1.Effect and mechanism of umbilical cord blood stem cells in the treatment of stroke

BACKGROUND: Studies have confirmed that umbilical cord blood stem cells can improve the neurologicalfunction after stroke, but the mechanism of umbilical cord blood stem cells in the treatment of stroke is unclear.OBJECTIVE: To review the effect and mechanism of umbilical cord blood stem cells in the treatment of stroke.METHODS: The first author searched PubMed and CNKI databases for relevant articles published from January2005 to December 2015 using key words of umbilical cord blood stem cells, stroke in English and Chinese,respectively. RESULTS AND CONCLUSION: With the further research on the neural differentiation of umbilical cord blood stem cells,umbilical cord blood stem cells have become a hot research topic in the field of neural system disease therapy. Atpresent, some experimental studies on cell transplantation for stroke have achieved ideal curative effects. However, themechanism of umbilical cord blood stem cells in the treatment of stroke is not clear. Known therapeutic mechanismsinclude the recombination of blood vessels, nerve loops and other damage tissue structures, secretion of variousnutritional factors to reduce endogenous apoptosis, promotion of endogenous blood vessels and nerve regeneration, etc.But the following problems need to be further studied, including whether the transplanted cells can be fully filled toreplace necrotic nerve cells and promote recovery of nerve function; how these survived stem cells establish the neuralconnections; how these transplanted cells reduce the apoptosis and necrosis of the host cells to restore the cell function.
2.Clinical study of plasma homocysteine, serum folic acid and vitamin B_(12) in ischemic leukoaraeosis

Objective To investigate the clinical importance and the relationship between the levels of plasma total homocysteine (Hcy), serum folic acid and vitamin B 12 , and the occurrence and development of ischemic leukoaraeosis (LA). Methods Ischemic LA group and normal control group with 100 cases for each were studied. The 100 cases in ischemic LA group were firmly diagnosed by CT or / and MRI. The levels of plasma total Hcy, serum folic acid and vitamin B 12 of the patients in ischemic LA group were measured, and compared with that in control group. Results The level of plasma Hcy in ischemic LA group (19.79?5.25?mol/L) was significantly higher than that in control group (12.02?2.11?mol/L), while the levels of serum folic acid and vitamin B 12 in ischemic LA group (2.08?0.84ng/ml and 140.13?45.49pg/ml) were lower than that in control group (4.39?1.30ng/ml, 297.45?120.23pg/ml). 3.Asian Resilience Scale’s Preliminary Revision, Reliability and Validity in Chinese College Students

Objective: To explore the Asian Resilience Scale’s preliminary revision, reliability and validity in college students of China. Methods: 646 students were investigated with the Asian Resilience Scale and the short form of NEO Five-Factor Inventory. Results: ①Three factors were extracted by exploratory factor analysis and explained 47.1% of variance. ②Confirmatory factor analysis showed good fit between the data and the construct of scale, ?2/df=2.36,RMSEA= 0.055,GFI=0.923,CFI=0.908,NNFI (TLI)=0.894. ③The Cronbach’s ? coefficients, split-half and retest reliability of the total scores were 0.865, 0.836 and 0.894 respectively. ④The total score of the Asian Resilience Scale was significantly negatively correlated with neuroticism and positively correlated with extraversion, openness to experience, agreeableness and conscientiousness. Conclusion: Reliability and validity of the Asian Resilience Scale’s revised Chinese version is corresponded with psychometric standard, and it could be used to assess the resilience of Chinese college students.
4.Expression of chemokine receptor CCR7 and VEGF-C is associated with prognosis of breast carcinoma

AIM: To explore the protein levels of chemokine receptor 7 (CCR7) and vascular endothelial growth factor ( VEGF)-C in breast carcinoma , and to investigate the effects of CCR 7 and VEGF-C on prognosis of breast carcinoma.METHODS:The protein expression levels of CCR 7 and VEGF-C in the breast carcinoma tissues and normal breast tissues were detected by the method of immunohistochemistry .At the same time, the relationship between clinico-pathologic characteristics and the protein expression of CCR 7 and VEGF-C in the breast carcinoma tissues was analyzed . The relationship between the protein expression of CCR 7 and VEGF-C and survival time of the breast cancer patients was estimated by Kaplan-Meier method.RESULTS:The positive expression rates of CCR 7 and VEGF-C in the breast carcino-ma tissues were significantly higher than those in the normal breast tissues (P<0.01).A positive correlation was observed between the protein expression of CCR7 and the protein expression of VEGF-C in the breast carcinoma tissues (r=0.613, P<0.01).The protein expression of CCR7 and VEGF-C was correlated with lymph node metastasis and TNM stage (P<0.05), but both were not related to patients ’ age, primary tumor size, estrogen receptor and progesterone receptor .The survival time of the patients with CCR 7 and VEGF-C positive expression was significantly shorter than that of the patients without the expression (P<0.05).CONCLUSION:The positive expression of CCR7 and VEGF-C proteins is associated with the prognosis of breast cancer , and combined detection of CCR 7 and VEGF-C protein expression levels may be helpful to judge the prognosis of breast cancer .
5.Effect of laparoscope anesthesia inducted by sevoflurane combined with different doses of propofol on VEGF-C and PGE2 in patients with hysteromyoma

