1.Test of Blood Donors for Human Cytomegalovirus Infection by Polymerase Chain Reaction

The test of 110 normal blood donors recruited by one hospital for human cytomegalovirus (HCMV) infection was conducted by polymerase chain reaction,with its findings being analysed, indicating that the HCMV-DNA positive rate among the mentioned above blood donors was 61. 8% (68/110) ,in the meantime the HCMV-DNA positive rate was raised as the age increased. In the view of high HCMV infection rate among blood donors, it was recommended that the donor blood should be tested and screened for HCMV,when the special vulnerable cohort such as the patients with immune deficiency and transplanted organs and infants wanted to be transfused with donor's blood.
2.Study of early comprehensive treatment for acute ruptured intracranfal aneurysm.

Objective To apply early comprehensive rescue including embolism and microsurgery for trea- ting acute ruptured intracranial aneurysm in order to lower mortality and disability.Methods 120 patients with rup- tured intracranial aneurysm were treated by early comprehensive rescue.An cases were given support treatment,e- mergency rescue and DSA after hospitalization.Embolism and microsurgery were done within 72 hours and releasing bleeding CSF were done after surgery to prevent complications.Results Good recovery was over 80%(excellent in 95 ases,light disability in 18 cases,heavy disability in 4cases,and death in 3 cases).2 cases relapsed during hospi- talization.5 cases were coma induced by serious cerebral vesospasm.Conclusion Early comprehensive treatment including embolism and microsurgery complication prevention may reduce rebleeding,serious vasospasm and hydro- cephalus,1ower mortality and improve prognosis.
3.Management of primary gallbladder cancer by combining laparoscopy and open surgery

Objective To explore the method and the effect of combined use of laparoscopy and open surgery in the treatment of primary gallbladder cancer. Methods Pathological frozen-section examinations were carried out in all cases suspected of gallbladder cancer during laparoscopic cholecystectomy (LC). Patients in Ⅰ~Ⅱ Nevin stage received surgical removal of loose connective tissue at hepatic portal and gallbladder bed under laparoscope, while patients in stage Ⅲ or above underwent conversions to open operations of radical or extended radical resections. Results Out of the 34 cases of gallbladder cancer receiving LC, conversions to open surgery were required in 6 cases, open operations were performed after laparoscopic operation in 7 cases, and 1 patient refused further treatment. Except for 1 case of port-site cancer implantation below the xiphoid process, no other severe intra- or post- operative complications occurred. Follow-up revealed a 5-year survival rate of 80% (12/15) (survival range, 2~8 years) in 15 cases of laparoscopic surgery, and a survival duration of 8 months~4 years in 8 cases of open surgery. Conclusions Combined use of laparoscopy and open surgery in the management of variable stages of primary gallbladder cancer is effective.
4.Construction and identification of mouse SK2 gene subclones

BACKGROUND: Small conductance,Ca2+-activated potassium(SK)channel,presents in various cell types and plays a crucial role in action potential profile.However,coupling and modulation of calcium and associated molecules to SK2 channel remains unclear.OBJECTIVE: To construct the recombinants of pGBAT7 and target fragments of SK2 gene,so as to observe the coupling and regulation of SK2 channel gene to calcium and other molecules.METHODS: Three pairs of primers of the target fragments of SK2 gene were designed and synthesized based on the full-length sequences of SK2.After being identified,they were individually sub-cloned into the yeast expressive plasmid pGBKT7 to construct pGBKT7-SK2 vectors.The recombinant pGBKT7-SK2 vectors were transformed into yeast AH109 by electroporation,and their activation was tested.The recombinants were extracted from yeast AH109 and verified by electrophoresis and sequencing.RESULTS AND CONCLUSION: The target fragments of SK2 gene by PCR were 411,546 and 729 bp,respectively.Three sub-clones of pGBKT7-SK2 were successfully constructed.Electrophoresis and sequencing showed that the constructed sub-clones of pGBKT7-SK2 met the expected requirements.The recombinant pGBKT7-SK2 vectors transformed into the yeast could be activated.The successful construction of the sub-clones of SK2 gene provides an important material basis for further study in the SK2 channel and function-associated molecules.
5.Effect of Miacalcic Combined with Calcium in the Treatment of Osteoporosis and Influence on Bone Metab-olism Index

