Objective:To determine the promoting effect of pyrrolidine dithiocarbamate(PDTC),an inhibitor of NF-?B,on the sensitivity of human ovarian cancer cell line SKOV-3 to chemoagent cisplatin and the possible mechanisms.Methods:SKOV-3 cells were treated with various concentrations of cisplatin and PDTC combination for different periods.Then the growth inhibition of SKOV-3 cells were observed by MTT assay,expression of I-?B? and P65 protein by Western blotting assay,and apoptosis of cells by flow cytometry.Results:Separate application of both cisplatin(0.1-10 ?g/ml)and PDTC(10-40 ?mol/L L)significantly inhibited the growth of SKOV-3 cells and caused cell apoptosis(P

Cerebral microbleed is a cerebral small vessel disease.The detection rate is 4.5% to 84.9%in different populations.Vascular cognitive impairment is the general name of various cerebrovascular diseases and their risk factors caused varying degrees of cognitive impairments.Studies have shown that cerebral microbleed is closely associated with vascular cognitive impairment.This article reviews the relationship between cerebral microbleed and vascular cognitive impairment in different populations,different degrees and different locations.
3.Impact of NF-?B on P-glycoprotein-mediated multidrug resistance in ovarian cancer cells

Objective To investigate the expression of multidrug resistance gene 1 product P-glycoprotein(P-gp) when nuclear factor kappa B is induced or inhibited.Methods Ovarian cancer cells SKOV-3 were treated with Docetaxel or combinded with pyrrolidine dithiocarbamate(PDTC),the inhibitor of NF-?B.Western blot assay、 flow cytometry assay and MTT assay were used to measure P65 protein,P-gp,apoptosis of cancer cells,and the survival rate respectively.Results Docetaxel increased P65 protein and P-gp expression,while combind use of PDTC reversed this function.Both Docetaxel(≥(1 ?g/mL))and PDTC(≥(10 ?mol/L)) significantly inhibited the growth of SKOV-3 cells,and caused apoptosis.The combined use of Docetaxel and PDTC at low concentrations significantly inhibited the growth of cancer cells and caused apoptosis as compared with those treated with Docetaxel only.Conclusion The expression of P-gp may be ralated to the activation of NF-?B;Inhibition of NF-?B can enhance the chemosensitivity of ovarian cancer cells to Docetaxel.

An early and accurate response evaluation is essential for clinicians to decide whether to continue the treatment with current thera-peutic regimen or to make necessary changes.This article briefly introduces the development of response evaluation criteria for tumors,elab-orates on the application of radiological evaluation criteria in local interventional therapy or sorafenib treatment of hepatocellular carcinoma (HCC),compares the four radiological evaluation criteria,i.e.,World Health Organization (WHO)criteria,Response Evaluation Criteria in Solid Tumors (RECIST),European Association for the Study of Liver (EASL)criteria,and the modified RECIST (mRECIST),and thinks that EASL criteria and mRECIST are better than WHO criteria and RECIST and can predict the prognosis of HCC patients at earlier time points.

Objective To analyze the epidemiological trend and distribution of stroke mortality in residents of Tianjin, China. Methods The study was based on 102 718 died cases with stroke in Tianjin between 1999 and 2006. The death cause and the stroke typing were coded according to the ICD -9 and ICD -10. Standardized mortality rates were calculated for stroke and its subtypes, adjusted for age and sex according to the world standard population of the Year 2000. The age, sex and geographic distribution of the stroke and its subtype mortality were analyzed. Chi-square tests were used to determine the statistical significance of mortality trends and differences. Results Stroke mortality rate in Tianjin declined from 133.52 / 100 000 / year in 1999 to 102.52 / 100 000/year in 2006. Cerebral infarction accounted for more than 50% of stroke mortality. 8.Effects of IGF-1 on PRNP expression and APP metabolism of PC12 cells

