Objective To study the relationship between Bcl-2/Bax protein expression and the degree of malignancy of astrocytoma.Methods Bcl-2/Bax protein expression positive rate and apoptotic cell rate were determined using immunohistochemical technique and TUNEL, in 10 normal human brain tissues and 80 human astrocytoma specimens of differently malignant degrees confirmed by pathology separatively. Results Bcl-2/Bax protein positive expression was not found in normal human brain tissues, and apoptotic cell rate was only (0 6?0 1)%. 2.Clinical Observation on Treatment of Osteoarthritis of Knee by Needle-Warming Method and Functional Training

Purpose: To observe the long-term and recent clinical effect of needle-warming method plus functional training in the treatment of osteoarthritis of the knee joint. Methods:After 106 cases of the patients were divided into the needle-warming group and comprehensive group by the order of their first visit, the needle-warming group was treated by the needle-wanning method and the comprehensive group was treated by the needle-warming method plus functional training. After ten treatments, the clinical data in the two groups were evaluated upon Lequesne scale before and after the treatments. Three months later, follow-up survey was given to process the statistic management of the reoccurrence rate of the symptoms.Results: In the two groups after the treatment, the symptoms of the knee joint and daily life ability were obviously improved than the respective conditions before the treatment, and the integral decreased (P<0.05), with no significant difference (P>0.05) between the two groups. In the follow-up survey, there was a difference (P<0.05) between the two groups in the positive rate of the recurrent symptoms. Conclusion: Certain therapeutic effect exists in the treatment of osteoarthritis of the knee joint by the needle-warming method. If functional training is combined,the therapeutic effect would be more stable, without easy reoccurrence.
3.EFFECTS OF TAURINE, QUINIDINE AND THEIR COMBINATION ON EXPERIMENTAL ARRHYTHMIAS INDUCED BY ACONITINE IN MICE

The constant iv. of aconitine can induce the varied arrhythmias in mice. Taurine 100mg/kg iv. or Quinidine 5 mg/kg iv. can delays the onset time of arrhythmias, and increases the dose of aconitine, respectively .But no effects were seen in mice pretreated with taurine 50mg/kg or quinidine 2.5mg/kg. However their combination can significantly decrease the incident of ventricular fibrillation and delay the onset time of death. Our results suggested that taurine, quinidine and their combination are effective on arrhythmias induced by aconitine in mice.
4.Changes of Caspase 3mRNA Expression After Rat Focal Cerebral Ischemia/Reperfusion Injury Treated by Mild Hypothermia

Objective To explore the relationship between the changes of Caspase 3mRNA expression and the injuried nervous cell apoptosis after rat focal cerebral ischemia/reperfusion injury who was then treated by mild hypothermia.Methods The middle cerebral arteries(MCA) of SD rats were occluded for 2 hours,and reperfused for 12 hours.Using FCM and semiquantitative RT-PCR technique,the DNA fragmentation rate and Caspase 3mRNA expression level were detected in the shamoperation group,the control group and the mild hypothermia group,respectively.Results The DNA fragmentaation rate and Caspase 3mRNA expression level in the sham operation group and the mild hypothermia group were obviously lower than those in the control group.Conclusions The significantly decreased level of Caspase-3mRNA expression may be related to the obviously decreased nervous cell apoptosis after rat cerebrall ischemia/reperfusion injury treated by the mild hypothemia.
5.Determination of cellular proliferative activity of lung cancer with flow cytometry

The cellular proliferative activity (CPA) of the surgical specimens of 35 patients with lung cancer,10 patients with benign lesions of the lungs,and 10 normal subjects was studied with flow cytometry.It was found that CPA was significantly higher in lung cancer than in benign pulmonary lesion and it was significantly higher in benign pulmonary lesion than in the normal lungs.The finding suggests that CPA is useful in the screening of lung cancer patients.
6.Role of intracellular protein tyrosine kinase activation in vascular smooth muscle cells migration mediated by angiotensin Ⅱ

Objective To investigate the role of intracellular protein tyrosine kinase (PTK) activation in vascular smooth muscle cells (VSMCs) migration mediated by angiotensin Ⅱ (AngⅡ). Methods Intracellular PTK activities were investigated by means of ?- 32 P incorperation in cultured rat VSMCs stimulated by AngⅡ. The migrated VSMCs were enumerated in a modified Boyden Chamber. Results Without any specific stimulus, only a few migrated cells were detected. However, the number of migrated cells were increased, under the condition of AngⅡ at the concentration of 1?10 -10 , 1?10 -9 , 1?10 -8 , 1?10 -7 and 1?10 -6 mol/L, by 3.7, 4.6, 5.9, 6.6 and 6.3 times respectively, compared with the control group. Intracellular PTK activities were also increased by 1.0,1.7,2.4,3.2 and 2.4 times compared with the control group, under the condition of AngⅡ at the concentration of 1?10 -10 , 1?10 -9 , 1?10 -8 , 1?10 -7 and 1?10 -6 mol/L, respectively. Statistical analysis showed that there was a significant positive correlation between AngⅡ-induced VSMCs migration and intracellular PTK activation. 7.Cleavage of Oncogene ki-rasG12V mRNA by Ribozyme and It' s Specificity

