1.Hyperbaric oxygen combined with Schwann cells transplantation for spinal cord injury in rats:electrophysiological and functional changes of the hind limbs

BACKGROUND:Hyperbaric oxygen therapy can improve the microenvironment of the injured spinal cord, and hyperbaric oxygen combined with Schwann cel transplantation is expected to improve the therapeutic efficacy on spinal cord injury. OBJECTIVE:To investigate the effect of Schwann cel transplantation plus hyperbaric oxygen on the neural functional recovery of rats with spinal cord injury. METHODS:A total of 80 female SD rats with spinal cord injury were randomized into 4 groups, with 20 in each group:blank control group, injection of L-DMEM via the tail vein at 6 hours after modeling;cel transplantation group, injection of 3×106 RESULTS AND CONCLUSION:The motor function of the lower limbs was better in the combination group than the cel transplantation and hyperbaric oxygen groups, as wel as better in the cel transplantation group and Schwann cel suspension via the tail vein at 6 hours after modeling;hyperbaric oxygen group, hyperbaric oxygen therapy at 1 hour after modeling;combination group, combined therapy of Schwann cel transplantation and hyperbaric oxygen. Inclined plane test, modified Tarlov score, Basso-Beattie-Bresnahan score for motor function evaluation of rat hind limbs were performed and measured at 1, 3 days, 1, 2, 3, 4 weeks after treatment. SRY gene expression in the spinal cord was measured at 4 weeks after transplantation using PCR method. Horseradish peroxidase tracer and electroneurophysiology detection was done at 8 weeks after transplantation.hyperbaric oxygen groups than the blank control group. SRY expression was detected in the cel transplantation group and combination group, but not in the blank control group and hyperbaric oxygen group. The number of nerve fibers positive for horseradish peroxidase was higher in the combination group than the cel transplantation and hyperbaric oxygen groups fol owed by the blank control group, and there were significant differences between different groups (P<0.01). In addition, the latencies and amplitudes of somatosensory evoked potential and motor evoked potential in the combination group were also better than those in the other groups (P<0.05 or P<0.01). These findings indicate that the combined therapy of Schwann cel transplantation and hyperbaric oxygen can promote the synaptic regeneration, improve limb motor function and electrophysiological function in rats with spinal cord injury, which is superior to hyperbaric oxygen or Schwann cel transplantation alone.
2.Association of angiotensinogen gene M235T polymorphisms with essential hypertension:a meta-analysis

Objective To evaluate the association of angiotensin (AGT ) gene M235T polymorphism with essential hypertension (EH) .Methods The literatures examining the association of the M 235T polymorphisms with EH were selected in electronic data-bases .Using RevMan5 .0 ,heterogeneity and pooled effect sizes were calculated .Results Totally ,29 articles ,with 5 971 cases and 6 443 controls were finally included in this study .The odds ratio (OR) value of TT+ TM∶MM genotype was 1 .13 (95% CI 1 .00-1 .25 ,P=0 .04) in worldwide populations .Subgroup analysis showed that the OR values of TT ∶MM genotype were 1 .13 (95% CI 0 .90-1 .42 ,P=0 .28) in subjects published before 2008 and 1 .29(95% CI 0 .94-1 .75 ,P=0 .11) in those after 2008(year of 2008 included) .The OR values of T ∶M in Han population was 1 .24 (95% CI 1 .01-1 .53 ,P=0 .04) .Conclusion TT+ TM genotype of the M235T polymorphisms is associated with EH in worldwide populations ,and T allele is also correlated with EH in Han popu-lation .
3.The effects of Rheum emodin on LPS-induced HIF-1α and COX-2 expression in intestinal epithelial cells

Objective To observe the level of hypoxia-inducible factor-1 alpha (HIF-1α) and its downstream target gene cyclooxygenase-2 (COX-2) in LPS-treated intestinal epithelial cells,and to explore the possible intervention targets of Rheum emodin.Methods Human intestinal epithelial cells were cultured in vitro treated with LPS to establish the experimental model.The protein level trends of HIF-1α and COX-2 were measured by Western blot in LPS dose-dependent and time-dependent manners.The protein level trends of HIF-1α,COX-2,Phospho-IκB-α and Phospho-NF-κB p65 were measured in LPS plus various concentrations of Rheum emodin treated groups.The expression of HIF-1α mRNA were detected by PCR after cells treated with LPS or LPS plus Rheum emodin,respectively.The effect of Rheum emodin on the proliferation of intestinal epithelial cells was measured by MTT assay in each group.Data were analyzed with ANOVA,and P <0.05 was considered significant.Results LPS induced the protein level of HIF-1α in a dose-dependent and a time-dependent manners.With increasing concentrations of LPS,the protein level of HIF-1α increased to the peak when cells were treated with LPS at 10-30mg/mL,and then gradually decreased (P <0.05).Firstly the protein level of HIF-1α reached the peak at 0.5 h after treatment,and then decreased to the lowest level at 4 h,and finally returned to a high level (P<0.05).The protein level trend of COX-2 went a similar way to that of HIF-1α (P <0.05).Rheum emodin inhibited the protein levels of LPS-induced HIF-1α,COX-2,Phospho-IκB-α and Phospho-NF-κB p65 with a significant dose-effect relationship (P < 0.05).The PCR showed Rheum emodin inhibited LPS-induced increasing expression of HIF-1α mRNA.MTT assay showed different concentrations of Rheum emodin (0 μmol/L,20 μmol/L,40 μmol/L,60 μmol/L,80 μmol/L) had no significant effect on cell proliferation (0.95 ± 0.02,0.89 ± 0.03,0.88 ± 0.04,0.91 ± 0.03,0.83 ± 0.03,P > 0.05).Although Rheum emodin produced biological effect at this concentration range,and it had no toxicity to intestinal cells.Conclusions LPS induces HIF-1α/COX-2 signaling pathway in a time-dependent and a dose-dependent manners in intestinal epithelial cells.Rheum emodin blocks the hypoxia pathway of LPS/HIF-1α/COX-2 and the inflammatory pathway of LPS/IκB-α/NF-κB/COX-2,which may play a protective effect on intestinal epithelial cells.
4.Fingerprint research on authentication of official and unofficial plants of Rheum L. by RAPD

