1.the Application of Homemade Stapler in Anus-retained Operation and it′s Relative Aspects.

Objective To investigate the application of homemade stapler in anus-retained operation on low and super-low colon or rectal cancer.Methods From August 1991 to August 2006,this technique was used in 333 patients with low colon or rectal cancer,among which 96 patients were right hemicolon excision,ileum colon anastomosis,colon sigmoideum excision,colon descendens rectum anastomosis and 183 patients were rectum pro-excision,low pro-excision,54 patients were low pro-excision,super low anastomosis.The other traditional operations(59 patients manipulation anastomosis) were used in 166 patients.Results Fourteen patients had anastomotic fistula(one rectovaginal fistula) in group1 and two in group2.Two patients had pro-sacral bone bleeding,and one dead.Conclusion The stapling device was a safe and feasible choice in anus-saving operation of low rectal cancer.
2.Distribution of free Ca~(2+) and characterization of Ca~(2+) channel in the cultured early-stage human embryonic retina

Objective To investigate the distribution of free Ca 2+ and characterization of Ca 2+ channel in the cultured 11~15 week human embryonic retina. Methods The cells from 11~15 week embryonic retina were dissociated enzymatically and cultured on coated cover slides. Three weeks later, Fluo3, a Ca 2+ indicator, was added in Ca 2+ containing or no Ca 2+ Hepes buffer and incubated with retinal cells for 30 min, meanwhile with or without verapamil, perdipine, or dexamethasone adding. Ca 2+ staining and Ca 2+ transit before and after 10 ?mol/L or 50 ?mol/L KCl stimulation was observed and recorded using Zeiss LM510 confocal laser scanning microscopy. Results In response to K + exposure, a rapid rise of free Ca 2+ in cytoplasma and nuclei of neuron and glial cell was observed, but was repressed with the treatment of verapamil, perdipine, or dexamethasone. The obvious Ca 2+ influx from cytoplasma into nuclei was observed even in the absence of external Ca 2+ . Conclusion L type Ca 2+ channel was expressed and functionally matured in the cultured early stage human embryonic retina.
3.Allele frequencies of 5 short tandem repeat loci of Kashin-Beck disease patients on chromosome 12

Objective To analyze the allele frequencies of 5 short tandem repeat(STR)loci(D12S313,D12S304,D12S1640,D12S1708 and D12S1583)on chromosome 12 among Kashin-Beck disease(KBD)patients and the control population living in the area suffered from KBD.Methods Fifty KBD patient8 and 50 non-KBD patients were chosen in endemic afea of Shaanxi Province,5 STR loci on chromosome 12 were genotyped by the technology of polymerase chain reacfion(PCR)and capillary electmphoresis.The pelymorphisms of STR in these popIllations were analyzed.The allele and genotype frequencies of each STR in the corresponding groups were caleulated and compared. Results In KBD group,the 5 STR loci had 8,6,7,5 and 11 types ofalleles and 17,11,15,8 and 28 genotypes, respectively;while in the control group,the number of aUele types of 5 STR loci were 6,8,6,4 and 10,the number of genotype of those loci were 13,21,14,8 and 23,respectively The allele frequence of D12S304 locus was statiBtically significant between KBD patients and controls(P<0.05),especially for the 319 bp allele(P<0.006 25). Conclusion There is an association between D12S304 locus and KBD.The 319 bp allele might play the key role.

8.Study on the relationship between the resting heart rate and target organ damage in elderly patients with metabolic syndrome

Objective To investigate the relationship between the resting heart rate (RHR) and target organ damage (TOD) in elderly patients with metabolic syndrome(MS). Methods 264 elderly patients with MS were divided into four groups according to the level of RHR: RHR1 group, RHR<65 beats/minute (bpm) (46 cases) ;RHR2 group, 65≤RHR<75 bpm (77 cases);RHR3 group, 75 bpm≤RHR<85 bpm (89 cases);RHR4 group, RHR≥85 bpm (52 cases).Electrocardiography, echocardiography, carotid uhrasonography, crcatinine clearance rate (Ccr) and quantitative assay of 24 hours' albuminuria were performed. Results (1) Compared with RHR1, RHR2 and RHR3 groups, RHR4 group showed higher levels of carotid intima-medial thickness (IMT), carotid arterial diameter (CAD), left ventricular mass index (LVMI) and albuminuria(P< 0.05 or P<0.01), and lower levels of left ventricular ejection fraction (LVEF) and Ccr (all P< 0.01). (2) The IMT, CAD, LVMI and albuminuria were positively correlated with RHR (r=0.33, 0.23, 0.61, 0.58, respectively, all P<0.01). However, the LVEF and Ccr were negatively correlated with RHR (r=-0.59, -0.51, all P<0.01). (3) Logistic multivariate analysis showed that RHR and pulse pressure (PP) had effects on myocardial hypertrophy, coronary heart disease, heart failure, cerebral stroke and renal dysfunction(P<0.05 or P<0.01). Except heart failure, PP played a more important role than RHR. Coneinsions RHR may be an independent risk factors for TOD in elderly patients with MS,and RHR regulation is important for the development of MS in the elderly.

Arnebia euchroma is the main source for medicinal herb Zicao. and its most important component shikonin compounds have high medicinal and industrial value. This research is aimed to build overexpression vectors and RNAi vectors for key secondary metabolism genes of A. euchroma, and bulid platform for constructions of related transgenic lines using GATEWAY technology. To build genetic material based genetic research platform is to provide a great convenience for digging and functional verification of the genes on secondary metabolic pathway, and also to fill the gaps in transgenic research of A. euchroma. This study is also important for the cultivation of shikonin high-yielding strains of A. euchroma.

10.The effect of Smad3 knockdown on the secretion of extracellular matrix induced by IGFBPrP1 in activated hepatic stellate cells

Objective To identify the effect of IGFBPrP1 on the secretion of extracellular matrix in hepatic stellate cells through the Smad3 signaling pathway. Methods (1)Two pairs of chemically synthesized siRNAs (siRNA1, siRNA2) targeting Smad3 were transfected into HSC-T6 cells,real-time PCR and Western blot were used to evaluate the silence efficiency, and the better siRNA was used. (2)HSC-T6 cells were divided into four groups: Negative control group, siRNA-Smad3 transfection group, siRNA-Smad3+IGFBPrP1 group and IGFBPrP1 group. The better siRNA was chosen to transfect into HSC-T6 cells. The protein expressions of Smad3, fibronectin and Collagen Ⅰ were evaluated by Western blot. Results (1)siRNA2-Smad3 inhibited Smad3 gene expression stronger than another siRNA. (2)After transfection of siRNA2-Smad3, the protein expression of Smad3 was significantly decreased compared to the negative control group(P<0.01). The protein expression of fibronectin and Collagen Ⅰ in IGFBPrP1 stimulating HSCs treated with siRNA2-Smad3 were significantly decreased compared to that in IGFBPrP1 stimulating HSC without siRNA2-Smad3 (P <0. 01 ).Conclusion IGFBPrP1 induces the secretion of extracellular matrix in hepatic stellate cells through the Smad3 signaling pathway.
