2.Effect of isoflurane on muscle relaxation produced by rocuronium

Objective To evaluate the potentiation of neuromuscular block produced by rocuronium during isoflurane anesthesia. Methods Sixty ASA Ⅰ -Ⅱ patients (39 male, 21 female), aged 18-56 yr, weighing 42-88 kg undergoing ENT, oral or facial plastic surgery were enrolled in this study. The patients were premedicated with atropine 0.01 mg?kg-1 . Anesthesia was induced with diazepam 0.1 mg?kg-1 , pethidine 1mg?kg-1 and droperidol 50?g?kg-1. Oro- or naso-tracheal intubation was accomplished under topical anesthesia. After intubation the patients received hydroxydione 2.5 g and fentanyl 5 ?g?kg-1 . The patients were mechanically ventilated. Anesthesia was maintained with inhalation of 50% N2O in O2 alone (groupⅠ) or combined with 0.6% isoflurane (group Ⅱ ) or 1.2% isoflurane (group Ⅲ ) . A bolus of rocuronium 0.6 mg?kg-1 was given during maintenance of anesthesia. Neuromuscular function was monitored using TOF response measured by accelerography (Biometer, Denmark). (1) Onset time (time from the end of injection to maximum depression of T1 ) , (2) the duration of T1 = 0, (3) time from the end of injection to recovery of T1 to 25% , 50% and 90% control, (4) recovery index (time from T, 25%-75%) were recorded. End-tidal isoflurane concentration was monitored.Results The onset time was significantly longer in group Ⅰ[ (1.7?0.2) min] than that in group Ⅱ[ (0.9?0.1) min] and groupⅢ[(0.8?0.1) min] ( P

Objective To investigate the association between single nucleotide polymorphisms (SNPs) of DOPA decarboxylase (DDC) and dopamine receptor-1 (DRD1) and clinical phenotype feature in autistic children.Methods TaqMan probes real-time polymerase chain reaction (PCR) was used to determine genotype and allele of SNPs of DDC gene (rs6592961) and DRD1 (rs251937) gene in 97 autism children.The Children Autism Rating Scale (CARS) was used to evaluate clinical phenotype feature.Results There was no significant difference in the distribution of the allelic frequency and genotype between mild-medium group and severe group of CARS scores (P > 0.05).For DDC gene (rs6592961),significant difference was found in subscale between genotypes G/G and A/A (P =0.043).For DRD1 gene (rs251937),significant difference was found in subscale between genotypes T/T and C/C (P =0.029).Conclusions In DDC gene (rs6592961),the children with G/G genotype were more obvious than the children with A/A genotype.In DRD1 gene (rs251937),the children with T/T genotype were more obvious than the children with C/C genotype.
4.Quantitative analysis of renal injury using CEUS in hypertensive rabbits model induced by L-NAME

Objective To assess the value of contrast-enhanced ultrasound (CEUS) on quantitative analysis of re?nal cortex perfusion in hypertensive rabbits model. Methods Hypertensive rabbit modal (n=10) were established by inject?ing N-nitro-L-arginin methylester (L-NAME). CEUS and Cystatin C (CysC) serum level analysis were performed at differ?ent time points:before and the 2nd, 4th, 6th and 8th week after injecting L-NAME. Time-intensity curve and area under curve (AUC) were analyzed quantatively while correlation of AUC and CysC were also analyzed. Results Serum level of Cys C in?creased significantly at the 6th week after L-NAME administration which is earlier than the increase of serum levels of Scr and BUN. AUC decreased at first then increased after L-NAME administration. Upon addition of L-NAME, rise time (RT) and peak intensity (PI) decreased while mean transit time (MTT), time from peak to one half (HPT) and time to peak (TTP) in?creased. Our study confirmed a positive correlation between AUC and Cys C (r=0.950, P<0.001). Conclusion Setting up rabbits model by L-NAME is convenient and reproducible, which is an useful tool in experimental study of preclinical and clinical phase of hypertensive renal injury. CEUS combining with CysC serum level analysis is considered as an effective technology for evaluating renal function in hypertensive patients.
5.Analysis of etiology and drug resistance of infectious diarrhea among children in Beijing area in 2009

