1.Concurrent PCF chemotherapy plus radiotherapy for middle and advanced cervical cancer

Background and purpose:Cervix cancer is the second most common cancer in gynecologic malignant tumors. Chemotherapy has been used more and more lately, and concurrent chemo-radiotherapy are receiving increasing attention. In this study, we investigated the clinical effect of concurrent PCF chemotherapy plus radiotherapy for moderate and advanced cervical cancer. Methods:One hundred and ninty-six patients with moderate and advanced cervical cancer were randomly divided into two groups: only radiotherapy group and chemo-radiotherapy group (98 cases for each group). Two groups underwent the same fractionation irradiation, the chemo-radiotherapy group was given PCF regimen that consisted of (DDP) at the dose of 20 mg daily intravenously for 1-5 days, (5-FU) at the dose of 500 mg daily for 1,3,5 days and (CTX) at the dose of 400 mg daily for 2,4 day for 2-4 cycles. Both groups received radiotherapy with 30 Gy first and were irradiated by the combination of external radiotherapy plus intracavitary irradiation.Results:The immediate response rates of the two groups were 100 %, there was no statistical difference. 2.Atherogenic index of plasma and dorsalis pedis artery atherosclerosis

Objective To investigate the relationship between Atherogenic Index of Plasma(AIP) and the dorsalis pedis artery atherosclerosis in type 2 diabetes. Methods One hundred and twenty-seven patients with type 2 diabetes and 33 healthy individuals were included in the study. The diabetic patients were devided into 2 groups, with( n = 66) or without ( n = 61 ) atherosclerosis. The intima media thickness (IMT) of dorsalis pedis artery were measured by B-mode Doppler ultrasonography. The triglyceride(TG) ,high density lipoproteincholesterol(HDL-C) and other glucolipid metabolic indices were measured in all patients. The AIP was defined as the log value of TG/HDL-C ratio. The one-way ANOVA test was performed to determine the differences among three groups. The relationships between IMT and the other indicators were assessed using Pearson bivariate correlations analysis. The optimal operating point of AIP to diagnose the dorsalis pedis artery atherosclerosis in type 2 diabetes was obtained by drawing ROC curve. Results The AIP value in atherosclerosis diabetic patients,no-atherosclerosis diabetic patients and healthy subjects were 0. 229 ± 0. 132,0. 112 ± 0. 074 and -0. 045 ± 0. 033 respectively. The IMT value were (0. 71 ± 0. 24 ) mm, (0. 49 ± 0. 09 ) mm and (0. 34 ± 0. 15 ) mm respectively. Both indices shown statistical differences among the three groups (Ps = 0. 001 respectively). After adjusting for potential influence factors, AIP showed positive correlation with IMT( r = 0. 315, P = 0. 001 ). The OOP of AIP was 0. 1671 ,the area under the curve was 0. 783. Conclusion AIP was well related to the dorsalis pedis artery atherosclerosis in type 2 diabetes,and the OOP of AIP had good diagnostic reference value in these patients.
3.Expression of the estrogen receptor ? and ? in mouse thymus

Objective:To detect estrogen receptor ? and ?(ER-?,ER-?) protein expression in different age of mouse thymus.Methods:Protein expression of ER-? and ER-? in thymus was analyzed via immunohistochemistry.Moreover,the relationship between ER-? and cytokeratin 18(epithelial cell marker) was further tested through fluorescence double-staining.Results:Immunohistochemical analysis revealed that both ER-? and ? protein was found in nuclei of some thymocytes of 3 month mice.However,expression of ER-? was absence while ER-? was still positive in aged mice,such as 12 months and 16 months old.Double staining further confirmed that lots of ER-?/? positive cells were Foxp3 negative cells.Conclusion:Expression of ER-? is absent while ER-? is still positive in thymus of aged mice,which indicates ER-? is the critical estrogen receptor that involves in thymic involution.Moreover,ER-?/? do not participate in Treg development within thymus.
4.Evaluation of Cica-Beta Test Kit for Detection of Metallo-β-Lactamase-Producing Pseudomonas Aeruginosa