Object In order to identify the resources of official and unofficial plants in Rheum L.at molecular level, a random amplified polymorphic DNA (RAPD) analysis was performed. Methods The amplification program of polymerase chain reaction (PCR) was optimized. Arbitrary decamer primers of 121 based on the individual selecting method, and 64 based on the BSA method were screened. Results Four primers among all screened primers were found to be useful to amplify specific strips to the official plants of Rheum L. Conclusion This method is reliable, accurate, quick and reproducible for authenticating the official and unofficial plants of Rheum L. at molecular level. Moreover, the experiment results indicate that it is more effective to screen the primers using BSA method than using individual selecting method.
5.Clinical significance of the serum-soluble intercellular adhesion molecule-1 in patients undergoing radical resection of lung cancer

To investigate the expressions and changes of serum soluble intercellular adhesion molecule-1 (sICAM-1) in lung cancer patients before and after operation and the roles of sICAM-1 in differential diagnosis, metastatic potential, and prognosis. Methods From 2002 to 2003, a total of 17 samples of lung carcinoma tissues and sera were studied. The other 11 non-cancer patients were employed as the controls. Cell surface sICAM-1 levels were analyzed by immunohistochemistry method and ELISA. Results The serum sICAM-1 concentration in the cancer patients was (432.0?124.4) ng/ml. In the controls, however, the serum concentration of sICAM-1 was (262.3?77.7) ng/ml. In patients with lung cancer, the serum concentration of sICAM-1 was significantly higher than that of the controls (P0.05). Conclusion The serum sICAM-1 concentrations in lung cancer patients may play an important role in staging and may also serve as a useful indicator of advanced disease.
6.Experimental research of shielding the patients during electron beam radiotherapy

Objective To explore the shielding effects of 1-4 layers of lead aprons (LPs) and body shielding devices (BSDs) against stray radiation (SR) outside the electron beam field of 6-15 MeV.Methods JR-115B LiF TLDs were used to measure the stray radiation doses (SRDs) to the patient undergoing treatment,before and after being shielding,for different distances,different energies,different applicators,variable layers of LPs,and different thickness of body shielding devices (BSDs),respectively,along long calculating and comparing the shielding ratios of LPs and BSDs against SR.Results When the applicator (10 cm × 10 cm) is unchanged,the shielding ratio increased with the increased distance from measuring point (r =0.717,P < 0.05) and decreased with the increased electron energy (r =-0.678,P < 0.05);when the energy was constant,there was no correlation between the shielding ratio and the size of applicator (P > 0.05);For lower energy electron beam of 6 and 9 MeV,the shielding ratio for 1 mm Pb-BSD was slightly higher than that for 2 layers of LA (t =2.519,2 662,P < 0.05),ranging from 81.5% to 95.3% and 55.4% to 84.6%,respectively.For 12 and 15 MeV higher energy electron beam,the shielding ratio for 2 mm Pb-BSD was slightly higher than that for 4 layers of LA (t=3.768,7.934,P<0.05),ranging from 64.6% to 93.4% and 51.1% to 92.4%,respectively.Conclusions LAs or BSDs are availavle for effectively reducing the doses from stray radiation,and may help reduce the secondary risks from stray radiation.BSDs have more obvious advantages than LPs with regard to shielding effect.
7.Molecular Mechanisms of Macrolide Resistance of Mycoplasma pneumoniae

OBJECTIVE To investigate macrolide resistance and main molecular mechanisms in Mycoplasma pneumoniae. METHODS Thirty two throat swabs from children infected with M. pneumoniae were cultured by modified Hayflick medium. Antibiotic susceptibility test was used to screen the macrolide-resistant M. pneumoniae. The 23S rRNA gene sequences of the strains were determined with polymerase chain reaction and sequencing. RESULTS Nineteen strains were isolated from 32 throat swabs successfully.Fifteen strains were resistant to macrolide antibiotics according to the results of antibiotic susceptibility test. Once the strain was resistant to one of macrolide antibiotics,it would be resistant to the others. Sequencing results of the sensitive strains and the standard strain FH were completely same. Fifteen resistant strains presented A2063G point mutation in 23 SrRNA region Ⅴ, in which 2 examples showed the coexistence of the sensitive strain and the resistant strain. CONCLUSIONS Macrolide-resistant M. pneumoniae is common and serious at present. The antibiotic resistant isolate carries point mutations of the 23S rRNA region Ⅴ.

