Objective To Observe the effects of combination regimen of gemcitabine and cisplatin in treating advanced non-small cell lung cancer (NSCLC).Methods 30 patients with advanced NSCLC were treated with GP(GEMZAR 1000mg/m2 d1,8;CDDP 25mg/m2 d1~3 ).The course was repeated every 3 weeks for at least 2 cycles.Results The overall response rate was 36.7%(11/30),whereas 13 patients had stable disease and 6 patients showed progressive disease.The response rate was 46% in untreated patients and 29% was obtained in treated ones.Significant difference was observed between the two groups (P
2.Clinical and imaging characteristics of posterior reversible encephalopathy syndrome

Objective To investigate the clinical and imaging characteristics of posterior reversible encephalopathy syndrome (PRES).Methods The clinical and imaging data of 7 patients with PRES were analyzed retrospectively.Results Among the 7 cases with PRES, 3 cases were secondary to pregnancy induced hypertension syndrome, 3 cases secondary to renal failure, 1 case secondary to hypertension.The clinical characteristics were that 7 cases presented with headache and disturbance of vision, 6 with epilepsy, 4 with nausea and vomitting, 1 with hemiparesis, 1 with ataxia.Cranial CT examination in 6 cases revealed occipital low density lesions in 3, widely white matter edema in 2, multiple small hematomas in 1, and no lesion in 2. Brain MRI showed that the occipital lobe was involved in 7 cases, cerebellar in 3, brain stem in 2, fronto-parietal subcortical white matter in 2, caudate nucleus in 1, thalamus in 1. The lesion showed gyrus-like abnominal signals in lobes and patchy abnormal signals in the other areas. The lesion showed low or iso-signal in T1WI, high signal in T2WI and Flair. 2 cases enhancement scannings showed no enhancement effect in 1 and gyrus, patchy and ring-like enhancement in another. The lesions demonstrated mild high signals in 2, low signal in 1 and no abnormalmality in 1 on DWI among 4 cases.Conclusions The headache, disturbance of vision and epilepsy are the principal clinical symptom in PRES . The imaging features of PRES are symmetrical long T1 and T2 signals on the bilateral posterior white matters of cerebrum.
3.Value of changes of serum complement, immunoglobulin and inflammatory cytokines in children with mycoplasma pneumonia

Objective:To investigate the clinical value of the changes of serum complement,immunoglobulin and inflammatory cytokines in children with mycoplasma pneumonia (MP).Methods:60 cases of children with MP infection were collected as MPP group,60 cases of healthy children as control group.The serum levels of complement (C3,C4) and immunoglobulin (IgA,IgG,IgM) were detected by INA,the serum levels of inflammatory cytokines (IL-8,IL-10,IL-13,TNF-α) were detected by ELISA.Results:①The serum levels of IgM,C3,C4,IL-8,TNF-et and IL-13 in MPP group in acute stage were significantly higher than those in the recovery stage and control group (P<0.05),the levels of IL-8,TNF-et,IL-13 in the recovery stage were significantly higher than those in control group (P<0.05);the serum levels of IgA,IL-10 in MPP group in acute stage were significantly lower than those in the recovery stage and control group,and these were still significantly lower than the control group (P<0.05);the IgG in MPP group in acute stage was not significant difference with control group (P>0.05),but it in recovery stage was significantly higher than the acute stage and the control group (P<0.05).②The serum levels of IgM,IgG,C3,C4,IL-8,TNF-et and IL-I 3 in MPP group in severe were significantly higher than those in the mild stage and control group (P<0.05),and the levels of IgM,IL-8,TNF-αt,IL-13 in the severe stage were significantly higher than those in control group (P<0.05);the serum levels of IgA,IL-10 in MPP group in severe stage were significantly lower than those in the mild stage and control group,and these in mild stage were still significantly lower than the control group (P< 0.05).Conclusion:Children with MP infection have a significant cellular and humoral immune dysfunction,the detection of the changes of serum complement,immunoglobulin and inflammatory cytokines is valuable to assessing the disease staging and degree.

Objective: Recent studies have suggested that congestive heart failure(CHF) is related to neuroendocrine system and cytokines,and its activation contributes to myocardial remodeling and leads to the development and progression of CHF.Tumour necrosis factor-?(TNF-?) is one of the most important factors.This study intends to establish the animal model of isoproterenol(ISO)-induced CHF in rats and to study the changes of serum TNF-?.Methods:Male SD rats were randomly allocated to the ISO-induced CHF group(n=50) receiving two subcutaneous injections of 170mg ISO per kilogram of body and the healthy control group(n=10) receiving two subcutaneous injections of 0.25ml normal saline.At 18th week,the hemodynamic parameters including HR,LVSP,LVEDP and ?dp/dtmax were assessed.TNF-? in plasma were all detected.Results:Compared with the control group,the LVSP、?dp/dtmax significantly decreased and the HR,LVEDP enlarged notably in CHF group(P
5.Effects of extracts of Dragon's blood on fibroblast proliferation and extracellular matrix hyaluronic acid.