Objective To evaluate Cica-Beta Test kit for detection of metallo-β-lactamase-producing Pseudomonas aeruginosa (PAE)in the clinical microbiology laboratory.Methods A total of 82 imipenem-resistant PAE clinical isolates from litera-ture[5]was dectected to metallo-β-lactamase (MBLs)by PAE-MHT and Cica-Beta Test kit.Results The sensitivity,speci-ficity and accuracy rate of PAE-MHT was 84.6%,97.2% and 97.6%,and the sensitivity,specificity and accuracy rate of Cica-Beta Test kit was 76.9%,100% and 96.3%,respectively.Two methods had a good consistency.Conclusion Two methods are simple,quick for detecting to metallo-β-lactamase-producing Pseudomonas in clinical laboratories.
5.Clinical observation for low-dose azithromycin with tiotropium bromide in the treatment of stable chronic obstructive pulmonary disease

Objective To study the efficacy and safety of low-dose azithromycin with tiotropium bromide in the treatment of stable chronic obstructive pulmonary disease.Methods A hundred and ten patients were randomized into three groups:tiotropium bromide group (36 cases,group A),azithromycin with tiotropium bromide group(38 cases,group B)and control group(36 cases,group C).Patients in group A were given tiotropium bromide (18 μg,q,d )in addition to conventional treatment.The patients in group B were given lowdose azithromycin (250 mg,twice a week) in combined with tiotropium bromide.The patients in control group were given the conventional treatment only.The courses of treatment lasted for six months.Results Compared with the control group,the frequency of acute exacerbation in patients treated with azithromycin and tiotropium bromide was reduced remarkably ( 2.1 ± 0.6 and 4.9 ± 0.7,t =18.5061,P < 0.05 ).The severity of clinic symptoms ( Cough 1.3 ± 0.5 vs.2.2 ± 0.6,P < 0.05 ),expectoration ( 1.0 ± 0.2 vs.1.7 ± 0.3,P < 0.05 ),anhelation ( 1.5 ± 0.8 vs.2.1 ± 0.6,t =3.6342,P < 0.001 ) ],life quality ( 29 ± 8 vs.42 ± 11,P < 0.05 ) and six-minutes walking distance( [ 370.00 ± 14.26 ] m vs.[ 290.00 ± 12.85 ] m,P < 0.05 ) of the azithromycin with tiotropium bromide group were improved significantly when compared with control.Compared with the tiotropium bromide group,the frequency of acute exacerbation ( 2.1 ± 0.6 vs.3.2 ± 0.8,P < 0.05 ),the severity of clinic symptoms (Cough 1.3 ±0.5 vs.1.8 ±0.4,P<0.05),expectoration( 1.0 ±0.2 vs.1.3 ±0.3,P <0.05) and anhelation( 1.5 ±0.8 vs.1.9 ± 0.6,P < 0.05 ) ],life quality ( 29 ± 8 vs.36 ± 10,P < 0.05 ) and six-minutes walking distance ( [ 370.00 ± 14.26 ] m vs.[ 330.00 ± 13.76 ] m,P < 0.05 ) were improved over those of tiotropium bromide group.Conclusion The long-term low-dose azithromycin in combinned with tiotropium bromide is good and safe in treating stable COPD.Therefore,it is worth of further clinical evaluation.

7.Glucocorticoids-induced osteonecrosis of the femoral head:adipogenic differentiation and treatment progress

BACKGROUND:Glucocorticoids may induce local bone trabecular and bone marrow necrosis, femoral head col apse and deformation, thus resulting in hip dysfunction. However the pathological mechanisms and treatment of glucocorticoids-induced osteonecrosis of the femoral head remain unclear, and the pathogenesis mechanisms are controversial. The current studies focus on the understanding of the pathological mechanisms.
OBJECTIVE:To summary the research progress of adipogenic differentiation theory of glucocorticoids-induced osteonecrosis of the femoral head and the treatment.
METHODS:The first author searched literature from CNKI and PubMed database from 1988 to 2010, by using the key words is“Glucocorticoids, osteonecrosis of femoral head, bone marrow stromal stem cells, adipogenic differentiation, differentiation factors, treatment, choices”in English, and“glucocorticoids induced osteonecrosis of femoral head, adipogenic differentiation, treatment, research progress”in Chinese. Articles regarding the adipogenic differentiation theory of glucocorticoids-induced osteonecrosis of the femoral head and the treatment were included.
RESULTS AND CONCLUSION:A total of 112 literatures were screened out, according to inclusion and exclusion criteria for literature screening, 54 articles were included. Modern researches emphasize the celland molecular biology level, and show that the biological base of glucocorticoids-induced osteonecrosis of the femoral head is abnormal adipogenic differentiation of bone cells, glucocorticoids cause the variations of adipogenic differentiation factors, leading to adipogenic differentiation of bone marrow stromal cells. But glucocorticoids affects multiple differentiation factors, it may cause great error in the evaluation of the pathogenesis of glucocorticoids-induced osteonecrosis of the femoral head purely from one factor. The abuse of glucocorticoids is the leading cause for the osteonecrosis of femoral head. Further studies are needed to investigate the pathogenesis of osteonecrosis of the femoral head and treatment programs.
8.Construction and expression of eukaryotic expression vector pcDNA3-ICOSIg

