1.Quantitative Determination of Sarsasapogenin in "Antivirotic Oral Liquid" by Duble-Wavelength TLC Scanner

Sarsasapogenin in "antivirotic oral liquid" made by different factories was determined quantitatively by duble-wavelength TLC scanner. The method is simple, accurate, sensitive,and reproducible. The average recovery was 102. 8%, coefficient of variation was 3. 7%, andcoefficient of correlation was 0. 9997. Results showed that this method is suitable for the quality control of "antivirotic oral liquid ".
2.Effect of insulin-like growth factor-1 on expression of stem cell factor from sepsis rat colonic smooth muscle cell

Objective To explore the effect and intracellular signal transduction pathway of insulin-like growth factor 1 (IGF-1) on the expression of stem cell factor (SCF) in spesis rats colonic smooth muscle cells (SMC).Methods The model of spesis rats was established,colonic SMC from spesis rats were cultured by enzymolysis and identified by α-actin immunofluoresence methods.Colonic SMC incubated with different concentrations (0,50,100 and 150 μg/L) of IGF-1 for different durations (0,12,24,36 and 48 h),and another SMC were pretreated with specific mitogen-activated protein kinase (MEK) inhibitor PD-98059 and phosphatidylinostiol 3-kinase(PI 3-kinase) inhibitor LY-294002 before IGF-1 induced.The expression of SCF in colonic SMC was examined by Western blot and quantitative reverse transcription polymerase chain reaction.Results (1) A very low level of SCF was expressed in colonic SMC cultured in bovine serum free medium.The protein and mRNA were 0.223 ± 0.047 and 0.149 ± 0.023 respectively.The expression of SCF protein and mRNA from SMC was increased to 0.398±0.033 and 0.243±0.013(P<0.05) when concentration of IGF-1 was 50 μg/L.The final effective concentration of IGF-1 in vitro was 100 μg/L,the levels of SCF protein and mRNA were increased to 0.719±0.017 and 0.360±0.006(P<0.05).(2) For the time point induced the SCF expression by IGF-1,the expression level of protein and mRNA in 0 h were 0.347±0.031 and 0.092 ±0.018 respectively.The time point of SCF peak level occurred at the 24 h,the expression level of SCF protein and mRNA were increased to 0.893 ± 0.049 and 0.396 ± 0.048 (P < 0.05).(3) The IGF-1-induced SCF expression was reduced significantly by a pretreatment of PD-98059 (SCF protein and mRNA were reduced 37% and 45%,P<0.05),and LY-294002 had no effect on the expression of SCF (P>0.05).Conclusion The expression of SCF from colon SMC in sepsis rat is stimulated by IGF-1 in both dose and time-dependent manners,in which ERKMAPK signal transduction may play an important role.
3.Design of MRI interventional RF coil

Objective To design a new type of RF endorectal coil so as to increase signal to noise ratio(SNR) and improve the efficiency of MRI diagnosis.Methods Through circuit and construction optimization,a MRI model of coil in vivo was designed.Results The images and spectroscopy of a special phantom were acquired and the results were compared to those of the commercial TORSO coil.Conclusion This coil gives a significantly higher SNR at the region of interest(ROI).The achieved high local SNR and resulting high spatial resolution can add more anatomic and biochemical information to the diagnosis of prostate diseases.
4.Effects of mesalamine on the expression of TNF-α, IL-6, IL-17 and intestinal barrier function with ulcerative colitis

