Objective To investigate the effect of microRNA-204(miR-204)on autophagy in U251. Methods Inhibition of miR-204 in U251 cell lines was done by transfection of miR-204 inhibitor(AMO-204) .Cell viability was detected by MTT assay.The autophagy of U251 was tested by immunofluorescence tech-nique.The protein level of Beclin 1,LC3 and Bcl-2 was detected by Western blot.Results Cell viability was markedly increased after inhibition of miR-204 in U251 cells.The number of autophagosome was decreased.The levels of Beclin 1 and LC3 were decreased,the protein level of Bcl-2 was significant increased by transfection of AMO-204 in U251 cells(P<0.05).Conclusion MiR-204 might at least in part promote glioma via inhibi-ton autophagy,indicating that miR-204 might be a potential target for the treatment of glioma.

Objective:To investigate the relation between eotaxin, STAT6 and allergic rhinitis through comparing expressions of eotaxin and STAT6 in nasal mucosa of normal guinea pigs and of guinea pig allergic rhinitis models. Methods: Twenty-four healthy guinea pigs were equally randomized into 2 groups: normal control and allergic rhinitis models. Guinea pig allergic rhinitis models were established by the ovalbumin challenge method. Protein expressions of eotaxin and STAT6 in the nasal mucosa of the 2 groups were detected by immunohistochemical technique. Results: Expressions of eotaxin and STAT6 were found in the nasal mucosa of both normal guinea pigs and allergic rhinitis guinea pigs. Eotaxin was mostly located in the cytoplasm of the serous and mucous glands,while STAT6 mostly in the cytoplasm of subepithelial infiltrative cells.Expressions of eotaxin and STAT6 in the nasal mucosa of allergic rhinitis guinea pigs were significantly higher than those of normal guinea pigs~(P

Objective Studing the relationship among matrixmetal-loproteinnse2 (MMP2), the tissue inhibitors of metalloprotein-ase2 (TIMP2) and chnicopathological characteristics such as histological grade, clinicopathological stage of the rectal cancer. Assessing the significance of MMP2 and TIMP2 expression in rec-tal cancer for prognosis and treatment. Methods To test the expression level of MMP2 and TIMP2 in rectal caneer patients with immunohistochemisty method. Results were analysised with χ2 test. The Kaplan-Meier (Log-rank test) was used for univariate survival analysis. Results The positive rates of MMP2 and T1MP2 ex-pression in rectal cance are higher in comparison with normal tissues. With MMP2 have a trendence of increas-ing the positive expression of TIMP2 decreases. Conclusion MMP2 and TIMP2 relate obviously with the clini-copathological character of the rectal cancer. They may play an important role in the proliferation, invasion and of the cancer. Testing the level of expression of MMP2 and TIMP2 may be helpful to prejudge the prognosis of the patients with rectal cancer. Some new therapic methods relating to MMP2 and TIMP2 may give patients with rectal cancer a more satisfying treatment.

Objective To investigate value of color Doppler flow imaging(CDFI) and Doppler power imaging(CDPI) in the diagnosis of choroidal detachment(CD). Methods Color Doppler ultrasound was performed in 30 cases with choroidal detachment. At the base of eyeball shape, lens, vitreous and retina examined by ultrasound, intraocular band-shaped echo was examined by CDFI and CDPI, to understand its blood flow. The observation was compared with that of surgical results. Results Of 30 cases with choroidal detachment, the diagnostic positive correspondonce of ultrasound was 80.0%, while ultrasound combined with CDFI and CDPI, the correspondence increased up to 96.7%. The sonographic features of CDFI and CDPI in choroidal detachment patients were abundant blood flow in band-shape or short stick-like. In 3 cases out of 4 cases with retinal detachment, blood flow was not abundant,their blood flow showed a sparked pattern. In addition, 14 cases combined with vitreous hemorrhage, 8 cases had vitreous opacity and 4 cases vitreous proliferation. Ultrasound diagnosis corresponded with that of clinical surgery in 27 choroidal detachment cases. Conclusions Ultrasound combined with CDFI and CDPI provide valuable references for diagnosis and treatment of choroidal detachment.

Objective To study the prevalence of chronic kidney disease (CKD) among patients with bronchial asthma in Zhengzhou.Methods A total of 655 patients (older than 14years) were interviewed and received physical,uria and blood examination.Bronchial asthma and CKD were diagnosed according to related definition.Results The prevalence of albuminuria,hematuria,reduced eGFR and CKD was 9.41%,10.37%,3.03% and 17.38% respectively in the above special patients,which was higher than that of the general population.Women had higher prevalence of CKD than men (21.47% vs 13.99%,χ2=6.060,P=0.014).Prevalence of 1 to 5 stage of CKD was 8.61%,5.10%,3.03%,0.48% and 0.16% respectively.In the acute phase of asthma,prevalence of CKD (24.42%) was significantly higher than that in chronic duration and clinical remission period (χ2=12.445,P=0.002) and the same significant differences were found in albuminuria and reduced eGFR (χ2=19.619,P＜0.01 and χ2=9.305,P=0.010).Conclusion The bronchial asthma patients have higher incidence of renal damage and should be estimated,especially in the acute phase.

Objective To establish an experimental autoimmune encephalomyelitis (EAE) model in Wistar rats with myelin basic protein fragment (MBP69-85) and observe its pathological changes. Methods MBP69-85 dissolved in normal saline was mixed with complete Freund's adjuvant (CFA) (including 6 mg bacille Calmette Guerin) to prepare the encephalitogenic emulsion. Ten out of 70 Wistar rats were chosen as a control group, the others were divided equally into group A,B,C according to the difference of the encephalitogenic emulsion. Rats in group A were immunized with 50 μg MBP69-85 +CFA (including 6 mg BCG). Rats in group B were immunized with 25 μg MBP69-85+CFA (including 6 mg BCG). Rats in group C were immunized with 25 μg MBP69-85+CFA (including 12 mg BCG). The pathological changes of brain and spinal cord tissues were examined by light microscopy after HE staining and immunohistochemistry of MBP and NF. Results Some of the Wistar rats immunized with 50 μg MBP69-85 showed disorder at 12～16 days after immunization. The clinical symptoms included tail acratia or paralysis of tail and limbs, head tilt, etc. and the mean score was 2.38±1.89. There were infiltration of inflammatory cells inside nervous tissue and perivascular cuffings in HE stained sections. The immunohistochemistry of MBP and NF showed demyelination in the white matter and axon injury. Conclusion To some extent, the establishment of EAE depends on the dose of the immunizing antigen. The BCG in CFA was not the major cause of morbility of the rats. The EAE model induced with MBP69-85 in Wistar rats, showing typical clinical symptoms and pathological changes of multiple sclerosis, is a reliable animal model for the study of pathogenesis and treatment of multiple sclerosis.

Objective To discuss the effect of microRNA -27a on U251 glioma cells.Methods Over-expression or inhibition of miR -27a in U251 glioma cells were done by transient transfection of miR -27a mim-ics or AMO-27a in vitro.Cell viability was detected by MTT assay .Invasion ability of U251 was detected by tr-answell invasion assay.The level of miR-27a and PPARγwere detected by real -time PCR.Results Under in-hibition of miR-27a condition,the proliferation and invasiveness of U 251 glioma cells were decreased .The level of PPARγwas significantly increased ,whereas the level of miR-27 a was decreased .Overexpression of miR-27 a increased the proliferation and invasiveness of U 251 glioma cells and decreased the level of PPARγ.Conclusion Inhibition of miR-27a is benefit for inhibiting the proliferation and invasiveness of U 251 glioma cells.
