1.Investigation of the value in clinical diagnosis of two dimers (D-D), fibrinogen (Fbg) and platelet parameters (PLT, MPV, PDW) level induced by postoperative deep vein thrombosis (DVT) in patients with severe trauma

Objective To investigate the D-in patients with severe trauma in two dimers (D-D),fibrinogen (Fbg) and platelet parameters (PLT,MPV,PDW) level induced by postoperative deep vein thrombosis (DVT) in the formation of value in clinical diagnosis.Methods Eighty cases of healthy people as control group,were compared before and after operation and level and 321 severe traumatic patients were measured before surgery,after 1 days and 3 days of D-D,Fbg and platelet parameters,trauma were divided into fracture,traumatic brain injury,multiple injuries in three groups.Results Different trauma patients with D-D,Fbg concentration and platelet index compared with the control group increased obviously significant (P < 0.05),especially the brain trauma and multiple injuries in two group increased significantly (P <0.01),the positive rate of D-D was 22.8%,35.2%,40.9%.Postoperative 1D in addition to brain injury group,trauma group D-D levels compared with those before operation were significantly increased with significant (P < 0.05),other indicators compared with preoperative changes little,the positive rate of D-D increased slightly,but without statistical significance.Levels of Fbg 3D after the trauma group decreased significantly,D-D increased significantly,decreased the number of PLT,MPV,PDW value increases,the value of preoperative has the remarkable significance (P < 0.01 or P < 0.05).The positive rate of D-D was increased up to 41%,59.2%,62.3%,compared with those before operation by chi-square test was significant (P <0.01).Conclusion The patients with severe trauma patients after two D-dimer concentration should be alert to the occurrence of DVT,contribute to the clinically as soon as possible to take measures to avoid,there is an important clinical significance in the treatment and prognosis of trauma patients.

3.Morphologic effect of huangjiao granule on apoptosis of cerebral neurocytes in elderly rats with middle cerebral arterial occlusion

BACKGROUND: Huangiao granule is pure Chinese drugs pharmaceu-tics, which can eliminate heat by purgatingfu-organs, clear unconscious-ness and stop endogenous wind as well as promote blood flow and removeblood stasis etc.OBJECTIVE: To observe the effect of huangjiao granule on apoptosis ofneurocytes in rat models with middle cerebral arterial occlusion, and probeinto the possible mechanism of huangjiao granule in the treatment of acutecerebral infarction.DESIGN: Complete randomized grouping design, control animal experi-ment.SETTING: Department of Neurology, Affiliated Hospital of TraditionalChinese Medicine Combined with Western Medicine, Tongji Medical Col-lege of Huazhong University of Science and Technology.MATERIALS: The experiment was conducted in the Central Laboratory,Hubei College of Traditional Chinese Medicine. Sixty old Wistar rats aged22 months were selected. After 3-day-feeding for adaptability, rats wererandomized into 6 groups according to body mass: Normal control group,model group, nimodipine gorup, 10 g/kg huangjia ogranule group and 5 g/kghuangjiao granule group with 10 rats in each group.METHODS: ①Rats were made into cerebral ischemic models with mid-dle cerebral arterial occlusion method. Thirty minutes before modeling, ratsin the 10, 5, 2.5 g/kg huangjiao granule (main gredients were radix et rhi-zoma rhei, cornu bubali etc. provided by Manufacturing Laboratory ofWuhan First Hospital with the batch number of 021120) groups respec-tively received gastric perfusion of 1, 0.5, 0.25 g/mL huangjiao water so-lution containing crude drug at the dose of 10, 5, 2.5 g/kg with each rats perfused once; Rats in the nimodipine group received gastric perfusion of1 mg/mL nimodipine water solution, containing crude drug at the dose of 10 mg/kg with each rats perfused once; Rats in the model group and normal control group received gastric perfusion of normal saline at the dose of 10 mL/kg with each rats perfused once. ② Apoptotic stainning sections in situ were observed under light microscope, 5 fields were observed under high power (20×10). Observed the number of mean positive cells of each highpower field, and then caculated the percentage of apoptosis. Histologic changes of local and peripheral ischemic cortexes were observed under light microscope. Changes of ultramicrostructure of cells in cerebral cortexes of rats were observed under electron microscope. ③Differences of measurement data were compared with F test and Q test.MAIN OUTCOME MEASURES: Morphologic effects of huangjiao granule on apoptotic rate of neurocytes in brain tissues of old rats with middle cerebral arterial occlusion (MACO) and cells in cerebral cortexes.RESULTS: A total of 60 Wistar rats were involved in the analysis of results. ①Number of apoptotic positive cells in brain tissues and apoptotic rate in rats of model group were significantly higher than those in the normal control group (P < 0.01); Those in each huangjiao granule groups and nimodipine group were obviously lower than those in the model group (P < 0.01); Differences in numbers of apoptotic positive cells in brain tissues and apoptotic rates in rats of 10 g/kg huangiao group and nimodipine groupwere not obvious (P < 0.05). Number of apoptotic positive cells in brain tissues and apoptotic rate in rats of 10 g/kg huangjiao granule group were markedly lower than those in the 5, 2.5 g/kg huangjiao granule groups (P < 0.05, 0.01).②Observation with light microscope and electron microscope showed that there were obvious apoptosis in local cells of brain tissues in rats of model group, which can significantly ameliorate by haungjiao granule.CONCLUSION: Huangjiao granule has some antagonistic effects on acute ischemic apoptosis of neurocytes, which may be one of the possible mechanisms of huangjiao granule in the treatment of acute cerebral infarction.
4.Effects of fluvastatin on the tubulointerstitium in progressive diabetic kidney disease

