2.Preparation of novel bulk carbon nano tubes dental materials for implant
Chinese Journal of Tissue Engineering Research 2008;12(1):180-184
BACKGROUND: Carbon nanotubes (CNTs) is about 1 mm in diameter and about 1-10 μm in length cylinder-like material, and it only contains the spiral of carbon atoms arranged on the surface. Because of its unique tubular structure, lower density than that of graphite and with a high enough intensity, CNTs are expected to substitute bone.OBJECTIVE: To primarily study the influence of the content of polycarbosilane (PCS) and the sintering pressure on CNTs, look for the most optimum condition for the preparation of chunk CNTs, analyze the physical properties and biocompatibility of the material, and provide a choice for candidate implant denture material.DESIGN: The content of cement and the sintering pressure are adjusted in the experiment to make sure the most optimum condition for the preparation, and the capabilities of the material are observed and analyzed to make sure if that the material is suitable for implant denture.SETTING: Department of Stomatology, Xiangya Hospital, Central South University; Prosthodontics Staff Room, Oral Medical College of Central South University. MATERIALS: 80% pure molecules of CNT (Nano Lab Company USA), 20-40 nm in diameter, 5-20 mm in length, was provided by Nacheng Technology Development Corporation of Beijing. PCS was compounded by 502 Development Room of Defense Technology University, and the molecular weight is 1 300; Ten male white Wistar rats with 8 weeks old were provided by Animal Experiment Center of Guangxi Medical University. The treatments with the animals accorded to animal ethical standard in the experiment.METHODS: The experiment was carried out in the Experimental Center of Material College, Central South University from January to August 2005. By using the spark plasma sintering (SPS), at the sintering temperature of 1 200 ℃, the pressure from 20 MPa to 60 MPa, PCS was used as the cement to produce the new chunk multi-wall CNTs.MAIN OUTCOME MEASURES: By using Archimedes method and adsorption of gases method, Japanese MitutoyoHM-101 little burden Vickers hardness meter was used to test the apparent bulk density, specific surface area, Vickers hardness at the different contents of PCS and sintering pressures. Scanning electron microscope (SEM) and transmission electron microscope (TEM) were used to observe the microstructure of the material.RESULTS: ① Microstructure: SEM and TEM indicated that the nanotube structure of the material was retained after sintered. ② Conforming phase of material: X-ray diffractometer indicated that carbon nanotubes were felt by PCS pyrolysis product of SiC; along with the PCS content and sintering pressure increased, the specific surface area was reduced, and Vickers hardness and density increased. Specific surface area decreased. ③ Histological compatibility: The inflammatory reactions of the CNTs/PCS were little more serious along with the increase of the content of PCS. The physical property of carbon nanotubes was prepared in this way, it was closed to bone tissues, it had the better biocompatibility, and it was the suitable candidate implant denture material.CONCLUSION: Appropriate to PCS for the bonding agent, through SPS, at 1 200 ℃, 20-60 MPa sintering pressure, CNTs/PCS which the material property is closed to bone tissues can be prepared, and CNTs/PCS subcutaneous implantation in rats does not show significant rejection; therefore, it is suitable candidate for dental implants material.

BACKGROUND: The vibration of high speed handpiece during tooth preparation may have adverse effect on the continuous force of the bonding interfaces among the cement, dentin, and post core. OBJECTIVE: To find the effect of different types of cements on the coronal microleakage of fiber post under different tooth preparation timings, and to provide evidence for the clinical operation. METHODS: The extracted mandibular premolars were divided into 3 groups, and they were cemented by Rely X luting, Panavia F and Paracora 5 mL, respectively. Each group was averagely divided into 3 subgroups: A, B, C groups which were prepared 15 minutes, 45 minutes, and 90 minutes after the cements mixed. All roots were then dyed and transparented. The teeth were observed under stereoscopic microscope and the dyeing scores were also recorded. RESULTS AND CONCLUSION: Rely X luting caused the highest coronal microleakage of fiber post, followed by Panavia F, while Paracore 5 mL, brought about the lowest under the same tooth preparation timing. Different tooth preparation timing caused no significant effect on the coronal microleakage of fiber post when the cement had been completely set.

Objective To study the effect of electroacupuncture (EA) combined with repetitive transcranial magnetic stimulation (rTMS) on endogenous neural atem cells, as well as learning and memory in rats with cerebral ischemia. Methods One hundred and twenty male Wistar rats were randomly divided into 5 groups: a normal group, a model group, an EA group, a rTMS group and an EA plus rTMS group. After establishing acute middle cer-ebral artery occlusion, the rats in the various experimental groups were treated with EA, rTMS or EA plus rTMS. The rats' ability to avoid an electric shock was observed using a jumping stand, Immunohistochemical techniques were used to measure the infarct area and the number of Brdu-positive cells. Results A small quantity of Brdu labelled cells was observed in the subgranular zone (SGZ) and the subventricular zone (SVZ) in rats in the normal group. In the EA, rTMS and EA plus rTMS groups, the numbers of Brdu labelled cells in the SVZ and SGZ was significantly higher than in the model group after both 7 and 14 days. The expression of Brdu around the SVZ and SGZ of the 3 treatment groups increased, especially in the EA plus rTMS group. At 7, 14 and 28 d there was significant improve-ment in the neural function and electric shock avoidance test scores in the EA, rTMS and EA plus rTMS groups com-pared with the model group, with the EA plus rTMS animals scoring best. Conclusion EA plus rTMS can promote the proliferation of nerve stem cells, facilitate neurological recovery and improve learning and memory in rats after cerebral ischemia.