Objective To evaluate hepatitis B virus large surfsce protein(LHBs) in monitoring of antiviral treatment.Methods LHBs.HBV serum markers and HBV DNA loads in 46 patients with adefovir dipivoxil(ADV) therapy were monitored for the whole course(60 weeks).Enzyme linked immunosorbent assay(ELISA),time differentiate immunofluoresence assay and real.time polymerase chain reaction(RT-PCR)were performed to detect LHBs,HBV serum markers and HBV DNA loads,respectively.And correlation analysis was also performed.Results Both LHBs and HBV DNA declined during the ADV treatment.and the correlation coefficient was 0.97.but LHBs declined after HBV DNA.There were 20 patients with HBV DNA<5×102/mL at 60th week.in which 8 were LHBs negative;in 14 recurrent patients,the HBV DNA turned to negative and became positive again,3 with negative LHBs;while in 12 patients resistant to the ADV therapy.2 were LHBs negative.Conclusion Dynamic monitoring of LHBs is useful in the evaluation of antiviral treatment.
9.Quantification of mitral apparatus geometry in mitral regurgitation with 3-dimensional echocardiography

Objective To evaluate the geometry of mitral valve and annulus in patients with mitral regurgitation, and to quantify the mitral annular remodeling and motion in ischemic mitral regurgitation (IMR) with mitral valve assessment (MVA) software. Methods Seventeen patients were divided into two groups: IMR group (n=10) and non-IMR group (n=7), and other 11 patients without significant mitral regurgitation (<2+) and mitral structure abnormality were regarded as control group. The imaging was analyzed offline with Siemens MVA software to measure the annular diameters, annular circumference, anterior and posterior annular length, annular area (MAA) and non-planar angel (NPA). The dynamic changes during the cardiac cycle were calculated. Results Compared with control group, the posterior annular length and NPA significantly increased in IMR group (P<0.05, P<0.01). During the systole, the posterior annular and annular circumference paradoxically prolonged in IMR group compared with control group (median Δ posterior annular length: -2.71 mm/m2 vs 0.52 mm/m2, P<0.05). In all patients of non-IMR group, the geometric character of mitral valve lesions shown in 3-dimensional echocardiography was accordant with surgical findings. Conclusion Three-dimensional echocardiography combined with MVA software provides a useful tool for assessment the geometric character of mitral valve lesions and quantification of mitral annulus remodeling, which is helpful to understand the mechanism of MR and guide surgical treatment.
10.The clinical efficacy of ω-3 fatty acids immune nutrition therapy in patients with acute ischemic stroke

Objective To explore the clinical value of ω-3 fatty acids (ω-3PUFA)immune nutrition therapy for the patients with acute ischemic stroke,and analyze its mechanism.Methods 40 patients with acute ischemic stroke were selected.They were divided into the treatment group and the control group,each group in 20 cases. The patients of the control group were given indwelling tube admitted to hospital within 24 hours,given the homogenized meal configured by nutriology department.The treatment group received ω-3PUFA enteral nutrition based on the control group.The neurologic recovery,nutritional status,lipid levels and the incidence of complications were evalua-ted.Results The total effective rate of the treatment group was 90.0%,which was higher than that of the control group (60.0%,χ2 =4.8,P <0.05).The incidence rate of complication of the treatment group was 30.0%,which was lower than the control group (65.0%,χ2 =4.91,P <0.05).The indicators such as NIHSS score,GCS score, TSF and AMC in the treatment group were better than those in the control group[(13.38 ±1.21)points vs.(10.12 ± 1.33)points;(12.9 ±2.6)points vs.(14.1 ±1.7)points;(14.06 ±7.16)mm vs.(14.8 ±4.18)mm;(26.32 ± 1.42)mm vs.(29.40 ±1.08)mm,t =3.51,6.44,3.22,5.19;all P <0.05].TG and LDL -C in the treatment group were lower than those in the control group[(1.60 ±0.71)mmol/L vs.(1.41 ±0.67)mmol/L;(3.11 ±0.60)mmol/L vs.(2.67 ±0.66)mmol/L,t =5.97,4.22;all P <0.05 ].Conclusion The clinical curative effect is better for application of ω-3PUFA on immune nutrition treatment for the patients with acute ischemic stroke.The patients'nerve function,blood lipid levels and nutritional status were significantly improved,and the complications were reduced.