Objective:To study the clinical efficacy of salmon calcitonin combined with calcium in the treatment of osteoporosis and the effect on bone metabolism index. Methods:Totally 360 cases of patients with osteoporosis were randomly divided into the ob-servation group and the control group with 180 ones in each. The patients in the control group were given compound calcium carbonate tablets, po, qd, and those in the observation group were given 50IU salmon calcitonin injection additionally, im, qd. After two months of treatment, the changes in pain score, bone density and bone metabolism related indicators of the two groups were compared before and after the treatment. The clinical efficacy and adverse reactions of the two groups were observed as well. Results: After the treat-ment, both groups showed lower pain scores (P<0. 01) when compared with those before the treatment, and the reduction in the ob-servation group was more significant than that in the control group. After the treatment, the bone density had no significant change in the two groups(P>0. 05). The bone metabolism parameters including serum BGP,β-CTX, PTH, BNALP and 25OHVitD were signif-icantly improved when compared with those before the treatment (P<0. 01), and the improvement in the observation group was better than that in the control group (P<0. 01). The incidence of adverse reactions in the observation group was significantly higher than that in the control group (P<0. 01). Conclusion:Salmon calcitonin combined with calcium can significantly improve the efficacy in the treatment of osteoporosis with improved pain scores and bone metabolism related indicators, and the combination is much better than calcium alone, however, the adverse reactions are significantly increased.
6.Evolutionary characteristics of envelope glycoproteins in Ebora virus

Objective To study the evolutionary and mutant characteristics of Ebora virus envelope glycoprotein.Methods A total of 100 Ebora virus envelope glycoproteins amino acid sequences isolated during 1976 and 2014 were collected from National Center for Biotechnology Information (NCBI).Multiple sequence alignment and phylogenetic tree analysis were performed to investigate the evolutionary and mutant characteristics of Ebora virus envelope glycoprotein.Results Glycoprotein amino acid sequences of Ebora virus isolated during 1976 and 2014 showed only 54.00%-65.00% homology among different subtypes,while 95.00%-100.00% homology in same subtypes.Ebola virus isolated from different regions in 2014 showed a 99.70%-100.00% homology of glycoprotein amino acid sequences in the same subtype.The homology of glycoprotein amino acid sequences of Ebola-Zaire virus isolated from Sierra Leone in 2014 was 100.00%,but three strains of Ebola-Zaire virus isolated from Guinea showed diversity in glycoprotein amino acid sequences.Glycoprotein amino acid sequences of Ebola virus with different subtypes were on different branches of phylogenetic tree.Glycoprotein amino acid sequences of Ebola-Zaire virus isolated from Sierra Leone in 2014 were on one branch,and those of Ebola-Zaire virus isolated from other countries during 1976 and 2014 were on the another branch.Conclusions Glycoprotein amino acid sequences of Ebora virus vary with time and region.Ebola-Zaire virus isolated from different regions in 2014 may be two variants with the same origin,and hybrid phenomenon is not observed among virus of different subtypes.
7.Visual function of senile cataract patients after implantationofan aspheric multifocalversusan aspheric monofocal intraocular lens