Objective: To design and confirm the cleavage activity of ribozyme Rz217 to oncogene ki-rasG12V messenger RNA and search for a new method for gene therapy targeting oncogene ki-ras. Methods: According to Symon' s principle,design an ribozyme specific for ki-rasc12v mRNA, both the constructs for transcription in vitro of ribozyme Rz217 and ki-ras exonl and the mammalian expression constructs of ribozyme Rz217 were constructed by DNA recombinant technique,ribozyme Rz217 and ki-ras exonl mRNA was obtained by transcription in vitro with T7 and SP6 RNA polymerase. Pancre atic carcinoma cell line PaTu8988 and human hepatocellular carcinomacell line BEL7404 were transfected with Rz217 mammalian expression constructs and the level of endogenous ki-rasG12V mRNA or ki-ras mRNA was determined by semiquantitative RT-PCR. Results: Not only in vitro but also in vivo, ribozyme Rz217 can cleave the mRNA of oncogene ki-ras (G12V) in site-specific manner and can not cleave the mRNA of wild-type ki-ras. Conclusion: Ribozyme Rz217 can cleave oncogene ki-rasc12v mRNA and the cleveage is specific for ki-rasG12V mRNA.
8.Changes in neural stem cell proliferation after rat cerebral ischemia/reperfusion injury is treated with mild hypothermia

Objective To study changes in neural stem cell proliferation in ischemic brain tissue after cerebral ischemia/reperfusion injury treated with mild hypothermia. Methods The middle cerebral arteries (MCA) of Sprague-Dawley rats were occluded for 2 hours, then reperfused for 3, 7, 11, 14, 18 and 28 days. Using immunohisto-chemical staining, the bromodeoxyur-idine ( BrdU)-positive cells in the brain tissue of the operationally intervened side were examined in a sham-operation group, a control group, and in rats treated using mild hypothermia. Results There were a few BrdU-positive cells in the sham-operated rats, but there were obviously more in the mild hypothermia group than in the control group. The peak period for proliferation of neural stem cells in the ischemic brain tissue was longer in the mild hypothermia group than that in the control group. Conclusion Mild hypothermia may promote proliferation of neural stem cells in ischemic brain tissue after cerebral ischemia/reperfusion injury.
9.HPLC-EC DETERMINATION ON THE CONTENTS OF DOPAMINE AND ITS METABOLITES IN BILATERAL STRIATA OF SUBSTANTIA NIGRA LESIONED RATS

The contents of dopamine (DA) and its metabolites, homovanillic acid (HVA) and 3,4-dihydroxyphenyl acetic acid(DOPAC), in both striata were determined with HPLC-EC technique in each of 28 albino rats, which were divided into 3 groups (bilateral lesioned, unilateral lesioned and healthy control groups). Among the unilateral lesioned rats, three indices such as DA, HVA/DA and DOPAC/DA in the lesioned striata were 22.27%, 420.00% and 199.75% of those of the intact striata, respectively. This suggests that there may be an enhanced compensatory DA release in the striatum ipsilateral to the lesion of the rat. Having compared the lesioned striata of unilateral lesioned rats with those of bilateral lesioned animals, and the intact striata of unilateral lesioned rats with those of the control ones, we did not find any significant differences in DA, HVA/DA or DOPAC/DA. The result implies that the compensatory DA release in the lesioned striatum probably accounts for the mechanism of the ipsilateral nigrostriatal system, not for the crossed nigrostriatal fibers.
10.Analysis of the variation trends of bed allocation and its equity at primary medical institutions in China

Objective Measure the variation trend and the problems of bed allocation and its equity at primary medical institutions and provide theoretical basis for optimizing the policy of health resources allocation for the government.Methods Analysis of the distribution and equity of the inpatient beds at primary medical institutions in terms of geographical area by means of the survey of regular overall statistics forms and the methods of Gini coefficient and Theil index.Results From 2009 to 2012,the number increase of beds at such institutions geographically was 7.72%,while that in eastern,central and western regions of China were respectively 5.35%, 3.97% and 27.88%.The number increase at community healthcare centers was 46.58%,while that in eastern,central and western regions of the country were respectively 18.09%,29.57%和 81.53%.The number increase at township hospitals was 2.33%,while that in eastern,central and western regions were respectively - 3.83%,- 0.43% and 22%.The Gini coefficient of bed allocation at such institutions was beyond 0.6,while that in eastern, central and western regions were respectively under 0.3,0.3-0.4 and 0.6 above.The Gini coefficient of bed allocation at community healthcare centers fell from above 0.4 to under 0.4.The Gini coefficient in the western regions increased from 0.3-0.4 to over 0.4.The variation trend of township hospitals was similar with the overall conditions at such institutions.The intra-regional Theil indexes and their contribution rates of bed allocation at such institutions were higher than those of inter-regional. Conclusions The overall variation trend of bed allocation at such institutions was on the rise,yet its variation trend of equity remains unchanged.The bed allocation and its equity at such institutions were higher than those at township hospitals.The growth rate in western regions was the highest,but the absolute number was the lowest.Moreover,the equity of bed allocation in such regions was not only the lowest,but also was found rising.The influence of bed allocation in intra-region terms was large,while its equity was not only lower but falling as well.
