Conclusion: The PCL-b-PLLA scaffold has a good compatibility with cells and is suitable for cartilage tissue engineering.
2.Expression and significance of tumor suppressor gene Kruppel-like transcription factor 6 mRNA and protein in gastric cancer

Objective To investigate the correlation between Kruppel-like transcription factor 6 (KLF6) and gastric cancer.Methods By using immunohistochemistry (IHC) and reverse transcription PCR,the expression of KLF6 protein and mRNA in human gastric carcinoma specimens and adjacent tissues from 73 patients were examined.Results The KLF6 protein expression rates were 39.7 % (29/73) and 90.41%(66/73) in 73 cases of gastric cancer and adjacent tissues,The KLF6 protein expression was significantly lower in human gastric carcinomas than the adjacent tissues by chi-square test (P < 0.01).The KLF6 expression rates were 25.0 % (10/40),57.6 % (19/33) in poorly and well differentiated cancer.The difference between the two groups was significant (P < 0.01).The optical density ratio of KLF6 mRNA were 0.1586±0.1071 and 0.8899±0.0638 in the gastric cancer and cancer adjacent tissue respectively,the difference between the two groups was significant (P < 0.01).Conclusion KLF6 expression is low in gastric carcinoma,and moreover,KLF6 expression is significantly lower in poorly differentiated cancers than the well differentiated gastric cancers,there are negative correlation between KLF6 and gastric cancer,therefore KLF6 could be a useful marker for gastric cancer.
3.Qualitative Prediction of Yeast Growth Process Based on Near Infrared Spectroscopy

To improve the yield of industrial fermentation, a method based on near infrared spectroscopy was presented to predict the growth of yeast.The spectral data of fermentation sample were measured by Fourier-transform near-infrared (FT-NIR) spectrometer in the process of yeast culture.Each spectrum was acquired over the range of 10000-4000 cm1.Meanwhile, the optical density (OD) of fermentation sample was determined with photoelectric turbidity method.After that, a method based on competitive adaptive reweighted sampling (CARS) was used to select characteristic wavelength variables of NIR data, and then extreme learning machine (ELM) algorithm was employed to develop the categorization model about the four growth processes of yeast.Experimental result showed that, only 30 characteristic wavelength variables of NIR data were selected by CRAS algorithms, and the prediction accuracies of training set and test set of the CARS-ELM model were 98.68% and 97.37%, respectively.The research showed that the near infrared spectrum analysis technology was feasible to predict the growth process of yeast.
4.Effects of penehyclidine hydrochloride preconditioning on hepatic ischemia-reperfusion injury in rats

Objective To investigate the effects of penehyclidine hydrochloride(PHC)precon-ditioning on hepatic ischemia-reperfusion injury (HIRI)in rats.Methods Fifty-four male Sprague-Dawley rats,weighing 230-250 g,were randomly divided into three groups (n =18 each):sham op-eration group (group S),HIRI group (group HR)and penehyclidine hydrochloride group (group PHC).In group S,the hepatoduodenal ligaments of rats were only pulled and separated,then abdo-mens were closed.In group HR,an atraumatic vascular clip was placed on the vessels blocking the portal venous blood supply to the median and left lateral lobes of the liver for 45 minutes,which re-sulted in approximately 70% rat liver ischemia injury.In group PHC,the rats were treated with 0.45 mg/kg penehyclidine hydrochloride at 30 minutes before the portal venous and hepatic arterial were blocked like group HR.Animals were killed at 2 h (T1 ,n =6),4 h (T2 ,n =6),24 h (T3 ,n =6) after HIRI or sham surgery.Liver tissues and blood samples were taken for analysis.The serum con-centration of ALT and AST were measured as the markers of hepatic functional damage.The TNF-a and IL-1βconcentration were measured by the enzyme linked immunosorbent assay (ELISA)tech-nique.The endothelial nitric oxide synthase (eNOS ) and hypoxia-inducible factor (HIF-1α) expression were measured by immunohistochemical staining.Results Compared with group S,the expressions of ALT,AST,TNF-αand IL-1βin groups HR and PHC increased at T1-T3 (P <0.05), which were similar to the change trend of HIR-1αand eNOS expressions (P <0.05).Compared with group HR,the expression of ALT,AST,TNF-αand IL-1βin group PHC decreased at T1-T3 (P <0.05).Nevertheless expression of HIF-1αand eNOS of groups HR and HPC were increased than that of group S,and experssion of HIF-1αand eNOS of group HPC were increased than that of group HR at T1 and T2 (P < 0.05 ).Conclusion PHC preconditioning can protect the liver from HIRI.The mechanism may be associated with the up-regulation of eNOS and HIF-1α,as well as reducing the in-flammatory response.
5.Secondary order mass spectrometry identification and biomarker of serum proteins in patients with Keshan disease

