1.Surface gene variants of hepatitis B virus of the co-existence of HBsAg and HBsAb in patients with chronic HBV infection

ObjectiveTo analyze the characteristics of S gene varivants of patients with chronic hepatitis B virus infection with coexistence of HBsAg and HBsAb.MethodsHBV DNA S gene regions were amplified and sequenced in 26 HBsAg-positive/HBsAb-positive patients (test group) and the newly diagnosed 39 HBsAg-positive/HBsAb-negative patients (control group).The sequencing results and amino acid variants of this region were analyzed.ResultsThe amino acid variants were significantly more frequent in test group than in control group among the complete S gene region (P<0.05).In addition,a significantly bigher amino acid variants arose in patients group versus control group in HBsAg major hydrophilic region (MHR),especially the first loop area of a-determinant (6.69% vs 0.89%,P<0.05).ConclusionThe coexistence of HBsAg and HBsAb in patients with chronic HBV infected might be related to the emergence of S gene variants in and around HBsAg a-determinant especially in the first loop area.
2.Pre-occlusion and Stump Pressure of the Internal Carotid Artery in Carotid Endarterectomy

Objective To investigate the value of the pre-occusion and stump pressure(SP)of the internal carotid artery in carotid endarterectomy(CEA).Methods During Januray 2007 to January 2009,12 patients with carotid arterial stenosis underwent CEA in our deparment.With cervical plexus block,we performed pre-occlusion test and measured the stump pressure(SP)by using vascular shunt selectively.The patients were monitored with clear consciousness during the whole procedure of CEA.The data of postoperative complications and results of pre-occlusion and SP as well as the follow-up were analyzed afterwards.Results During the operaion,three patients had transient increase of heart rate and blood pressure.The lowest SP was detected in two patients,who had moderate or severe stenosis of the contralateral carotid artery complicated with lesions in the vertebral basilar artery.Two patients showed positive results in pre-occusion test,and vascular shunt was used in four cases.After the operaion,one patients developed hyperperfusion syndrome.Nine patients showed improvement of symptoms of dizziness and decreased eyesight.This series were followed up for a mean of 11 months(ranged from 1 to 26 months).During the period,two of the patients showed re-stenosis of the carotid artery;none of them had cerebral infarction.Conclusions Cervical plexus block combined with pre-occusion test ensures the safty and effectiveness of minimally invasive carotid endarterectomy.We recommend selective vascular shunt according to the results of pre-occlusion test,SP,and the severity of the lesions in the contralateral carotid artery and vertebral basilar artery.
3.A clinical study on varicose veins of the lower extremity and Cockett syndrome

Objective This study was to investigate the incidence of Cockett syndrome in patients with varicose veins of the lower extremity and the relationship between Cockett syndrome and varicose veins and deep venous insufficiency of the left lower extremity. MethodsDuring 1999~2001,a total of 148 patients with varicose veins of the lower extremity were investigated by descending deep phlebography and iliography preoperatively. Results (1) There were 40(65.6%), 37(68.5%), 13(39.4%) cases respectively with abnormalities of the left common iliac vein(ALCIV) in patients with varicose veins of the bilateral, left and right leg, respectively. Objective:The paper provided the basis of morphology to treat severe obstructive sleep apneasyndrome (OSAS) by applying hard palate short uvulopalato-pharyngoplasty (HPS-UPPP) in clinic.Methods:The curve length of hard palate and soft palate,the distance between the greater palatine foramenwere measured with vernier caliper and the position relation of the nerve and vessels passing throughgreater palatine foramen was observed in 100 oranium and 50 cadaver. Results:The curve length of hardpalate was 49.3± 0. 28 mm;the curve length of soft palate was 26.1±0. 30 mm;the distance between thegreater palatine foramen was 27.3±0. 24 mm. Conclusion:The results have the guiding significance in re-moval of length of hard palate and soft palate ,and the way of operation.
6.Construction and expression of multivalent bispecific single chain Fv antibody gene specific for human ?-seminoprotein and CD_3 molecule

Objective To construct multivalent bispecific single chain Fv antibody(Bi scFv) gene specific for human ? seminoprotein (? Sm) and CD 3 molecule and to detect its expression in HeLa cells. Methods Human IgG3 upper hinge/human p53 tetramerization domain fusion gene was obtained by recursive polymerase chain reaction (PCR),and was inserted into pUC19 to construct cloning plasmid pUC19/IgG3/p53.The anti CD 3 scFv and anti human? Sm scFv was cloned into pUC19/IgG3/p53 subsequently to construct multivalent bispecific anti human ? Sm /anti CD 3 scFv fusion gene,scFv 2(? Sm/CD 3),which was then subcloned into the pSecTag2 B expression plasmid.The pSecTag2 B plasmids including the fusion gene were then transfected into HeLa cells.The expression products were analyzed by both SDS PAGE and Western blot and were purified with Ni 2+ NTA superflow affinity chromatography.The binding affinity for PC 3 cells and peripheral blood mononuclear cells (PBMCs) was measured by flow cytometry. Results The multivalent scFv 2(? Sm/CD 3) gene consisted of 1638 bp encoding 546 amino acid residues,and was the same as that reported previously.The expression products of the multivalent scFv 2(? Sm/CD 3) with a relative molecular mass ( Mr ) of about 67 000,as confirmed by SDS PAGE and Western blot. After purified with Ni 2+ NTA superflow affinity chromatography,the multivalent scFv 2(? Sm/CD 3) showed significantly stronger binding to PC 3 cells and PBMCs than Bi scFv. Conclusions The multivalent scFv 2(? Sm/CD 3) which could bind to PC 3 cells and PBMCs has been successfully gained providing a laboratory foundation for clinical use.
7.An in-vitro study of Lovastatin's biphasic effects on proliferation and adhesion of human umbilical vein endothelial cells

