Objective To investigate the expression of multidrug resistance gene 1 product P-glycoprotein(P-gp) when nuclear factor kappa B is induced or inhibited.Methods Ovarian cancer cells SKOV-3 were treated with Docetaxel or combinded with pyrrolidine dithiocarbamate(PDTC),the inhibitor of NF-?B.Western blot assay、 flow cytometry assay and MTT assay were used to measure P65 protein,P-gp,apoptosis of cancer cells,and the survival rate respectively.Results Docetaxel increased P65 protein and P-gp expression,while combind use of PDTC reversed this function.Both Docetaxel(≥(1 ?g/mL))and PDTC(≥(10 ?mol/L)) significantly inhibited the growth of SKOV-3 cells,and caused apoptosis.The combined use of Docetaxel and PDTC at low concentrations significantly inhibited the growth of cancer cells and caused apoptosis as compared with those treated with Docetaxel only.Conclusion The expression of P-gp may be ralated to the activation of NF-?B;Inhibition of NF-?B can enhance the chemosensitivity of ovarian cancer cells to Docetaxel.

Objective Probe application value of CHN(the standards of skeletal maturity of hand and wrist for Chinese-CHN method)bone age in judicial identification,and analyze factors and related problems which influence its accuracy.Methods 522 patients participated in judicial identification of living age estimation.Two posteroanterior radiographs for the left wrist were shot with respective exposure value for all patients.And two oblique radiographs were added.Bone ages were estimated according to CHN method.Result Among 522 cases were 489 males and 33 females.Bone ages of 129 males reached to tiptop of CHN-18.4 and average bone age of other male patients is 16.5.Bone ages of 14 females reached to tiptop of CHN-17.3 and average bone age of other female patients is 15.7.Among 522 cases there were 388 patients with their CHN bone ages above 16.0,about 74.3%.The group of data indicates that multiple factors can influence accuracy of bone age estimation.Among them change of radiograph position and angle influenced estimation accuracy most.Conclusion If radiographs of high quality can be acquired,CHN method can be proficiently used,and radiologists can pay attention to the effect on estimation accuracy of epiphysis classification by change of radiograph position and angle,then CHN bone age can mostly meet requirements of living age estimation.

Objective To conduct the global assessment of methylated DNA status of CpG islands. Methods The modified AIMS (amplification of inter-methylated sites) technology was adopted, which mainly included 3 groups of isoschizomers (methylation-sensitive and methylation-insensitive restricted enzymes). The comparison analysis of methylome among 3 concentrations (6%, 8%, 10%) of polyacrylamide with 8M urea denaturing sequencing gel and the digestion effect among 3 groups of isoschizomers were carried out. The results of personal molecular imager FX system and autoradiography were also compared. Results Genome-wide detection of methylome of CpG islands might be the technical basis for the subsequent analysis. The resolving power and scope bands in 6% polyacrylamide denaturing sequencing gel were superior to those in gels with the other two concentrations. 3 groups of isoschizomers could meet the need of genome-wide detection of methylome of CpG islands. Personal molecular imager FX system and autoradiography could be used together, and they compensated each other. Conclusions The optimized AIMS technology is a kind of high-throughput method to analyze methylome, which is simple, specific, and easy to handle. The 3 groups of isoschizomers can cover most of CpG islands for genome-wide detection and get the ideal results. Only a small quantity of genome DNA is enough to meet the need of clinical detection.

Objective To study the protective effect of resveratrol on endothelial cells injury induced by oxidized low-density lipoprotein (oxLDL) and its protective mechanism. Methods The injured model was established by endothelial cell treated with oxLDL. In the experiment, endothelial cell were cultured in vitro and divided into seven groups which were: control group; solvent (DMSO) group, oxLDL group, quercetin plus oxLDL group, Reseveratrol-L (0.5 mol/L), M(5 mol/L), H(50 mol/L) plus oxLDL group respectively. Objective To analyze the epidemiological trend and distribution of stroke mortality in residents of Tianjin, China. Methods The study was based on 102 718 died cases with stroke in Tianjin between 1999 and 2006. The death cause and the stroke typing were coded according to the ICD -9 and ICD -10. Standardized mortality rates were calculated for stroke and its subtypes, adjusted for age and sex according to the world standard population of the Year 2000. The age, sex and geographic distribution of the stroke and its subtype mortality were analyzed. Chi-square tests were used to determine the statistical significance of mortality trends and differences.