Objective To investigate effect of laparoscope anesthesia inducted by sevoflurane combined with different doses of propofol on vascular endothelial growth factor-C (VEGF-C) and prostaglandin E2(PGE2)in patients with hysteromyoma.Methods 77 cases of patients with uterine fibroids were selected and divided into two groups.40 cases in group A were anesthetized inducted by 2.0 mg/kg propofol combined with sevoflurane, 37cases in group A were anesthesia inducted by 1.0 mg/kg propofol combined with sevoflurane.VEGF-C and PGE2 in serum were compared before and 48h after the operation.Results Compared with preoperation, VEGF-C and PGE2in serum were lower (P<0.05), VAS score was lower (P<0.05);Compared with group B, VEGF-C and PGE2 in serum of the group A were lower (P<0.05), VAS score was higher (P<0.05).Conclusion Different doses of propofol on serum VEGF-C and PGE2 in patients with laparoscopic uterine fibroids have different effects, 1.0 mg/kg has a sedative effect, 2 0 mg/kg with anesthesia induction.
6.Proteomic analysis of energy metabolism-related proteins of mice midbrain

Objective To identify energy metabolism-proteins of mice midbrain by using the proteomic technique,and to investi-gate the relationship between these proteins and neural diseases.Methods Two-dimensional gel electrophoresis was used to sepa-rate totally soluble proteins extracted from mice midbrain.Some protein spots on two-dimensional gel electrophoresis gels were ana-lyzed by using matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS).Results 24 protein spots related with energy metabolism were separated by two-dimensional gel electrophoresis and identified by using MALDI-TOF MS successfully.Conclusion The establishment of energy metabolism-related proteins map of mice midbrain lays a foundation for the research on the involvement of these proteins in neural disease pathogenesis.
7.Reliability and validity of the Chinese revised version of the intolerance of uncertainty scale

Objective To explore the reliability and validity of the Chinese revised version of the Intolerance of uncertainty scale(IUS).Methods A total of 930 college students completed the IUS.45 college students participated in the retest 3 weeks later.The self-rating anxiety scale (SAS) and the self-rating depression scale (SDS) were used as criterions for assessment of the validity of the Chinese revised version of IUS.Results The Chinese revised version of IUS contained 21 items.Exploratory factor analysis found a two-factor model (negative belief and emotional experience,behavioral dysfunction and negative self-reference) with a cumulative variance contribution rate of 42.09%.And the results of Confirmatory factor analysis (x 2/df =2.82,GFI =0.90,TLI =0.90,CFI =0.91,RMSEA =0.065) indicated that this model provided a reasonable good fit for Chinese college students.The Cronbach α of internal consistency reliability were good for IUS and two subscales,which were 0.929,0.885 and 0.864,respectively.The test-retest reliabilities for IUS and two subscales were 0.782,0.766 and 0.714 respectively.The IUS scores were positively correlated with the scores of SAS and SDS (r =0.489 ~0.575,P < 0.001).Conclusion It suggests that the Chinese revised version of IUS has good reliability and validity,which is qualified enough for relevant research and applications for Chinese adults.
8.Preparation and identification of monoclonal antibody specific to the epithelial cell adhesion molecule

Objective To prepare and identify the monoclonal antibody (mAb) specific to epithelial cell adhesion molecule (EpCAM) and explore the function of the mAb.Methods The EpCAM antigen expressed by the prokaryotic expression systems was used to immunize the BALB/c mice,and then the splenic cells from the mice were fused with the Sp2/0 cells to produce hybridomas secreting specific mAb.The positive clones were screened by the ELISA.The western blot analysis was used to identify the reactivity of the mAb to the antigen.Then the immunohistochemistry staining was used to detect EpCAM expression in the 3 primary colorectal carcinoma tissues.Results Three mAb specific to EpCAM were obtained by ELISA tests.Western blot results indicated that these three kinds of antibodies could react to the EpCAM antigen,but no response to the GST tag.Immunohistochemical staining results identified that these mAb could give positive signals to the primary colorectal carcinoma tissues from patients.Conclusion Three mAb specific to EpCAM are obtained and identified,which contributes to the diagnosis and therapy of the carcinoma in the future.
9.Strain-dependent effects of fluoxetine on tail suspension test in mice

Objective To study the strain difference in response to the antidepressant fluoxetine in mouse tail suspension test.Methods Two outbred mouse strains (KM and ICR) and three inbred mouse strains (C57BL/6,Balb/c and DBA/2) were used in this study.They were treated with the selective serotonin reuptake inhibitor (SSRI) fluoxetine or saline and the immobility time in tail suspension test was recorded.Results There was significant difference of baseline immobility time among different stains with C57BL/6 the most immobile((145.0 ±16.8) s) and DBA/2 the least ((34.5 ± 6.1)s).Fluoxetine significantly decreased the immobility time in C57BL/6 ((116.0 ± 10.3) s vs (145.0 ± 16.8) s) of control),Balb/c ((44.3 ± 6.2) s vs (75.3 ± 10.3) s) of control) and DBA/2 mice ((16.6 ± 4.3) s vs (34.5 ± 6.1) s) of control),while the immobile time of KM and ICR mice was not influenced by fluoxetine.Conclusion The effects of fluoxetine in tail suspension test are strain dependent.Fluoxetiue exhibits antidepressant effects in C57BL/6,Balb/c and DBA/2 mice,but not in KM and ICR mice.
10.Statistical concepts related to negative clinical results.

It is a trend that clinical researchers overemphasize on hypothesis testing and the P value. However, P value has some limitations in the presentation of study results. Power is a tool for study design and results explanation, especially for negative results. Confidence interval contains more information for clinical results than P value. P value, power, and 95% confidence interval should be provided in the clinical reports to facilitate the interpretation of the results.
