Purpose:To evaluate the protection of astragalus injection for Doxorubicin (adriamycin)-associated cardiotoxicity. Methods:58 patients were randomized into two groups for this clinical trial. 30 patients were in the test group ,and 28 in control group. Patients in the test group received astragalus injection in 5% glucose solution once every day for two weeks starting three days before adriamycin-based chemotherapy. 28 patients in the control group began to receive oral vitamin E 100 mg twice a day and coenzyme Q10 20 mg three times a day for two weeks. Chemotherapy is the same as that in the test group. Electrocardiogram (ECG) and echocardiogram were employed to evaluate the cardiac function. Results:There was a substantial difference in ejection fracton between the two groups, but the difference was not statistically significant. Statistically significant differences were observed in the two groups with regard to all other parameters, including abnormal changes in ECG and LVIDD、LVISD、A/E、FS. Conclusions:Astragalus injection is a good drug which can prevent the occurrence of acute cardiotoxicity associated with adriamycin. It can also reduce the occurrence of chronic cardiotoxicity.

Objective To study the effects of costimulatory blockade with anti-inducible costim- ulator antibody(ICOS mAb)in combination with CTLA4Ig on prevention of islet allograft rejection. Methods Experimental animals were randomly divided into 4 groups(10 rats in each group).CT- LA4Ig + ICOS mAb group(group A):intraperitoneal injection of CTLA4Ig on day 0,2,4 and ICOS mAb on day 1,3,5 after islet transplantation;ICOSmAb group(group B):intraperitoneal injection of ICOS mAb on day 1,3,5 after islet transplantation;CTLA4Ig group(group C):intraperitoneal injection of CTLA4Ig on day 0,2,4 after islet transplantation;control group(group D):simple islet transplantation. The islet allograft survival and pathological changes in the transplanted islets after transplantation were observed.By using RT-PCR,the expression of IL-2 and IL-10 mRNA in the transplanted islets was detected. The expression of CD4~+ and CD8~+ T cell was detected by flow cy- tometry. Results In group A,the survival time was obviously prolonged as compared with other three groups and the transplanted islets were near normal under a light microscope. As compared with other three groups, the expression of IL-2 mRNA was significantly decreased in group A(P0.05).The expression of CD4~+ and CD8~+ T cell was not obviously up-regulated on the day 21 after transplantation. Conclusion The blockade of costimulatory signals with ICOS mAb in combination with CTLA4Ig has a favorable effects to restrain the rejection of islet transplantation.

Objective:To study the anti-proliferative effect of lupeol on human bladder cancer T24 cell line and the regulating mechanism for p53/miR-34a signaling. Methods:CCK-8 assay was performed to evaluate the effects of lupeol at different concentra-tions on cell viability in 24 h and 48 h. Caspase inhibitors were used to identify the subtypes of Caspase during lupeol induced cell death. The effects of lupeol on the expression of total p53 protein and miR-34a were evaluated by western blot and real-time PCR, re-spectively. The effects of lupeol on downstream targets of miR-34a were quantified by real-time PCR. Results:Lupeol could inhibit the proliferation of T24 cells in a dose-dependent manner. The IC50 of lupeol was (77. 23 ± 6. 78) μmol·L-1 in 24 h. Compared with the control group, lupeol could elevate the expressions of p53 and miR-34a (P<0. 01). Moreover, the mRNA expression of miR-34a tar-gets, Bcl-2, CD44 and c-Myc were significantly down-regulated after the treatment with lupeol (P<0. 01). Conclusion:Lupeol can inhibit T24 cell proliferation, which is related with the regulating effects on p53/miR-34a signaling.
