1.Uric acid, hyperuricemia and ischemic stroke

Uric acid is an end product of purine metabolism.Hyperuricemia is defined as serum uric acid level >420 μmol/L in man,and 360 μmol/L in woman.Several mechanisms can cause elevated serum uric acid levels.Systematic reviews and Meta-analyses have shown that hyperuricemia is significantly correlated with the increased morbidity and mortality in patients with ischemic stroke.However,as a powerful antioxidant and a radical scavenger,uric acid seems to have a neuroprotective effect in ischemic stroke.Uric acid administered early can enhance the effect of recombinant tissue plasminogen activator (rtPA) thrombolytic therapy in rats with thromboembolic stroke.Therefore,it is necessary to conduct a randomized controlled trial for the clinical effects of rtPA thrombolysis in combination with uric acid in patients with acute ischemic stroke.A phase 3 clinical trial is ongoing,and its results are worth waiting for.
2.Influence mechanism and intervention strategies of psychological resilience in patients with cerebral infarction

Objective To analyze the effect of self efficiency,social support and coping style on the psychological resilience.Methods A total of 386 patients with cerebral infarction in a grade 3A hospital of Tangshan City were served as the subjects.The resilience scale,self efficiency scale,social support scale and medical coping questionnaire were taken as the survey tools.The Pearson correlation analysis,multiple linear regression model and structure equation model(SEM) were applied.Results The resilience score was (59.72 ±12.17) points,the resilience was positively correlated with the self efficiency,social support,facing coping style and avoidance coping style(P<0.01),and negatively correlated with the surrender coping style(P<0.01).The total efficiency coefficients of self efficiency sensation,subjective support,facing coping style and surrender coping style for affecting the resilience were 0.720,0.264,0.121 and-0.092 respectively.Conclusion Self efficiency,social support and coping style have significant influences on the psychological resilience.
3.Effect of hydroxyl camptothecinum on pharmacokinetics of cyclosprine A and the membrane lipid fluidity of red cell in rabbit

AIM To investigate the effect of hydroxyl camptothecinum(HCPT) on pharmocokinetics of cyclosprine A(CsA) and the membrane lipid fluidity (LFU)of red cell in rabbits. METHODS The whole blood concentration of CsA in rabbits was monitored using method of FPIA. The LFU of red cell was measured using method of DPH fluorescence polarization technique. RESULTS The pharmocokinetic parameters of CsA in group CsA+HCPT such as a,V(c),K12 and CL(s) were significantly lower and T1/2(a) was significantly higher (P<0.05) than in group CsA. The concentration of CsA of group CsA+HCPT was higher after injection 0.25, 0.5 and 1.0 h, but have significantly high at 1.0 h point (P<0.05). After injection 0.5,1.0, 1.5 h the LFU of red cell in group CsA+HCPT was higher than in group CsA (P<0.05). The LFU in group CsA was lower that in control at 0.5,1.0 and 1.5 h point (P<0.05). CONCLUSION HCPT can enhance the whole blood concentration of CsA in rabbits. CsA administrated with HCPT is feasible.

5.Proteomic screening of the proteins associated with early lung squamous carcinoma

Objective:To screen the early biomarkers associated with lung carcinoma by using proteomic method. Methods:The proteins of early lung squamous cell carcinoma tissues and adjacent normal tissues were separated by two-dimensional gel electrophoresis (2-DE) and differentially expressed proteins were screened by mass spectrometry (MS). The differential expressions of part proteins were verified by immunohistochemical method. Results:The proteomic profiles of lung squamous cell carcinoma and the adjacent normal tissues were established and the average matched protein spots in the two group were 626 and 602, respectively. Ten protein spots with higher expression in lung squamous cell carcinoma tissues were identified as Annexin 1(Anx-A1),heat shock proteins (HSP27), etc. The proteins were mainly related with cell cycle and signal transduction. The results of immunohistochemistry showed that the positive expression rates of Anx-A1 and HSP27 in lung squamous cell carcinoma tissues were significantly increased (P<0.05). Conclusion:Proteomic technology was an effective method for preliminary identification of the proteins associated with early lung carcinoma. The identified proteins laid a solid foundation for further screening the molecular markers for early diagnosis and treatment of lung cancer.
6.Real-time fluorescence PCR-molecular beacon assay for detecting mutations in rpoB genes of rifampin-resistant Mycobacterium tuberculosis

Objective To establish a rapid method to detect mutations in rpoB genes of rifampin-resistant Mycobacterium tubereulosis in dinical specimens using Real-time fluorescence PCR molecular beacon assay.Methods 174 strains of Mvcobacterium tuberculosis clinical isolates were analyzed using real-time fluorescence PCR molecular beacon assay foilowed with DNA sequencing while 12 strains of NTM and 4 strains of bacteria other than Mycobacterium tuberculosis were used as the contrast.Results Eighty-two 89.1 of 92 rifampin (RIF)-resistant strains and 3 of 82 RIF-sensitive strains were found to harbor mutation in the rpoB gene using real-time fluorescence PCR-molecular beacon assay.The specificity, sensitivity,and accuracy of this assay were 96.3%,89.1%,and 92.5%,respectively-Eithty-three of 92 RIF-resistant strains and 1 of 82 RIF-sensitive strains were found to harbor mutation in the rpoB gene using the direct DNA sequencing.The specificity,sensitivity,and accuracy of the direct DNA sequencing were 98.8,90.2%,and 94.2%,respectively.As compared with real-time PCR molecular beacon assay,171 of 174(98.3%)strains of myeobactefium tuberculosis clinical isolates had the salne results.Conclusion Real-time fluorescence PCR-molecular beacon assay can be used as a rapid screen method to detect RIF-resistant isolates.
7.Pharmacokinetics of levofloxacin-methane sulfonic acid in patients with pulmonary tuberculosis