OBJECTIVETo investigate the effects of Dragon' s blood extract on proliferation and secret extracellular matrix function of fibroblasts in vitro.
METHODSDragon' s blood was extracted by chloroform, acetoacetic ester, alcohol. Human fibroblast were cultured in vitro in media containing gradient dilutions of Dragon' s blood extracts (0.002, 0.02, 0.2, 2, 20 mg/ml) , which was followed by cell proliferation assessed with MTT assay on 0, 12, 24, 36, 48, 60, 72 h. Under the optimal concentration, the cell growth curves were drawn and the flow cytometry (FCM) was used to determine the changes of cell cycle. On 0, 12, 24, 36, 48, 60, 72 h, the concentration of hyaluronic acid in the supernatant of fibroblast culture was measured by radioimmunoassay.
RESULTS0.2-2 mg/ml Dragon' s blood extracts enhanced the proliferation of fibroblasts in a dose-dependent manner. 2 mg/ml was the optimal dilution of Dragon's blood extract, and it increased the ratio of S cells in cell cycle [(25.80 ± 3.10)%] than control group [(7.50 ± 0.70)%, P < 0.01]. From 12 h to 72 h, in 2 mg/ml Dragon's blood group, concentration of Hyaluronic acid secreted by fibroblasts gradually increased, but were less than control (P < 0.01).
CONCLUSIONSDragon's blood acetoacetic ester extract improved the proliferation of cultured human fibroblasts in vitro, might be beneficial to promote wound healing.

6.In Silico Cloning and Bioinformatics Analysis of AP2/ERF Family Gene from Brassica Oleracea

Objective:To obtain AP2/ERF genes from Brassica oleracea by using in silico cloning. Methods: AP2/ERF genes were cloned by retnieving the EST database and using the bioinformatics software with the Arabidopsis thaliana AP2/ERF as a querying probe. Results:Two AP2/ERF family transcriptional regulators (BoAP2/ERF1 and BoAP2/ERF2) were isolated from Brassica olera-cea by the in silico cloning method. Some characters of AP2/ERF protein were analyzed and predicted by the tools of bioinformatics in the following aspects including the composition of amino acid sequence, hydrophilicity and hydrophobicity, subcellular localization, secondary and tertiary structure of protein and function. Conclusion:Bioinformatical analysis shows BoAP2/ERF1 and BoAP2/ERF2 gene encode 40. 85kDa and 39. 44kDa protein with 371 and 352 amino acids. The domain is predicted to locate on nucleus. Sequence analysis indicates the protein may be involved in signaling transducer and stressing response roles in plantbiotic stresses.
7.Simultaneous Determination of 17 Amino Acids in Sarcocarp of Northeast Empetrum nigrum by HPLC-ELSD

OBJECTIVE:To determine the content of 17 amino acid in sarcocarp of northeast Empetrum nigrum simultaneous-ly. METHODS:HPLC-ELSD method was adopted. The determination was performed on Waters Symmetry C18 column with mobile phase consisted of 5.0 mmol/L heptafluorobutyric acid solution(containing 0.7% trifluoroacetic acid)-acetonitrile(gradient elution) at the flow rate of 2.0 L/min,the column temperature was 37 ℃,the drift tube temperature was set at 60 ℃. RESULTS:The line-ar ranges of glycine,serine,aminosuccinic acid,aminopropionic acid,threonine,glutamic acid,cystinol,diaminocaproic acid, histidine,arginine,proline,valine,methionine,oxyphenylaminopropionic acid,isoleucine,aminocaproic acid,phenylalanine were 0.0059-0.1877,0.0082-0.2628,0.0104-0.3328,0.0070-0.2227,0.0093-0.2978,0.0115-0.3678,0.0094-0.3004,0.0114-0.3655,0.0121-0.3880,0.0136-0.4355,0.0090-0.2878,0.0092-0.2930,0.0117-0.3730,0.0142-0.4530,0.0103-0.3280, 0.0103-0.3280 and 0.0129-0.4130 mg/mL(all r>0.9990). RSDs of precision,stability and repeatability tests were not all less than 2.0%. Recoveries were 98.0%-101.2%(RSD<2.0%,n=6). CONCLUSIONS:This method is simple,precision,stable and repeatable,and can be used for simultaneous determination of 17 amino acid in sarcocarp of northeast E. nigrum.
8.Effects of Dragon’s blood extracts on fibroblast proliferation and procollagen type III