BACKGROUND:So far, inducible co-stimulator is the important costimulatory molecule family member. Inducible co-stimulator can promote the activation of T cel s proliferation and secretion, regulate Th1/Th2 cel polarization dependence, enhance B cel function which depend on the T cel s. So, blocking the inducible co-stimulator may result the inactivation and no reaction of cloning in T cel s, thus inducing the immune escape of tumor on the body.
OBJECTIVE:To build a plasmid expression of inducible co-stimulator Ig, in order to observe the expression in rat body.
METHODS:cDNA encoding the extracel ular domain of human inducible co-stimulator was prepared. The encode of the domain was fused with the gene of immunoglobulin IgG constant fragment (Ig) of encoding mouse, in order to build the inducible co-stimulator Ig fusion gene and the secreted eukaryotic expression vector pcDNA3-inducible co-stimulator Ig. Enzyme digestion of the recombinant and sequencing was performed, and then the positive liposome coated pcDNA3-inducible co-stimulator Ig was transferred into the muscle tissue of mouse right thigh. Western blot was used to detect the level of inducible co-stimulator Ig.
RESULTS AND CONCLUSION:The sequencing confirmed that the size of target gene fragment pcDNA3-inducible co-stimulator Ig plasmid was exactly the same with the sequence of inducible co-stimulator published on Genebank, which indicated the successful of plasmid construction. After transferred into the mouse for 7 days, the liposome coated pcDNA3-inducible co-stimulator Ig was positively expressed in the mice serum, which showed that pcDNA3-inducible co-stimulator Ig could be expressed in the rat muscle cel s. The results suggest that gene synthesis and recombinant technology can successful y construct the eukaryotic expression vector pcDNA3-inducible co-stimulator Ig.
9.Clinical Study of Using Cinobufotalin and Chemotherapeutic Agents by Transcatheter Arterial with Oily Chemoembolization in the Treatment of Primary Liver Cancer

Objective To evaluate the effect of cinobufotalin and chemotherapeutic agents by transcatheter arterial with oilychemoembolization(TACE) in the treatment of primary liver cancer.Methods 144 patients with HCC proved histopathologically were divided into 2 groups.76 of them(group A) were treated by transcatheter arterial infusion(TAI) with cinobufotalin 100 ml,DDP and 5-FU,then embolism with iodized oil mixed ADM;while the other 68 patients(group B) were treated by TAI with DDP and 5-FU,then embolism with iodized oil mixed ADM.The serum T lymphocytes,HBV DNA,AFP and CT scan were acquired before and after treatment.Results The effective rate(PR+MR) of group A was 86.64%,the lymphocyte transformation rate(LTT),T lymphocytes CD_3~+,CD_4~+proportion and CD_4~+/CD_8~+ratio markedly increased;HBV DNA descended in 21 cases,unchanged in 46 cases,and elevated in 9 cases;1 and 2 year survival rate was 86.84%(66/76)and71.05%(54/76) respectively.The effective rate(PR+MR) of group B was 72.73%,LTT,T lymphocytesCD_3~+,CD_4~+proportion and CD_4~+/CD_8~+ratio markedly descended;HBV DNA descended in 2 cases,unchanged in 20 cases and elevated in 46 cases;1 and 2 year survival rate was 72.73%(48/68) and 54.41%(37/68) respectively.There were significant statistical differences between the two groups(P

Objective To investigate if atorvastatin is effective in reducing carotid intima media thickness (CA-IMT) in normocholesterolaemic patients with transient ischemic attack (TIA). Methods Forty-two patients with TIA were recruited. Patients were randomised to receive atorvastatin(5-10 mg/d) or placebo daily. Serum concentration of cholesterol and CA-IMT were measured before randomisation and at 6-and 18-month intervals. Results CA-IMT was significantly lower in atorvastatin group than that in placebo group at 18-month (P
