Objective To establish the method of assaying total flavonoids in Lecithin Pollen Pellet.Methods Suo-shi pheresis was wsed to detect wave length of 500nm with Eldrin being as control article and sodium nitrite-aluminum nitrate-sodium hydroxide as developer.Results There was good linear correlation between Absorbability A and Eldrin mass concentration C rangmg from 0 to 48?g/ml;Average recovery rate of spotting was 98.7%(RSD=1.41%).Conclusion With the character of handling simply,accurate result and good reproduction quality,the method was a scientific method for quality control of assaying effective composition Lecithin Pollen Pellet.
2.Clinical Observation of Batroxobin in the Treatment of Senile Sudden Deafness

OBJECTIVE:To observe therapeutic efficacy and safety of batroxobin in the treatment of senile sudden deafness. METHODS:Totally 102 senile patients with sudden deafness were randomly divided into control group(51 cases)and observation group (51 cases). Control group was given routine treatment as Ginkgo biloba extract injection 105 mg+Dexamethasone sodium phosphate injection 30 mg+Methotrexate tablets 6 mg orally. Based on control group,observation group was additionally given Ba-troxobin injection 10 BU added into 0.9% Sodium chloride injection 100 mL intravenously,running out within 1 h,every 2 days, and then given 5 BU added into 0.9% Sodium chloride injection 100 mL intravenously,running out within 1 h. Both groups were treated for 10 d. Clinical efficacy,hearing recovery time,ear drowsiness disappearance time,dizziness disappearance time and tin-nitus disappearance time of 2 groups were observed. The levels of pure tone threshold,plasma specific viscosity(ns),high shear rate of whole blood viscosity(nbh),low shear rate of whole blood viscosity(nbl),hematocrit(HCT),erythrocyte sedimentation rate (ESR),Fb,endothelin (ET) and nitric oxide (NO) were observed before and after treatment as well as the occurrence of ADR. RESULTS:The total response rate of observation group was significantly higher than that of control group;hearing recovery time,ear drowsiness disappearance time,dizziness disappearance time and tinnitus disappearance time were significantly shorter than control group,with statistical significance(P<0.05 or P<0.01). Before treatment,there was no statistical significance in the levels of pure tone threshold,ns,nbh,nbl,HCT,ESR,Fb,ET and NO between 2 groups(P>0.05). After treatment,the levels of pure tone threshold,ns,nbh,nbl,HCT,ESR,Fb and ET in 2 groups were significantly lower than before;the observation group was significantly lower than the control group. The levels of NO in 2 groups were significantly higher than before;the obser-vation group was significantly higher than the control group,with statistical significance(P<0.01). During treatment,no obvious ADR was found in 2 groups. CONCLUSIONS:Based on routine treatment,batroxobin is effective in the treatment of sudden deaf-ness in the elderly,can effectively relieve the symptoms,improve the recovery of local blood flow and inner ear microcirculation disturbance with good safety.
3.Study of the Effect and Functional Mechanism of Pine Pollen on Reducing the Blood-fat Level of the Experimental Mice

Objective To study the effect of lecithin pollen on hyperlipidemia model experimental animals.Methods Mice were feed with high-fat feeds in order to establish the hyperlipidemia models.The contents of TC,TG and HDL-C in the serum of the mice were tested before the experiment and the mice were divided into 5 groups according to their TC level,induding the control group,the model group and 1.25,2.50and 3.75g/kg lecithin pollen groups.Lecithin pollen was given preventively for 30 days,and then the TC,TG and HDL-C level of the mice were retested.Results Compared with the model group, the TC level of the middle and high dose group decreased. The TG level of the high dose group also decreased and the HDL-C level of the high dose group increased. The difference was significant. Conclusion The lecithin pollen is effective in preventing the formation of hyperlipidemia in the experimental mice.
4.Effect of static magnetic fields on the cellular proliferation and cycle of the Human bone marrow mesenchymal stem cells

