1.Expression of ATP synthase 6 in breast cancer cell line, T47D cells, induced by heregulin β1

Objective : To explore expression of related genes in breast cell line T47D induced by heregulin β1. Methods: Suppression subtractive hybridization was performed using cDNA sub traction kit to detect expression of heregulin-responsive genes in T47D cells. Results: ATP syn thase 6 was up-regulated by heregulin β1 in T47D cells at 1 and 6 hours. Conclusion: Heregulin β1 participates in the regulation of expression of both ATP synthase 6 and oxidative phosphoryla tion of T47D cells.
2.Detection of Panton-Valentine Leukocidin Gene and Analysis of Homology in 50 Meticillin-resistant Staphylococcus aureus Isolates

OBJECTIVE To investigate the antibiotic resistance pattern of MRSA,homology and carriage of Panton-valentine leukocidin(PVL) in 50 meticillin-resistant Staphylococcus aureus isolates to guide the clinical treatment.Results of homology provide the proof for molecular epidemiology.METHODS According to CLSI′s guideline,antimicrobial susceptibility tests were performed with disk diffusion method.PVL gene and mecA gene were detected by PCR.The homology was analyzed by repetitive estra-genic palindromic elements(Rep)-PCR method.RESULTS MecA gene was detected in all 50 MRSA isolates.All isolates MRSA were resistant to oxacillin and cefoxitin.But they were all sensitive to vancomycin and teicoplanin.The rates of sensitivity to clindamycin,ciprofloxacin,piperacillin and sulbactam,SMZ-TMP,gentamicin,cefazolin and erythromycin were 7.5%,1.9%,24.5%,92.5%,20.8%,1.9% and 3.8%,respectively.The resistance rates of MRSA were higher than that of MSSA.Twenty PVL genes were detected from 50 MRSA isolates.CONCLUSIONS The resistance rates of MRSA are higher than MSSA.There is no isolate resistant to vancomycin and teicoplanin.40.0% MRSA carry PVL gene and part isolates have high homology.
3.The safety of preoperative use of dexmedetomidine in preschool children

Objective To explore the degree of cooperation and the effect of hemodynamics in preschool children who received preoperative use of dexmedetomidine.Methods One hundred preschool children who scheduled for lower abdominal operation were randomly divided into two groups,each 50patients.The treatment group was given bolus infusion dexmedetomidine 0.5 μ g/kg (20 ml),the control group was given bolus infusion 0.9% sodium chloride 20 ml.They were completed in 20 min.The cooperation was assessed by induction compliance checklist (ICC) score,and the level of mean arterial pressure (MAP),heart rate(HR),pulse oxygen saturation(SpO2),end-tidal pressure of carbon dioxide(PnCO2) at monitoring ward before bolus infusion (T1),end of bolus infusion (T2),before induction (T3),after induction (T4),incised skin (T5) and end of operation (T6) were compared.Results The scores ofICC in treatment group was significantly lower than that in control group [(2.73 ± 0.12) scores vs.(6.17 ± 0.91) scores] (P < 0.05).In treatment group,48 patients ICC score was 0,2 patients ICC score >5 scores,and in control group,there was 48,2 patients,there was significant difference(P < 0.05).The level of MAP,HR at T1,T2,T4between two groups had no significant difference (P > 0.05).The level of MAP,HR at T3,T5,T6 in treatment group were significantly lower than those in control group(P < 0.05).In control group,the level of MAP,HR at T3,T5,T6 were significantly higher than those at T1 (P < 0.05).In treatment group,the level of MAP,HR at T2-T6 had no significant difference compared with T1 (P > 0.05).The level of SpO2,PnCO2 between two groups and interclass had no significant difference (P >0.05).Conclusion Bolus infusion dexmedetomidine before operation can improve the induction of anesthesia and operation smoothly,and improve the patients early rehabilitation.
4.Clinical study of recombinant human interleukin-11 combined with ciclosporin in treatment of glucocorticoid invalid idiopathic thrombocytopenic purpura

