1.Design of Pure Capacitance Phonocardiogram Sensor Based on Phantom Power Supply

Objective To research a pure capacitance sensor for acquisition of cardiac phonocardiogram. Methods Under the phantom power supply (48V), the faint vibration signal of the heart was processed into electric signal by larger pre - treatment circuit module, shielded cable and the sensor pre-amplifier of circuit module. Results With some experiments, it can be proved that the sensor won't have affect on the sound field which frequency band under 1 000Hz and can detect the level of sound field like 20dB:30dB and 150dB or higher. Conclusion So with the sensor, the precise signal which can exactly reflect the status of patients' hearts can be detected, and it suits to work in complex condition for its high sensitivity, wide frequency band and low distortion.
2.Clinical analysis of 10 patients with severe acute pancreatitis with peripancreatic necrosis treated by minimally invasive technique of retroperitoneal approach

Objective To evaluate the efficacy of minimally invasive technique by retroperitoneal approach in the treatment of severe acute pancreatitis with peripancreatic necrosis.Methods The clinical data of 10 patients with severe acute pancreatitis combined with peripancreatic necrosis who underwent retroperitoneal drainage by aspirating and placing tube under B ultrasound guidance on the basis of routine therapy in the First Affiliated Hospital of University of South China from January 2016 to September 2016 were analyzed.Results All of the 10 patients successfully underwent puncture.Drainage was good after puncture and tube placement in 7 patients.The amylase activity was normal within 3 ~ 7 days,and the symptoms of abdominal pain and bloating were alleviated in 2 ~ 4 days.Among them,3 cases had poor drainage after catheterization due to a large necrotic area,and the percutaneous nephroscope was performed to remove the necrotic tissue of the pancreas.Bacteria was detected in the drainage fluid of 7 patients.The complications of pneumothorax,hemothorax and organ injury were not found in all the patients.The average hospitalization time was 19.4 days (10 ~ 34 days).Conclusions Minimally invasive retroperitoneal approach is safe and effective in curing severe acute pancreatitis with peripancreatic necrosis with good efficacy,which is worth for clinical recommendation and application.
3.Role of apoptosis related gene Bcl-2 and Bax during periodontal ligament remodeling in rats

Objective: To study the influence of occlusal force changes on apoptosis related gene Bcl-2 and Bax in rat periodontal ligament fibroblasts (PDLF). Furthermore, to explore the possible mechanism in remodeling of periodontal ligaments. Methods: Animal model of occlusal force changes was established by extracting the right first, second and third maxillary molars in male SD rats. Rats were sacrificed at 6,12 hours and 1, 2, 3, 5, 7, 14 and 28 days after teeth extraction(n=6), and their alveolar bone tissues were harvested. HE staining was used to observe the morphologic changes of alveolar bone tissues and immunohistochemistry were used to detect Bcl-2 and Bax expressions in PDLF. The rats of normal occlusal forces were used as control(n=6). The results were assigned a mean score based on the expressions and the intensity of cell positively stained for Bcl-2 and Bax. Results: Animal model of occlusal force changes was successfully established. HE staining showed that the control group had more pycnotic periodontal ligament, orderly aligned fibres, and flatter alveolar bone than model group. 4.Analysis of methylation level of bone marrow mesenchymal stemcells during senescence by ultra performance liquidchromatography-tandem mass spectrometry

Aim To measure the methylation rate of bone marrow mesenchymal stem cells (BMMSCs) using ultra-performance liquid chromatography-tandem mass spectrometry, and determine the methylation rate in the process of senescence.Methods The DNA extracted from BMMSCs would be digested into individual deoxynucleosides using enzymatic hydrolysis.To quantify the global genomic DNA methylation rate,we developed a method using ultra-performance liquid chromatography-tandem mass spectrometry with multiple reaction monitoring to simultaneously measure the levels of 5-methyl deoxycytidine and deoxyguanosine in digested genomic DNA.Results The DNA methylation rate could be analyzed within two minutes after hydrolyzing 1μg DNA for an hour.The detection limit of DNA methylation rate was 5.00×10-4.The coefficient of variance of the intra-day and inter-day precision was within 7.12×10-2 and 0.119, respectively.With the subculture of BMMSCs, the methylation rate gradually decreased.The methylation rate of stem cells was the lowest in 4~6 generation, and gradually increased with the subculture.Conclusions Using this method, the preliminary data on DNA methylation of BMMSCs in the process of senescence are obtained.The method is simple and convenient for sample preprocessing, and the method is fast and accurate with good sensitivity and repeatability.
5.Intravascular occlusion and clinical manifestation for eyes of carotid cavernous fistula

Objective To evaluate the effect of intravascular occlusion for carotid cavernous fistula (CCF) by analying the clinical features of CCF. Methods Twelve cases with CCF were analysed on their medical histories, clinical features in the eyes, clinical examinations and the therapeutic effects of balloon occlusion in internal carotid artery.Results Ten cases were treated successfully(83.33%), in which 2 cases were treated once more. There was no operation complication. The visual acuity of one case increased from no light perception to finger movement within 1 week preoperatively. The visual acuity of 10 cases improved 1~4 lines (Snellen chart). Some clinical symptoms and signs such as the lid, conjunctiva swelling, diplopia, the movement of eyeball and retinal hemorrhage were decreased remarkably. Carotid fistula of 12 cases with CCF were found to be occlusioned by digital subtraction angiography after operations.Conclusion The early diagnosis and treatment for CCF are necessary. Color doppler flow imaging, CT and DSA are useful examination methods for diagnosis of CCF. Intravascular occlusion is the best therapeutic method for CCF nowadays.
6.A study on the mechanisms involved in early liver graft regeneration following cold ischemia in a rat liver transplantation model