Objective: To observe the effect of static magnetic fields(SMF) on the proliferation of bone mesenchymal stem cells(MSC) in human. Methods: The MSC were obtained by using gradient centrifuge method, and then selected by the adhesive method. The third generation cells were irradiated by use of static magnetic fields at different intensities for 5 days(8 h/d). The method of MTT was employed to evaluate the level of proliferation. The parameters regarding the variation of the cell cycle were detected with the flow cytometry(FCM). Results: As compared to the control group, the proliferative rate of the MSC exposed to 0.05 mT SMF was significantly higher; there was no difference between the 0.10 mT group and control group; howere, cell proliferation was attenuated significantly when SMF intensity was 0.50 mT and 1.00 mT. No abnormal ploidy was found in any group. Conclusion: The effect of SMF on the proliferation of MSC is dependent on the magnetic intensity. 0.05 mT SMF can accerate the proliferation of MSC. 0.10 mT SMF have no effects on the growth of MSC. Wherease, 0.50 mT and 1.00 mT SMF can attenuate the growth of MSC.
5.Construction of EGFP-labled recombinant plasmid of VEGF165 gene and its expression in bone marrow mesenchymal stem cells

Objective To construct a EGFP-labled recombinant plasmid of VEGF165 (vascular endothelial growth factor) gene, and to study the transfection and expression of VEGF165 eukaryotic expression plasmid in mesenchymal stem cells (MSCs). Methods pIRES2-EGFP-VEGF165 recombinant plasmid was constructed, which was then transfected into rat MSCs. ELISA and MTT were used to detect the expression level and biological activity of VEGF in the conditioned medium after transfection. Results There was a significant increase in VEGF protein in the MSCs after being transfected with pIRES2-EGFP-VEGF165. The conditioned medium after transfection showed the biological activity of stimulating the proliferation of endothelial cells. Conclusions The pIRES2-EGFP-VEGF165, a eukaryotic expression plasmid for VEGF165 gene, is constructed. High levels of VEGF protein expression can be obtained in the MSCs transfected with pIRES2-EGFP-VEGF165. The expressed protein has the biological activity of VEGF.
6.Nanoparticle as a new gene transferring vector in VEGF gene transfection

Objective To evaluate the possibility and efficiency of nanoparticles as a new vector in vascular endothelial growth factor (VEGF) gene transfection. Methods Nanoparticle-VEGF (Np/VEGF)complex was prepared with poly (D, L-lactide-co-glycolide) (PLGA) loading VEGF165 gene using the multiple emulsion (w/o/w) technique. The envelopment efficiency and size of the complex were determined. Rat myocardial cells were cultured in vitro, and the Np/VEGF was transfected into the cultured myocardial cells. Then RT-PCR and ELISA were used to evaluate whether the Np/VEGF increased the level of gene expression. Four New Zealand rabbits were used, the suspension of Np/VEGF was injected into myocardial tissue of rabbits after thoracotomy. 96h after the operation, the tissue sections of the implant sites were observed with transmission electron microscope (TEM) to determine the process of nanoparticles as vectors for gene transfer to cardiac myocytes. Results The envelopment efficiency and size of the Np/VEGF complex thus prepared were 1.87% and 25-300nm respectively. RT-PCR and ELISA showed that VEGF gene could be successfully transfected into myocardial cells by nanoparticle, and NP/VEGF significantly enhanced gene transfection efficiency, and it was more effective than plasmid. 96h after the operation, a great number of nanoparticles were observed in myocardial cytoplasm and nucleus with TEM, and many nanoparticles began to dissolve and degrade, suggesting that the DNA was released slowly from the nanoparticles localized in the cytoplasmic compartment, and was then transferred into the nucleus. Conclusions NP/VEGF can act as a vector to transfect VEGF gene in vitro and in vivo, it significantly enhanced gene transfection efficiency, and it was more effective than plasmid.
7.Effects of chemical processing time on the reproducibility of film dosimetry and its optimization