ObjectiveTo observe the effect of recombinant human interleukin-11 combined with ciclosporin in treatment of giucocorticoid invalid idiopathic thrombocytopenic purpura.MethodsFrom August 2009 to August 2011,35 cases with glucocorticoid invalid idiopathic thrombocytopenic purpura patients were divided into two groups according to the treatment method:13 cases in control group were treated with ciclosporin and 22 cases in observation group were treated with recombinant human interleukin-11 at basis of above method.The clinical effect and platelet counts between two groups were compared.Results The total effective rate in observation group was 90.9% (20/22),which was significantly higher than that in control group [53.8% (7/13)] (P <0.05).After treatment,the platelet counts in both two groups were significantly increased [control group:( 112.5 ± 15.4) × 109/t vs.(13.2 ± 1.8 ) × 109/L; observation group:( 132.7 ± 22.3 ) × 109/L vs.(12.9 ± 1.6) × 109/L] (P < 0.05 ).Moreover,the platelet counts after treatment in observation group was significantly more than that in control group (P < 0.05).ConclusionRecombinant human interleukin-11combined with ciclosporin in treatment of glucocorticoid invalid idiopathic thrombocytopenic purpura is a good treatment scheme,which can be applied in clinic.
5.Peropective comparison of hydroxyapatite orbital implant between primary and second drilling procedure

Objective To evaluate the clinical effect of the procedure that involves drilling and inserting a motility peg at the time of hydroxyapatite orbital implant.Methods 89 patients who needed hydroxyapatite orbital implant were divided into groups A and B.The patients in group A were drilled and inserted a motility titanium peg when hydroxyapatite implantation.The patients in group B were drilled and inserted a motility titanium peg 6 months after hydroxyapatite implantation.Data were recorded.including the rate of postoperative pain and conjunctive edema,postoperative facial appearance,motility of the artifieial eye,the rate of hydroxyapatite exposure,excursion and extrusion of titanium peg and other complications.Results The mobility of the prosthesis was 44.97%in group A,and 45.34%in group B.There was no difference in appearance and the motility of their artificial eye between the two groups.The rate of excursion of titanium peg was 4.2% in group A and 7.3%in group B and no statistical difference was found between the two groups.There were no prosthesis exposure,extrusion of titanium peg,secondary infeetion and other complication in one-year follow-up.There were no difference in the rate of postoperative pain and eonjunctive edema in first operative stage.The postoperative reaction in group A was lighter than that in group B in secondary operanve stage.Conclusion The hydroxyapatite orbital implant primary procedure drilling is simple,safe,convenient and effective.It is worth to recommend that procedure that involves drilling and inserting a motility peg at the time of hydroxyapatite orbitaI primary implant.

Objective To explore the situation of respiratory virus co-infection with EV71 and CA16 in patients with hand,foot and mouse disease(HFMD) ,and analyze the influence of co-infection on clinical aspects.Methods From June to October of 2010,there were 348 patients enrolled in the study,with 248 hospitalization cases and 100 mild outpatients.All the patients were diagnosed as HFMD in Beijing You-an Hospital.The viral RNA from the pharynx swab samples were extracted and reversely transcribed by RT-PCR.All the samples were detected with the EV71 and CA16 by real-time fluorescence quantitative PCR.Twelve kinds of respiratory viruses were detected by a commercial multiplex-PCR method.The PCR products were confirmed by electrophoresis.Chi square test was used in the data analysis.Results Of the 348 HFMD patients,36 subjects were detected as positive for respiratory virus co-infection.In the 248 hospitalization cases,111 cases were positive for EV71 or CA16,with eight cases identified with respiratory virus co-infection(7.2%); the other 137 cases were negative for EV71 and CA16,with eleven cases identified with respiratory virus co-infection(7.4%).There was not significant difference between respiratory virus co-infection and the identification of EV71 /CA16(x2 = 0.059,P > 0.05).In the 100 mild outpatients positive for EV71 or CA16,seventeen cases were identified with respiratory virus co-infection(17%).The rate of respiratory virus co-infection in the mild outpatients was much higher than in the severe hospitalization patients(x2 = 4.830,P< 0.05).Among the 111 EV71(+) or CA16(+) inpatients,there were 101 cases diagnosed as severe cases(91.0%); similarly,there were 132 cases diagnosed as severe cases(96.4%) among the 137 EV71(-) CA16(-) cases.There was not difference between the identification of EV71/ CA 16 and illness of HFMD(x2 = 3.099,P > 0.05).The leading respiratory virus being identified were HRV A/B,PIV3 and FLU A in the 348 HFMD patients.Conclusions Co-infection with respiratory virus exists in the HFMD patients. However,the respiratory virus infection has no significant influence to the state of HFMD illness.
8.Changes of cardiac geometry and function in hypertensive patients with left ventricular concentric remodeling