BACKGROUND:Monofocal intraocular lens can restore distant vision of cataract patients, butit is not suitable fornear vision recovery, so most ofpatientsstil need to wear glasses. In contrast, multifocal intraocular lens can make up for the shortcomings of the single lenstoim provenear vision and increase the rate oftaking off the spectacles.
OBJECTIVE:To compare the visual function of senile cataract patients after aspheric multifocal and monofocal intraocular lens implantation.
METHODS:Totaly 30 senile cataract patients with 50 affected eyes were randomly divided into two groups: the eyes were implanted with aspheric multifocal and monofocal intraocular lens as experimental group and control group, respectively. At 3 months after implantation, distance visual acuity at a distance of 5 m, the best corrected distance visual acuity,as wel asnear and intermediate visual acuity at a distance of 30, 60 and 100 cm were detected, respectively. Besides, contrast sensitivity at a spatial frequency of 0.8, 1.5, 6 and 18 cpd under light and dark environment was measured.
RESULTS AND CONCLUSION:At 3 months after implantation, the distance visual acuity at a distance of 5 m in the two groups was significantly better than that before surgery (P< 0.05); no significant differences were found in near and intermediate visual acuitiesat a distance of 5 m and 100 cm between two groups, but the near visual acuitiesat a distance of 30 and 60 cm in the experimental group were significantly higher than those in the control group (P< 0.05). In addition, the contrast sensitivity at different spatial frequencies under light environment had no significant differences between two groups; and compared with the control group, contrast sensitivity at the frequency of 6 cpd under dark environment was significantly decreased in the experimental group (P< 0.05), but no significant differences were found in the other spatial frequencies. These results show that aspheric multifocal intraocular lens can significantly improve the near and intermediate visual acuity of senile cataract patients compared with the monofocal lens.
8.Systemic lupus erythematosus complicated with Pneumocystis carinii pneumonia

Objective To explore the clinical manifestation of systemic lupus erythematosus (SLE) complicated with Pneumocystis carinii pneumonia (PCP).Methods Retrospective analysis of the clinical data of two cases of SLE complicated with PCP.Results The two patients were both aged 31 years and have had SLE disease for 4 and 7 years respectively.Both had lupus nephritis and were previously treated with high dose corticosteroid and cyclophosphadmide.Both patients had ceased immunosuppressive drug treatment for 6 and 12 months respectively at the time of diagnosis of PCP infection.Both patients presented with acute fever with dry cough,shortness of breath at rest,progressive hypoxia and diffuse reticular shadows on chest X ray. Pneumocystis carinii were identified in the bronchoalveolar lavage fluid (BAF) in both cases.SMZ was used to treat the PCP infection.The first patient recovered with improvement in her clinical well being and chest X ray appearance,while the second failed to respond and eventually succumbed.Conclusion One should be alert to the occurrence of PCP when patients with SLE present with rapidly progressive hypoxemia and type I respiratory failure.The mortality rate is high in SLE patients complicated with PCP and early diagnosis and treatment are the key to improving survial in these patients.Recurrence of PCP is common and adequate duration of treatment is essential.

Objective To observe the effect of RNAi that silences MTA1 gene on invasion and migration of esophageal carcinoma 9706 cells. Methods The siRNA expression vector that silences MTA1 gene was transfected into EC9706 cells by liposome. MTA1 mRNA and protein expressions were detected through quantitative RT-PCR and Western blot, respectively. The invasion and migration of EC9706 cells were evaluated by scrape wound healing assay and cell invasion assay in vitro. Results MTA1 gene expression significantly decreased. Objective To investigate the association of primary liver cancer(PLC)with the mutations of HBV precore and basic core promoter(BCP)genes.Methods The serum markers of hepatitis B and the quantities of serum HBV DNA were detected in 144 HBsAg-positive PLC patients.The precore and BCP gene mutations in patients with HBeAg-negtive and HBV DNA-positive were detected by real-time PCR.One hundred and twenty chronic hepatitis B(CHB)patients were randomly selected to serve as the conol.Results There were 46(3 1.94%)patients with HBeAg-positive and 98(68.06%)patients with HBeAg-negative.In 98 HBeAg-negative patients,56(57.14%)were HBV DNA-positive,in which 43 (76.79%)were with precore 1896 gene mutations,50(89.29%)were with BCP1762/1764 gene mutations.and 38(67.86%)were with both gene mutations.Precore 1896 and BCP1762/1764 gene mutation rates in PLC patients were much higher than those in CHB patients(χ2=9.36 and 5.77,P<0.05).Conclusion PLC may be associated with the mutations of HBV precore anti BCP genes.