Objective To monitor the performance constancy of the HS-126E automatic X-ray film processor and to investigate the effects of chemical processing time on the reproducibility of film dosimetry.Methods With films having identical exposure,15 ready-pack Kodak X-omat-V films from the same batch were divided into 5 groups and exposed one by one under the identical conditions.One group of test film was processed per day from Monday to Friday with identical temperature and time.All films were digitized with a Vidar VXR-16 bit film digitizer. The average optical density(A) at irradiation field center was analyzed with RIT113 film dosimetry system software.At the same time,another 10 films in 5 groups from the same batch were exposed with vertical method and processed to get the calibration data and sensitometric curves.The film sensitivities,i.e.,ratio of A to delivered dose,were calculated and compared between groups.Another 15 films in 5 groups were exposed with horizontal method and processed similarly to the test films,the point dose at 1.5?cm depth on the central axis was calculated with sensitometric curve and compared with the value measured by ion chamber.Results Variations of A value in each group was less than 2%.During the processing period,a trend of decrease along with time in mean A value decreased day by day was observed.Both the standard deviations and coefficients of variation in all groups were within 2%.One-way ANOVA showed that there was a significant difference of the mean A value among all films from Monday to Friday(P
8.A phase Ⅰ-Ⅱ clinical trial of radiotherapy combined with hyperthermia for ≥5 cm non-small cell lung cancer

Objective To observe the results and side effects of radiotherapy(RT) combined with hyperthermia(HT) for non-small cell lung cancer(NSCLC)with lesion(T) ≥5?cm in diameter.Methods Patients conforming to T≥5?cm or T volume ≥125?cm~3 in the lung and other required eligibilities were given RT and HT.The temperature of HT in the center of Ts was automatically programmed between 43.0℃ and 43.5℃ by a computer.RT dose of D_T60-66?Gy was delivered in 30-33 fractions with in 1 hour after HT.Results From October 2001 to April 2004,30 patients were registered for this study.Among all patients,23 males and 7 female with median age of 61 years(32-81);22 squamous cancer and 8 adenocarcinoma type;6 stage Ⅰb,1 Ⅱb,10 Ⅲa,8 Ⅲb and 5 Ⅳ lesions.The mean volumes of T was 275.45?cm~3(125-900?cm~3).The mean number of HT given was 7.2 times(2-14).The mean irradiation dose was 59.10?Gy(23.96-9.67?Gy).Five out of 30 patients were dropped from this trial due to reasons un-related to acute side effects of RT or HT.None complained of breathlessness or aggravation of respiratory symptom during heating.No change had been observed in blood pressure,pulse or frequency of breath before or after HT,nor was there any hardening of subcutaneous fatty tissue as well as lipid nodules or severer than grade Ⅲ acute irradiation side effects.Complete response was observed in 23%,partial response in 50%,no change or progression in 27%.Loco-regional control rate was 83% with a median of median locoregional duration of 19.2 months.Eleven patients had died,including 2 of hemoptysis.The median survival duration was 19.7 months and 1-year survival rate was 75%.Conclusions Radiotherapy combined with hyperthermia modal can be used for NSCLC with primary lesion≥5?cm as it carries better efficiency and loco-regional control without server side effects.
9.Impact of three threshold segmentation of 18FDG PET image on target volume delineation and radiotherapy treatment planning of non-small cell lung cancer

