1.Apoptosis Changes in Rat Retina after Ischemia-reperfusion Injury

Objective To establish the rat ischemia-reperfusion injury model and learn the rat apoptosis changes after ischemia-reperfusion injury.Methods Ligate rat left common carotid artery 1h,reperfuse and exam retinal apoptosis in 1,6,12,24,48 and 72h groups.Result Apoptosis mainly occurs in retina ganglion cell layer and inner nuclear layer,occasionally in outer nuclear layer.Apoptosis occurs in 6h after reperfusion.In 24h,apoptosis achieves the peak,then decrease gradually.However,in 72h after reperfusion,there is apoptosis in rat retina.Conclusion rat retina ischemia-reperfusion injury can be observed after ligating common carotid artery.Apoptosis may play an important role in rat retina ischemia-reperfusion injury.In 72h after the operation,the injured retina didn't recover completely.
2.Changes in serum C-reactive protein level and erythrocyte sedimentation rate in patients after use of orthopedic metal implants

BACKGROUND:Most orthopedic surgeries use metal implants. Although this approach wil benefit for patients, it simultaneously makes patients infected by some bacteria. Serum C-reactive protein level and erythrocyte sedimentation rate are two indices for clinical y determining the inflammation. OBJECTIVE:To investigate the changes in serum C-reactive protein level and erythrocyte sedimentation rate in patients after use of orthopedic metal implants. METHODS:The clinical data of 100 patients who used metal implants between October 2012 and October 2014 at the Department of Orthopedics, Linqu People’s Hospital of Shandong Province of China were retrospectively analyzed. Among these patients, 36 patients received implantation of intramedul ary nails, 34 patients artificial joint, and 30 patients steel plates. At 1, 3, 7, 14, and 21 days after implantation, infection was determined according to serum C-reactive protein level and erythrocyte sedimentation rate, as wel as other laboratory indices, wound healing, and body temperature. According to these indices, the patients were assigned to two groups:no infection (n=52) and infection (n=48). Disease progression was analyzed for providing reliable evidence for rational use of antibiotics. RESULTS AND CONCLUSION:There were significant differences in serum C-reactive protein level and erythrocyte sedimentation rate in patients between infection and non-infection groups at 1 (t=3.43, P<0.05;t=3.78, P<0.05), 3 (t=3.35, P<0.05;t=3.68, P<0.05), 7 (t=3.28, P<0.05;t=3.54, P<0.05), and 14 days after use of orthopedic metal implants (t=2.56, P<0.05;t=3.02, P<0.05). No significant differences in these two indices were observed between infection and non-infection groups at 21 days after use of orthopedic metal implants (t=1.76, P>0.05;t=1.98, P>0.05). These findings suggest that after use of orthopedic metal implants, serum C-reactive protein level and erythrocyte sedimentation rate in patients should be closely monitored and simultaneously other indices like wound healing and body temperature should be determined to determine whether infection occurs after use of orthopedic metal implants so as to develop effective repair strategies. This wil provide reliable clinical evidence for rational treatment and prevention of orthopedics-related diseases.

4.Effect of dexmedetomidine combined with propofol for gastroscopy in elderly patients

Objective To evaluate the efficacy and safety of dexmedetomidine combined with propofol for gastroscopy in elderly patients.Methods Ninety ASA physical status Ⅰ-Ⅲ patients,aged 65-80 yr,weighing 45-80 kg,scheduled for elective gastroscopy,were randomized to receive either dexmedetomidine combined with propofol (group DP,n =45) or propofol (group P,n =45).In group DP,dexmedetomidine 0.4 μg/kg was infused over 5 min and propofol 1.0 mg/kg was injected intravenously 25 min later.Propofol 2.0 mg/kg was injected intravenously in group P.When body movement occurred during operation,additional propofol 0.2 mg/kg was given intermittently.Mean arterial pressure (MAP),heart rate (HR),pulse oxygen saturation (SpO2) and Narcotrend index were monitored during the whole procedures.The consumption of propofol and vasoactive agents,adverse cardiovascular events,and respiratory depression were recorded.Results Compared with group P,the consumption of propofol,incidences of respiratory depression and tachycardia,and degree of body movement were significantly decreased (P < 0.05),Narcotrend index was increased during operation (P < 0.01),and no significant change in the incidence of circulatory depression was found in group DP (P > 0.05).Conclusion Small dose of dexmedetomidine 0.4 μg/kg combined with small dose of propofol 1.0 mg/kg provides good efficacy and safety for gastroscopy in elderly patients.