Objective To investigate the etiology, epidemiologic features and drug resistance tendency of acute infectious diarrhea among children in Beijing area. Methods Enteric pathogenic bacteria were isolated, cultured and identified for serotype from the stool specimens of children with the initial clinical diagnosis of acute bacterial diarrhea in the intestinal clinic from January to October in 2009 ,and the antibiotic susceptibility of bacteria was tested by disk diffusion method. The cluster A rotavirus antigen was also detected by the qualitative technique of immunochromatographic double-antibody sandwich assay. Results Of the 256 stool specimens, 87 strains of 4 species of pathogenic bacteria were detected with the detectable rate of 34. 0% ,of which 2 strains were detected in one stool sample ,including 40 strains of salmonella (46. 0%) ,23 strains of shigella (26. 4%) ,2 strains of diarrheogenic escherichia coli (2. 3%) and 22 strains of staphylococcus aureus (25.3%). The positively detected patients consisted of 54 males and 32 females with the ratio of 1.69∶1 ,of whom 55 cases (64. 0%) were under 2 years of age. Of the 13 strains of shigella, 13 were sonnei shigella (56. 5%). And of the 22 strains of staphylococcus aureus,20 were detected among the infants under 1 year of age. The rates of crug resistance to certain antibiotics were lower in salmonella than in shigella (ampicillin :65. 0% vs. 95.7% ;compound sulfamethoxazole:20. 0% vs. 78. 3% ;ciprofloxacin:7.5% vs.8. 7% ;ceftriaxone: 15.0% vs. 73.9%). Of the 256 stool specimens ,47 were found positive for cluster A rotavirus,of whom 13 were also positive in stool bacteria culture. Conclusion Salmonella is the major pathogen among children with bacterial diarrhea in Beijing in 2009, and sonnei shigella is the main epidemic strains of shigella diarrhea. lnfants under 2 years of age are the susceptible population of the above two species of bacteria, while staphylococcus aureus mainly infect the infants under 1 years of age. Multi-resistance in shigella is still serious. The incidence of mixed infections of bacteria and rotavirus increases in children with infectious diarrhea.
6.Animal models of laparoscopy assisted natural orifice translumenal endoscopic surgery

Objective To explore the manipulation,feasibility and safety of laparoscopy assisted natural orifice translumenal endoscopic surgery(NOTES).Methods The technique was performed in 4 female porcine models.Diagnostic laparoscopy was performed first,and followed by transgastric access through the anterior gastric wall under the monitor of laparoscopy.The stoma was extended with the dilation ballon and the endoscope was sent to the abodominal cavity.Such procedures as endoscopic peritoneoseopy,fallopian tube ligation,oophorectomy,eholeeysteetomy and closure of gastrostomy were performed.The drainage tube and antibiotics of 3 days were used after operation according to the bleeding and potential risk of fistula and peritonitis.The animals were feeded on a semiliquid diet.Two weeks later,routine gastroseopy was pedbrmed to examine the healing of luminal incision.Subsequently,all models were sacrificed for the evaluation of the validity of NOTES.Results With the assistance of the laparoscope,endoscopic peritoneoseopy and,fallopian tube ligation,oophorectomy and cholecystectomy,twice for each,were successfully performed in all models.Abdominal drainage were used in two animals'after eholecystectomy.All porcines survived for 2 weeks postoperatively with no weight loss.Autopsic examination showed good healing of transgastric incisions,with little adhesion,no massive heinan'hage,abcesses,or injury to adjacent organs.Conclusion Combined laparoscopy and endoscopy for NOTES is feasible with relative safety.Laparoscopic assistance could facilitate the NOTES procedures at the present stage.

Objective To explore the expressions ofmiR-124* and laminin-8 in different grades of human gliomas,and analyze the potential regulatory relationship between them.Methods Thirty human glioma specimens of different stages and two healthy brain tissues,collected in our hospital from January 2011 to December 2012,were used in our study; Western blotting was performed to detect the laminin-8 and miR-124* protein expressions; plasmid vectors carried psiCHECKTM-2-wild-type (WT) laminin β1 chain 3 'untranslated region (3'UTR) and psiCHECKTM-2-mutant (MUT) laminin β1 chain 3'UTR were established,and they were,respectively,co-transfected with the miR-124* mimics,negative control ofmiRNA mimics,miR-124* inhibitors,negative control ofmiRNA inhibitors into the U87 cells;besides,cotransfection with psiCHECKTM-2 into U87 cells was used as blank control group; the fluorescence intensity of each group was determined by dual-luciferase assay 48 h after the cotransfection.Dual-luciferase assay was developed to verify the regulatory effects ofmiR-124* on laminin-8.The nu/nu nude mice were given subcutaneous injection of U87 cells to establish giloma animal models; the expression of β1 chain-containing laminin-8 protein was detected in gliomas by Western blotting; the expression of vascular endothelial cell marker CD31 was observed by immunohistochemical staining.Results Western blotting showed high expressions of laminin-8 protein in high-grade gliomas (WHO grade Ⅲ/ⅣV),but low expressions in low-grade gliomas (WHO grade Ⅰ/Ⅱ); real-time fluorescent quantitative PCR showed that the expression of miR-124* in high-grade gliomas was significantly lower than that in low-grade gliomas.Dual-luciferase assay confirmed that fluorescence intensity of U87 cells carried psiCHECKTM-2-WT laminin β1 chain 3'UTR and miR-124* mimics (2.13±0.25) was significantly weaker than that of cells carried psiCHECKTM-2-WT laminin β1 chain 3'UTR and negative control of miRNA mimics (2.71±0.08,P<0.05); that of those carried psiCHECKTM-2-MUT laminin β1 chain 3'UTR and miR-124* inhibitors (3.18 ±0.22) was significantly stronger than those carried,psiCHECKTM-2-MUT laminin β1 chain 3'UTR and negative control of miRNA inhibitors (2.70±0.17,P<0.05).The animal model results indicated the laminin-8 and CD31 expressions in miR-124* treatment group were obviously lower than those in control group.Conclusion MiR-124*,which can affect the malignant degrees of human gliomas through playing its negative regulation role in laminin-8 expression,maybe a target gene for the diagnosis and therapy of gliomas.

