1.Effects of ginsenoside Rg3 on vasculogenic mimicry of breast cancer MCF-7 cell line

Objective To investigate the inhibitory effects and mechanism of ginsenoside Rg3 on vasculogenic mimicry of human breast cancer MCF-7 cell line. Methods The MCF-7 cells at logarithmic growth phase were obtained, and were cultured with different concentrations (0, 20, 50, 100, 150 and 300 mg/L) of ginsenoside Rg3. Cells cultured without Rg3 were served as controls. The IC50 were determined by CCK8 assay and anti-angiogenic effects were performed for testing the potential of tube-like structure (TLSs) formation. The expression levels of VEGF-A, MMP 9 and HIF-1αwere detected by Western blotting and real-time PCR. The secreted contents of VEGF-A and MMP9 were detected by enzyme linked immunosorbent assay (ELISA). Results The ginsenoside Rg3 suppressed the proliferation of MCF-7 cells in a dose-dependent manner, in which IC50 was (115.34±8.50) mg/L. The formation numbers of TLSs in MCF-7 cells were significantly inhibited by Rg3 in concentration dependent manner in 50 mg/L, 100 mg/L and 150 mg/L for (19.0 ± 1.0), (15.0 ± 1.5), and (10.0±1.7) vs. controls (22.0±1.8, F=150.805, P<0.05). The mRNA and protein expression levels of VEGF-A, MMP9 and HIF-1αprotein were inhibited by 50 mg/L,100 mg/L and 150 mg/L Rg3 vs. controls (P<0.05). Meanwhile, the contents of VEGF-A in MCF-7 cell supernatant was down-regulated by 50 mg/L,100 mg/L and 150 mg/L Rg3 vs. controls (P<0.05). The contents of MMP-9 in MCF-7 cell supernatant was down-regulated by 100 mg/L and 150 mg/L Rg3 vs. controls (P<0.05). There was no significant difference in MMP-9 expression between 50 mg/L group and control group. Conclusion The ginsenoside Rg3 is able to inhibit the vasculogenic mimicry of MCF-7 cells, which may be related with the down-
regulation of VEGF-A, MMP9 and HIF-1α.
2.Clinical Observation of Yiqi Huayu Prescription for 52 Cases of Diabetes Peripheral Nerve and Blood Vessel Pathological Change

Objective To observe clinical therapeutic effect of Yiqi Huayu Prescription for diabetes peripheral nerve and blood vessel pathological change. Methods One hundred patients with diabetes peripheral nerve and blood vessel pathological change were randomly divided into two groups. The patients in control group (n=48) were treated by routine treatment. The patients in treatment group (n =52) were treated by oral use of Yiqi Huayu Prescription and routine treatment. Their therapeutic effects were compared after three courses of treatment, which was 30 days. Results The total effective rate was 92.9% in the treatment group and 61.1% in the control group. There was significant difference between the two groups (P 0.05). 3.Design of Pure Capacitance Phonocardiogram Sensor Based on Phantom Power Supply

Objective To research a pure capacitance sensor for acquisition of cardiac phonocardiogram. Methods Under the phantom power supply (48V), the faint vibration signal of the heart was processed into electric signal by larger pre - treatment circuit module, shielded cable and the sensor pre-amplifier of circuit module. Results With some experiments, it can be proved that the sensor won't have affect on the sound field which frequency band under 1 000Hz and can detect the level of sound field like 20dB:30dB and 150dB or higher. Conclusion So with the sensor, the precise signal which can exactly reflect the status of patients' hearts can be detected, and it suits to work in complex condition for its high sensitivity, wide frequency band and low distortion.
4.Effect of miR-210 on angiogenesis of renal clear cell carcinoma