Objective:To investigate the expression of acetyl-CoA carboxylase 1 (ACC1) in ovarian cancer tissues and cells, and the related mechanisms of the effect of ACC1 on cell migration and lipogenesis in ovarian cancer. Methods:Samples including 1 case of normal ovarian tissue, 1 case of ovarian cancer primary lesion tissue and 1 case of ovarian cancer omentum metastatic tissue diagnosed by pathology examination of patients undergoing surgery resection who admitted to Linyi Cancer Hospital between January 2019 and December 2021 were collected. Immunohistochemistry was used to detect the protein levels of ACC1 and Yin Yang protein 1 (YY1) of all tissues. The PROMO database was used to predict the possible binding sites of YY1 and ACC1 promoter region. Through the assembled viral vector, the HEY cells of human ovarian cancer with ACC1 or YY1 expression [the untreated cells were treated as the negative control (NC)], or knocked down ACC1 or YY1 (the interference sequence sh1, sh2, sh3 was transferred to the target gene, and the negative control sequence shNC was transferred to the interference sequence). Double luciferase reporter gene assay was used to verify the binding sites of YY1 and ACC1 promoter and the activity of transcriptional regulation. Real-time fluorescence quantitative polymerase chain reaction (qRT-PCR) and Western blot were used to detect the mRNA and protein expression levels of ACC1 and YY1 in the treated HEY cells, respectively. Transwell assay was used to detect the migration ability of HEY cells. Oil red O staining and Nile red staining were used to detect the lipid droplets in HEY cells. Results:The immunohistochemical scores of ACC1 and YY1 were 0, 2, 8 scores and 0, 4, 6 scores, respectively in normal ovarian tissue, primary lesion of ovarian cancer, and omentum metastatic tissue. Transwell assay showed that the number of invasive HEY cells in ACC1 overexpression group was more than that in NC group [(87.7±7.4) vs. (52.2±4.2), t = 5.19, P = 0.003]. The number of invasive HEY cells in ACC1-sh1 group, and ACC1-sh2 group with the knockdown of ACC1 was less than that in shNC group [(21.2±1.5), (29.7±2.3) vs. (56.2±5.3); t value was 6.41, 3.77; P < 0.001, P < 0.005]. The number of lipid droplets in HEY cells in the ACC1 overexpression group was more than that in the control NC group [Oil red O staining: (301±25) vs. (215±21); Nile red staining: (287±15) vs. (207±10); all P < 0.05]; the number of lipid droplets in HEY cells in ACC1-sh1 and ACC1-sh2 group with the knockdown of ACC1 was less than that in ACC1-shNC group [Oil red O staining: (113±8), (119±12) vs. (195±18); Nile red staining: (82±8), (117±11) vs. (165±17); all P < 0.05]. The result of dual luciferase reporter assay showed that overexpression of YY1 promoted the luciferase activity of the wild type ACC1 promoter region report gene ( P = 0.003), while the luciferase activity of the report gene was inhibited compared with the wild type after the mutation of binding sites of YY1 in ACCI promoter region ( P = 0.008). Western blot results showed that the expression levels of YY1 and ACC1 protein in HEY cells with YY1 overexpression group were higher than those in NC group, which indicated a synergistic increasing trend of both YY1 and ACC1; the expression levels of YY1 and ACC1 protein in YY1-sh1 group, YY1-sh2 group and YY1-sh3 group with the knockdown of YY1 were lower than those in the control YY1-shNC group, which indicated a synergistic decreasing trend of both YY1 and ACC1. Conclusions:ACC1 and YY1 are highly expressed in ovarian cancer metastatic tissues and both show a positive correlation trend. The expression level of ACC1 in vitro has an impact on cell migration and lipogenesis in ovarian cancer via YY1 transcriptionally regulating ACC1.

Chinese Journal of Physical Medicine and Rehabilitation 2022;44(9):779-783
Objective:To compare the effect of age and gender on mastication efficiency between healthy volunteers and dysphagic stroke survivors and to document any correlation of mastication efficiency with mandible movements.Methods:Thirty-two stroke survivors with dysphagia and 84 healthy volunteers were asked to chew two-color gum. Their chewing efficiency was indicated by the degree of color mixing after chewing (SDHue). The SDHue value was evaluated using ViewGum software. The healthy volunteers were further divided into an age 20-40 years group, a 41-60 years group and an over-60 group. The SDHue values were compared among the 3 groups. The SDHue values were correlated with clinical mandibule movement scores. Results:Among the healthy volunteers, those older than 60 years had significantly higher SDHue values on average than those between 20 and 40 years. However, no significant gender difference was observed. The dysphagic stroke survivors had, on average, significantly higher SDHue results than the healthy volunteers, and those results were positively correlated with their mandible movement scores. Conclusions:Mastication efficiency tends to be age-related but not gender-related. Among those with post-stroke dysphagia mandible movement is positively correlated with mastication efficiency.