Objective To investigate Lovastatin's effects on proliferation and adhesion of human umbilical vein endothelial cells (HUVECs). Methods Culture medium with different concentration of Lovastatin(0.001、0.01、0.1、1.0、10μmol/L) was prepared, HUVECs was cultured in 96 well-plate with the different medium. AT the point of 24,72 and 120 h, the cell's activity and quantity was assessed by MTT. HUVECs was cultured with Lovastatin in 6 well-plate for 24 hours, then collected the cells by trypsin digestion. The cells were seeded in 24 well-plate with 2×104/ml and adhering for 30 mins. Then counting the adhered cells in different wells. Results At 24 h, Lovastatin (0.01、 0.1 μmol/L ) promoted proliferation of HUVECs ( P < 0.05 ); at 72 h, Lovastatin ( 1.0μmol/L) was positive accelerating cell growth(P< 0.05 ). While Lovastatin ( 10μmol/L) inhibited the proliferation significantly ( P <0.05 ) at 120 h. As HUVECs was cultured with Lovastatin for 24 hours, Lovastatin (0.1、1μmol/L) inhanced the adhesion capability of HUVECs significantly( P< 0.05 ). Conclusion Lovastatin had biphasic effects on proliferation and adhesion of HUVECs dependent on the concentration. Lovastatin (0.1、1.0 μmol/L) could promote the proliferation and adhesion, while at higher concentration ( 10.0μmol/L) it inhibits cell proliferation and adhesion.
8.Silencing IDH-2 gene by siRNA-IDH-2 inhibits human small cell lung car-cinoma growth

[ABSTRACT]AIM:Toinvestigatetheeffectofsilencingisocitratedehydrogenase2(IDH-2)genebysmallinter-fering RNA (siRNA) on the biological characteristics of human small cell lung cancer cell line NCI -H446.METHODS:IDH-2 expression was knocked down in human small cell lung cancer cell line NCI -H446 by siRNA-IDH-2.The expression level of IDH-2 was determined by real-time PCR and Western blotting .The cell proliferation was measured by CCK-8 as-say , the protein expression of MAPK p 42 was detected by Western blotting , and the cell cycle was analyzed by flow cytome-try.The migration was observed using Transwell cell migration system .BALB/c nude mice were subcutaneously injected on the back with NCI-H446 cells transfected with siRNA-IDH-2/negative control siRNA or non-transfected cells to study the tumor growth .RESULTS:siRNA-IDH-2 remarkably down-regulated the expression of IDH-2 and MAPK p42 in the NCI-H446 cells.siRNA-IDH-2 inhibited both the proliferation and migration abilities of NCI-H446 cells, and the cell cycle was arrested in S phase as compared with negative control group .Additionally, the volume of xenograft tumors in siRNA-IDH-2 group was significantly decreased as compared with control group .CONCLUSION:siRNA-IDH-2 down-regulates the expres-sion of IDH-2 in NCI-H446 cells, reduces the cell migration efficiency and inhibits the tumor growth in vitro and in vivo.
9.Study of the Influencing Factors of Myocardial Infarction Patients Care Burden of Caregivers

Objective Through the evaluation of acute myocardial infarction patients care burden of caregivers,to understand caregiver stress con?dition,and to investigate the effective predictors of acute myocardial infarction patients care burden of caregivers,so as to provide the basis for im?provement of the care burden. Methods A cross?sectional study was conducted through convenience sampling using Zarit Caregiver Burden Inter?view as the measuring tool. Questionnaires were collected by face?to?face structured interviews towards caregivers. Care burden of caregivers was eval?uated on these primary caregivers of patients with acute myocardial infarction. Results Caregivers of patients with myocardial infarction endure high care burden level. There were statistically significance among patients with different ages or revenue,the relationship of caregivers with patients,nurs?ing hours per day,psychological alignment and patients′self?care ability. Conclusion Caregivers of patients with myocardial infarction endure high care burden level. The relationship between caregivers and patients,the patient′s gender,nursing total time,nursing time every day,and the pa?tient's life self?care ability are contributed to be predictive factors. Future interventions should be strengthen on the psychological ability of the align?ment as well as measures to improve the self?care ability of the patient's life.
