviously observed in group C and group D.②The cytology appearance of BALF: In group C and group D the numbers of eosinophils, neutrophils, lymphocytes were significantly fewer (2 h,4 h,8 h,P<0.05),however,the number of macrophage was significantly more(2 h,4 h,8 h,P<0.05)than in group B.Conclusion:The anti-TNF-α and IL-1β IgY can obviously alleviate pathological extent of inflammatory reaction in allergic bronchial asthma of guinea pigs by nebulization inhalation therapy. The therapeutic effect of anti-TNF-α and IL-1β IgY between 0.1% and 1.0% concentration is not obvious difference for pathology changes.

AIM: To investigate the anti-DIC effect of the domestic recombinant hirudin (rH) in order to provide preclinical pharmacological basis for its new drug application. METHODS: DIC was induced by constant iv infusion of thrombin (100 U?kg -1 ?h -1 ?1 h) plus PAMBA (50 mg?kg -1 ?h -1 ?1 h). Platelet count (PLT), PT and fibrinogen (FIB) determination as well as plasmaprotamine paracoagulation (3P) test served as diagnostic indexes of DIC, while pathlogical examination of microthrombosis of lungs and kidneys was followed. RESULTS: rH 125,62.5,31.25 and 15.625 ?g?kg -1 iv. gtt demonstrated a dose-dependant inhibitory effect on DIC. The inhibitory effects on DIC of rH(%) were 89.38%, 52.60%, 49.71% and 25.58% for PT, and 100.01% , 61.86%, 58.40% and 32.68% for PLT, and 85.22%, 73.01%, 33.60% and 14.86% for FIB respectively at the descending order of rH doses. CONCLUSION: Domestic rH being tested has powerful anti-DIC effect which is comparable with refludan—an r-hirudin product approved by European Community for marketing.

Objective To optimize the extraction procedures of compound Three-Flower-Tea (CTFT) by orthogonal design. Methods After having established the methods of quantifying the chlorogenic acid by TLC-UV, the orthogonal design was used to select the best extraction procedures, using chlorogenic acid content and extract yield as the measuring index, through controlling four different factors, naming extraction temperature (A), extraction time (B), watervolume added (C) and extraction number of times (D), and three levels at each factor. Result The effective factors which influenced the extraction procedures were A, C and D. Conclusion The optimum extraction procedures for CTFT should be 15-fold water volume, three-time decocting with 20 minutes per time at temperature of 100 ℃.

1. The subnucleus caudalis of the spinal trigeminal nucleus of cat was injected with 30% HRP (Sigma, type Ⅵ). Well localized groups of labeled neurons were observed in the dorsolateral horn of the contralateral red nucleus. This dorsolateral horn, being present in the rostral and middle thirds of the nucleus, consisted mainly of middle-sized and small neurons.2. Injections were also made into the second and third segments of the cervical spinal cord and the caudal part of the bulbar reticular formation. The labeled neurons appearing in these experiments were diffusely distributed in the red nucleus, yet limited in the rostral third of it.3. Summarizing our findings and the data from the literature, we got the impression that the rostral two thirds of the red nucleus gave projections to the central structures rostral to the upper cervical spinal cord, and the caudal two thirds of it to those caudal to the lower cervical cord, which indicating a somatotopic organization of the red nucleus neurons giving descending projection fibers.

Acta Anatomica Sinica 1955;0(03):-
HRP solution was injected into the right subnucleus caudalis of the trigeminal nerve in the cat, and the labeled cells in the nucleus raphe dorsalis of six cases were examined and analyzed. The labeled cells appeared more in the middle portion and its rostral adjacent parts of the nucleus raphe dorsalis, decreased in number rostrally and caudally, and were entirely absont in the caudal end of the nucleus. In the ventral part and the ventral areas of the lateral parts of the nucleus labeled cells were more than in the other areas of the nucleus, no difference was found between the two sides. In the dorsal part and the dorsal regions of the lateral parts of the nucleus only a few labeled cells were seen. The labeled cells varied in forms, but in the ventral part and the ventral regions of the lateral parts they were mainly fusiform in shape. In addition, in nucleus raphe magnus, nucleus raphe pallidus, and nucleus raphe obscurus, small numbers of neurones were labeled as well.As the nucleus raphe magnus takes part in the inhibition of the response to noxious stimuli in the spinal dorsal horn, and as both the nucleus raphe magnus and nucleus raphe dorsalis belong to the more developed raphe nuclei and contain more 5-HT neurones, it seems that there might be somatotopic relations between the nucleus raphe dorsalis which projects mainly to subnucleus caudalis and the nucleus raphe magnus which projects mainly to the spinal dorsal horn.