BACKGROUND:Dragon’s blood is the main ingredient of traditional medicine prescription for promoting granulation, which has been used in clinical treatment of a variety of refractory wounds and achieved the exact effects. But the Dragon’s blood effect on colagen secretion from normal fibroblasts has not been reported. OBJECTIVE: To investigate the effects of Dragon’s blood extract on the proliferation and secret function of fibroblastsin vitro. METHODS: Dragon’s blood was extracted by extracts chloroform, acetoacetic ester, and alcohol in turn. Normal human fibroblasts were respectively cultured in Dragon’s blood extracts of chloroform, acetoacetic ester, and alcohol, DMEM containing 1% dimethyl sulfoxide, and normal culture medium. Then, the fibroblasts were cultured in vitro in different media containing gradient dilutions of Dragon’s blood extracts (0.002, 0.02, 0.2, 2, 20 g/L), which was folowed by cel proliferation determination assessed with MTT assay. Under the optimal concentration, the cel growth curves were drawn and the flow cytometry was used to determine the changes of cel cycle. The concentration of procolagen type III in the supernatant of the fibroblast culture systems was measured by radioimmunoassay. RESULTS AND CONCLUSION:0.2 g/L-2 g/L dilution of Dragon’s blood extracted by acetoacetic ester enhanced the proliferation of fibroblasts in a dose-dependent manner. The 2 g/L was the optimal dilution of Dragon’s blood extracted by acetoacetic ester, and it increased the ratio of S cels in cel cycle than control group and decreased procolagen type III. These findings indicate that Dragon’s blood acetoacetic ester extract can improve the proliferation of cultured human fibroblastsin vitro, and decrease the secretion of procolagen type III of fibroblasts, and it can be beneficial to improve wound healing and inhibit hypertrophic scar.
9.The effect and mechanisms of 20-HETE on myocardial ischemia reperfusion injury

Objective To investigate the effect of 20‐HETE on the isolated myocardial ischemia reperfusion injury and to ex‐plore its underlying mechanisms .Methods Experiments were performed in isolated rat hearts subjected to 35 min of ischemia fol‐lowed by 40 min of reperfusion in Langendorff preparations .HET0016 (1 μmol/L) and various concentrations (10 ,30 or 50 nmol/L) of 20‐HETE were infused 10 min before the onset of ischemia and throughout the reperfusion period .Cardiac hemodynamic changes and myocardial contractility were continuously recorded with the Powerlab /8P system .Myocardial infarct size was meas‐ured by TTC staining .The level of ROS and the protein carbonyl content were determined by DHE fuorescence and DNPH method , respectively .Results Perfusion with HET0016 significantly improved myocardial ischemia reperfusion injury reduction in cardiac contractility ,after inhibited the production of 20‐HETE significantly reduced the occurrence of myocardial infarction area (P<0 .05) ,but exogenous join 20‐HETE aggravated I/R‐induced myocardial injury (P<0 .05) .Myocardial ischemia reperfusion injury significantly increased production of ROS and oxidative stress ,both of which were significantly inhibited by HET 0016 and enhanced by 20‐HETE administration(P< 0 .05) .Conclusion 20‐HETE stimulates ROS production and enhance protein carbonylation , which aggravates myocardial ischemia reperfusion injury .
10.Clinical and imaging features of vertebrobasilar dolichoectasia in patients with ischemic stroke: a retrospective case series study

Objective To investigate the clinical and imaging features in patients with vertebrobasilar dolichoectasia (VBD) through a comparative study in patients with ischemic stroke with or without VBD.Methods The patients with acute cerebral infarction were divided into either a VBD group or a non-VBD group according to magnetic resonance angiography.The VBD group was further divided into an anterior circulation infarction subgroup and a posterior circulation infarction subgroup.The cardiovascular risk factors,the diameter of basiar artery (BA),bifurcation height,and horizontal displacement were compared in all groups.Results A total of 269 patients with acute cerebral infarction were included,28 had VBD,accounting for 10.41% of the patients with acute cerebral infarction during the same period.The proportion of male patients (78.6% vs.66.8% ;x2 =4.392,P =0.036),age (70.38 ± 10.58 years vs.62.86 ± 12.20 years; t =2.870,P =0.009),and the proportion of hypertension (89.3% vs.47.7% ; x2 =17.367,P =0.000) in the VBD group were significantly higher than those in the non-VBD group.The multivariate logistic regression analysis showed that the advanced age (odds ratio [OR] 1.248,95% confidence interval [CI] 1.137-1.371; P=0.000),hyperglycemia (OR 1.599,95% CI 1.181-2.164; P =0.002),hypertension (OR 1.251,95% CI 1.020-1.534; P =0.032) and increased triglyceride level (OR 1.876,95% CT 1.021-3.445; P =0.043) were the independent risk factors for VBD,while female gender (OR 0.133,95% CI 0.024-0.735; P =0.021) was the independent protective factor for VBD.Of the 28 cerebral infarction patients with VBD,9 had anterior circulation infarction and 19 had posterior circulation infarction.There were significant differences in BA diameter ([5.40 ± 0.49] cm vs.[6.00 ± 0.77] cm; t =2.046,P =0.041),and the proportions of high score in bifurcation height (x2 =6.768,P =0.037) and horizontal displacement (x2 =5.241,P =0.042) between the 2 groups (all P <0.05).The multivafiate logistic regression analysis showed that the BA bifurcation height was an independent risk factor for posterior circulation infarction (OR 1.347,95% CI 1.069-2.457; P =0.038) in patients with VBD.Conclusions VBD accounted for 10.41% of the patients with acute cerebral infarction during the same period.Advanced age,hyperglycemia,hypertension and increased triglyceride level were the independent risk factors for VBD.Female gender was the independent protective factors for VBD,and the BA bifurcation height was an independent risk factor for VBD occurring posterior circulation infarction.