Objective To study the molecular mechanism involved in early liver regeneration following cold ischemia after rat orthotopic liver transplantation(OLT). Methods Syngeneic(Lewis to Lewis)rat OLT with hepatic artery reconstruction was performed.Graft survival and liver regeneration following 1 h and 16 h cold ischemic injury were evaluated.Specimen were collected at predetermined intervals from 0,1,2,4,16,24,48,72 h post-reperfusion.The patterns of IL-6 and TNF-α expression,STAT3 activation,and upregulation of immediately early genes(IEGs)including c-fos,c-myc,and c-jun were determined in liver grafts with 1 h and 16 h cold ischemia times.Quantitative analysis of IL-6 at 2 h after reperfusion was performed for grafts preserved for 1 h and 16 h.Progression of hepatocyte replication was confirmed by bromodeoxyuridine(BrdU)immunohistochemistry.Positively BrdU-stained nuclei were quantitated and analyzed between the two groups at 48 h after reperfusion. Results Survival rate in the two groups was 100%.Compared to 1 h cold ischemia group,IL-6 and TNF-α expression and STAT3 activation in 16 h cold ischemia group were markedly increased.Extensive hepatocyte replication was present.Upregulation of immediate early genes of c-fos,c-myc,and c-jun was also observed.Grafts with 1 h ischemic iniury had lower levels of IL-6 than that when grafts suffering from ischemia for 16 h(t=27.7,P<0.05).Number of positively stained nuclei in 16 h group were more than that in 1 h group at 48 h after transplantation(t=13.4,P<0.05). Conclusions Severe cold ischemia initiated liver regeneration after rat OLT.TNF-α and IL-6 are important factors in early liver regeneration.IL-6/STA3 pathway in liver regeneration is an important part in the recovery of grafts following cold ischemia injury.
7.Liver graft regeneration after liver transplantation in IL-6 knockout mice

Objective To observe recipient survival time and liver graft regeneration in an orthotopic liver transplantation model using IL-6 knockout mice as a donor.Methods A model of liver transplantation in C57BL/6 WT and IL-6 KO mice with C57BL/6 background was established.Thirty eight mice were divided into three groups:C57BL/6 wild type→C57BL/6 wild type control group(n=10),IL-6 KO→IL-6 KO group(n=14)and IL-6 KO→C57BL/6 WT group(n=14).Hepatocyte replication with BrdU uptake after liver transplantation was examined by immunohistochemistry.Resuits The survival time in the control group was>16 days.and that was 2 days and 1.6 days in the IL-6 KO→IL-6 KO and IL-6KO→C57BL/6 WT,respectively.The difierence in survival time in all three groups was statistically significant(F=190.09,P<0.01).The liver grafts in control group showed minimal injury and no necrosis and mild increase of BrdU uptake at 48 hours.Patchy areas of necrosis and hepatocyte ballooning were observed in the two groups using IL-6 KO mice as donors,there was minimum increase in BrdU uptake at 48 hours post transplantation.Conclusion IL-6 KO liver grafts fail to regenerate after liver transplantation.IL-6 is an important factor in liver regerative response.
8.The influence of ShengJi-hongfen Cream on wound healing histology and bFGF content of rabbits

Objective To investigate the mechanism of Shengji-hongfen Cream(SJHFC)affecting basic fibroblast growth factor(bFGF)in wound and hastening wound healing.Methods Rabbit models of cutaneous deficiency and infection were setup.Self-contrast observation was used for comparing the two wound sides of model rabbit,which were administrated with SJHFC(treatment group)and vaseline(control group)respectively.Microstructure of wound surface and changes of bFGF weTe observed and measured.Results The content of fibroblasts,phlogocyte,collagen and blood vessel in the SJHFC group increased markedly,compared with the control group after one week.The content of bFGF was significantly changed than the control group at the 3rd and 7th day(P<0.05).Conclusion SJHFC Can promote bFGF content of the wound,that maybe the mechanism of its hastening wound healing.
9.The effect and mechanism of microRNA in the invasion and metastasis of pancreatic cancer

Evidence has shown that the abnormal expression of specific microRNAs (miRNAs) is associated with the occurrence,development and prognosis of pancreatic cancer,which is the potential target of forecast about invasion and metastasis of pancreatic cancer.There is close relation between miRNAs and biological characteristics of pancreatic cancer invasion and metastasis.Therefore,miRNAs are expected to become the new research targets of the mechanism of invasion and metastasis of pancreatic cancer.
10.Diagnosis and treatment of gastritis cystica profunda (6 cases)

Objective To analyze the clinical manifestations, computed tomography scan (CT), gastroscope, endo-scopic ultrasonography (EUS), and therapy method of gastritis cystica profunda. Methods Retrospectively analyzed clinical manifestations, CT, gastroscope, EUS, and pathological results of 6 cases of gastritis cystica profunda. Results In these 6 cases, 3 of them were doubted gastric carcinoma, 3 cases were considered stomach mass by CT. Gastroscope hinted apophysis lesions, but all cases were suggested gastritis cystica profunda by EUS. And all cases were removed through endoscopic submucosal dissection (ESD). Pathology were confirmed the diagnosis. Conclusion EUS combined with endoscopic mucosal resection (EMR) or ESD technique can improve the diagnostic rate. For gas-tritis cystica profunda which are not associated with malignant tumor can be treated through ESD.