Objective To investigate the effects of fluvastatin on the tubulointerstitium damage in progressive diabetic kidney disease. Methods A rat model of type 2 diabetic nephropathy (DN) was developed successfully by combination of dietary induced insulin resistance and low dose STZ induced hyperglycemia after unilateral nephrectomy. Female SD rats were randomly divided into three groups: control rats, type 2 diabetic rats and type 2 diabetic rats treated with fluvastatin (2mg/kg/d). After 6 weeks, blood glucose, serum insulin, serum triglyceride and cholesterol, serum creatinine, and urinary protein were measured respectively. The protein expressions of c Jun and tansforming growth factor (TGF) ? 1 were studied by immunohistochemistry. TGF ? 1 gene expression was studied with a RT PCR technique. 5.Genotyping of K-ras exon 2 codons 12 and 13 mutations in colorectal cancer by pyrosequencing

ObjectiveTo investigate the clinical significance of pyrosequencing assay for determining K-ras mutations in exon 2 codons 12 and 13 in clinical colorectal cancer tissues.Methods Genomic DNA,extracted from K-ras mutant cell lines SW480 (homozygous,c.35G > T), DLD-1 (heterozygous,c.38G > A) and wild-type HT-29,was first used as the sequencing template respectively to test the accuracy of pyrosequencing methodology.The SW480 and DLD-1 DNA was separately mixed with wild-type HT-29 DNA in proportions of 2%,3%,5%,10%,20%,30% and 50%,the sensitivity for mutation detection was measured separately by pyrosequencing assay and directed Sanger DNA sequencing in the serial DNA mixture samples.The pyrosequencing assay results were compared with the corresponding Sanger sequencing and the datas were analysized by Fisher exact test.Pyrosequencing analysis was then performed for screening K-ras exon 2 mutations at codons 12 and 13 on DNA isolated from a panel of 30 colorectal cancer samples derived fromclinicalformalin-fixed andparaffinembedded(FFPE)tissues.ResultsCancer cell lines with known K-ras mutations ( SW480 and DLD-1 ) were readily detectable by pyrosequencing-based analysis.When the proportions of mutant colorectal cancer cell line DNA were 5% and 10% content,the mutation rates of K-ras gene detected by conventional Sanger DNA sequencing were 33.3% (4/12) and 58.3% (7/12) respectively,whereas the mutation rates detected by pyrosequencingbased assay were 91.7% (11/12) and 100%(12/12) respectively,there were significant differences between those two sequencing methodology ( P <0.05).Furthermore,we found 10 patients with K-ras exon 2 point mutations at codons 12 and 13 by pyrosequencing-based assay from 30 colorectal cancer FFPE tissues,the point mutation rate was 33.3% (10/30) and all of the mutations determined were heterozygous.The codon 12 was most frequently affected [30% (9/30)].Mutations with the highest frequency were G > A transitions [ 50% ( 5/10 ) ],followed by G > T transversions [ 30% ( 3/10 ) ].Conclusion The pyrosequencing assay provides an accurate and sensitive method for mutation screening of K-ras exon 2 codons 12 and 13 in routine diagnostic specimens,thereby allowing the selection of the cancer treatment in clinical individualized practice.
6.Expression of AQP1 and AQP5 is decreased in the alveolar-capillary membrane in rats with acute lung injury