Objective To evaluate the accuracy, of different threshold segmentation of 18FDG PET for target volume delineation of non-small cell lung cancer(NSCLC) and the potential influence on radiotherapy treatment planning. Methods Eight NSCLC patients who had tumor with clear margin on CT scan and the amplitude of tumor movements not more than 5 mm were enrolled. PET scans were carried out at 1 h after intravenous injection of 18FDG with CT image for attenuation revisement. Gross target volume (GTV) delineated on CT image ( GTVCT ) was used as the standard. Then, GTVs were delineated on PET image with three different threshold segmentation of 42% Imax(total) (42% of maximum voxel intensity within the tumor) ,Iback + 20% Imax-back(max) (mean background intensity + 20% of normalized background-subtracted maximum voxel intensity within the tumor) and Iback -20% Imax-back(slice) (mean background intensity + 20% of normalized background-subtracted maximum voxel intensity of each slice within the tumor) ,the corresponding GTV was named as GTV42%, GTV20%max and GTV20%slice. Both the size of GTV42%, GTV20%max, GTV20%slice and GTVCT,and the coverage over GTVCT for each GTV were compared. A three dimensional margin of 1 cm were added to GTVCT, GTV42%, GTV20%max and GTV20%slice to form corresponding PTVCT, PTV42%, PTV20%max and PTV20%slic e. Three dimensional conformal radiotherapy treatment plans were designed based on PTVCT,PTV42% , PTV20%max and PTV20%slice respectively for each patient. The prescription dose of all PTVs was 66 Gy in 33 fractions in 6.6 weeks. Both the volume accepting dose less than 95% of prescription dose within PTVCT ( VPTV ) and the lung V20 were compared among the four plans based on different PTVs. Tumor control probability(TCP) as well as lung normal tissue complication probability (NTCP) were also compared. Resuits Eight patients were enrolled in this study. Median deviation of volume between GTVPET and GTVCT were -54.1% , -21.5 % and 5.3 % for GTV42% , GTV20%max and GTV20%slice, respectively. Median coverage over GTVCT of GTV42% , GTV20%max and GTV20%slice was 45.9% ,78.0% and 95.3% respectively( F = 57.50,P<0.01). Median 7.5% of VPTV was observed for radiotherapy treatment plan based on PTV42% ,which meant that it might induce median 1% decrease of TCP comparing with that of radiotherapy treatment plan based on PTVCT. Whereas,there were only 1.3% and 0.0% of VPTV for treatment plans based on PTV20%max and PTV20%slice respectively. As far as TCP was concemed, both PTV20%max group and PTV20%slice group were superior to PTV42% group,there was no significant difference among PTV20%max group, PTV20%slice group and PTVCT group. Lung V20 and lung NTCP showed no significant difference among all groups. Conclusions The threshold segmentation of Iback + 20% Imax-back(slice) , being slice specialized, might be an optimal threshold segmentation for target volume delineation of lung caner. Independent of information of target volume provided by CT scan in advance,it is recommended to use for the target volume delineation of NSCLC with atelectasis.
10.Study of different registration methods for on-line kilovoltage cone-beam CT guided lung cancer radiation

Objective To select the optimal registration method for on-line kilovoltage cone-beam CT (KVCBCT) guided lung cancer radiation and evaluate the reproducibility of the selected method. MethodsSixteen patients with non-small cell lung cancer were enrolled into this study.A total of 96 pre treatment KVCBCT images from the 16 patients were available for the analysis.Image registration methods were bone-based automatic registration,gray-based automatic registration,manual registration and semi-auto matic registration.All registrations were accomplished by one physician.Another physician blindly evaluated the results of each registration,then selected the optimal registration method and evaluated its reproducibili ty.Results The average score of the bone-based automatic registration,gray-based automatic registration, manual registration and semi-automatic registration methods was 2.4,2.7,3.0 and 3.7,respectively.The score of the four different groups had statistics significant difference (F = 42.20,P < 0.001).Using the semi-automatic registration method,the probability of the difference between two registration results more than 3 ram in the left-right,superior-inferior,and anterior-posterior directions was 0,3% and 6% by the same physician,0,14% and 0 by different physicians,and 8%,14% and 8% by physician and radiation therapist.Conclusions Semi-automatic registration method,possessing the highest score and accepted re producibility,is appropriate for KVCBCT guided lung cancer radiation.