OBJECTIVE To study the pharmacokinetics of levofloxacin-methane sulfonic acid (LF-MSA)in 8 patients with pulmonary tuberculosis after administration 400 mg orally.METHODS The concentrations of LF-MSA in serum were measured by HPLC.The pharmacokinetic paramters were obtained using 3P97 program.RESULTS The results showed that a one-compartment model with 1 st order absorption were fitted for LF-MSA.The average values of tmax was 1.22 h.The concentrations of LF-SA in serum was (4.52±0.72) mg*L-1.The mininal concentrations of LF-AS at 24 hr after a single dose was (0.52±0.26) mg*L-1.The average area under the curve was (48.35±5.38) mg*h*L-1.The elimination half-life was (6.49±1.70) h.The apparent volume of distribution was (77.48±8.33)L.CONCLUSION According to our study,it was suggested that 400 mg twice daily may be given in order to maintain the concentrations of LF-SA in serum around 24 hours above the minimal concentrations of bactericide.
8.Effects of Jianpi Yishen Recipe on recurrence, metastasis and life span of mice transplanted with proventriculus squamous carcinoma after tumorectomy

OBJECTIVE: To observe the effects of Jianpi Yishen Recipe (JPYSR), a compound Chinese herbal medicine, on recurrence, metastasis and life span of mice transplanted with proventriculus squamous carcinoma cells after tumorectomy. METHODS: JPYSR was orally administered to 615 mice transplanted with proventriculus squamous carcinoma cells in the palma of left hind limb after tumorectomy. The local tumor recurrence, lung metastasis and life span of the mice were evaluated and compared with those of the 5-FU-treated mice and untreated mice. RESULTS: The pulmonary metastasis rate was 94.4% and the recurrence rate was 94.44% in untreated group. The pulmonary metastasis rate was 68.4%, the inhibition rate of pulmonary metastasis was 27.5%, the recurrence rate was 78.95%, and the inhibition rate of tumor recurrence was 65.35% in JPYSR-treated group. The average and median life spans were obviously prolonged in JPYSR-treated group, as compared with those in untreated group. The life-prolonging rate was 100%. CONCLUSION: JPYSR can effectively inhibit the local recurrence and pulmonary metastasis of the transplanted proventriculus squamous carcinoma in mice after tumorectomy, and prolong the life span.
9.Influencing factors for treatment compliance of patients with multidrug-resistant tuberculosis

Objective To investigate the influencing factors for post-discharge treatment compliance of patients with multidrug-resistant tuberculosis (MDR-TB).Methods MDR-TB patients who were hospitalized in a tubercu-losis hospital between November 2011 and January 2013 were chosen,post-discharge follow-up was conducted regu-larly through telephone call.Medicine-taking and re-examination of patients was inquired,factors influencing pa-tients’treatment compliance were analyzed.Results 299 patients were included in the study,the total treatment compliance rate was 81 .94% (n=245);249(83.28%)patients regularly took medicine,50(16.72%)didn’t regu-larly take medicine;254 (84.95%)were re-examined on time,45 (15.05%)were not re-examined on time;37 (12.37%)discontinued treatment,260 (86.96%)continuously treated till the survey deadline.Univariate analysis revealed that treatment compliance (including regular medication rate,timely re-examination rate,interrupted treat-ment rate,and total compliance rate)was significantly different among MDR-TB patients of different ages,education levels,treatment time,and with or without adverse reactions(all P <0.05 ).Logistic regression analysis revealed that treatment compliance of MDR-TB patients was negatively correlated with treatment time(β=-1 .47,Wald χ2=24.28,P <0.05)and adverse reactions(β=-2.02,Waldχ2 =24.24,P <0.05 ),while positively correlated with education levels(β=0.79,Wald χ2 =6.50,p <0.05 ).Conclusion Prolonged treatment time and adverse reactions can reduce the treatment compliance of MDR-TB patients,the higher education levels of MDR-TB patients have, the better treatment compliance they implement.
10.Effect of Antipsychotics plus Skill Training on Schizophrenics at Recovery

ObjectiveTo explore the influence of antipsychotics combined with skill traning on quality of life and social function of patients with schizophrenia at recovery.Methods80 patients with schizophrenia were randomly divided into study group (n=40, antipsychotics plus skill training)and control group (n﹦40, only antipsychotics) for 1-year treatment. At pre-treatment and end of treatment, both groups were assessed with Positiveand and Negative Symptom Scale (PANSS), Generic Quality Of Life Inventory-74 (GQOLI-74) and Social Disability Screening Schedue(SDSS) respectively to evaluate their effects, quality of life and social funtion.ResultsAfter 1-year treatment, the general scores of PANSS and each factors in both groups decreased significantly compared with the baseline(P<0.05~0.01), and the significant difference was found between two groups(P<0.01). Improvement of each factor score of FAD(except factor of the material life) and SDSS in study group were more better than those in control group(P<0.05~0.01).ConclusionTreatment with antipsychotics plus skill traning is helpful to release the mental syndrome, improve quality of life and social funtion, and facilitate the recovery of the patients with schizophrenia at recovery.