Objective To determine if aquaporin1 ( AQP1) and aquaporin5 ( AQP5) are expressed in the alveolar-capillary membrane in rats, and to investigate the changes of AQP1 and AQP5 expression in the rat with acute lung injury.Methods The distribution of AQP1 and AQP5 in alveolar capillary membrane was investigated by immunohistochemistry and immunoelectron microscopy with affinity-purified antibodies to human AQP1 and AQP5.The possibility that alveolar capillary membrane AQP1 and AQP5 undergo altered regulation was studied by a rat model established using intra-tracheal instillation of lipopolysaccharide (LPS).Results Immunolabelling showed that AQP1 was stained primarily in the microvascular endothelium of normal lungs, while AQP5 was expressed in type I pneumocytes. Immunohistochemical analysis showed a significant decrease in the expression of AQP1 and AQP5 in injured lungs at 4 -48 h after LPS instillation.AQP1 protein was resumed partly at 24 h after LPS instillation and steroid administration, whereas AQP5 was unchanged.Conclusions The decreased expressions of AQP1 and AQP5 in injured lungs suggest that both of them may play a role in abnormal fluid transportation.
7.Association analysis of IL-23R polymorphisms in allergic rhinitis.

OBJECTIVE:
Polymorphism of interleukin-23 receptor (IL-23R) gene was found being related with the susceptibility to several immune-related diseases. For the terminal differentiation of IL-17-producing effecter T-helper cells in vivo, the IL-23R gene is very important. As proved recently, Th17 cells might have a great influence to the pathogenesis of allergic airways disease. Our intention was paid to find any association between the polymorphisms in the IL-23R gene and allergic rhinitis(AR) in Chinese population.
METHOD:
A group of patients was involved in a case-control comparison, consisted of 239 AR patients and 271 control Chinese subjects. The study claimed to take some blood samples for DNA extraction and select 3 single-nucleotide polymorphisms in IL-23R individually genotyped by the PCR-RFLP method.
RESULT:
Comparison with the controls, a great growing prevalence of the homozygous rs7517847 GG genotype and G allele appeared in AR patients (P < 0.05, respectively). Besides, a great increased frequency of the GGA haplotypes was presented in AR patients in comparison with the controls (P < 0.05).
CONCLUSION
The results demonstrated a important association pattern between polymorphisms in IL-23R and AR in Chinese population. Between rs7517847 in an SNP of IL-23R and AR, a great association was identified.

8.Downregulation of lanosterol synthase gene expression by antisense RNA technology in Saccharomyces cerevisiae.

