1.Expression of human papillomavirus DNA in keratoacanthoma tissue

Objective To detect the infection rate of human papillomavirus (HPV) types in keratoacanthoma (KA) tissue, and to study the distribution of HPV DNA-positive cells. Methods In situ hybridization was performed to detect HPV subtypes 6/11, 16/18, and 31/33 in paraffin-embeded tissue samples from 46 patients with KA and 34 nomal human controls. Results Mucosal HPV DNA was positive in 31 (67.39%)of the 46 KA samples, and mixed infection rate of HPV amounted to 83.87% (26/31) in these positive samples.The DNA of tested mucosal HPV types existed mainly in episomal status. Positive reactions were observed within or at the edge of nuclei. HPV infected cells showed a stripe-, slice-like or focal distribution mainly in superficial middle lamina of spinous layer at the bottom or lateral margin of crater-shaped epidermal depression. No mucosal HPV DNA was noted in normal control samples. Conclusions Compared with normal controls, patients with KA are infected with mucosal HPV at a higher frequency, and mucosal HPV DNA mainly exists in episomal status in KA tissue, hinting that HPV infection play a certain role in the pathogenesis of KA.
2.Study on Complement and Activated Complement Factors in Peripheral Blood and Skin in Patients with Psoriasis

Objective To investigate the levels of complement and activated complement factors in peripheral blood and skin in patients with psoriasis and their significance in the development of psoriatic lesion.Methods Enzyme linked immunosorbent assay (ELISA) and immuno turbidi metric analysis were used to determine the levels of C3d, sC5b-9, C3 and C4 in sera from 57 patients with psoriasis vulgaris.Immunohistochemistry was employed to assess in situ expression of SC5b-9 in lesional and non lesional skin of 37 patients.Results There was a significant decrease of C3 and C4, and increase of C3d and sC5b-9 in patients with psoriasis in comparison with those of normal controls (t=10.88~ 24.80, P<0.01).The levels of C3 and C4 were significantly lower, while the levels of C3d and sC5b-9 were significantly higher in progressive stage than those in static stage (t=4.55~ 6.77,P< 0.01).In situ expression of sC5b-9 was significantly higher in stratum corneum and dermo epidermal junction of lesional skin than that in non lesional skin and normal controls (χ2=62.90,45.45;P< 0.01).There was no significant difference of C5b-9 expression in stratum corneum and dermoepidermal junction of lesional skin between progressive and static stage.Conclusions There is significant correlation between the development of psoriatic lesion and local deposit of activated complement factors.

4.Detection of T cell receptor gamma gene rearrangements in mycosis fumgoides

Objective To assess the diagnostic significance of T cell receptor gamma gene rearrangemerits in mycosis fungoides (MF), so as to develop a sensitive diagnosis tool. Methods A total of 50 specimens were collected, including 33 skin lesion specimens and 2 lymph specimens from 30 patients with MF,15 skin lesion specimens from 15 patients with inflammatory dermatoses. PCR was performed with specific primers targeting TCR V gamma 8, 9, 10, 11 to detect T cell receptor gamma gene rearrangement. Results Monoclonai rearrangements of TCR gene was observed in 88% (29/33) of specimens from patients with MF and 33% (5/15) of samples from patients with inflammatory dermatoses. Conclusions The detection of TCR gene rearrangements, as an ancillary test, is useful in the diagnosis and differential diagnosis of MF.
5.Mediating effect of self-identity on relationship between interpersonal adaptation and mobile phone addiction tendency in college students

Objective:To explore the mediating effect of self-identity on the relationship between interpersonal adaptation and the tendency of mobile phone addiction in college students.Methods:Totally 498 college students (217 males and 281 females) were selected and assessed with the College Student Interpersonal Adaptability Inventory (CSIAI),Mobile Phone Addiction Tendency Scale (MPATS) and Personal Identity Scale(PIS).The bias-corrected percentile bootstrap method was used to analyze the mediating effect of self-identity between interpersonal adaptation and mobile phone addiction tendency in college students.Results:The MPATS scores of male students were significantly lower than female students [(2.6 ± 0.7) vs.(2.7 ± 0.6),P < 0.05].The MPATS scores were negatively correlated with the scores of CSIAI and PIS (r =-0.31,-0.36,Ps < 0.01),the CSIAI scores were positively correlated with the PIS scores (r =0.50,P <0.01).Self-identity played a partly mediating role between interpersonal adaptation and mobile phone addiction tendency,which accounted for 43.6% of the total effect.Conclusion:It shows that the mobile phone addiction tendency may be related to interpersonal adaptation and self-identity,interpersonal adaptation could affect the mobile phone addiction tendency through the self-identity.
6.EFFECTS OF EXOGENOUS ATP ON CELL PROLIFERATION AND CYTOSKELETAL STRUCTURES OF HUMAN STOMACH CARCINOMA CELLS (MGC-803)