Objective To investigate the effect of mesalazine on the expression of TNF-α, IL-6, IL-17 and intestinal barrier function in patients with ulcerative colitis. Methods 76 patients with UC from January 2015 to April 2016 in The Fifth Central Hospital of Tianjin were collected and randomly divided into control group and observation group with 38 cases in each group. After admission were given maintenance of water and electrolyte, basic acid-base balance and nutritional support treatment, and the control group were treated with conventional drug sulfasalazine (SASP) treatment, two tablets each time, four times once day, four weeks for each course, the observation group was treated with mesalazine, four tablets each time, three times once day, four weeks each course of treatment. The levels of TNF-α, IL-6, IL-17, L/M and Baron endoscopic scores in the two groups were observed after two courses of treatment. The efficacy and side effects of the two groups were compared. Results No dropout or dropout cases in this study, two groups after the treatment of TNF-α, IL-6, IL-17, L/M, Baron scores were significantly decreased (P<0.05), but the observation group after treatment the above indexes were all significantly lower than the control group (P<0.05), the total effective rate of the observation group was 89.47%(34/38) higher than that of the control group 71.05%(21/38), the adverse reaction rate of 13.16% (5/38) was lower than that in the control group 34.21% (13/38), the differences were statistically significant(P<0.05). Conclusion Compared with conventional drug SASP, the effect of on the expression of TNF-α, IL-6 and IL-17 in serum of patients with UC and the improvement of intestinal barrier function can be effectively suppressed, which has the advantages of clinical efficacy and low adverse reaction rate.
5.Application of BHP9504 fluorescent analytic instrument

Objective To introduce the clinical application of BHP9504 fluorescent analytic instrument.Methods The chemical immunofluorescent test was used in high-precision,high-stability photo-signal test by photon counting technology.Results Photo-signal test could examine strong photo-signals in dark background so that it was able to improve sensitivity of experiments.Conclusion This technique widens the range of application and reduces specimen amount.
6.Association between circulating endothelial cells and atherosclerosis in maintenance hemodialysis patients

Objective To explore the association between circulating endothelial cells (CECs)and atherosclerosis in maintenance hemodialysis(MHD)patients. Methods A crosssectional study was performed to investigate the association between CECs and carotid atherosclerotic change in 65 MHD patients,25 non-hemodialysis patients with chronic kidney disease(CKD)of stage 4 or 5(CKD-non-HD)and 24 age-and Sex-matched healthy controls. CECs in peripheral blood were determined by multiparametrie flow cytometry(FCM).CECs were labeled with CD3-PerCP and CD146-PE before FCM and identified as CD3dim,CD146bright.Atherosclerosis in both groups Was assessed by the measurement of common carotid arery intimamedia thickness (CCA-IMT)and plaque of the common carotid arteries with ultrasound scanner. Results CECs were significantly higher in pre-dialysis patients[(151.52±98.24) cell/ml]and CKD-non-HD patients[(183.00±81.38)cell/ml ] compared with control group[(106.50± 24.14)cell/ml](P<0.05 and P<0.01,respectively).But the number of CECs was not significantly different between MHD and CKD-non-HD patients.CCA-IMT was also significantly higher in MHD patients[(0.94±0.36)mm]and CKD-non-HD patients [(1.02±0.37)mml compared with control group[(0.75±0.15)mm](P<0.05 and P<0.01,respectively).The number of pre-dialysis CECs was positively correlated with CCA-IMT in MHD patients(r=0.328,P<0.01).Multivariate analysis showed that CEC level was a strong independent risk factor of CCA-IMT. Conclusion InMHD patients, CEC level is associated with carotid atherosclerosis and may be used as a marker to evaluate the endothelial damage.
7.Brain natriuretic peptide rs198388 polymorphism and essential hypertension in Hunan Han people