The cyclization of 2,3-oxidosqualene is the key branch point of ergosterol and triterpenoid biosynthesis. Downregulation of 2,3-oxidosqualene metabolic flux to ergosterol in Saccharomyces cerevisiae may redirect the metabolic flux toward the triterpenoid synthetic pathway. In our study, primers were designed according to erg7 gene sequence of S. cerevisiae. Three fragments including 5' long fragment, 5' short fragment and erg7 coding region fragment were amplified by PCR. 5' long fragment consists of the promoter and a part of erg7 coding region sequence. 5' short fragment consists of a part of promoter and a part of erg7 coding region sequence. These fragments were inserted reversely into pESC-URA to construct antisense expression plasmids. The recombinant plasmids were transformed into S. cerevisiae INVSc1 and recombinant strains were screened on the nutritional deficient medium SD-URA. The erg7 expression level of recombinant strains, which harbored antisense expression plasmid of erg7 coding region, was similar to that of INVScl by semi-quantitative PCR detection. But erg7 expression level of recombinant strains, which harbored 5' long antisense fragment and 5' short antisense fragment, was significantly lower than that of the control. The results of TLC and HPLC showed that the ergosterol content of recombinant strains, which harbored 5' long antisense fragment, decreased obviously. The ergosterol contents of the others were almost equal to that of INVSc1. Lanosterol synthase gene expression was downregulated by antisense RNA technology in S. cerevisiae, which lays a foundation for reconstructing triterpenoid metabolic pathway in S. cerevisiae by synthetic biology technology.
9.The basic strategies and research advances in the studies on glycosyltransferases involved in ginsenoside biosynthesis.

Traditional herbal medicines, Panax ginseng, Panax quinquefolium and Panax notoginseng, attract our attention for their extensive and powerful pharmacological activities. Ginsenosides are the main active constituents of these medicinal herbs. The related glycosyltransferases involved in ginsenoside biosynthesis are the key enzymes which catalyze the last important step. Modification of ginsenoside aglycones by glycosyltransferases produces the complexity and diversity of ginsenosides, which have more extensive pharmacological activity. At present, ginsenoside aglycones and compound K have been obtained by synthetic biology. As the last step of ginsenoside biosynthesis, glycosylation of ginsenoside aglycones has been studied intensively in recent years. This review summarizes the basic strategies and research advances in studies on glycosyltransferases involved in ginsenoside biosynthesis, which is expected to lay the theoretical foundation for the in-depth research of biosynthetic pathway of ginsenosides and their production by synthetic biology.
10.Prevalence and prevention of Kaschin-Beck disease in different ecotypic areas in Gansu Province

Objective To grasp the epidemic features of Kaschin-Beck disease in different ecotypic areas in Gansu Province,in order to provide ecological basis for Kashin-Beck disease control.Methods Totally 37 counties with Kashin-Beck disease were divided into four ecological areas,villages with historical serious condition in township and townships with historical serious condition in county were investigated.Clinical examination and Xray of right hand of all 7-12-year-old children in the villages were carried out,while investigation of prevention and control measures was performed.Results Among 27 966 children from four ecological areas,the clinical detection rate was 0.05% (14/27 966) and the X-ray positive rate was 1.26% (353/27 966),metaphyseal rate was 1.25% (350/27 966),bone-side positive rate was 0.01% (3/27 966) and no case of epiphyseal and carpal were discovered.The clinical detection rate in children in the four ecological areas was not statistically significant (x2() =7.757,P >0.05),the Loess Plateau-gully region of Longdong [0.09% (10/11 604)] > the Anyon area of Qinling of Longnan [0.04% (3/7 969)] > Alpine grassland meadow area of Gannan [0.02% (1/4 021)] > the Loess Plateau-hills region of Longzhong [0 (0/4 372)].The detected rate of X-ray in four ecological areas was statistically significant (x2 =18.133,P < 0.05),the Anyon area of Qinling of Longnan [1.49% (119/7 969)] > the Loess Plateau-gully region of Longdong [1.41% (164/11 604)] > the Loess Plateau-hills region of Longzhong [0.89% (39/4 372)] > Alpine grassland meadow area of Gannan [0.77% (31/4 021)].Comprehensive prevention and control measures on Kaschin-Beck disease were different in different ecotypic areas.Relocation,long-distance education and selenium supplement measures had not been implemented in the four ecological areas.Conclusion The epidemic situation of Kaschin-Beck disease is different in different ecotypic areas in Gansu Province,which maybe related to ecological environment,but is under a state of control.
