1.Correlation between HDL-C/TG ratio and cardiovascular risk in elderly patients with hypertension

Objective To investigate the correlation between HDL-C/TG ratio and cardiovascular risk in elderly patients with hypertension, in order to set up an evaluation criterion which may objectively assess the cardiovascular risk in elderly population. Methods A survey was carried out among 2011 subjects aged 60 or over, who received health examination in the General Hospital of PLA in May 2008. Hypertension was diagnosed by using the definition recommended by WHO/ISH. The subjects were divided into 4 groups (male hypertesion group, male control group, female hypertension group and female control group) according to their gender and with or without hypertension. The parameters of hematology (systolic pressure, diastolic pressure, pulse pressure and mean arterial pressure) and the indicators of lipidemia [total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C)] were measured, and HDL-C/TG ratio was calculated. Logistic regression analysis was used to analyze the correlation between the indicators of lipidemia and cardiovascular risk. Results In both male and female hypertension groups, systolic pressure, diastolic pressure, pulse pressure and mean arterial pressure showed significant differences compared with those of the control groups gender-wise (P0.05). Other indicators of lipidemia showed no significant differences between hypertension group and control group. 2.Inhibitory Kinetic-Spectrophotometric Determination of Trace o-dinitrobenzene

In HCl medium,trace o-dinitrobenzene can sensitively inhibit the discoloration reaction of neutral red oxidized by KIO4. The optimum neagent is 0.55 mL neutral red, 0.60 mL HCl, 0.6 mL KIO4, respectively. The neaction ttmperatune is 80℃, neaction time is 6 min. The optimum condition and kinetic parameters are investigated in detail,a new method for determination of trace o-dinitrobenzene is proposed. The detection limit is 7.7×10-7 g/L and the linear range is 0.0~4.0 μg/L o-dinitrobenzene. It has been applied to the determination of trace o-dinitrobenzene in environmental water sample with satisfactory results.

To study the changes in levels of tumor necrosis factor ? (TNF ?), interleukin 6 (IL 6) and interleukin 1? (IL 1?) produced by the peripheral blood mononuclear cells (PBMC) of patients with congestive heart failure (CHF), and to investigate the effects of captopril, one of angiotensin converting enzyme inhibitors in active form, on production of these cytokines. PBMC from 35 CHF patients and 12 healthy controls were isolated by density centrifugation with Ficoll Hypaque, and were cultured with captopril at 10 -4 ~10 -2 mol/L in the presence of lipopolysaccharide (LPS). After incubation for 24 hours, TNF ? and IL 6 contents in the culture supernatants were measured by an enzyme linked immunosorbent assay (ELISA), and IL 1? by radioimmunoassay (RIA). 5.Vasodilating characteristics of iptakalim hydrochloride

Objective To observe the pharmacological characteristics of a novel antihypertensive drug iptakalim hydrochloride (Ipt) by dilating large and small arteries. Methods The vasodilating action of Ipt was studied using isolated rat tail artery helical strips and aortic rings. Results The vasodilating action of Ipt at 10 7-10 3mol/L was observed in rat tail artery strips precontracted with potassium chloride (KCl) in a concentration and endothelium dependent manner, but no significant vasodilating effect was seen on rat aortic rings. Ipt-induced relaxation in tail artery was significantly greater in spontaneous hypertensive rats than that in normotensive rats. The vasodilating actions of Ipt in small arteries could be markedly antagonized by glibenclamide, an inhibitor of ATP-sensitive potassium channels. Conclusion The results suggest that iptakalim hydrochloride has a selective vasodilating action in small arteries, and the action is related to activation of ATP-sensitive potassium channels.

7.ANALYSIS OF PLATELET ACTIVATION AND ITS CLINICAL SIGNIFICANCE IN OLDER HYPERTENSIVE PATIENTS

To investigate the changes of activated platelet marker and its significance in older hypertensive patients, glycoprotein Ⅱb/Ⅲa(PAC 1)and P selectin(CD62P) were used as molecular markers for activated platelets,which were analyzed by flow cytometric 3 color immunofluorescence. The results showed that PAC 1 expression on platelet membrane surface in older hypertensive patients was higher than that in healthy individuals and was not related with mean blood pressure.However, the expression of CD62P on platelets membrane surface did not differ in the two groups.These results suggest that PAC 1 may be regarded as a sensitive and specific molecular marker for activated platelets. Platelet activation occurrs in older hypertensive patients, and is of great importance for the antiplatelet therapy in hypertensive patients.
8.CHANGES IN L-ARGININE/NO PATHWAY OF PLATELET IN ELDERLY PATIENTS WITH ESSENTIAL HYPERTENSION

To observe the changes in L Arginine /NO pathway of platelet in elderly patients with essential hypertension 20 elderly patients with essential hypertension and 15 healthy volunteers were entered for the study. The changes in L Arg transport, the activity of NOS and the production of NO - 2 were measured. Results showed that the activity of NOS and the production of NO - 2 were decreased, compared with those of healthy volunteers. The maximum transport velocity (Vmax) of L Arg transport in elderly patients with essential hypertension was 70% of healthy volunteers. No changes in Michaelis constant (Km) were found. This results suggested that function of L Arginine/NO pathway of platelet in patients with hypertersion is decreased. L Arginine/NO pathway of platelet played an important role in antithrombosis. Not only drugs to lower blood pressure should be given but also antiplatelet drugs to prevent thrombosis.

10.Relativity between Duration of Type 2 Diabetes Mellitus and Pathological Changes of Coronary Heart Disease

Objective To investigate the correlation of diabetes duration and coronary arterial pathological changes by coronary angiography in patients with coronary heart disease and type 2 diabetes mellitus.Methods 206 patients with coronary heart disease admitted into the hospital were divided into 4 groups:no diabetes(n=62),the duration of diabetes ≤5 years(n=41),>5~≤10 years(n=38)and >10~≤20 years(n=45).The interventional therapy history and type 1 diabetes mellitus were excluded.The pathologic changes of coronary artery evaluated by coronary angiography and the results among different groups were analyzed with the diabetes duration.Results There was no significant differences in the pathological changes of coronary artery between the group of no diabetes and the groups of ≤5 years duration and >5~≤10 years duration.However,the group of >10~≤20 years duration showed more serious pathologic changes of coronary artery than other groups including the multi-branch lesion,diffuse lesion and obstruct lesion(P<0.01).Conclusion Following the process of the diabetes,the coronary lesions in patients with type 2 diabetes mellitus tend to be more widespread and severe.