Objective To investigate the relation between brain natriuretic peptide (BNP) rs198388 polymorphism and the susceptibility of essential hypertension in Han population of Hunan. Methods A total of 567 patients with hypertension (the hypertension group) and 555 healthy volunteers (the control group) were enrolled. Gender, age, smoking and drinking history of the 2 groups were not significantly different. Blood pressure was measured in the 2 groups. After fasting for 12 h or more, blood glucose, total cholesterol, triglycerides, high density lipoprotein cholesterol and low density lipoprotein cholesterol were measured. DNA polymorphism analysis was done by polymerase chain reaction-restriction fragment length polymorphism method, and genotype was determined by agarose gel electrophoresis. Results The GG, GA, and AA genotypeswere detected.The frequencies of GA and AA genotypes and A allele were significantly lower in the hypertension group (GA and AA:12.3%;A:6.9%) than those in the control group (GA and AA:18.4%; A:9.7%; P=0.009, and P=0.014, respectively). Conclusion BNP rs198388 polymorphism may be associated with essential hypertension in Han people in Hunan. Carrying rs198388 GA and AA genotypes and A allele may be the reason for low risk of hypertension.
8.Association between osteopontin and intact parathyroid hormone in maintenance hemodialysis patients

Objective To explore possible associations between osteopontin(OPN) and intact parathyroid hormone(iPTH),to investigate effects of them on the progression of carotid artery calcification in patients receiving long-term hemodialysis.Methods Forty-eight maintenance hemodialysis (MHD) patients and 28 age-and sex-matched healthy volunteers were recruited.The concentration of OPN in peripheral blood was determined by enzyme linked immunosorbent assay (ELISA).Levels of iPTH and presence of plaques in the common carotid arteries were also measured.The demographics were recorded.Results Compared with controls,levels of OPN[(137.4±80.8) ng/L vs (31.6±6.7) ng/L,P < 0.01] and iPTH[(456.4±326.4) ng/L vs (66.9±19.3) ng/L,P < 0.01] were higher in MHD patients before hemodialysis,the numbers of calcific plaques in the common carotid arteries were increased in MHD patients (P < 0.01).There was a positive correlation between pre-dialysis OPN levels and iPTH levels (r =0.620,P < 0.01) in MHD patients.Higher levels of OPN and iPTH correlated with greater numbers of calcific plaques in the common carotid arteries after division into three subgroups of MHD patients based on calcific plaques.In multiple linear regression analysis,the correlation between the pre-dialysis OPN and iPTH levels remained the same even if adjusting for confounding effects[β =0.468,95%C1 (0.036,0.195),t =2.936,P =0.005].Conclusion OPN level is positively correlated with iPTH level in hemodialysis patients,which suggesting that both of them play important roles in the progression of carotid artery calcification.

10.Clinical analysis of headache in patients with cerebral venous sinus thrombosis

Objective To analyze the characteristics of the common first attack of headache in patients with cerebral venous sinus thrombosis (CVST). Methods The clinical data of 51 patients who were diagnosed as CVST with MR venography (MRV)or DSA were collected retrospectively. The patients were divided into either a acute group (≤3 week,n= 32)or a chronic group (>3 weeks, n=19). Results (1)The age of onset of symptoms in these patients was 20 to 40 years,and most of them were females. Of all the first symptoms,headache ranked first,accounting for 84. 3%(43/51 );headache only accounted for 52. 9%(27/51 ),headache with other symptoms (ophthalmic symptoms, hemiplegia,and aphasia,etc. )accounted for 31. 4%(16/51 ),and other symptoms such as epilepsy, paralysis,and ophthalmic symptoms accounted for 15. 7%(8/51). (2)The proportion of headache only as the first symptom in the acute group was higher than that in the chronic group (65. 6% vs. 31. 6%,P<0. 05), and the proportion of chronic headache with other symptoms as the first symptom in the chronic group was higher than that of the acute group (P<0. 05). In addition,the proportion of the first symptom including ophthalmic manifestations in the chronic group was higher than that of the acute group (P <0.01 ). (3)Forty-three patients with headache,dull headache or distensible pain accounted for 72. 1%(31/43), thunderclap headache accounted for 11. 6%(5/43),and other kinds of headache accounted for 16. 3%(6/43),however,there were no significant differences about the types of headache between the two groups (all P>0. 05 ). Conclusion Young patients without previous history of migraine,especially women of childbearing age with sudden onset and progressive worsening headache,and the patients with idiopathic intracranial hypertension,CVST should be considered as an important possibility.