Objective To investigate the effect of miR-210 on angiogenesis of human renal clear cell carcinoma . Methods Detect the expression of miR-210 in 40 cases of renal clear cell carcinoma tissues , and analyze the rela-tionship between the expression of miR-210 and microvessel density ( MVD) .miR-210-ASO was lipotransfected in-to renal clear cell carcinoma cell line 786-O.RT-qPCR was used to verify the transfection effect .The effect of the supernatant of control group , negative control group and miR-210-ASO group tumor cells on the lumen formation of human umbilical vein endothelial cells ( HUVEC) was observed in a 3-D culturesystems .The transplantation tumor model of nude mice was established , and the effect of miR-210 on the formation of the transplanted tumor micro vessel was observed by endomucin and VEGF immunofluorescence staining under laser scanning confocal micro -scope.Results The expression of miR-210 was positively correlated with microvessel density in renal clear cell carcinoma ( P<0.05 ) .The expression of miR-210 was significantly decreased in 786-O cells transfected with miR-210-ASO( P<0.05 ) .The lumen formation of HUVEC cells co cultured with miR-210-ASO group cell supernatant was significantly less than that of control group and negative control group ( P<0.05 ) .The tumor volume of miR-210-ASO group was less than that of the control group , and the number of the micro vessel and the VEGF expres-sion were significantly less than that of the control group ( P<0.05 ) .Conclusions Inhibition of miR-210 can suppress blood vessel formation in renal clear cell carcinoma .
5.Brain activation of acupuncture stimulation at Zusanli acupoint on heroin addicts

0bjective To observe the different brain activation of acupuncture and electroacupuncture on healthy subjects and healthy subjects.Methods The different brain activation involved in heroin addiction between healthy subjects and addicts was detected by fMRI.Acupuncture point used in present study Was Zusanli (ST 36).Results Different brain activations between healthy subjects and addicts during electroacupuncture were hypothalamus(X0,Y2,Z9,t=7.36,P<0.01),anterior cingulate(X5,Y49,Z8,t=4.11,P<0.01),tempo-ral gyrus(X61,Y12,Z8,t=3.05,P<0.01).The difference of activated regions during conventional acupuncture between healthy subjects and heroin addicts was thalamus(X2,Y16,Z12,t=2.87,P<0.01),parahippocampus (X17,Y52,Z3,t=3.14,P<0.01),and hypothalamus(X0,Y2,Z9,t=6.98,P<0.01).Conclusion Regions with significant activation detected by fMRI are different during acupuncture in heroin addicts and in the healthy subjects.Notably,the hypothalamus activation is more robust in the addicts than in the healthy subjects during ac-upuncture stimulation.
6.DETERMINATION OF BIOTIN IN FOODS AND FEEDS

Objective: To establish microbiological method for determining biotin in foods and feeds. Methods: Using biotin dependent microorganisms Lactobacillus plantarum(ATCC 8014), the biotin content was detected indirectly by the growth of cultrued bacteria spectrophotometrically.Results: The detection limit was 0.03 ng. The relative standard deviations(RSD) of within and between run assays were 2.2%-3.8%,2.1%-5.3% respectively. The recovery of added biotin was 93.4%-104.6%, and RSD was 3.1%-4.1%. Conclusion: This assay is sufficiently accurate and repoducible for determining biotin in foods and feeds.
7.Impact of deferoxamine intervention on perihematoma tissue in rats with intracerebral hemorrhage

