Objective To investigate the application effect of the ankle foot orthoses (AFO)in the rehabilitation of patients with hemiplegia and abnormal gait after stroke Methods Sixty consecutive stroked patients with hemiplegia admitted to Meizhou People′s Hospital,Guagdong Province from January in 2013 to June in 2015 were enrolled retrospectively, and they were divided into either an AFO group or a non-AFO group (n = 30 in each group)according to the odd or even admission numbers. The patients in the non-AFO group were treated with conventional rehabilitation training and those in the AFO group were treated with AFO. Before and after treatment, the Berg balance scale (BBS)was used to assess the balance ability of the patients,10 m maximum walking speed (MWS)was used to assess the walking speed of the patients, and the physiological cost index (PCI)was used to assess the walking efficiency of the patients. Results After treatment, there was significant difference in Brunnstrom grade between the AFO group and the non-AFO group (P < 0. 05). The BBS score and MWS of the patients in the AFO group were 39 ± 5 and 0. 97 ± 0. 38 m/ s respectively after treatment, and they were higher than those before treatment (33 ± 4 and 0. 28 ± 0. 07 m/ s)and those of non-AFO group (36 ± 4 and 0. 54 ± 0. 31 m/ s)after treatment. There were significant differences (all P <0. 05). The PCI was 0. 84 ± 0. 30 in the AFO group after treatment was 0. 84 ± 0. 30. It was lower than that before treatment (1. 32 ± 0. 31)and that of non-AFO after treatment (0. 96 ±0. 33). There was significant difference (all P < 0. 05). Conclusion The application of APO in stroked patients with hemiplegia and abnormal gait has better clinical efficacy. It can significantly im-prove the balance state of patients and improve the walking speed and efficiency.

A high-performance liquid chromatography/electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) method was developed and validated for the determination of mizoribine in human serum using thiamphenicol as internal standard (IS). The serum samples of mizoribine were precipitated with acetonitrile and separated by HPLC on a reversed phase C18 column with a mobile phase of 0.1% ammonium acetate water solution-methanol (47:53, v/v). Mizoribine and IS were detected in the multiple reaction monitoring mode with precursor/product ion transitions of m/z 258.2/126.0 and 354.1/185.2, respectively. The calibration curves were linear over the range of 0.02-2 microg mL(-1) for mizoribine. The limit of quantification (LOQ) was 0.02 microg mL(-1) with acceptable precision and accuracy. The validated method was successfully applied for the evaluation of a bioequivalence study on Chinese healthy volunteers. The main pharmacokinetics parameters after oral administration of 100 mg mizoribine test or reference formulation were as follows: Cmax (1.00 +/- 0.21), (1.00 +/- 0.22) microg mL(-1); AUC(0-infinity) (6.72 +/- 1.39), (6.48 +/- 1.44) microg h mL(-1); t1/2 (2.77 +/- 0.26), (2.66 +/- 0.29) h; tmax (2.95 +/- 0.78), (2.84 +/- 0.50) h.

AIM:To investigate the effects of taurine on lipopolysaccharide ( LPS)-induced myocardial damage in rats.METHODS:Healthy male SD rats ( n=30) were randomly divided into control group ( CON) , LPS model group ( LPS) and taurine treatment group ( TAU) .The rats in CON group and LPS group were intravenously injected with normal saline, and the rats in TAU group were injected with taurine (100 mg/kg).After 2 h, the rats in LPS group and TAU group were intraperitoneally injected with LPS at 10 mg/kg, and the rats in CON group were injected with normal saline . Six hours after injection of LPS , the blood samples were collected for determination of superoxide dismutase ( SOD) activi-ty, malondialdehyde (MDA) content, and tumor necrosis factor α(TNF-α) and interleukin-6 (IL-6) levels. The myocar-dial tissues were processed for histological examination and the analysis of Western blot .RESULTS:Compared with CON group, LPS significantly reduced SOD activity in the serum and heme oxygenase 1 ( HO-1) protein expression in the myo-cardial tissues, increased the serum content of MDA and levels of TNF-αand IL-6.LPS also significantly elevated the lev-els of TNF-αand IL-6, and up-regulated the cyclooxygenase-2 ( COX-2) expression and phosphorylation of nuclear factor kappa B ( NF-κB) in the myocardial tissues .Taurine pretreatment significantly elevated SOD activity and HO-1 protein ex-pression level, decreased the levels of COX-2, TNF-α, IL-6 and phosphorylated NF-κB.Histological observation showed that taurine reduced inflammatory response in the myocardial tissue .CONCLUSION: Taurine attenuates LPS-induced myocardial damage in rats .The beneficial effects of taurine may be associated with its reduction of p-NF-κB/COX-2 signa-ling by activation of HO-1/CO.