Objective To investigate the changes of geometry and function of left ventricle (LV) and left atrium(LA) in hypertensive male patients with concentric remodeling.Methods Consecutive male patients were divided into two groups: normal control group(NC group,n=146) and concentric remodeling group(CR group,n=274).The whole patients were checked with echocardiogram.Echocardiographic parameters included: thickness of interventricular septum(IVST) and LV posterior wall(PWT),LV end-diastolic diameter(LVIDd),media-lateral diameter(LVID),LV length(LVL),LA anterior-posterior diameter,media-lateral diameter,superior-inferior diameter,LA area,LV end-diastolic volume,end-systolic volume,stoke volume,ejection fraction(EF),early diastolic peak velocity (E) and atrial contraction peak velocity(A) of the mitral valve flow.LV mean diameter(LVMD) and E/A were calculated.Student t test was used to compare the variables between the two groups.Results IVST,PWT,IVST/LVPWT ratio,LA diameters and LA area in CR group were larger than those in NC group.LVIDd was shorter and LVID was larger in CR group than in NC group.LVL in CR group was larger than that in NC group.LVMD/LVL ratio was smaller than that in NC group.There was no significant difference in LVEF between two groups while E/A ratio in CR group was smaller than that in NC group.Conclusions The concentric remodeling in hypertensive patients makes the LV a thin,long but thick-walled cavity.The LV diastolic function is impaired in CR group but the systolic function reserves well.
9.The effects of glutamine dipeptide on the improvement of endotoxemia in severely burned patients.

OBJECTIVETo explore the influence of glutamine dipeptide on the plasma endotoxin levels in severely burned patients.
METHODSThirty burned patients with TBSA of 30 - 70% and III degree burn area more than 20% were randomly divided into control (C) and study (S) groups. Glutamine dipeptide powder in dose of 0.5 g/kg/day was given orally in bolus to those patients in S group during 1 - 12 postburn days (PBDs). The plasma levels of glutamine were determined during 1 - 12 PBDs. Simultaneously, the plasma endotoxin level was detected on 1, 3, 6 and 12 PBDs. The wound healing rate at 30 PBD and total hospital stay days were recorded.
RESULTSThe plasma glutamine levels at 1 PBD in C and S groups were obviously lower than normal level (659.5 +/- 35 micromol/L), but there was no difference between these two groups (P > 0.05). The plasma glutamine levels in C group was much lower than that in S group at 12 PBD (P < 0.05). The plasma endotoxin concentration on 1 PBD in these two groups increased evidently compared with the normal value (P < 0.05), and there was no difference between the two groups (P > 0.05). The plasma endotoxin level in S group was much lower than that in C group on 3 PBD (P < 0.05). As for the wound healing rate at 30 PBD, it was markedly higher in S group than that in C group (91% vs 85%). On the other hand, the hospital stay days in S group were evidently lower than that in C group (52 vs 67).
CONCLUSIONOral intake of glutamine dipeptide in burn patients could be beneficial to the maintenance of the plasma concentration of glutamine and in decreasing plasma endotoxin level. It would also enhance the wound healing rate at 30 PBD and shorten the hospital stay days.

10.Synthesis and biological evaluation of H2S donor memantine derivatives

H2S has a role of protecting neurons from ischemia-reperfusion injury and significantly reducing the cerebral infarction area, but high concentration of H2S can induce neurotoxicity. Memantine, a N-methyl-D-aspartic acid(NMDA)receptor antagonist, could reduce the neurotoxicity of H2S at high concentration. Nine novel structures(compounds I1-I9)were designed by coupling(4-hydroxy phenyl)-3H-1, 2-dithiole-3-thione(ADT-OH)with memantine through alkanes as linkers and synthesized by four-step reactions from ADT-OH. Their neuroprotection against damage induced by glutamate on HT-22 cells was evaluated by MTT method. The results indicated that these compounds markedly increased the survival rates of damaged HT-22 cells at the concentration of 1 μmol/L(P< 0. 01), which suggested that these compounds could preferably protect neurons against damage induced by glutamate.