Objective·To investigate the effects of insulin-like growth factor 1 on prion encoding gene (PRNP) expression and amyloid precursor protein (APP) metabolism of PC12 cells. Methods·After PC12 cells were treated with 20, 40, 80 ng/mL IGF-1 for 24 h, real-time PCR was used to detect the mRNA expression levels of PRNP, and Western blotting was used to detect the protein levels of AKT, phosphorylation of AKT (pAKT), ERK and phosphorylation of AKT (pERK). The level of β-amyloid 42 (Aβ42) in supernatant fluid was detected by ELISA. Results·Compared with the blank control group, the expression of PRNP mRNA in Alzheimer's disease (AD) model group was increased significantly (P<0.01). The expression of PRNP mRNA was significantly increased after cells were treated with 40 and 80 ng/mL IGF-1 for 24 h (P<0.01). There was no significant difference in APP protein expression among AD model group and three IGF-1 treatment groups (P>0.05). Compared with the blank control group, the level of Aβ42 in supernatant fluid of model group was decreased significantly with the increasing of IGF-1 concentration. The expression level of Aβ42 was decreased significantly in 40 and 80 ng/ml IGF-1 treatment group (P<0.05). The expression of AKT/pAKT, ERK/pERK in AD model group was significantly increased along with the increasing of IGF-1 concentration (P<0.05). Conclusion·IGF-1 could regulate the expression of PRNP gene and effect the metabolism of APP, which may be associated with PI3K/AKT, MAPK/ERK1/2 signaling pathway.
9.Protective effects of resveratrol on oxidized low-density lipoprotein-induced injury on endothelial cells

Objective To study the protective effect of resveratrol on endothelial cells injury induced by oxidized low-density lipoprotein (oxLDL) and its protective mechanism. Methods The injured model was established by endothelial cell treated with oxLDL. In the experiment, endothelial cell were cultured in vitro and divided into seven groups which were: control group; solvent (DMSO) group, oxLDL group, quercetin plus oxLDL group, Reseveratrol-L (0.5 mol/L), M(5 mol/L), H(50 mol/L) plus oxLDL group respectively. Endothelial cells were incubated with 200?g/ml oxLDL for 18 hours in the absence or presence of various concentrations of resveratrol and quercetin. The apoptotic index was detected by flow cytometric. Meanwhile, the content of Malondialdehyde (MDA), and the activity of SOD, GSHPX in the culture medium and homogenate of endothelial cells were measured. Results Incubation of endothelial cells with oxLDL for 18 h elicited a significant increase in the percent of apoptotic endothelial cells, in control group, the percent of apoptotic cells is 7.6?0.6, however in oxLDL group, it increased to 41.4?2.3, furthermore, oxLDL induced the increase of the contents of MDA, and accompanied by the decrease in the activity of SOD, GSHPX in the endothelial cells, the difference of was significant as compared with control group. Pretreatment of resveratrol with 0.5~50 mol/L decreased these oxLDL-induced changes, in Reseveratrol with 50 mol/L group, the percent of apoptotic cells decreased to 13.0?1.5, the difference of was significant as compared with oxLDL group (P
10.Detection of sperm protein 17 in serum and its significance in patients with gynecology cancer

Objective To quantify the level of human sperm protein 17(Sp17)in serum of the healthy and patients with gynecological cancers and to evaluate whether Sp17 is a suitable serum marker for the diagnosis of gynecological cancers.Methods The recombinant human Sp17 was radioiodinated by the chloramine T method.125I-rhSp17 was used to develop a radioimmunoassay to determine Sp17 in serum of the healthy and the patients.Meanwhile,Sp17 levels in serum were compared with the expression of Sp17 in tissues from gynecological cancers.Results The level of serum Sp17 was(15.60?7.66)?g/L in healthy subjects(n=59),which was significantly lower than those in twenty-two patients(64.36?34.44)?g/L.Sera from nine of twenty-two patients(40.9%)were positive for Sp17 by radioimmunoassay assay.Tumor tissues from twelve of twenty-two patients(54.5%)were positive for Sp17 by immunohistochemistry assay.Conclusions Markedly elevated serum levels of Sp17 were found in patients with gynecological cancers.However,there was no significant correlation between serum level of Sp17 and expression of Sp17 in tumor tissues in patients with gynecological cancer.