Objective To assess the working memory and explore the activation of brain areas for children with primary nocturnal enuresis (PNE) with fMRI scan. Methods Twenty three right-handed children with PNE and 20 age-matched right-handed healthy children as the controls were recruited.Intelligence tests were performed by means of Wechsler Young Children Scales of Intelligence (C-WISC) in children with PNE and normal controls. The full intelligence quotient (FIQ), verbal IQ (VIQ),performances IQ (PIQ) and the memory/caution (M/C) factor of PNE children and the controls were measured. After Intelligence tests, an enent-related fMRI scan was performed using the categorial N-Back working memory task. Percent of correct responses (PCR) and mean reaction time to correct response (mRT) were recorded and analyzed by the student t test. The fMRI data were analyzed using Statistical Parametric Mapping 2 (SPM2), the differences in activation were compared between two groups. Results The data of 15 PNE children and 15 healthy children were evaluated. The FIQ, VIQ and PIQ in PNE group were in a normal range and no statistical significance with the control group ( P > 0. 05). M/C factor in the PNE group(90.4 ±10.2)was significantly lower than that in the control group (99. 6 ± 11.9) (t =2. 260,P < 0. 05). In the N-Back test, PNE children had significantly less PCR [(72.7±6.3 ) % vs. ( 86. 3 ±6. 7) %, t = 5. 727,P < 0. 01] and longer mRT [ (625. 8 ± 72.5) ms vs. (534. 8 ± 63. 3 ) ms, t = 3. 684,P < 0.01] than the healthy controls. The activation regions of PNE patients and healthy children were mainly in the dorsal right frontal lobe, right parietal lobe, left temporal lobe gyrus fusiformis and bilateral cerebellum posterior lobe. The activation level in left posterior cerebellar lobe in PNE children was significant lower than that in healthy controls (P<0.01). Conclusion The children with PNE have deficits in working memory which might be associated with the dysfunction of the left cerebellum.

Objective:This work aims determine the expression of the neurotensin (NTS) gene in hepatocellular carcinoma (HCC) subgrouping using immunohistochemical staining (IHC) as well as to evaluate the correlation between the activation of NTS/IL-8 pathway in HCC and inflammatory response in microenvironment and epithelial mesenchymal transition (EMT) in cancer and in the prognosis of patients. Methods:Tumor tissues and corresponding adjacent normal tissue were collected from 64 cases of HCC patients. The expression levels of NTS protein and multiple inflammation and EMT-related proteins, including IL-8, VEGF, MMP9, CD68, E-Cadherin,β-Catenin, and Vimentin, were examined in 64 cases of paraffin-embedded HCC tissues using the immunohistochemistry (IHC) staining method. The clinical outcome and overall survival (OS) among 64 cases of HCC patients were compared. Results:We found that the frequency of NTS-expressing tissues among all HCC samples was 17.19%(11/64). Significantly increased IL-8 protein was confirmed in 90.91%of NTS+HCC samples and was positively correlated with the levels of NTS protein in cancer tissues (P=0.036), which implied the dysfunctional activation of NTS/IL-8 pathway in HCC. The levels of VEGF and MMP9 were significantly correlated with the co-expression of NTS and IL-8 in HCC. Evident features of EMT, including decreased membrane expression of E-Cadherin and increased accumulation of cytoplasmicβ-Catemin and Vimentin, were found in NTS+IL-8+samples. The co-expression of NTS and IL-8 in cancer was significantly correlated with the clinical outcomes of patients, as the mortality rate of NTS+IL-8+HCC patients is 2.5-fold higher than that of others after surgery (P=0.022).Accordingly, the OS of NTS+IL-8+HCC patients significantly decreased (24.65±4.45 m vs. 75.79±16.32 m, P=0.013), and these patients are at a higher risk of death at an expected hazard ratio (HR) of 3.457. Conclusion:The NTS/IL-8 pathway is dysfunctionally activated in a subgroup of HCC samples. Highly expressed NTS is associated with increased inflammatory response in microenvironment, enhanced EMT in cancer, and worse prognosis in HCC patients.
