Chinese Journal of Organ Transplantation 1998;19(1):42-44
Allotransplantation with fetus parathyroids was performed in 4 cases of hypoparathyroidism after the grafts had been radiated with 6 Gy of 60Co and cultured in Balb/c nude rats for 14 davs. The recipients took 4 days of cyclosporine A(dose of 20~25mg·kg-1/d),one day preoperation and 3 days postoperation. The patients were followed up for 5~60 months.Traced observation on the patients'clinical symptoms and signs, the changes in the doses of Calcium and Vitamine D and in the levels of serum Calcium and parathyroid hormone was conducted to determine the grafts'survival. The results were satisfactory with all cases surviving for the follow-up period. It is considered that this method is much better than any we did previously.
2.Detection of alpha-fetoprotein messenger RNA in peripheral blood of patients with hepatocellular carcinoma
Chinese Journal of Current Advances in General Surgery 1999;0(03):-
Objective:To determine whether alpha-fetoprotein(AFP) messenger RNA(mRNA),a specifically hepatocyte-expressed gene, could be a marker of metastasis in hepatocellular carcinoma. Methods:Peripheral blood samples were obtained from 30 patients with hepatocellular carcinoma (HCC).A sensitive reverse transcription polymerase chain reaction( RT-PCR) technique was used to detect the human AFP mRNA in the blood. Results:Of the 30 HCC patients,14(46.7%) patients had positive AFP mRNA.The presence of AFP mRNA correlated with the TNM clinical stages, the size of tumor, extrahepatic metastasis or with portal vein carcinothrombosis(P0.05).Conclusion:The positive of AFP mRNA in the peripheral blood can indicate the presence of hematogenous metastasis in patients with HCC.

The aim of this study is to determine whether seeded Dacron with autologous venous tissue fragments shows significant 6-keto-prostglandin F 1? (6 K PGF 1? )production after in vivo implantation. The Dacron grafts seeded with autologous venous fragments were implanted into the inferior vene cava(IVC)of the 13 canines as seeded group;and the control group (8 canines),in which grafts were only preclotted with fresh blood. Plasma 6 K PGF 1? and TXB 2 were assessed at different time. All of the specimens explanted at exsanguination were observed with light microscopy and sanning electron microscopy and the amounts of 6 K PGF 1? from the luminal surface were assessed using ridioimmunoassay. The results showed that the total patency rate of the explanted vessels was higher in seeded group (61.5%) than that in control group ( 25% ),and that endothelial cells lined the whole luminal surface of Dacron at the 14th day after operation in seeded group, but thrombus covered the surface of Dacron at the 14th day in control ones. The level of 6 K PGF 1? from plasma and the luminal surface of Dacron grafts in seeded group was higher, but the level of plasma TXB 2 was lower than that in control group. It can be concluded that seeding Dacron with autologous venous fragments makes a new endothelial lining possible at two weeks, and can release more 6 K PGF 1? and thus lead to an improved patency rate of canine IVC reconstructions,so the graft patency might be determined according to the changes of plasma 6 K PGF 1? and TXB 2 level.

Abstract: Objective To investigate the allelic mutations of kdr and ace-1 in Anopheles gambiae s.l., providing a scientific basis for mosquito-borne disease prevention and control in the region. Methods Mosquitoes were collected from six villages in two malaria-prone areas, Est-mono (Ogou koulidé, Kepssi, Igbomedji) and Anié (Kolocopé, Bana, Afolé). All mosquitoes were used for DNA extraction, species identification, and genotyping of kdr and ace-1. Results An. coluzzii and An. gambiae, two closely related species within the An. gambiae complex, were identified across six collection sites, with An. gambiae being predominant, constituting approximately 96.53% of the samples. Among all mosquito samples detected, high-frequency mutations were observed at the kdr 1014 locus in the An. gambiae complex, with mutation frequencies reaching 100.0% in Ogou koulidé, Bana, and Afolé, 97.5% in Kepssi and Igbomedji, and 91.1% in Kolocopé district. The allelic frequency of 1014F ranged from 85.5% to 100%. Mutation frequency of An. coluzzii 1014 locus in all three regions was 100%, with an allelic frequency of 1014F ranging from 90% to 100%, indicating a high rate of homozygosity. The kdr 1575 mutation frequency in An. gambiae ranged from 6.6% to 24.3%, with the allelic frequency of 1575Y ranging from 3.2% to 12.2%, all detected mutations were heterozygous, and no mutations were detected at the 1575 locus in An. coluzzii. Mutations at the ace-1 G119 locus were also observed in the An. gambiae complex, with mutation frequencies in An. gambiae ranging from 8.8% to 15% and the allelic frequency of 119S from 4.4% to 7.5%. In An. coluzzii, mutation frequencies ranged from 0 to 20.0%, with an allelic frequency of 119S from 0 to 10%, and all detected 119S mutations were heterozygous. Conclusions The high frequencies of the Anopheles gambiae kdr mutation observed suggest that malaria mosquito vectors in Togo may have developed a high level of resistance to pyrethroid insecticides and possibly a moderate resistance to organophosphorus or carbamate insecticides. This indicates that local vector management authorities should prioritize insecticide resistance issues in future malaria vector control and resistance management planning, and conduct regular monitoring.

Journal of Clinical Pediatrics 2013;(10):905-909
Objectives To investigate the clinical features of progressive familial intrahepatic cholestasis type 2 (PFIC2) and to illustrate the importance of genetic diagnosis. Methods The mutations in 27 exons of ABCB11 encoding bile salt export pump (BSEP) were identiifed using polymerase chain reaction (PCR) and direct DNA sequencing in 6 children with suspected PFIC2. The pathogenicity of the newly identiifed mutations were predicted by SIFT, PolyPhen-2, SNPs&GO software. The clini-cal features and laboratory examinations were reviewed. Results Four disease-causing mutations, p.R928*, p.E554K, p.R575Q and p.Y337H were identiifed, and the last three mutations were novel. These three kinds of novel mutations can cause the disease. Two children with genetic diagnosis had such manifestations as onset within a month after birth, jaundice, hepatosplenomegaly, upset, increased levels of total bilirubin and direct bilirubin, GGT<100 U/L and high levels of total bile acid. Conclusions Genetic diagnosis is a potent tool for clinical diagnosis of PFIC2.