This study was carried out to investigate the effects of ATP on cultured human stomach carcinoma MGC-803 cells. After ATP(0.16—0.23 mg/ml, 24—120 hrs)treatment, inhibition of cell growth rate was observed. The inhibition rate reached 80% after 96 hr exposure to ATP. At the same time, anti-tubulin immunofluorescent staining showed increase of organized microtubules (MT) in ATP-treated MGC-803 cells, in contrast with the poor MT immunofluorescence in control MGC-803 cells. By using rhodamie phalloidin combined staining, increased number of actin bundles, took the place of actin dots and patches which were seen in control MGC-803 cells. Immunofluorescent staining of fibronectin in ATP-treated MGC-803 cells was also increased evidently as compared with the control MGC-803 cells. Concomitant with the improvement of MT and stress fiber organization, the cell Concohas become more flattened. It appeared that the increase of MT and stress fibers was closely related to the recovery of cell morphology in ATP treated tumor cells. It is therefore concluded that ATP not only may inhibit tumor cell proliferation, but also may induce differentiation of cell morphology of tumor cells through its action on cytoskeletal structures.
7.Formula Optimization of Levodopa Microcapsules Floating Tablets

To optimize the formula of levodopa microcapsules floating tablets. Methods:The contents of levodopa and benserazide in the microcapsules floating tablets were determined by HPLC simultaneously. The release rate as the index, an orthogonal design was used to optimize the formula and preparation technology of levodopa microcapsules floating tablets. The release property of the microcapsules floating tablets was evaluated. Results:The HPLC method for the in vitro determination of levodopa and benserazide in the floating tablets met the methodological requirements. The selected formula was as following:the amount ratio of stearic acid, the drugs, acrylic resin and HPMC was 2∶5∶2∶1. The average weight of the tablets was 550 mg. The results of validated tests showed that the microcapsules floating tablets had floating and sustained release property, which could be used by divided dose. Conclusion: The optimized formula of the microcapsules floating tablets is reasonable, and the production process is stable and feasible.
8.Proteomic screening of the proteins associated with early lung squamous carcinoma

Objective:To screen the early biomarkers associated with lung carcinoma by using proteomic method. Methods:The proteins of early lung squamous cell carcinoma tissues and adjacent normal tissues were separated by two-dimensional gel electrophoresis (2-DE) and differentially expressed proteins were screened by mass spectrometry (MS). The differential expressions of part proteins were verified by immunohistochemical method. Results:The proteomic profiles of lung squamous cell carcinoma and the adjacent normal tissues were established and the average matched protein spots in the two group were 626 and 602, respectively. Ten protein spots with higher expression in lung squamous cell carcinoma tissues were identified as Annexin 1(Anx-A1),heat shock proteins (HSP27), etc. The proteins were mainly related with cell cycle and signal transduction. The results of immunohistochemistry showed that the positive expression rates of Anx-A1 and HSP27 in lung squamous cell carcinoma tissues were significantly increased (P<0.05). Conclusion:Proteomic technology was an effective method for preliminary identification of the proteins associated with early lung carcinoma. The identified proteins laid a solid foundation for further screening the molecular markers for early diagnosis and treatment of lung cancer.
9.Effects of the Zusanli or Sanyinjiao point electroacupuncture of obese rats on fat metabolism

Objective To study the electroacupuncture effects of the single point Zusanli or Sanyinjiao on simple obese rats and to observe the regulation of fat metabolism.M[ethods Before and after the treatment,the body weights and lengths of rats in each group were measured tO calculate the Lee's index;besides,total cholesteriol (TC) and triglycerides (TG) in the rat blood were also determined.Finally,the rats were treated and observed for tWO courses,and SPSS11.5 statistical software was employed to conduct statistical analysis of the experimental data.Results A significant difference was observed about the effects on the height,weight,Lee's index,TC and TG between treatment group and the model group or normal control group (P<0.05 or P<0.01),but no significant difference between treatment group and normal control group (P>0.05).Conclusion Electroacupuncture of Zusanli and Sanyinjiao points has effects on simple-obesity rats,which can make weight loss and decrease Lee's index,TC and TG levels.The underlying mechanism may relate to the improvements of fat metabolism.
10.Robustness analysis of adaptive neural network model based on spike timing-dependent plasticity.

To explore the self-organization robustness of the biological neural network, and thus to provide new ideas and methods for the electromagnetic bionic protection, we studied both the information transmission mechanism of neural network and spike timing-dependent plasticity (STDP) mechanism, and then investigated the relationship between synaptic plastic and adaptive characteristic of biology. Then a feedforward neural network with the Izhikevich model and the STDP mechanism was constructed, and the adaptive robust capacity of the network was analyzed. Simulation results showed that the neural network based on STDP mechanism had good rubustness capacity, and this characteristics is closely related to the STDP mechanisms. Based on this simulation work, the cell circuit with neurons and synaptic circuit which can simulate the information processing mechanisms of biological nervous system will be further built, then the electronic circuits with adaptive robustness will be designed based on the cell circuit.