Objective To evaluate the effect and safety of point injection and em bedding therapy on postoperative pain of hemorrhoid patients. Me thods Totally 105 hemorrhoid patients were rand omized into a treatment group (54 cases) and a control group (51 cases). The tre atment group was treated by point injec tion and embedding therapy at Changqiang point (GV 1) under the local anesthesia . The control group was treated by Indometacin Suppository after operation. On the fir st, second, four th and seventh day of the treatment, the pain occurrence, pain severity, and com prehensive effect scores were recorded for the evaluation of therapeutic effect and safety. 7.Application of isola system in reconstruction of lumbosacral joint after resection of high-level sacral tumors

[Objective]To investigate the operation indications and techniques of the ISOLA spinal system in reconstruction of the lumber-pelvis continuity after resection of high-level sacral tumors. [Method]A retrospective study was performed in 43 patients with high-level sacral tumor (HLST) between July 2001 to January 2007. Symptoms, signs, histological findings of the tumors and their outcomes were analyzed. All patients underwent resection of HLST and spinopelvic reconstruction with ISOLA spinal system,11 unilateral and 32 bilateral.[Result]Among the 43 patients, 33(77.1%)had chordoma, giant cell tumor of bone and neurogenic tumors. Thirty-four patients alive were followed-up for an average of 35 months(12~62 months), 5 patients had a recurrence of chordoma and 29 patients with HLST were under control. The pain originated from the lumbosacral joint and the spinal nerve root was obviously relieved.The patients could stand and ambulate normally. No loose screw or broken rod were found.X-rays showed no signs of the enlarged screw canal or lowered lumbar spine.The effect was 76.5%(26/34) excellent judged by the ISOLS system.[Conclusion]Spinopelvic reconstruction by ISOLA system after resection of HLST is characterized by easy manipulation, less complications and instant stable fixation. It may be widely used in the clinical practice.
8.Research progress of MicroRNA-34a in hepatocellular carcinoma

Primary hepatocellular carcinoma (HCC) is one of the most common malignant tumors worldwide.MicroRNAs (miRNAs) are a kind of endogenous non coding singlestranded RNAs,which can function as a tumor promoter or suppressor in the development and progression of human malignant tumors.Studies have shown that miRNAs play an important role in the occurrence and development of HCC.Numerous studies have found that the expression of miR-34a in various tumors is reduced and it participates in the carcinogenesis.Currently,the role of miR-34a in the tumorigenesis and development of HCC has attracted widespread attentions.Studies have confirmed that miR-34a can regulate the expression of its target genes and thus affect some key biological processes such as cell proliferation,apoptosis,invasion and metastasis of HCC.In this paper,we mainly reviewed the recent research progress on the role of miR34a in HCC.
9.Effects of ginsenoside Rg3 on vasculogenic mimicry of breast cancer MCF-7 cell line

Objective To investigate the inhibitory effects and mechanism of ginsenoside Rg3 on vasculogenic mimicry of human breast cancer MCF-7 cell line. Methods The MCF-7 cells at logarithmic growth phase were obtained, and were cultured with different concentrations (0, 20, 50, 100, 150 and 300 mg/L) of ginsenoside Rg3. Cells cultured without Rg3 were served as controls. The IC50 were determined by CCK8 assay and anti-angiogenic effects were performed for testing the potential of tube-like structure (TLSs) formation. The expression levels of VEGF-A, MMP 9 and HIF-1αwere detected by Western blotting and real-time PCR. The secreted contents of VEGF-A and MMP9 were detected by enzyme linked immunosorbent assay (ELISA). Results The ginsenoside Rg3 suppressed the proliferation of MCF-7 cells in a dose-dependent manner, in which IC50 was (115.34±8.50) mg/L. The formation numbers of TLSs in MCF-7 cells were significantly inhibited by Rg3 in concentration dependent manner in 50 mg/L, 100 mg/L and 150 mg/L for (19.0 ± 1.0), (15.0 ± 1.5), and (10.0±1.7) vs. controls (22.0±1.8, F=150.805, P<0.05). The mRNA and protein expression levels of VEGF-A, MMP9 and HIF-1αprotein were inhibited by 50 mg/L,100 mg/L and 150 mg/L Rg3 vs. controls (P<0.05). Meanwhile, the contents of VEGF-A in MCF-7 cell supernatant was down-regulated by 50 mg/L,100 mg/L and 150 mg/L Rg3 vs. controls (P<0.05). The contents of MMP-9 in MCF-7 cell supernatant was down-regulated by 100 mg/L and 150 mg/L Rg3 vs. controls (P<0.05). There was no significant difference in MMP-9 expression between 50 mg/L group and control group. Conclusion The ginsenoside Rg3 is able to inhibit the vasculogenic mimicry of MCF-7 cells, which may be related with the down-
regulation of VEGF-A, MMP9 and HIF-1α.

Objective To observe the protective effect of Ulinastatin in rat islet isolation and advance an improved method in rat pancreas islet isolation.Methods Rat pancreas was perfused with Hanks solution containing collagenase.