Objective To investigate the role of long noncoding RNA nuclear paraspeckle assembly transcript 1 (NEA T1) in regulating the activation of microglias.Methods (1) Microglias BV2 were routinely cultured in vitro and NEA T1 mRNA expression was detected by real-time PCR (RT-PCR) after 0,0.1,0.5 and 1 μg/mL lipopolysaccharide (LPS) stimulation for 6 h;NEAT1 mRNA expression was detected by RT-PCR after 1 μg/mL LPS stimulation for 0,6,12 and 24 h.(2) NEAT1 siRNA (NEA T1-si) was transfected into BV2,and RT-PCR and Western blotting were employed to detect the LC3 mRNA and protein expressions.(3) The BV2 cells were divided into control group,NEAT1-si group,LPS group and NEAT1-si+LPS group;RT-PCR was used to detect the tumor necrosis factor-α (TNF-oα) and inducible nitric oxide synthase (iNOS) mRNA expressions;morphological changes of BV2 cells were observed under inverted microscope.(4) The BV2 cells were divided into control group,negative control+Torin-1 group and NEAT1-si+Torin-1 group;Western blotting was used to detect the LC3 protein expression and LC3,TNF-α and iNOS mRNA expressions were detected by RT-PCR.Results (1) NEA T1 was significantly up-regulated in LPS-stimulated BV2 cells in dose-and time-d ependent manners;significant differences were noted between each two groups (P<0.05).(2) The LC3-Ⅱ mRNA expression in the NEAT1-si group was significantly decreased as compared with that in the control group (P<0.05);LC3-Ⅱ/Ⅰ protein ratio (0.7) in the NEAT1-si group was significantly lower than that in the control group (1.03,P<0.05).(3) As compared with those in the control group,the TNF-α and iNOS mRNA expressions in the NEAT1-si group were decreased;As compared with those in the LPS group,the TNF-oα and iNOS mRNA expressions in the NEA T1-si+LPS group were significantly decreased (P<0.05).(4) LC3-Ⅱ/Ⅰ protein ratio in the control group,Torin-1 group and Torin-l+NEAT1-si group were 0.7,1.14 and 0.97,respectively;LC3-Ⅱ,TNF-α and iNOS mRNA expressions in the Torin-1 group were significantly higher than those in the control group (P<0.05);LC3-Ⅱ,TNF-α and iNOS mRNA expressions in the NEAT1-si+Torin-1 group were significantly higher than those in the Torin-1 group (P<0.05).Conclusion Knockdown of long noncoding RNA NEA T1 could attenuate microglia activation through inhibiting autophagy,and NEA T1 maybe the key molecule for the mitigation and cure of the neuroinflammation related diseases.
10.Exercise-induced muscle damage and physiotherapy: a visualized analysis

ObjectiveTo conduct a visualized analysis of the researches related to the physiotherapy for exercise-induced muscle damage, identify the present situation, and predict the hotspots and developing trends. MethodsRelevant literature about physiotherapy of exercise-induced muscle damage were collected in Web of Science Core Collection from the establishment of the database to October 31, 2022, and CiteSpace 6.1.R3 software was used for visualized analysis. ResultsA total of 357 articles were included, involving 51 countries/regions, 532 authors and 346 institutions. The annual number of documents issued showed an overall upward trend. The country with the highest number of documents was the United States, the organization was Edith Cowan University, and the author was Howatson Glyn. Keywords with high attention in the last three years were performance, eccentric exercise, recovery and soreness. Cited document clustering words were exercise-induced muscle damage, delayed-onset muscle soreness, foam rolling and exercise performance. ConclusionExercise-induced muscle damage is a muscle micro-injury, and the most important concerns are post-injury muscle soreness, myofascial pain and ultrastructural changes. Researches about physiotherapy of exercise-induced muscle damage are gradually increasing. Main physiotherapy treatments include cryotherapy, cold water therapy, massage, foam rolling and eccentric exercise. Presently, eccentric exercise is a hot topic in this field, and foam rolling would be hot in the future.