Objective To investigate the clinical diagnostic value and pathogenesis of serum protein identification in Keshan disease (KD).Methods A total of 65 chronic KD patients were selected as the patient group in KD endemic areas,while 29 cases of dilated cardiomyopathy (the DCM group),62 healthy cases from KD endemic areas (control 1 group) and 28 healthy cases from non-endemic areas (control 2 group) were selected as controls.Liquid chip time of flight mass spectrometry (ClinProtTM MALDI-TOF-MS) was used to determine the expression of proteins/peptide peaks.ClinProTools 2.2 software was used to analyze the protein profiles to determine differentially expressed proteins/peptide peaks.The Genetic Algorithm (GA),QuickClassifer Algorithm (QC) and Supervised Neural Network Algorithm (SNN) methods were used to screen marker proteins.Matrix-assisted laser desorption/ionization time-of-flight Mass Spectrometry technique (MALDI-TOF/TOF) was also used as a secondary mass spectrometry to identify differentially expressed peptides.Results Between the KD and control 1 groups,34 differentially expressed proteins/peptides and 5 marker proteins were identified,while 52 differentially expressed proteins/peptides and 5 marker proteins were identified between the KD and control 2 groups,and there were 67 differentially expressed proteins/peptides and 5 marker proteins between the KD and DCM groups.During secondary mass spectrometry,two peptides for mass-to-charge ratio (m/z) 2 079 and 1 465 were obtained,peptide of matching β-globin showed low expression while peptide of matching fibrinogen showed high expression in the KD patients.Conclusions Serum marker proteins can be used as biomarkers for diagnosis and differentiation of KD.β-globin and fibrinogen play an important role in the development of KD myocardial injury.
6.Delay aversion test between the male children with obsessive-compulsive disorder and ones with attention-deficit/hyperactivity disorder

Objective:To clarify the characteristics and differences between obsessive-compulsive disorder (OCD)and attention-deficit/hyperactivity disorder (ADHD)in male children.Methods:Forty boys with OCD and 40 boys with ADHD were selected from out-patient department,and 40 normal boys matched with age,gender and IQ were recruited as the controls.Patients'diagnosis was made according to the International Statistical Classifica-tion of Diseases and Related Health Problems,Tenth Revision (ICD-10).The choice delay tasks (CDT1 /2)were assessed respectively for the measurements of normal or high ecological validity of delay aversion.In CDT1 /2, waiting time and scores of the tasks were indicators reflecting the delay aversion.Results:The scores of CDTs were higher in children with OCD than in the controls [CDT1,(30.9 ±3.2)vs.(27.6 ±3.8);CDT2,(31.3 ±4.4)vs. (28.5 ±4.9);P <0.01].The scores of CDTs were lower in children with ADHD than in the controls [CDT1, [(25.0 ±4.3)vs.(27.6 ±3.8)];CDT2,[(26.2 ±4.6)vs.(28.5 ±4.9);P <0.001].Conclusion:The results in-dicate that the children with OCD have a trend to choose the task with longer waiting time and higher score,and the children with ADHD trend to choose the task with shorter waiting time and lower score.The results suggest that the pathogenesis of hyperactivity/impulse control may be different between OCD and ADHD in delay aversion.
7.Response inhibition and fine-motor coordination in male children with Tourette syndrome comorbid attention-deficit/hyperactivity disorder

Objective:To explore the differences and similarities of the neuropsychological functioning defi-cits in children between Tourette syndrome (TS)and attention-deficit/hyperactivity disorder (ADHD). Methods:Thirty boys with TS-only,36 with TS-plus-ADHD,36 with ADHD were selected from out-patient department,and 50 normal boys (NC)matched with gender,age and IQ were recruited as the controls. Patients'diagnosis was made according to the International Statistical Classification of Diseases and Related Health Problems,Tenth Revision (ICD-10). They were assessed with the Stroop Color-Word Interference Test (Stroop)and Purdue Pegboard Test (Purdue)to evaluate the response inhibition and fine-motor coordination respectively. Results:The Stroop scores were higher in children with ADHD than in other groups (P<0. 05 ),there was no difference between children with TS-only and TS-plus-ADHD (P>0. 05 ). All Purdue scores were higher in children with disease than in normal children (P<0. 05 ). Conclusion:The results indicate that the response inhibition deficit may be found in children with ADHD,but not in those with TS-only and TS-plus-ADHD. The neural compensatory mechanism may be re-sponsible for the response inhibition function in children with TS whilst the fine-motor coordination deficit was as-sociated with the disease groups.
8.Treatment for upper ureteral calculi of solitary kideny with Holmium laser lithotripsy through modular flexible ureteroscope