Objective To investigate the effect of the abnormally accumulated iron ions in the perihematoma tissue lesions and the impact of deferoxamine intervention. Methods A total of 135 SD rats were randomly assigned to sham-operation (n = 15), intracerebral hemorrhage (n =60) and deferoxamine (n =60) groups. A rat model of intracerebral hemorrhage was induced by the infusion of autologous blood. The neurological deficit score, brain water content (dry/wet weight method), blood-brain barrier permeability (Evans blue extravasation method), DNA fragmentation (TUNEL staining), and the impact of deferoxamine intervention were observed at different time points in all groups.Results One to seven days after intracerebral hemorrhage, the neurological deficit score, perihematomal water content, the blood-brain barrier permeability, and the numbers of TUNEL positive cells in the intracerebral hemorrhage group were significantly higher than those in the sham-operation group (P <0.01, P <0.05, P <0.01, and P <0.05). All the observation indexes in the deferoxamine and intracerebral hemorrhage groups showed the same trend, however, the neurological score, perihematomal water content, the blood brain barrier permeability, and the numbers of TUNEL positive cells were significantly lower than those in the intracerebral hemorrhage group (P < 0.05, P < 0.05, P < 0.05, and P < 0.05). Conclusions The abnormal accumulation of iron ions involves in the pathological injury of perihematoma tissue after intracerebral hemorrhage, and deferoxamine may reduce this injury.
8.Comparison of clinical effect of perforated gastroduodenal ulcer treatment by laparoscopy and traditional open repair

Objective To compare the clinical effects of perforated gastroduodenal ulcer treatment by laparoscopy and traditional open repair.Methods 70 cases of patients with perforated diagnosed as gastroduodenal ulcer were selected and divided into laparoscopic group and open group by different operation type.Laparoscopy was used in laparoscopic group,while traditional open repair was used in open group.The mean time in operation,intraoperative blood loss and intraoperative and peostoperative complications were observed and recorded.Results There was no difference in operative mean time between the two groups(P > 0.05),but intraoperative blood loss was less in laparoscopic group than that in open group (P < 0.05),exhaust time and hospitalization time in laparoscopic group were shorter than that in open group(P < 0.05),and no complication was found in laparoscopic group(0.00%),while 10 cases in open group(22.73%),the rate in the former group was lower than that in the latter group(P <0.05).Conclusion Using laparoscopy to repair perforated gastroduodenal ulcer had little trauma,less pain,less complication,shorter recover time and safe.It is worthy of clinical.
9.Chemical Monitoring Testing Package Application for High Pressure-steam Sterilization:An Observation and Analysis

OBJECTIVE To observe and compare the application result of two kinds of chemical indicator(CI)card contained in high pressure-steam sterilization chemical monitoring testing package for providing correct credible evidence of supplying material in each batch after high pressure-steam sterilization and to avoid resource lost because of fault estimation.METHODS The chemical monitoring testing package was made according Sterilization Criteria published in 2002,with the 3M 1250 and 1243 CI cards and 1292 biology indicator(BI) to observe the results in tested package after sterilization.RESULTS The BI in chemical monitoring testing package was all qualified,the qualified rate of 1250 was 70%,while of 1243 was 100%.CONCLUSIONS If there is no BI for monitoring,1243 CI card should be chosen in high pressure-steam sterilization chemical monitoring testing package for supplying material in each batch after high pressure-steam sterilization which is not influenced by moisture and the contacted material and is easy to read and evaluate,the resource lost caused by fault estimation can be avoided.
10.Effect of anti-HER-2 engineered antibody chA21 on angiogenesis in nude mice xenografts of human ovarian cancer SKOV3 cells

Purpose To investigate the effect of anti-HER-2 engineered antibody chA21 on the angiogenesis in nude mice xenografts of human ovarian cancer SKOV3 cells that overexpresses HER-2.Methods An animal model with SKOV3 cells involved in nude mice was established and the mice were randomized into control group and chA21 group;chA21 was administrated via caudal vein for 5 weeks, meanwhile the same volume of chA21 buffer was injected in control group.The mice were terminated after 5 weeks and the xenografts dislodged,VEGF and DLL4 expression in two groups were quantitatively analyzed by immunohistochemistry on tissue microarray sections combined with a micro-image analyzing system, and microvessel density (MVD) values were compared between the two groups by immunohistochemical staining of CD31.Results The expression of VEGF, DLL4 and MVD values in chA21 group were lower than those in control group (P<0.05).Conclusion chA21 inhibits the angiogenesis of transplantations of human ovarian cancer SKOV3 cells effectively.
