Clinical Medicine of China 2010;26(3):229-233
Objective To explore the role of IL-1β in inhalation injury repairment through its effects on the expression of prostaglandin E_2 in cultured airway fibroblasts of mouse. Methods The cultured airway fibroblasts in vivo from male Kunming species mouse were randomly divided into control group and treatment group. The superna-rant and cells were collected at different time after treated with IL-1β. The expressions of PGE_2, COX-2 and mPGES1 protein in supernatant and cells were measured by ELISA and Western Blot. Results ①PGE_2 levels in the cultured airway fibroblast supematant at 8 h(148.2±28.3) ng/L,16 h(267.6±45.4)ng/L,24 h(210.5±38.6) ng/L and 48 h(198.7±36.5) ng/L were higher than that in the control group (P < 0.01), and peaked at 16 h;COX-2 ex-pressions at 8 h(0.478±0.029) COX-2/β-actin, 16 h(0.672±0.047) COX-2/β-actin,24 h(0.617±0.036) COX-2/β-antin, and 48 h (0.593±0.034) COX-2/β-actin were higher than that in control group (P < 0.01), and peaked at 16 h;mPGES1 at 8 h(0.300±0.018) mPGES1/β-actin, 16 h(0.549±0.034) mPGES1/β-actin, 24 h (0.497±0.030)mPGES1/β-actin and 48 h(0.443±0.026)mPGES1/β-actin were higher than that in the control group(P < 0.01), and peaked at 16 h. ②PGE_2 in 0.1 μg/L IL-1β group (142.9±2.3) ng/L, 1.0 μg/L IL-1β group (267.6±45.4) ng/L and 10.0 μg/L IL-1β group (587.3±106.9) ng/L were higher than those in the control group (58.5±16.0) ng/L (P < 0.01), and there was significant difference among the three groups (P < 0.01);COX-2 in 0.1 μg/L IL-1β group(0.525±0.031) COX-2/β-actin, 1.0 μg/L IL-1β group(0.672±0.047) COX-2/ β-actin and 10.0 μg/L IL-1β group(1.012±0.064)COX-2/β-actin were higher than that in the control group (0.309±0.019) COX-2/β-actin (P < 0.01), and there was difference among the three groups (P < 0.01);mPG-ES1 in 0.1 μg/L IL-1β group(0.380±0.021) mPGES1/β-actin, 1.0 μg/L IL-1β group (0.549±0.034) mPG-ES1/β-actin, and 10.0 μg/L IL-1β group(0.879±0.045) mPGES1/β-actin were higher than those in the control group(0.199±0.012)(P<0.01),and there was difference among the three groups(P<0.01).Conclusions IL-1β may upregulate PGE_2, COX-2 and mPGES1 expressions in airway fibroblast, indicating that IL-1β influences PGE_2 synthesis through COX-2 and mPGES1 expression and participates in airway inhalation injury course. Airway fibroblasts may be a main source cell of inflammatory mediators in injury repair.
2. Effects of transfection of hGM-CSF gene into HepG2 cells mediated by hydroxyapatite nanoparticles on the growth of HepG2 cells
Tumor 2008;28(3):224-227
Objective: To observe the impact of transfection of human granulocyte macrophage colony stimulating factor (hGMCSF) mediated by hydroxyapatite(HA) nanoparticles on the growth of HepG2 cells in order to lay the foundation for further studying gene-modified tumor vaccines of hepatoma in clinic. Methods: The proliferation of HepG2 cells was measured by MTT assay. The plasmids containing hGM-CSF gene were transfected into HepG2 cells mediated by HA nanoparticles. Subsequently, resistant cells were selected by G418 screening. The monoclonal cells were selected by limiting dilution method. RT-PCR was used to identify the integration of hGM-CSF into HepG2 cells and transcription of hGM-CSF. ELISA was performed to detect the level of secreted hGM-CSF in cultured medium and the lasting time. The effect of hGM-CSF on cell cycle and apoptosis were assessed by flow cytometry (FCM) analysis. Results: MTT assay demonstrated that Nano-HA suspension liquid had no significant effects on the growth of HepG2 cells at the concentration below 80 μg/mL. RT-PCR demonstrated the hGM-CSF gene was successfully integrated and steadily expressed in the transfected HepG2 cells. ELISA indicated stable secretion of hGM-CSF from the stable transfected HepG2 cells [mean (216.22 ± 45.78) ng/106 cells per 24 h]. FCM analysis showed that hGM-CSF had no significant effects on the cell cycle and apoptosis of HepG2 cells. Conclusion: hGM-CSF gene could be safely transfected and stably expressed in HepG2 cells mediated by HA nanoparticles. Transfection of hGM-CSF gene mediated by HA nanoparticles has no effects on the growth of HepG2 cells. This study may lay a foundation for the further study of gene-modified hepatoma vaccines.

AIM: To determine the median effective dose(ED50) of spinal anesthesia 0.75% ropivacaine for cesarean section. METHODS: Thirty-two ASAⅠ-Ⅱ parturients undergoing elective cesarean section delivery with use of combined spinal-epidural anesthesia were enrolled in this study. The initial dose of was 12.8 mg of 0.75% ropivacaine, and the dosing intervals were 98.4% of the preceding dosage. Doses for subsequent parturients were determined by the response of the previous parturient. Successful anesthesia was defined as a loss of sensation to an acupuncture stimulus at the T7 level, full motor blockade within 10 minutes after administration of the local anesthetic, analgesia of incision and the satisfaction of skelaxin evaluated by one operator. ED50 and its 95% confidence interval (CI) was determined with use of Dixon method. RESULTS: Five parturients were rejected out the study because of doubtful anesthsia. According to the rest twenty-seven parturients' data, the ED50 of intrathecal 0.75% ropivacaine was 11.97 mg (95% confidence interval [CI], 11.75-12.19 mg). CONCLUSION: The ED50 of spinal 0.75% ropivacaine for cesarean delivery under the conditions of this study were 11.97 mg.

Chinese Journal of Urology 2001;0(09):-
Objective To evaluate the improvement of diagnosis and treatment of hypercatecholaminism. Methods A total of 95 cases of hypercatecholaminism were reviewed.Of them 74 cases were of pheochromocytoma and 21 cases were of adrenal medullary hyperplasia (AMH).In pheochromocytoma group,67 cases (90.5%) had symptom of hypertension.Increase in 24 h urinary VMA occurred in 62 cases (83.8%) and elevated urinary catecholamine level in 67(90.5%).All thses had positive findings of ultrasound,CT scan and MRI.In AMH group, all the patients were hypertensive and their 24 h urinary VMA was increased. Most of them (15/16) had higher urinary catecholamine level than normal;14 out of the 21 cases had positive findings of B-ultrasound examination. Results Surgical operations were performed in the 74 patients with pheochromocytoma including 9 cases of extra-drenal pheochromocytoma.Of them 5 cases were confirmed as malignant pheochromocytoma. In the 21 cases of AMH,18 undewent surgical operation.Of them 12 received unilateral resection and 6 received bilateral resection of adrenal gland. The pathological examination confirmed the diagnosis of AMH. Conclusions The diagnosis of pheochromocytoma should be focused on endocrinological examination,ie,qualitative examination, whereas it is diffucult to localize the lesion in AMH compared with pheochromocytoma. The establishment of notion of AMH as an isolated clinicopathological entity broadens and perfects the theory of hypercatecholaminism.