Objective To evaluate the efficiency and clinical value of modular flexible ureteroscope combined with Holmium laser litho-tripsy on the treatment for upper ureteral calculi of solitary kiden. Methods There were 26 cases of upper ureteral calculi of solitary kidney, including 12 cases of left ureter calculi and 14 cases of right ureter calculi. The diameter of the calculi ranged from 0. 6~2. 2 cm, averagely (1. 38 ± 0. 22) cm. F6 double-J tubes were indwelled postoperatively for 4 weeks, and catheters were indwelled postoperatively for one week. Examine with KUB one day after operation to evaluate the effective rate. Results The Ureteral guiding sheath were successfully implanted in 23 cases, 2 cases underwent second lithotripsy after indwelling D-J tubes 2 weeks later, D-J tubes failed to be implanted only in one case, which has changed to percutaneous nephrolithotomy (PCNL) for ureter stricture. The stone free rates was 88. 5%(23/26)after the first ses-sion,and it was 100%(26/26)at the end of the session. The operation time was 15~45 min, meanly (27 ± 3. 2) min. One patient had ureter perforation during the operation, and one patient had severe infection after operation. Conclusion The modular flexible ureteroscopy com-bined with Holmium laser lithotripsy is an effective and safe treatment, especially for upper ureteral calculi of solitary kideny, and it should be considered as the first choice for solitary kidney calculi.
9.Expression and significance of autophagic regulatory factor mTOR and Beclin1 in benign and malignant pleomorphic adenoma

Objective To explore the expression and significance of main autophagic regulatory factors of mTOR and Beclin 1 in benign and malignant pleomorphic adenoma .Methods The expression of Beclin1 in 35 cases of carcinoma in parotid pleomorphic adenoma(CPA) ,37 cases of benign pleomorphic adenoma(PA) and 20 cases and normal parotid tissues was measured by adopting the immunohistochemical staining method .The expression of p-mTOR and Beclin1 in CPA ,PA and normal parotid tissues was measured by Western blot .Results The positive expression of p-mTOR in normal parotid tissue ,parotid pleomorphic adenoma and parotid pleomorphic adenoma showed the increasing trend ,and the difference among them was statistically significant (P<0 .05) . The Beclin1 level in 3 kinds of tissue showed the descending trend ,the difference among the three tissues was statistically signifi-cant(P<0 .05) .Conclusion mTOR signal factor is over-expressed in CPA tissue ,while the positive expression of Beclin 1 is inhibi-ted in CPA tissue .

Objective To explore the brain's metabolism about the hippocampus, anterior cingulate and thalamus of first episode depressive patients. Methods Using proton magnetic resonance spectroscopy and 1 HMRS analyzing technology to detect multiple metabolic indices of the hippocampus, anterior cingulate and thalamus of 20 patients and 20 healthy persons, including N-acetylaspartate ( NAA), choline ( Cho ), myoinositol ( MI ), creatine(Cr). Results The ratios of MI/Cr of the bilateral thalamus of the depressive patients group decreased ( Left (0.23 ±0.13) ,(0.25 ±0.09), t=2.25, P=0.03,Right(0.33 ±0.15,0.34 ±0.04), t=2.42, P=0.02).The ratios of NAA/Cr and Cho/Cr of the bilateral hippocampus of the depressive patients group were both lower (NAA/Cr: Left t=3.06, P=0.004,Right t=3.54, P=0.001. Cho/Cr: Left t=2.94, P=0.006,Right t=4.02,P=0.001 ). The ratios of Cho/Cr and MI/Cr of the anterior cingulate of the depressive patients group were both significantly higher (NAA/Cr: t=3.12, P=0. 003;Cho/Cr: t=2.62, P=0.01). Conclusion The metabolic abnormalities of hippocampus, anterior cingulate and thalamus maybe constitute the nervous biochemical basis of first episode depression.
